Necrotic enteritis (NE) continues to present major challenges to the poultry industry, and the etiologic agent Clostridium perfringens is the fourth leading cause of bacterially-induced food- borne illnesses in the US. This study was designed to evaluate the effects of a probiotic during naturally occurring NE. On day of hatch, 1080 Cobb 500 male broilers were randomly allocated to three groups (12 replicate pens/treatment, 30 birds/pen) including 1) negative control (NC): corn-soybean meal diet; 2) positive control (PC): NC + 20 mg virginiamycin/kg diet (0.450 kg Stafac®20/ton); and 3) NC + PrimaLac (1.36 and 0.91 kg/ton from 1–21 and 22–42 days, respectively). One day (d) post placement, all birds were challenged by a commercial live oocyst coccidia vaccine as a predisposing factor to NE. Body weight and feed intake were measured at the onset of NE (d 8) and end of each feeding phase. On d 8, small intestines of two birds/pen were examined for NE lesions, and jejunum samples from one bird were collected for mRNA gene expression analysis of tight junction proteins, cytokines, and nutrient transporters. Data were analyzed using the Jump (JMP) software and significance between treatments identified by LSD (P < 0.05). Compared to NC, supplementation of probiotic reduced d 1–42 mortality; however, PC was the only group with significantly lower mortality. Despite significantly improved feed conversion ratio (FCR) in PC and probiotic groups during d 1–42, average daily gain was only higher in PC (77.69 g/bird) compared with NC (74.99 g/bird). Furthermore, probiotic and PC groups had significantly reduced lesion scores in the duodenum and jejunum compared to NC. Expression of claudin-3 was higher, while expression of zonula occluden-2 tended (P = 0.06) to be higher in probiotic-supplemented birds compared to NC. Moreover, birds fed the probiotic diet had significantly higher expression of IL-10, IL-17, AMPK-α1, and SGLT1 mRNA compared to NC birds. The expression of PepT1 was higher for the probiotic-supplemented group compared to PC. IFN-γ expression was lower in PC compared to NC, while there was no difference between probiotic and NC. There were no differences in gene expression of sIgA, TNF-α, IL-1β, and IL-22 among treatments. Collectively, these data indicate that in a naturally occurring NE model, supplementation of a probiotic helps to improve FCR and reduce lesions, potentially due to the improvements in mRNA expression of tight junctions, cytokines, and nutrient transporters.

The withdrawal of antibiotic growth promoters from poultry feed has increased the risk of necrotic enteritis (NE) outbreaks. This study examined the effects of a probiotic (PROB) or probiotic/prebiotic/essential oil supplement (PPEO) during a subclinical NE challenge. On day (d) of hatch, 960 male broilers were randomized to four groups (8 pens/treatment, 30 birds/pen) including (1) negative control (NC): corn-soybean meal diet; (2) positive control (PC): NC + 20 g Virginiamycin/ton diet; (3) NC + 227 g PROB/ton diet; and (4) NC + 453 g PPEO/ton diet. One d after placement, birds were challenged by a coccidia vaccine to induce NE. Feed intake and body weights were measured on d 8 (NE onset) and end of each feeding period. On d 8, the small intestines of three birds/pen were examined for NE lesions. Jejunum samples and ileal mucosal scrapings from one bird/pen were respectively collected to measure mRNA abundance (d 8 and d 14) and profile the microbiota (d 8 and d 42). Data were analyzed in JMP or QIIME 2 and significance between treatments identified by LSD (P &lt; 0.05). PROB and PPEO had significantly lower mortality (d 0–14) and NE lesion scores compared to NC. Feed conversion ratio was significantly lower in PC, PROB, and PPEO, while average daily gain was higher in PPEO and PC groups compared to NC from d 0–42. On d 8 and d 14, mRNA abundance of claudin-3 was higher in PPEO compared to NC. On d 14, compared to NC, mRNA abundance of sIgA and PGC-1α in PROB and PPEO were lower and higher, respectively. Compared to NC, PPEO increased mTOR abundance on d 14. On d 8, relative abundance of Clostridium sensu stricto 1, Ruminiclostridium9, Prevotellaceae, Prevotellaceae UCG-014, ASF356, and Muribaculaceae was higher in NC compared to PPEO and PROB, while Lactobacillus was lower in NC. Escherichia-Shigella had higher abundance in PC compared to PPEO and PROB. Collectively, these data indicate that during a subclinical naturally occurring NE, supplementation of PROB or PPEO supports performance and reduces intestinal lesions, potentially through modifying tight junction proteins, gut microbiota, immune responses, and cell metabolism.

