AbstractThe roots of Morus species are well described in the Pharmacopoeia of the People's Republic of China (ChP) for its traditional use in treating liver fibrosis due to its hepatoprotective property. However, little is known about the hepatoprotective effect of the roots of Morus indica L. (RoMi), and the pharmacological mechanism(s) are uncertain due to its intricacy. Therefore, this study evaluates the hepatoprotective activity of the ethanolic extract of RoMi (eRoMi) against the CCl4-induced in-vivo animal model at different dosages (100 and 200 mg/kg BW) in comparison with silymarin as a positive control. The hepatoprotective activity of eRoMi was evaluated by measuring the levels of serum biomarkers, hepatic antioxidant enzymes and was verified by histological studies. Interestingly, 1,2-bis(trimethylsilyl) benzene, 1,4-phenylenebis (trimethylsilane), 2,4,6-cycloheptatriene-1-one, 3,5-bis-trimethylsilyl and α-amyrin were the active components found in eRoMi as detected by GC–MS. Oral administration of eRoMi (200 mg/kg BW) to rats significantly protected serum biochemical parameters (increased ALT, AST, LDH, bilirubin and GGT as well as depletion of antioxidant enzymes and hepatic GSH) and elevation in hepatic lipid peroxidation as compared to CCl4-treated rats. The hematological indices such as erythrocytes, hemoglobin, monocytes and lymphocytes were also normal in eRoMi-treated rats. The histopathological evaluation indicated a significant restoration of liver structure as compared to silymarin. This study is the first scientific validation for the traditional use of eRoMi to understand its hepatoprotective activity.

The current study was conducted to assess the hepatoprotective activity of different extracts of M. rubicaulis (Lam.) at (200 and 400 mg/kg) doses b.w. against CCl4 induced liver intoxication in Albino Wistar rats. Male or female Wistar rats about 150 and 200 gm body weight were selected for present study. The animals were divided into thirteen groups, six rats in each group. The extracts and Silymarin-treated animal groups significantly reduced the activities of various biochemical markers such as SGPT, SGOT, ALP, and TB which were elevated by carbon tetrachloride (CCl4) intoxication. The extracts of M. rubicaulis (Lam.) showed a dose dependent hepatoprotection activity. Among all the extracts, ethanolic extract produced maximum hepatoprotection (SGPT-83.15 %, SGOT76.82%, ALP-79.33%, TB-80.00%) at a 400mg/kg dose. After CCl4 administration, the levels of hepatic-antioxidant enzymes such as Glutathione (GSH) and Catalase (CAT) were reduced, whereas the level of hepatic lipid peroxidation (-LPO) increased. These hepatic antioxidant enzymes were also restored to normal levels by extracts and Silymarin treatment. The results of the present investigation indicate that all the extracts of M. rubicaulis (Lam.) possess hepatoprotective activity which may be due to the presence of various chemical constituents.

The purpose of this study was to see if ethanolic and aqueous extracts of Bauhinia acuminatea (Linn.) had any hepatoprotective activity against CCl4-induced albino rats. The levels of biochemical parameters (SGOT, SGPT and ALP) were reduced and the levels of hepatic antioxidant enzymes such as GSH and CAT were decreased whereas the level of hepatic lipid peroxidation (MDA) was elevated byCCl4 induction in Albino rats when compared with the normal group. The ethanolic and aqueous extract of Bauhinia acuminata (Linn.) and Silymarin treated animal groups showed significant decrease in activities of different biochemical parameters like SGOT, SGPT, ALP and lipid peroxidase i.e., MDA level which were elevated by carbon tetrachloride (CCl4) intoxication, at doses of 200 and 400 mg/kg and increased the level of antioxidant enzymes such as GSH and CAT. The high dose of ethanolic extracts (400 mg/kg) was more effective as compared to low dose (200 mg/kg). So, it was concluded from the result that the ethanolic extract of Bauhinia acuminata (Linn.) possesses significant hepatoprotective activity compared to aqueous extracts against CCl4 induced hepatotoxicity in rats.

ABSTRACT: The study is designed for the evaluation of in-vivo Hepato protective and in-vitro Anti oxidant activity of ethanol extracts from the bark of Shorea robusta (Dipterocarpaceae) by CCl4 induced hepatotoxicity in rats. Ethanol extracts from the bark Shorea robusta (EESR) was evaluated for hepatoprotective activity in rats by inducing liver damage by CCl4. The anti oxidant activity of EESR was assayed by various in-vitro antioxidant methods and activities were compared to standard ascorbic acid. Ethanol extracts at an oral dose 200mg/kg and 400mg/kg exhibited a significant (*p<0.005) protective effects by lowering the level of SGOT, SGPT, ALP, Serum bilirubin, total cholesterol and increasing the level of total proteins as compared to Silymarin (50mg/kg) used as positive control. The extracts exhibit significant anti oxidant activity in various in vitro anti oxidant models.  From these studies we are concluding that, the ethanolic extracts of S.robusta have potent hepatoprotective effects and have anti oxidant properties, hence can be used as a natural product against liver damage.KEY WORDS: Anti oxidant, Carbon tetra chloride,  Hepatoprotective,  Shorea robusta

