Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.

ABSTRACT: In the present study, antibacterial and antioxidant [1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity] activities of a weed, slender amaranth (Amaranthus viridis L.) were investigated. Extracts of different plant parts were prepared in n-hexane, chloroform and ethyl acetate. Antibacterial activity was measured by using 100 mg mL-1 concentration extracts against 4 deadly phytopathogenic bacterial species viz. Pseudomonas syringae Van Hall, Ralstonia solanacearum Smith, Erwinia carotovora (Jones), Holland and Xanthomonas axonopodis Hasse. In antioxidants assays, 10, 20 and 30 mg mL-1 extracts were used keeping DPPH as control. In these bioassays, ethyl acetate fraction of A. viridis leaf exhibited the best antibacterial and antioxidants activity. Ethyl acetate leaf fraction showed the highest inhibition zone diameter (IZD) where it caused 21 mm IZD against P. syringae and 19 mm IZD against E. carotovora. This extract also showed 22, 52 and 84% antioxidant activity at 10, 20 and 30 mg mL-1 concentrations, respectively. Previously there is no report available that describes antibacterial activity of root extract of A. viridis against P. syringae. Moreover, antioxidant activity of stem and root extracts in n-hexane, chloroform and ethyl acetate was investigated first time in the world. It was concluded that the biological activities observed during the present investigation may be due to the presence of bioactive constituents that can be harnessed as natural antibacterials and antioxidants.

A hydromethanolic root extract of Ampelocissus martinii Planch. (A. martinii) was analyzed by standard methods for its phytochemical content, antioxidant activity, α-amylase and α-glucosidase inhibitions and antibacterial activities. The root extract exhibited the highest content of saponins, followed by phenols, proanthocyanidin and flavonoids, respectively. It showed high antioxidant activity in FRAP and CUPRAC assays. The root extract and standard Trolox had similar antioxidant activities in the DPPH and ABTS assay. It also showed much higher α-amylase and α-glucosidase inhibitory activity compared to standard acarbose. Moreover, the root extract inhibited all tested Gram-positive bacteria with the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of 6.25 mg/mL. These results indicate that A. martinii root can be pharmaceutically used as active ingredients to prevent bacterial infection and radical-related diseases especially diabetes.

Chokeberry (Aronia Medik.) is a non-traditional fruit plant known as a rich source of biologically active compounds and inhibits the numerous biological activities. We compared the antioxidant activity and phenolic compounds of fruits between widely cultivated Aronia mitschurinii (AM-TCH, from Tchekhov district; AM-D, from Dmitrov district; AM-OZ, from Orekhovo-Zuevsky district of Moscow region, Russia) and introduced North American Aronia species (Aronia arbutifolia (AA-M), A. melanocarpa (AML-M), A. × prunifolia (AP-M), which have not been planted yet in the arboretum of Main Botanical Garden of the Russian Academy of Sciences (Moscow). Studying samples were collected in their secondary distribution range. Ethanolic extracts were determined for antioxidant capacity (antioxidant activity by DPPH and phosphomolybdenum methods, the total content of polyphenols, flavonoids, phenolic acids) and measured spectrophotometrically. As standards were used Trolox (TE) for antioxidant activities, gallic acid (GAE) for polyphenol content, quercetin (QE) for flavonoid content, caffeic acid (CAE) for phenolic acid content. The antioxidant activity by DPPH method in ethanol extracts of investigated plants was from 6.96 (AM-D) to 8.89 (AM-OZ) mg TE.g-1 DW. Reducing the power of investigated extracts exhibited activity from 151.47 (AM-OZ) to 297.8 (AA-M) mg TE.g-1 DW. The content of polyphenol compounds determined from 25.98 (AM-TCH) to 54.39 (AA-M) mg GAE.g-1 DW, phenolic acids content was from 7.76 (AP-M) to 11.87 (AM-D) mg CAE.g-1 DW and the content of flavonoids detected from 8.12 (AM-OZ) to 16.62 (AM-D) mg QE.-1 DW. Obtained data showed a strong correlation between the content of polyphenol compounds and reducing the power of extracts (r = 0.700), between flavonoids and phenolic acids (r = 0.771) and also between phenolic acids and reducing power (r = 0.753) in Aronia ethanol extracts. Fruits of investigated species of Aronia can be propagated as a source of polyphenol compounds with antioxidant activity and obtained results may use for farther pharmacological study.

