Abstract The objective of this research was to determine the antagonistic activity of Trichoderma spp. isolated from patchouli rhizosphere (Pogostemon cablin Benth.). Another objective was to perform antagonistic screening of these fungi to inhibit the growth of the wilted pathogen Fusarium spp. In vitro research was conducted in the Laboratory of Plant Pathology, Universitas Syiah Kuala, from January to June 2020. The study used a completely randomised design with five treatments and three replications. The antagonistic screening was carried out by using the dual culture method of Trichoderma spp. against Fusarium spp. with the medium of Potato Dextrose Agar (PDA). The result showed that five isolates of Trichoderma have different antagonistic percentages in inhibiting the Fusarium. The highest antagonistic activity was found from isolate 2 and the lowest value was shown by isolate 3.

Trichoderma species are well known for their biocontrol activity which colonize many soil and tuber-borne and foliage plant pathogens. In this study, 12 native isolates of Trichiderma spp were collected from various crop rhizosphere soil samples and characterized them phenotypically based on morphological and cultural features and genotypically based on sequence analysis of internal transcribed spacer (ITS) region-PCR amplification. The results obtained from phenotypic and genotypic observation revealed that isolates were belonged to five different species namely T. asperellum, T. harzianum, T. longibrachiatum, T. koningii and T. koningiopsis. All Trichoderma isolates produced ~600 bp amplicon and phylogenetic analysis revealed that all isolates were grouped with respective species. Further, the antagonistic potential of all the isolates was evaluated against Fusarium spp. following in vitro dual culture method. The results showed that isolates of T. harzianum exhibited maximum growth inhibition activity. The highest rate of inhibition was recorded with T. harzianum isolate TBT6 (87.1%) followed by TBT7 (82.2%), while the least inhibition was observed in T. longibrachiatum isolate TBT10 (59.7%) after 7 days of incubation. The antagonistic T. harzianum isolate TBT6 can be used for development of Trichoderma based bio-formulation and served as bio-control agent against Fusaium spp. under field conditions.

Nutmeg (Myristica fragrans Houtt) is a multipurpose plantation commodity crop that almost all parts of the plant have strategic economic value to be cultivated. In Indonesia there are the largest nutmeg commodity centers, including the Maluku islands, North Sulawesi and Aceh. The biggest problem with nutmeg plants is the attack of pests and diseases. One of the most severe diseases in nutmeg is white root mushroom (Rigidoporus lignosus). This study aims to 1) determine the fungus of white root fungus antagonists (Rigodoporus lignosus) from the Aceh nutmeg rhizosphere (Myristica fragrans Houtt), 2) identify the content of antibiotic fungus JAP antagonist compounds in Aceh nutmeg plants. The study was conducted by taking soil samples from nutmeg plants at three nutmeg plantations in South Aceh. The antagonist fungus isolation activity uses a dilution method. Isolation of JAP from infected roots using direct planting method. Furthermore, it is identified macroscopically and microscopically. The antagonist test was carried out using the dual culture method with RAL. From the rhizosphere in the nutmeg area, 5 candidate antagonist isolates were obtained, having high in vitro antagonism ability to the pathogen R. lignosus. Identification results showed that isolates 1, 2 and 3 were Trichoderma spp, and isolates 4 and 5 were Gliocladium virens. Late blight disease on potato caused by a plant pathogenic fungus (Phytophthora infestans)  is the most important disease in Indonesia.  The use of antagonist fungi Trichoderma is an environmentally friendly technology to control the potato disease. The purpose of this study was to determine the ability of Trichoderma spp. to control P. infestans in vitro. Trichoderma spp. have been isolated from suppressive soil at central potato areas in district of Lembang, Bandung, West Java Province. To determine the inhibition ability of Trichoderma spp against P. infestans, a dual culture method was performed.  Variable observed was inhibition zone of Trichoderma spp. against P. infestans. The results showed that 9 isolates of Trichoderma were successfuly isolated from suppressive soil, i.e. T. viride (2 isolates), T. atroviride (1 isolate), T. harzianum (1 isolate) and T. aureoviride (5 isolates). All the Trichoderma isolates revealed growth inhibition abality against P. infestans and. The highest growth inhibition (68.6%) was observed by T. viride isolate ( Ti 9).

<p>Layu yang disebabkan oleh Fusarium spp. merupakan salah satu penyakit penting tanaman stroberi (Fragaria x ananassa<br />Dutch.) di daerah subtropika, yang dapat menggagalkan panen. Penelitian bertujuan untuk mempelajari potensi Trichoderma spp.<br />dalam mengendalikan Fusarium spp. Isolat Trichoderma spp. diisolasi dari rizosfer tanaman stroberi dan Fusarium spp. diisolasi<br />dari tanaman stroberi yang mengalami layu fusarium. Isolat cendawan dimurnikan, dikarakterisasi, dan dibandingkan dengan isolat<br />cendawan acuan. Uji antagonis dilakukan secara in vitro dan in vivo. Uji in vitro dilakukan dengan metode dual culture dan slide<br />culture. Uji in vivo dilakukan di rumah kasa menggunakan dua varietas stroberi, yaitu Santung serta California. Hasil penelitian <br />in vitro memperoleh dua jenis isolat cendawan antagonis, yaitu Trichoderma sp.1 dan Trichoderma sp.2, dan dua jenis cendawan <br />patogen Fusarium, yaitu Fusarium sp.1 dan Fusarium sp.2. Isolat Trichoderma sp.1 memiliki kemampuan antagonisme lebih tinggi<br />dibandingkan dengan isolat Trichoderma sp.2. Isolat Trichoderma sp.1 mampu menghambat pertumbuhan Fusarium sp.1 dan<br />Fusarium sp.2 secara berturut- turut, yaitu 49,7% dan 49,6%. Isolat Trichoderma sp.2 mampu menghambat pertumbuhan Fusarium<br />sp.1 dan Fusarium sp.2 lebih rendah, yaitu sebesar 45,8% dan 43,4%. Mekanisme antagonis yang terjadi antara cendawan antagonis<br />dan patogen pada uji in vitro, yaitu pembelitan dan intervensi hifa. Hasil pada uji in vivo pada perlakuan Trichoderma sebelum<br />Fusarium menunjukkan keefektifan pengendalian paling baik (41,72%) dibanding perlakuan lain. Varietas Santung lebih tahan<br />terhadap serangan patogen dibandingkan varietas California. Implikasi dari hasil penelitian ini adalah, agens hayati Trichoderma<br />spp. lebih optimal digunakan sebagai pencegahan (preventif) tanpa menunggu tanaman terinfeksi penyakit layu fusarium.</p>

