Candidiasis is one of the frequently encountered opportunistic infections in the oral cavity and can be found in acute and chronic presentations. The study aimed to develop fluconazole-loaded sesame oil containing nanotransfersomes (FS-NTF) by the thin-layer evaporation technique to improve the local treatment of oral candidiasis. Optimization of the formulation was performed using the Box‒Behnken statistical design to determine the variable parameters that influence the vesicle size, entrapment efficiency, zone of inhibition, and ulcer index. Finally, the formulated FS-NTF was embedded within the hyaluronic acid‒based hydrogel (HA-FS-NTF). The rheological behavior of the optimized HA-FS-NTF was assessed and the thixotropic behavior with the pseudoplastic flow was recorded; this is desirable for an oral application. An in vitro release study revealed the rapid release of fluconazole from the HA-FS-NTF. This was significantly higher when compared with the fluconazole suspension and hyaluronic acid hydrogel containing fluconazole. Correspondingly, the ex vivo permeation was also found to be higher in HA-FS-NTF in sheep buccal mucosa (400 μg/cm2) when compared with the fluconazole suspension (122 μg/cm2) and hyaluronic acid hydrogel (294 μg/cm2). The optimized formulation had an inhibition zone of 14.33 ± 0.76 mm and enhanced antifungal efficacy for the ulcer index (0.67 ± 0.29) in immunocompromised animals with Candida infection; these findings were superior to those of other tested formulations. Hence, it can be summarized that fluconazole can effectively be delivered for the treatment of oral candidiasis when it is entrapped in a nanotransfersome carrier and embedded into cross-linked hyaluronic acid hydrogel.

Elderly individuals are at increased risk of oral thrush (oral candidiasis) due to decreased saliva secretion. Due to their antimicrobial properties, edible oils can be effective natural agents for oral care. The objective of the present study was to compare the effects of sesame oil, which is widely used for cooking in Asian countries, and two other edible oils on the growth of both mycelial and yeast forms of five clinical isolates of Candida albicans, a causative microorganism of oral thrush. We assessed the effect of each oil in concentrations of 0.078%, 0.156%, and 0.313% on growth of the mycelial forms of the clinical isolates over 24 hr using the crystal violet method. We also evaluated the effect of each oil on growth of the yeast forms by counting the number of viable yeast cells after culturing in the oils for 24 hr. Sesame oil inhibited the growth of both mycelial and yeast forms. Safflower and olive oil also inhibited the growth of both forms of C. albicans but to a lesser extent than sesame oil. The ability to inhibit the growth of the mycelial form correlated with sesame oil concentration. Roasting influenced growth inhibition ability and high-roasted sesame oil most effectively inhibited the yeast form. The growth inhibitory effect differed among the five isolates. We hypothesize that the sesamin and fatty acid components of sesame oil are involved in its antifungal activity.

It has been previously demonstrated that DMBA-induced rat mammary carcinoma originates in the terminal end bud (TEB) of the mammary gland by proliferation of intermediate type cells (1). The earliest lesion identified is the intraductal proliferation (IDP), which gives rise to intraductal carcinomas. These evolve to cribriform, papillary and comedo types (2). In the present work, we report the ultrastructural changes that take place in the IDP for the formation of a cribriform pattern.Fifty-five-day-old Sprague Dawley virgin female rats were inoculated intra- gastrically with 20 mg 7,12-dimethylbenz(a)anthracene (DMBA) in 1 ml sesame oil. Non-inoculated, age-matched females were used as controls. Mammary glands from both control and experimental rats were removed weekly from the time of inoculation until 86 days post-inoculation. The glands were fixed and processed for electron microscopy (2).The first change observed in IDP's was the widening of intercellular spaces and the secretion of an electron dense material into these spaces (Fig. 1).

The aim of this study was to investigate the ability of NLC in increasing photostability of tomato extract in term of antioxidant activity. Photostability testing on antioxidant activity of samples were conducted by accelerating method using UVB radiation 32.400 joule for 21 hours radiation. Antioxidant activity was measured by DPPH method. NLC was made by High Shear Homogenization (HPH) method at 24000 rpm for 4 cycles, while conventional creame was made by low speed at 400 rpm. The product were characterized include: pH, viscosity, and particle size. There were had difference characters and physical stability. NLC had smaller size, more homogenous and more stable than conventional creame. It was known that stability of antioxidant activity of tomato extract in NLC system higher than in conventional creame. That was showed with k value, as constanta of rate scavenging activity decreasing in antioxidant power between time (Sigma 2-tail less than 0.005) of NLC and conventional creame were: 2.03x10-2 %/hour ±0.08 (3.94) and 4.71x 10-2 %/ hour ±0.23 (4.88) respectively.

<p><strong><em>Latar Belakang: </em></strong>Merokok berhubungan dengan jamur rongga mulut yang dapat mengakibatkan <em>oral candidiasis</em>. <em>Stichopus hermanii</em><em> </em>mengandung efek antioksidan, antifungi dan immunostimulator. <strong><em>Tujuan: </em></strong>Mengevaluasi efek proteksi ekstrak <em>Stichopus hermanii </em>terhadap jumlah limfosit pada tikus Wistar yang terpapar asap rokok dan diinduksi <em>C.albicans.<strong> Bahan dan Metode: </strong></em>Rancangan penelitian ini adalah <em>post test-only control group</em> <em>design</em><strong><em>. </em></strong>42 ekor tikus Wistar jantan, dibagi menjadi 7 kelompok, Kelompok1 (saline 0,1mL, udara segar, CMC-Na 0,2%), Kelompok2 (saline 0,1mL, asap rokok, CMC-Na 0,2%), Kelompok3 (<em>C.albicans </em>0,1mL, udara segar, CMC-Na 0,2%), Kelompok4 (<em>C.albicans </em>0,1mL, asap rokok, CMC-Na 0,2%), Kelompok5 (saline 0,1mL, asap rokok, ekstrak <em>Stichopus hermanii</em> 0,02mg/kgBB), Kelompok6 (<em>C.albicans</em> 0,1mL, udara segar, ekstrak <em>Stichopus hermanii </em>0,02mg/kgBB), Kelompok7 (<em>C.albicans </em>0,1 mL, asap rokok, ekstrak <em>Stichopus hermanii </em>0,02mg/kgBB). Tikus Wistar diinduksi <em>C.albicans</em> 1 minggu, terpapar asap rokok 8 minggu, dan diberi ekstrak <em>Stichopus hermanii</em> 8 minggu. Selanjutnya, tikus Wistar dikorbankan setelah 2 bulan perlakuan. Jumlah limfosit dihitung melalui metode hapusan darah dengan <em>different counting</em> dibawah mikroskop cahaya dengan pembesaran 1000x. Data yang diperoleh dianalisis menggunakan uji <em>Kruskal-Wallis</em> dan <em>Mann-Whitney</em>.<strong><em> Hasil:</em></strong> Kelompok yang terpapar asap rokok dan diinduksi C.albicans memiliki dapat menurunkan jumlah limfosit, kelompok suplementasi menggunakan ekstrak ethanol <em>Stichopus hermanii</em> dapat meningkatkan jumlah limfosit<em>. </em><strong><em>S</em></strong><strong><em>impulan:</em></strong><strong> </strong>Suplementasi ekstrak <em>Stichopus hermanii</em> memiliki efek protektif untuk memicu proliferasi limfosit pada tikus Wistar setelah paparan asap rokok dan induksi <em>C.albicans</em>.</p>