Aims: This study was carried out to investigate the antimicrobial activity of Chrysophyllum albidum leaves extract on selected Gastro-instestinal bacteria such as Salmonella typhimurium, Shigella dysentariae, Vibrio cholera, Escherichia coli and Clostridium perfringens. Methodology: The leaves were extracted using ethanol, methanol and distilled water; the concentration of the extracts employed were 100 mg/ml, 200 mg/ml, 400 mg/ml and 500 mg/ml respectively; however the   leaf extracts of Chrysophyllum albidum were screened for anti-microbial activity using the in vitro cup-plate method of agar diffusion technique with concentration of 10-5cells/ml of the selected bacteria. Simultaneously, 30 µg tetracycline and 30 µg metronidazole were used as positive control. Results: The result showed that the most active among them is Tetracycline; followed by ethanolic extract, aqueous extract, methanolic extract and metronidazole extract respectively on the tested bacteria. Conclusion: This research justifies the traditional use of the leaves of Chrysophyllum albidum for the therapeutic purposes; hence can be commercialized by pharmaceutical outfit; if not for anything but its availability and readily for human consumption.

Introduction: Diabetes mellitus is an endocrine and metabolic disorder characterized by the defect in insulin secretion or insulin action or both. The association between diabetes mellitus and oxidative stress is well established. Cucurbitaceae is the largest family containing 120 genera mostly grown for its sweet and juicy fruit in warm climates all over the world. In the present work ethanolic extracts of the seeds of Trichosanthes cucumerina Linn (Chichinno), Lagenaria siceraria (Lauka), Cucurbita pepo (Pharsi), Luffa aegiptiaca (Ghiraula), and Benincasa hispida (Kubindo) collected from Western Nepal has been evaluated for their in vitro antioxidant activity and in vivo hypoglycemic effects. Methods: In vitro antioxidant activity was assessed by using DPPH free radical scavenging activities and their IC50 values were calculated. In vivo hypoglycemic effect was examined on normoglycemic rats. The clinical significance of ethanolic extract at the doses of 250 and 500 mg/kg body weight was investigated in 0, 30, 60, 120 and 180 minutes of oral administration. Metformin treated group was used as the positive control. Results: For DPPH radical scavenging action, the IC50 values of Trichosanthes cucumerina, Luffa aegiptiaca, Benincasa  hispida, Cucurbita pepo, Lagenaria siceraria were found to be 60.72, 127.73, 49.63, 98.16 and 52.46 µg/ml respectively which were compared to the IC50 value of ascorbic acid (Positive control) which was found to be 38.11 µg/ml. The extracts of Benincasa hispida, and were having higher antioxidant activity and tested for in vivo hypoglycemic activity.In vivo administration of two doses of ethanolic extract of Lagenaria siceraria reduced the level of blood glucose while the best result was obtained at 250 mg/kg. Conclusions: Present study revealed promising antioxidant and hypoglycemic activity of ethanolic extract of Lagenaria siceraria. The further exploration of Lagenaria siceraria for its effective use in the traditional medicinal system is essential.

The present study aimed to analyze the phytoconstituents of Neptunia triquetra (Vahl) Benth. Anti-inflammatory and hepatoprotective activities of ethanol (EE), chloroform (CE) and dichloromethane (DCME) of stem extracts were evaluated using in vivo experimental models. The extracts were analyzed for phytoconstituents using GC-HRMS. Anti-inflammatory activity of CE, EE and DCME was accessed using carrageenan-induced paw oedema, cotton pellet-induced granuloma and the carrageenan-induced air-pouch model in Wistar albino rats. The hepatotoxicity-induced animal models were investigated for the biochemical markers in serum (AST, ALT, ALP, GGT, total lipids and total protein) and liver (total protein, total lipids, GSH and wet liver weight). In the in vivo study, animals were divided into different groups (six in each group) for accessing the anti-inflammatory and hepatoprotective activity, respectively. GC-HRMS analysis revealed the presence of 102 compounds, among which 24 were active secondary metabolites. In vivo anti-inflammatory activity of stem extracts was found in the order: indomethacin > chloroform extract (CE) > dichloromethane extract (DCME) > ethanolic extract (EE), and hepatoprotective activity of stem extracts in the order: CE > silymarin > EE > DCME. The results indicate that N. triquetra stem has a higher hepatoprotective effect than silymarin, however the anti-inflammatory response was in accordance with or lower than indomethacin.