Incremental levels of the phytase, OptiPhos® Plus, were fed to 150 weaned piglets (five treatments of 30 pigs) to determine the effect on piglet performance, nutrient digestibility and bone mineralisation. Piglets were fed a basal diet from four to six weeks and a pre-starter diet from six to ten weeks of age. The basal diets contained reduced digestible phosphorus (P) and calcium (Ca) (1.5 g/kg digestible P, and Ca 5.5 g/kg and 5.6 g/kg for weaner and pre-starter, respectively) to provide a negative control. Phytase was added at 0, 125, 250, 500 and 1000 phytase units (FTU)/kg feed. Faecal samples were collected at nine weeks of age and analysed for P, Ca and crude protein (CP) content to calculated digestibility. Metacarpal bone IV mineralisation was assessed by ash content and the weight and length of the bone. Incremental increases of phytase improved final body weight (BW) (P<0.001), average daily gain (ADG) (P<0.001) and feed conversion ratio (FCR) (P<0.01). The statistical model predicted a linear increase for daily feed intake (DFI), ADG and BW with increased phytase increments. BW at the end of the trial increased by 824 g per increase of 250 FTU/kg (P<0.001). ADG and DFI increased by 19 g/day (P<0.001) and 18 g/day (P<0.05) respectively, per increase of 250 FTU/kg. As the effect on ADG and final BW was higher at lowest doses, a quadratic effect for these parameters was observed, within the measured range of phytase levels (P=0.029 and 0.028, respectively). A reduction of 0.03 in FCR was observed for each 250 FTU/kg increase (P<0.01). For each increase in 250 FTU/kg, bone dry weight increased by 131 g (P<0.05) and bone ash concentration by 1.9% of fat free dry matter (P<0.001). The results showed that the addition of phytase to piglet diets low in P led to better utilisation of phytate-P and consequently better piglet performance.

SummaryThe effect of a Buttiauxella phytase, in a dose range of 0-1000 FTU/kg in feed, on production performance in growing/finishing pigs fed European type wheat, corn, barley and SBM based diet was determined. Five dietary treatments were tested including a positive control (PC), a negative control (NC) without inorganic P and with reduced Ca (–0.12%) and metabolisable energy content (–0.14 MJ ME/kg) or NC supplemented with Buttiauxella phytase (Axtra® PHY, Danisco Animal Nutrition, DuPont Industrial Biosciences, Marlborough, UK) at 250, 500 or 1000 FTU/kg respectively. A total of 100 pigs (mean initial BW of 30 kg, Topig x Pi, 10 weeks of age) were used, with 20 replicates per treatment (50% gilts and 50% barrows). Pigs were individually housed in pens and randomly allocated on the basis of body weight (BW) and gender to the five treatments. Feed, in mash form, and water were offered ad libitum. Basal diets had an intrinsic phytase level of approximately 300 and 440 FTU/kg in NC grower and NC finisher diets respectively. Mean average daily gain (ADG), average daily feed intake (ADFI) and feed conversion ratio (FCR) were calculated for the feeding phases of 30–85 kg (grower), 85–125 kg (finisher) on trial and for overall period. During the grower phase, increasing phytase dose resulted in a linear increase in ADG. Phytase inclusion at 250, 500 and 1000 FTU/kg improved ADG by 3.5, 7.2 and 8.1% respectively compared to NC and by 0.8, 4.5 and 5.3% respectively compared to those fed the PC. Performance in the finisher phase, overall period and slaughter parameters were not different between dietary treatments. The data showed that it was beneficial to increase phytase dose up to 1000 FTU/kg in grower pigs up to 85 kg BW. In the finisher phase, in pigs with BW above 85 kg, 500 FTU/kg was sufficient to replace inorganic P and maintain performance and carcass characteristics comparable to pigs fed the PC.

Two 21-day experiments were conducted to investigate the effects of exogenous enzymes on growth performance, tight junctions, and nutrient transporters, jejunal oligosaccharides and cecal short-chain fatty acids (SCFA) of broiler chickens challenged with mixed Eimeria. Two different basal diets: high fiber-adequate protein (HFAP; Expt. 1) or low fiber-low protein (LFLP; Expt. 2) were used in the two experiments. In each experiment, birds were allocated to four treatments in a 2 × 2 factorial arrangement (with or without protease and xylanase combination; with or without Eimeria challenge). In Expt. 1, with HFAP diets, Eimeria upregulated (p < 0.05) the expression of claudin-1, but downregulated (p < 0.05) glucose transporters GLUT2/GLUT5. On the contrary, enzymes downregulated (p < 0.05) claudin-1 and alleviated the Eimeria-depressed GLUT2/GLUT5 expression. In both experiments, Eimeria decreased (p < 0.05) cecal saccharolytic SCFA and increased (p < 0.05) cecal branched-chain fatty acids. The challenge × enzyme interaction (p < 0.05) showed that enzymes reversed the Eimeria effects on fermentation pattern shift. In conclusion, Eimeria altered tight junctions and nutrient transporters expression promoted cecal proteolytic fermentation and inhibited saccharolytic fermentation. Exogenous enzymes showed the potential of alleviating the Eimeria-induced intestinal gene expression changes and reversing the unfavorable cecal fermentation pattern.

