This study aimed to determine the effectiveness of antibacterial formulations derived from herbal plants against mastitis-causing bacteria in dairy cattle. Herbal extracts composed of betel leaves extract, kecombrang flower extract, and turmeric extract were tested against Staphylococcus aureus, Streptococcus agalactiae, and Escherichia coli. The research was carried out in a Completely Randomized Design (CRD) using ten treatments and four replications at the Laboratory of Research and Testing, Faculty of Animal Husbandry, Padjadjaran University. The results showed that herbal antibacterial formulations could inhibit the growth of Staphylococcus aureus, Streptococcus agalactiae, and Escherichia coli bacteria. The most inhibitory power was obtained from P8 treatment (50% betel leaves + 50% kecombrang flower + 50% turmeric) categorized in "strong inhibition." The percentage of antibacterial inhibition of P8 formulation compared to chloramphenicol was 32% against Staphylococcus aureus, 33% against Streptococcus agalactiae, and 31% against Escherichia coli. Conclusively, the best herbal antibacterial formulation was a combination of 50% betel leaves extract, 50% kecombrang flower extract, and 50% turmeric extract.

Bovine mastitis is the predominant cause for antimicrobial use on dairy farms and is a major source of economic losses in the dairy industry. In this study, the antimicrobial susceptibility profiles of common mastitis-causing pathogens, Staphylococcus aureus (n = 62), Streptococcus agalactiae (n = 46), and Escherichia coli (n = 129), were determined for dairy cattle with mastitis across 142 Ukrainian farms. The results showed that there were more gentamicin resistant S. aureus isolates (16.95%) identified in this study than previously reported for Ukrainian dairy cattle. Moreover, low levels of amoxicillin susceptibly (13.51%) were observed for St. agalactiae, which contrasted a previous study showing susceptibility levels of >50%. St. agalactiae resistance to tetracycline was observed in 80% of the isolates. Cephalosporin use was most ineffective against E. coli, with 43.27–56% of the isolates exhibiting this resistant trait. Overall, this study performed a preliminary analysis of antimicrobial resistance on mastitis isolates from Ukrainian farms. However, given the limited numbers of the isolates tested in this study and that the publications on antimicrobial resistance in animal husbandry of Ukraine are very few, more extensive investigations are needed to comprehensively examine susceptibility patterns of mastitis-causing pathogens in dairy cattle in Ukraine.

Modern medicines that are developing now come from active ingredients isolated from plants that require large amounts of plants. The development of new drugs from endophytic fungi found obstacles in the amount of pure compounds produced. Therefore further research is needed by using endophytic fungi as a new antimicrobial producer. This study aims to see the ability or activity of pure compounds produced by Sporothrix sp endophytic fungi from Dahlia tuber (Dahlia variabilis). Test the activity of pure compounds produced by Sporothrix sp. Endophytic fungi on E. coli and Staphylococcus aureus determined by disc diffusion method. With doses of 10, 30 and 50μg/disk. In Escherichia coli bacteria doses 10 and 50μg/disk gave significant inhibition of pure compounds from isolation compared to the positive control of ciprofloxacin, which was marked by a statistically significant test result (p <0.05). In contrast to Staphylococcus aureus there was no significant difference in doses of both doses of 10.30 and 50μg/disk. Determination of pure compounds was carried out by HPLC and Infra Red Spectrophotometry.

The main approach to curbing mastitis is to prevent the entry of microorganisms from the outer environment into the mammary gland, which is achieved by the use of papilla disinfection following every time of milking. The objective of this work was to examine the antimicrobial activity of the disinfectant dodicin hydrochloride, in fact the preparation that contains this disinfectant (DESU? M), against bacteria, the most frequent causes of mastitis in cows. The efficacy of modified DESU? Mwas examined under laboratory conditions using the quantitative test against the following microorganisms: Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae and Streptococcus uberis, under the conditions in the field. The udders of an experimental group of 20 cows of the Holstein-Friesian breed were immersed in a solution of the modified preparation DESU ? M following every time of milking over a period of three months. In the second group of 10 cows of the Holstein-Friesian breed, marked as the control group, no disinfection was applied following the milkings. The preparation DESU? Mexhibited satisfactory antimicrobial efficacy against the most frequent causes (Escherichia coli, Staphylococcus aureus, Streptococcus agalactiae and Streptococcus uberis) of mastitis in cows under laboratory conditions. With the application of the preparation DESU? M, following every time of milking over a period of three months, the number of somatic cells was reduced by almost one half in comparison with their number at the start of the experiment. No residue of the modified preparation DESU ? M were found in any sample of milk from the experimental group of cows.

The possibility of adherence of Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli or Streptococcus agalactiae to the epithelium of the mammary gland was investigated by inoculating them into this gland of mice. S. aureus, S. epidermidis and E. coli did not adhere to alveolar epithelium in suckling or non-suckling mice. S. agalactiae adhered to alveolar epithelium in non-suckling mice but adhesion was not sufficiently strong to withstand suckling. Bacterial adherence probably does not play a significant role in the establishment of mastitis by these organisms.

Najčešći izolati bili su Staphylococcus aureus u 21,74 % uzoraka i Streptococcus agalactiae u 17,39 % izolira- nih uzoraka, koagulaza-negativni stafilokoki (13,04 %), Klebsiella pneumoniae (13,04 %), Ente- rococcus spp. (8,70 %), Escherichia coli (8,70 %), Streptococcus spp., Enterobacter, Serratia spp. i Yersinia enterocolitica (4,35 %).

ABSTRACT Putative N-acetylmuramyl-l-alanine amidase genes from LambdaSa1 and LambdaSa2 prophages of Streptococcus agalactiae were cloned and expressed in Escherichia coli. The purified enzymes lysed the cell walls of Streptococcus agalactiae, Streptococcus pneumoniae, and Staphylococcus aureus. The peptidoglycan digestion products in the cell wall lysates were not consistent with amidase activity. Instead, the structure of the muropeptide digestion fragments indicated that both the LambdaSa1 and LambdaSa2 lysins exhibited γ-d-glutaminyl-l-lysine endopeptidase activity. The endopeptidase cleavage specificity of the lysins was confirmed using a synthetic peptide substrate corresponding to a portion of the stem peptide and cross bridge of Streptococcus agalactiae peptidoglycan. The LambdaSa2 lysin also displayed β-d-N-acetylglucosaminidase activity.