<p class="AbstractContent"><strong>Objective:</strong> Diabetes Mellitus is a leading illness to the society and despite of a lot of researches, not a single cure has been discovered. Most of the time, people focused on fighting diabetes and medicinal plants were utilized. The purpose of this study was to investigate the hypoglycemic effect of Scarlet Spiral Flag (<em>Costus woodsonii</em>) and to be reviewed for preclinical trials.</p><p class="AbstractContent"><strong>Methods: </strong>20 male Swiss mice were made diabetic by inducing Alloxan monohydrate. Then separated randomly into 4 groups with different treatments administered daily for 7 days; Control, Positive Control (600 µg/kg Glibenclamide), T1 (250mg/kg Scarlet Spiral Flag ethanolic extract) and T2 (500mg/kg Scarlet Spiral Flag ethanolic extract). Their blood sugar was monitored onset after 1<sup>st</sup> treatment (6<sup>th</sup>, 12<sup>th</sup> and 24<sup>th</sup> hours) and during the week within the 1<sup>st</sup>, 4<sup>th</sup> and 8<sup>th</sup> day.</p><p class="AbstractContent"><strong>Results:</strong> Both the onset and fasting blood sugar monitoring showed a significant decrease in the mean average blood glucose level of the mice.<strong></strong></p><p class="AbstractContent"><strong>Conclusion: </strong>The ethanolic extract of Scarlet Spiral Flag (<em>Costus woodsonii</em>) has a potent antidiabetic effect in Alloxan-induced diabetic mice. Background: The present research reported isolation, characterization of protopine from Hedyotis corymbosa and investigation of in-vivo hepatoprotective activity of protopine against simvastatin induced hepatotoxicity in experimental rodents. Methods: Protopine was isolated from H. coryambosa by column chromatography using chloroform: methanol: diethylamine (9:1:1) as the mobile phase and structural characterization was done by UV, FTIR, 1H-NMR and 13C-NMR and mass spectroscopy, followed by determination of in-vivo liver protective effect of protopine against simvastatin (20 mg/kg, p.o.), induced hepatotoxicity in experimental rodents. The liver protective activity was assessed by interpreting distinct biochemical parameters like SGOT, SGPT, cholesterol, urea, total bilirubin, total protein and albumin along with the haematological and histopathological studies. Results: The reports of spectroscopic techniques confirmed that the isolated compound is protopine, an isoquinoline alkaloid. The treatment with protopine significantly at (P<0.05-P<0.001) and dosedependently reversed simvastatin induced elevated level of SGOT, SGPT, cholesterol, urea, total bilirubin and restored the total protein and albumin level in rodents. Furthermore, protopine also signifies the blood parameters at a dose of 11 and 22 mg/kg and restored the defence mechanism of the body. The histological examination revealed that protopine at a dose of 22 mg/kg showed the regeneration of hepatocytes around central vein with near normal liver architecture. Conclusion: The results of the current study confirm the liver protective effect of protopine against simvastatin induced hepatotoxicity and therefore, scientifically support its traditional use.

In this study we investigated the in vivo Hepatoprotective activity of ethanolic extract of Garuga pinnata (EEGP) leaves in Carbon tetrachloride (CCl4) induced hepatotoxicity using wistar rats of either sex as model. Hepatotoxicity was induced by the administration of CCl4 intraperitoneally (0.125ml CCl4 in liquid paraffin (1:1) per 100g body weight). Garuga pinnata leaves extract at different dose levels (200 and 400mg/kg, p.o.) showed the dose dependant hepatoprotective effect and was compared with well known standard hepatoprotective Silymarain (100mg/kg). When groups were treated with CCl4, significant increase in serum biochemical parameters such as Serum Glutamate Oxaloacetate Transaminase (SGOT), Serum Glutamate Pyruvate Transaminase (SGPT), Alkaline phosphate (ALP), Acid Phosphate (ACP), Creatinine and alteration of tissue biochemical parameters such as reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), lipid peroxidation (LPO), and the total proteins were observed. The histopathological examination of the CCl4 treated groups showed sinusoidal congestion, centrilobular necrosis, marked vacuolations and congestion. However, pretreatment with extract of leaves of Garuga pinnata significantly reduced the increased serum levels of biochemical parameters and restored antioxidant defense enzymes level to its normal. Moreover, histopathology of leaves extract treated groups showed normal architecture with minimal sinusoidal congestion. Taken together, our study concludes that EEGP to be a more potential agent for caring liver from CCl4 induced damage.

Ziziphus rugosa Lam. belongs to the family Rhamnaceae and is found chiefly in deciduous and semi evergreen forest of Western Ghats. The present research was undertaken to establish in vitro antioxidant and in vivo hepatoprotective activity of ethanolic extract of Z.rugosa Lam. leaves. The powdered leaves of Z. rugosa were extracted with ethanol and preliminary phytochemical screening was performed for the presence of various phytoconstituents. DPPH assay and β-glucuronidase inhibition assay were selected for the free radical scavenging activity. For the assessment of hepatoprotective activity, alcohol and CCl4 induced hepatotoxicity model were used. The phytochemical analysis of ethanolic extract showed the presence of alkaloids, saponins and flavonoids. The extract exhibited concentration dependent radical scavenging activity with an IC50 value of 61.88 μg/ml and β –glucoronidase inhibition activity with an IC50 value of 70.61 μg/ml. It was speculated that the Z. rugosa Lam. ethanolic extract shows dosedependent hepatoprotective activity which is equivalent with the standard drug Silymarin. The inhibition of free radicals or free radical scavenging activity is significant in the protection against CCl4 and alcohol induced hepatopathy. Hence, it is likely that the antioxidant activity of ethanolic extract of Z. rugosa Lam. might contribute to the hepatoprotective action.