An experiment was conducted to study the influence of isoamylase supplementation to maize-soybean diets on productive performance and amylase activities of broilers from 1 to 42 d of age.The results showed that different dose of isoamylase(300g/T,600 g/T,900 g/T, containing 3000IU of isoamylase/g)in the maize - soybean meal diet significantly affected the performance of broilers within 1-42 days. Within 1-21 days , the average daily gain(ADG) of experimental groups( experiment I (E I) experiment II (E II) experiment III (E III) ) improved by 3.2% -10.32%(P>0.05) compared with the control groups(negative control(NC) and positive control(PC)), average daily feed intake(ADFI) improved by 0.63%-7.43%(P>0.05), while feed conversion ratio decreased by 0.55%-4.43%(P>0.05); Within 21-42 days , the ADG of experimental groups decreased by 1.38%-4.84% and ADFI by 11.89%-15.58% (P<0.05), feed conversion ratio by 5.41%-7.54% compared with that in the NC group; Within 1-42 days , the ADG of experimental groups improved by 0.54%-4.06%(P>0.05), while ADFI decreased by 0.75%-11.83% (P<0.05), and feed conversion ratio by 3.97%-7.52% (P>0.05) compared with those in the negative control. Isoamylase supplementation influenced amylase activity of various parts of the chyme in digestive tract and the tissues of small intestine and pancreas. 21 d, the amylase activity in experimental groups improved by 4.67%-79.29%(P<0.05) compared with PC, α-amylase activity in the majority experimental groups were lower than PC group, but the difference was not significant(P>0.05).

Abstract The effect of a Buttiauxella sp. phytase on performance and bone ash in piglets was evaluated. A negative control (NC) diet was formulated without inorganic phosphate [0.11% digestible phosphorus (P)] and lower in calcium (Ca, 0.5%). NC was supplemented with the phytase at 0, 500 and 1,000 FTU/kg diet or with mono-calcium phosphate (MCP) at 4 levels to add 0, 0.7, 0.14 and 0.18% digestible P and with additional limestone to maintain Ca to P ratio (1.2 to 1.3). The latter was the positive control (PC) and contained 0.7% Ca and 0.29% digestible P, meeting NRC requirement. A total of 108 Pietrain × (Landrace × Yorkshire) piglets were fed adaptation diets from 21 (weaned) to 42 d of age. Then, they were divided into 9 blocks according to body weight (BW) (9 replicates/treatment, mean initial BW 10.2 kg, mixed sex) and allocated at 2 piglets per pen. Piglets were fed mash test diets (based on corn, soybean meal with broken rice, rice bran and sugar beet pulp) ad lib for 28 d. At the end, 1 piglet per pen was euthanized and the right foot collected to determine metacarpal ash, Ca and P content. Data were analyzed using JMP 14.0. The NC reduced (P &lt; 0.05) average daily gain (ADG), average daily feed intake (ADFI), bone ash, Ca and P, and increased feed conversion ratio (FCR) vs PC (Table 1). Increasing phytase and MCP linearly increased (P &lt; 0.05) ADG, bone ash, Ca and P and reduced FCR. Linear regression analysis using increasing digestible P from MCP as reference indicated that phytase at 1,000 FTU/kg can replace 0.16, 0.18 or 0.16% digestible P from MCP, using bone ash, ADG or FCR as response parameter, respectively. In conclusion, Buttiauxella phytase improved performance and bone ash in piglets fed diet without added inorganic phosphate.

