The present study was planned to enable in vitro conservation of Vanda testacea, a highly medicinal orchid species through in vitro asymbiotic seed germination technique in Mitra orchid medium supplemented with cytokinins (Kn - 4.65 μM, BAP - 4.44 μM), and auxin (NAA- 5.37 μM). The germination frequency and initiation of germination was higher in NAA augmented medium and seedlings developed in 12.50 ± 0.50 weeks. Coconut water (20%) proved optimum for the multiplication of protocorm like bodies. Activated charcoal successfully checked the release of brownish exudates in the cultures. Nothodoritis zhejiangensis Z. H. Tsi is a rare and endangered epiphytic orchid restricted to a narrow distribution in China. The species is threatened with extinction as a result of loss of suitable habitats. An efficient propagation system is part of this species’ conservation plan. Seed germination reached 64.7% on Knudson's C (KC) medium containing 1.0 mg·L−1 α-naphthaleneacetic acid (NAA), 10% coconut water, and 0.1% activated charcoal. After 50 days culture, most callus (71.3%) from seed-derived protocorms formed on KC medium with 1.0 mg·L−1 2,4-dichlorophenoxyacetic acid and 10% coconut water. Callus could be subcultured 12 times during ≈2 years with more than a 3.0-fold increase from the third to the twelfth subculture. Furthermore, 84% of callus from the tenth sub-culture on KC media supplemented with 1.0 mg·L−1 NAA, 5% coconut water, and 0.1% activated charcoal formed protocorm-like bodies (PLBs). Many (57%) protocorms on KC medium containing 1.0 mg·L−1 6-benzylaminopurine and 10% coconut water formed PLBs. Both callus and PLBs formed simultaneously from different protocorms on KC medium containing 0.01 to 1.0 mg·L−1 thidiazuron and 10% coconut water. PLBs were incubated 12 times during ≈2 years with an approximate 2.4-fold increase per sub-culture. Both callus and PLBs maintained their competence to regenerate plantlets. Hyponex N026 medium supplemented with 1.0 mg·L−1 NAA, 50 g·L−1 banana homogenate, and 0.1% activated charcoal was suitable for plantlet formation and growth of 95.5% of plantlets that developed from PLBs. The roots of plantlets 2 cm in height or taller were wrapped in Chilean sphagnum moss and fixed to a fir bark block; 69.3% of plantlets survived after 180 days in a greenhouse. This protocol is an efficient means for the large-scale propagation of this endangered orchid.

The present study on in vitro seed germination of Rhynchostylis retusa obtained on nutrient medium made clear pathway for the in vitro propagation of orchids by seed culture. The presence of coconut water in the medium enhanced seed germination and growth of seedlings. For enhancing the growth of seedlings further the individual seedlings were cultured on MS medium supplemented with 3 mg/l BAP and 1g/l activated charcoal. These seedlings were sub cultured on rooting medium (IBA) supplemented with activated charcoal. The plantlets were transferred to soil with 90% success. Hence the present investigation made possible for the large scale multiplication of R.retusa and thus to conserve this endemic orchid.

An efficientin vitroregeneration protocol from seed culture has been established successfully forDendrobium chrysotoxum, an epiphytic orchid having tremendous ornamental and medicinal values. Seed germination response was encouraging in Mitra (M) medium enriched with different combinations of auxins and cytokinins. Medium supplemented with 0.4% activated charcoal (AC), 2 mg/L 6-benzyl amino purine (BAP), and 2 mg/L indole-3-acetic acid (IAA) produced best seed germination percentage in 2 weeks of culture. Incorporation of higher concentration of kinetin (KN) or BAP in combination with low auxin in medium induced pronounced shooting and leaf formation. Reduction in leaf development was evident when cytokinins exist singly in medium indicating synergistic effect of auxin and cytokinin in leaf induction. Presence of elevated level of indole-3-butyric acid (IBA) or 1-naphthalene acetic acid (NAA) with low cytokinin content in medium generated morein vitrorooting, though IBA was found to be more effective in rooting induction as compared to NAA. Thein vitroprotocol for asymbiotic seed germination developed from the present investigation can be used for rapid mass propagation of this highly importantDendrobiumorchid species.

Orchid seeds are very small, dust like in appearance, fusiform in shape, lacking endosperm and have undifferentiated embryo covered by transparent seed coat. Mycorrhizal association is required for seed germination of naturally growing orchids. In this symbiotic association, orchid species are dependent on mycorrhizal association for supply of mineral nutrients. In in-vitro condition, such demand of minerals may be compensated by external supply of sugar and mineral nutrients that are required for seed germination of orchid. Several orchids are responded by this asymbiotic seed culture and have commercial importance. Therefore, formulation of efficient in-vitro protocol is important for commercially important orchid species as well as endangered orchid species for conservation. This review paper is focused on various aspects of asymbiotic seed germination of orchids and the role of organic additives in successful seed germination.

Seeds from mature seed pods of Cypripedium calceolus var. parviflorum were germinated on 1/4 MSMO (Sigma) + 100ml/l coconut water + 1% sucrose +/- 8g/l agar (pH 6.0), and with or without prechilling at 5C for 8 weeks. Protocorm with apex (stage 3) was use as an index of germination. Seeds sown on agar medium withou chilling treatment resulted in a 40% germination rate in 120 days but the germination was very uneven. Seeds germinated on agar medium with prechilling developed more synchronously with 92% germination in 60 days (ie. about 120 days after sowing). Suspension culture of seeds without prechilling resulted in 85% germination after 90 days. The synchronization of seed germination in suspension culture was intermediate between that on agar with and without prechilling. Protocorms germinated in suspension culture appeared morphologically identical to those germinated on agar medium. All stage 3 protocorms developed further on the same agar medium in darkness. Both agar and suspension culture in media containing coconut water provided reliable seed germination methods for this orchid species.

Cattleya trianae (Linden & Reichb.f., 1860), Colombian national flower, is in danger of extinction due to the destruction of its natural habitats and excessive collection for horticultural purposes. Therefore, in vitro culture is a tool for the conservation of threatened species. In this study we determined the most suitable culture medium for asimbytic seed germination and in vitro propagation of C. trianae. Initially, mature capsules were collected, the seeds were subsequently disinfected and seeded with the syringe method (Vendrame et al., 2007), to evaluate the effect of five media on the development of C. trianae after 20 weeks. The seedlings were transplanted and acclimated using different substrates. The best percentage (54.2%) of seedling formation after 20 weeks was found in MS + JP medium with significant differences (P <0.05: Tukey HSD). In this research, it is reported that the addition of organic additives to the MS medium improves the efficacy of this, and therefore, allows a greater growth and development of C. trianae under in vitro conditions.