Summary While there is a large number of scientific papers reporting chemical composition and biological activities of Aronia melanocarpa, there is a lack information regarding the commercially available bagged tea. In order to supply new information on the antioxidant activity of the Aronia melanocarpa tea infusions, the aim of this study was to evaluate individual phenolic compounds which could be responsible for antioxidant activities of these beverages. Selected anthocyanins (cyanidin-3-O-galactoside, cyanidin-3-O-glucoside, cyanidin- 3-O-arabinoside, and cyanidin-3-O-xyloside), gallic acid, caffeic acid, rutin, morin, and protocatechuic acid were simultaneously detected from commercially available tea infusions using a High Performance Liquid Chromatographic (HPLC) method. The antioxidant activity was measured using five in vitro spectrophotometric methods: 1,1-diphenyl- 2-picrylhydrazyl radical scavenging activity (DPPH), 2,2'-azino-bis (3-ethylbenzthiazoline- 6-sulphonic acid) radical cation scavenging activity (ABTS), ferric reducing-antioxidant power (FRAP) and reduction power (RP)Fe(III) to Fe(II). Obtained results showed that anthocyanins, predominantly of cyanidin-3-O-galactoside, are the major class of polyphenolic compounds in tea infusions. Among phenolic acids the most abundant is caffeic acid. A significant correlation between DPPH and ABTS and FRAP and RP suggested that antioxidant components in these beverages were capable scavenging free radicals and reducing oxidants. Generally, these beverages had relatively high antioxidant capacities and could be important dietary sources of antioxidant phenolics for the prevention of diseases caused by oxidative stress.

Plants produce wide variety of phytochemical compounds which have been known to perform most biological activities such as antimicrobial, anticancer, antioxidant activities etc. Plants which possess these medicinal properties are known to be medicinal plants. In this study, Annona muricata, Piper betle and Mentha spicata were collected from Chennai, Tamil Nadu, India. The crude extracts were taken using various solvents like acetone, ethanol, chloroform, toluene and petroleum ether. All the extracts were subjected for qualitative analysis for the phytochemicals. All the samples were also subjected for antibacterial activity and TLC bioautography analysis for antioxidants. Since they found to possess antioxidant activity, quantitative assay for antioxidant activity like DPPH and FRAP assays were performed.Keywords: Annona muricata, Piper betle and Mentha spicata

Objective: This study focuses on the antioxidant potential of Djibouti brown seaweed and their phenolic contents. Methods: We evaluated the antioxidant potential by DPPH method (1,1-diphenyl-2-picrylhydrazyl) and their phenolic contents of seven Djibouti seaweed: Cytoseira myrica, Padina pavonica, Sargassum fluitans, Sargassum ilifolium, Sargassum sp, Turbinaria triquetra and Turbinaria turbinata. Also, we searched the secondary metabolites of these seaweeds. Results: We obtain a higher antioxidant activity at 60,7±0,9 % and a higher phenolic content at 199,01±0,5 μg equivalent phloroglucinol (PGE)/g dry matter for Padina pavonica. A good linear correlation (R2 = 0,898) is observed between the antioxidant activity and the phenolic content of the seaweed studied. Also, two Padina pavonica collected in two different locations have different biochemical concentrations and antioxidant activity, suggesting the influence of the marine environment on the biosynthesis of secondary metabolites and the biological activities of seaweed. The present study shows the presence of tannins, saponosides, flavonoids and steroid-terpenes. Conclusion: The species studied show interesting antioxidant activities and can be consumed to prevent oxidative stress.