AbstractThis study evaluated the effects of three different thermostable phytase variants, based on the AppA gene from E. coli (AppAT1, AppAT2 and AppAT3) on growth performance, nutrient digestibility and bone mineralisation in broiler chickens at inclusion levels of 250 and 500 FTU/kg. The eight treatment groups included a positive control (PC) which was sufficient in Ca and P, a negative control (NC, the same basal formulation as the PC, but reduced in Ca and P), and NC supplemented with AppAT1 at 250 and 500 FTU/kg (AppAT1-250 and AppAT1-500), AppAT2 at 250 and 500 FTU/kg (AppAT2-250 and AppAT2-500) and with AppAT3 at 250 and 500 FTU/kg (AppAT3-250 and AppAT3-500). Over the entire feeding period, body weight (BW) and average daily gain (ADG) were significantly higher in the PC group, with all phytase supplemented groups being statistically the same, compared to the NC group. Feed conversion (FCR) for the PC-fed birds (1.479) was significantly (P&lt;0.05) better compared to the NC birds (1.582) and those fed the AppAT3-250 diet (1.523). Reduced levels of Ca and P in the NC group led to significantly (P&lt;0.05) lower tibia ash (40.9%) compared to the PC group (47.4%). Birds fed the phytase diets had significantly higher tibia ash compared to the NC birds, with those from the AppAT2-500 and AppAT3-500 groups being statistically the same as the PC group. Diets AppAT1-500, AppAT2-250, AppAT2-500 and AppAT3-500 significantly increased Ca digestibility compared to the NC. Apparent total track digestibility (ATTD) of P was improved for AppAT1-500 and AppAT2-250. The ATTD of Ca and P for all of the phytase supplemented groups reached the same level of the PC and AppAT1-500 group. It was concluded that adding any of the phytases tested, especially when included at 500 FTU/kg to a feed reduced in Ca and P, led to improved performance and bone mineralisation back to the same levels as seen for the Ca and P sufficient diet.

The following study evaluated effects of a xylanase and beta-glucanase combination on growth performance of broilers fed energy reduced versus nutritionally adequate maize-soybean meal-based diets. A total of 648, one-day-old male broilers (Ross 308) were assigned to floor-pens (24 birds/pen, nine pens/treatment, three treatments) in a randomised block design. Treatments included: (1) a nutritionally adequate positive control diet (PC); (2) a negative control (NC) diet in which energy, crude protein and digestible amino acids were reduced by 3.4% (-105 kcal apparent metabolisable energy), 2.3% and 1.2 to 3.0% vs PC, respectively; and (3) NC plus a xylanase and beta-glucanase combination that supplied 1,220 U xylanase and 152 U beta-glucanase per kilogram of final feed. All diets contained a background of 500 FTU/kg phytase and were offered to birds ad libitum. Birds fed NC showed reduced average daily gain (ADG) by -6.1% (P<0.05); increased feed conversion ratio (FCR) by 9.2 points (P<0.05), and overall (d 1-35) body weight corrected FCR which was increased by 9.4 points (P<0.05) vs the PC group. Enzyme supplementation increased final BW (+4.2%, P<0.05), ADG (+5.4%, P<0.05) and tended to reduce FCR (+7.5 points, P=0.054) from d 22-35 vs NC, without affecting average daily feed intake. Improvements in performance due to the enzyme combination were equivalent to performance on the PC diet in all cases. The results suggested that significant improvements in growth performance of broilers fed maize-soybean meal-based diets which had been reduced in energy and nutrients can be realised by supplementation with xylanase in combination with beta-glucanase.

The aim of this study was to evaluate the impact of a probiotic Bacillus licheniformis strain (DSM 28710; B-Act®) on growth performance and its capacity to mitigate necrotic enteritis (NE; induced via a Clostridium perfringens challenge) in poultry. A broiler trial was conducted, examining three treatments for 42 days under an induced NE challenge; a negative control (basal diet only); an antibiotic treated group (oxytetracycline hydrochloride (OXT), therapeutic dose of 105 mg OXT/litre in drinking water, for three days after C. perfringens challenge); and a B-Act group (500 g B-Act/tonne of feed, equalling 1.6×1012 colony forming units B. licheniformis DSM 28710/tonne of feed, supplemented from start until finish). Despite the induced NE challenge, weight gains of the B-Act and OXT groups were similar to each other but significantly higher compared to the control at the end of the study (P<0.05). Weight gain of the B-Act group was already significantly higher compared to the control on day 21 (P<0.05), indicating a potential benefit of the probiotic even before clinical establishment of NE. Feed conversion ratio (FCR) values followed a similar pattern throughout the study, with a significantly lower overall FCR for the B-Act and OXT groups compared to the control (P<0.05; d0-42). Birds fed B-Act had significantly (P<0.05) lower NE lesions compared to the control and OXT group on day 21, although OXT was not supplemented to the animals at this stage yet. Both B-Act and OXT groups had significantly (P<0.05) lower NE scores than the control on day 28, demonstrating the effectiveness of the antibiotic treatment and the mitigating effect of B-Act on the effects of a Clostridium perfringens induced NE challenge.