A method for the separation and quantification of acteoside by reverse-phase high-performance liquid chromatography (HPLC) was developed and validated. Hot water and ethanolic extracts of Osmanthus fragrans leaves and flowers were analyzed for acteoside content. Excellent linearity was obtained, with an r2 higher than 0.999. The precision, specificity, and accuracy of our method were excellent, suggesting that it can be conveniently used for the quantification of acteoside in the crude extract of O. fragrans. The hot water and ethanol extracts were analyzed, and their biological activities were tested. The extraction yields, marker (acteoside) contents, and antioxidant activities of the leaf and flower extracts were analyzed. The antioxidant activity was confirmed by measuring the 2,2-diphenyl-2-picrylhydrazyl radical scavenging activity, reducing power, and total phenolic content. The acteoside content tended to be higher in the 100% ethanol extract of O. fragrans compared to those with the other extraction conditions tested. Overall, almost all extracts prepared with ethanolic solvents tended to produce better antioxidant activity than those prepared with hot water. These results suggest that the ethanolic extract of O. fragrans could serve as a potential antioxidant and anti-inflammatory pharmaceutical source, and our validated method would be useful for the quality control of O. fragrans extracts.

AbstractThe aim of the study was to assess the effect of three weeks of storage on the chemical and rheological properties of apple juices obtained from Idared and Jonatan apples variety. Total antioxidant activity, levels of bio-active compound groups and the viscosity were measured to characterize the investigated juices. The method applied for the determination of ascorbic acid concentration was with 2, 6-diclorophenolindophenol. Total phenols (TP) in apple juices were determined using the Folin-Ciocalteau method and antioxidant activity by the use of DPPH free radical method. The viscosity of apple juices was investigated by a rotational viscometer, Brookfield viscometer (Brookfield Engineering Inc., Model RV-DV I Prime) with RV spindles. During three weeks of storage, different rates of all measured properties have been observed decreasing for both studied apples varieties juices. The juices from Jonatan apples have higher antioxidant activities that are correlated with the higher content in polyphenols and lower values of viscosity.

Background: Lactoferrin (LF) exhibits multiple beneficial biological activities and thus has been used as a health food and additive. To broaden its application in the food industry, the porcine LF (pLF) gene has been engineered into rice to produce recombinant LF (rpLF) for use as a food additive. The iron-binding and antimicrobial activities of rpLF and its positive effects on early weaned piglets have been previously evaluated, yet several features, such as the signal peptide removal, glycosylation sites and antioxidant activity of rpLF, have not been fully characterized. Objective: In this work, the rice-produced rpLF was purified and its biochemical structure and antioxidant activities characterized. Methods: HPLC, Western blot, PAS/VVL/PNA staining, Edman degradation assay, MALDI-TOF, LC-MS/MS and antioxidant activity assays were performed. Results: The results showed that this purified rpLF is a mature form of LF; its signal peptide was correctly removed, and two N-glycosylation sites located at N365 and N472 were identified. The molecular mass heterogeneity of rpLF could be eliminated by treatment with PNGase glycosidase, suggesting that different degrees of N-glycosylation occur in rpLF. A series of assays including the iron chelating activity, reducing power assay, lipid peroxidase activity and radical-scavenging activity showed that the antioxidant activity of rice-produced rpLF was equivalent to that of bovine LF. Conclusion: Rice-produced rpLF was correctly processed post-translationally and displayed antioxidant activity equivalent to that of bovine LF; thus, rice-produced rpLF can be recognized as a plant-based antioxidant to be used as a functional additive in animal feed and for the food industry.

Kersen trees (Muntingia calabura L.) are included in the Elaeocarpaceae family and are mostly used as a protective tree. The kersen leaves were an environmental waste (trash) and still lacks implementation in food processing. This study was aimed to determine the chemical characteristics and antioxidant activity of kersen leaves at different altitudes and their potential as an herbal tea. The difference in the height of the place to grow will be used as a factor to be studied so that this study uses a completely randomized design with a one factor pattern (height the place of growings). Kersen leaves were taken from three districts, namely Jember (83 amsl/above mean sea level), Situbondo (5 amsl), and Bondowoso (255 amsl). The results showed that the chemical characteristics of kersen leaves consisting of moisture content, ash content, fat content, and carbohydrate content were not significantly different between regions, while protein content and antioxidant activity levels were significantly different between regions.