Several species of Fusarium are causal agents of onion rot in field and storage. Most prevalent are F. oxysporum f. sp. cepae and F. solani, and recently F. proliferatum, a toxigenic species. Most frequently isolated fungi in our field experiments were F. solani and F. proliferatum with different pathogenicity. Certain differences in antagonistic activity of Trichoderma asperellum on different isolates of F. proliferatum and F. solani have been found in in vitro study in dual culture, expressed as a slower inhibition of growth of the former, and faster of the latter pathogen. Antagonistic abilities of species from genus Trichoderma (T. asperellum) are important, and have already been exploited in formulated biocontrol products in organic and conventional production, in order to prevent soil borne pathogens inducing fusarium wilt and rot. The importance of preventing onion infection by Fusarium spp., possible mycotoxin producers, has been underlined.

Tobacco production is a key line in the Cuban economy and generates the largest income in the agricultural sector. Within phytopathogens affecting this plant, Fusarium spp., constitutes a pest of interest, due to vascular involvements that cause deterioration of the commercial value of the leaf. Therefore, the research was carried out with the aim of determining the antagonistic activity in vitro of native strains of Trichoderma harzianum and T. viride on isolates of Fusarium oxysporum and F. phyllophylum, from Nicotiana tabacum L. in Granma province, Cuba. The work was carried out at the Laboratory of Agricultural Microbiology, University of Granma. Half dextrose potato agar was prepared to establish the dual crops of Trichoderma spp. vs. Fusarium spp. Once the treatments were established and the incubation time had elapsed, the percentage inhibition of mycelial growth, antagonistic capacity and mycoparasitic activity was determined. A fully randomized design with four replicas per treatment was used and a two factorial ANOVA and Tukey test (p≤0.05) were used for statistical processing. It was shown that the strains of Trichoderma spp., recorded intermediate values of phytopathogenic inhibition, the competitiveness of the antagonist was mostly located in class two of the Bell scale and it was found that penetration, winding, vacuolization, deformation and granulation constitute the main forms of mycoparasitism. Which points to this biocontrol method as an alternative to consider for the management of Fusarium spp., in tobacco agroecosystems.

Tanaman budi daya merupakan tanaman yang sering diserang oleh cendawan pathogen, sehingga mengakibatkan penurunan populasi dan produksi tanaman. Pengendalian hayati dengan cendawan antagonis merupakan salah satu metode yang paling efektif dan lebih ramah lingkungan dalam menekan pertumbuhan patogen tanaman. Penelitian ini bertujuan untuk menguji aktivitas antagonis cendawan isolat lokal Riau terhadap beberapa cendawan patogen pada tanaman budi daya. Uji antagonis dilakukan secara in vitro dengan metode dual culture menggunakan lima belas cendawan isolat lokal Riau terhadap Fusarium oxysporum f.sp. lycopersici, Ganoderma philippii, G. boninense, Rigidoporus microporus dan Colletotrichum sansevieria. Hasil penelitian menunjukkan bahwa Trichoderma sp. PNE 4 memiliki aktivitas antagonis tertinggi dan isolat FER C1 serta isolat LLB07 hanya memiliki aktivitas antagonis yang tinggi dalam menekan pertumbuhan cendawan patogen. Trichoderma sp. PNE 4 mampu menghambat pertumbuhan miselium F. oxysporum sebesar 85,30%, G. Philippii (100%), G. boninense (100%), dan C. sansevieria (100%). Isolat FER C1 hanya menghambat R. Microporus (50,39%) dan isolat LLB07 menghambat G. philippii (52,20%). Trichoderma sp. PNE 4 merupakan cendawan uji yang terpilih sebagai cendawan antagonis, karena memiliki kemampuan daya hambat  >70%.Abstract Cultivated plants are often attacked by pathogenic fungi resulting in a decline of population and crop production. Biocontrol with antagonistic fungi is one of the most effective and environmentally friendly methods in suppressing the growth of plant pathogens. This study aims to examine the antagonistic activity of local isolates fungi Riau against some pathogenic fungi on cultivated plants. The antagonistic test was performed in vitro by dual culture method using fifteen local isolates fungal Riau against Fusarium oxysporum f.sp. lycopersici, Ganoderma philippii, G. boninense, Rigidoporus microporus and Colletotrichum sansevieria. The results showed that Trichoderma sp. PNE 4 isolate exhibited highest activites and  FER C1 and LLB07 isolates exhibited high activities suppressed the growth of the fungal pathogen. Trichoderma sp. PNE 4 isolate inhibited mycelial growth F. oxysporum (85.30%), G. philippii (100%), G. boninense (100%) and C. sansevieria (100%). FER C1 isolate only inhibited R. microporus (50.39%), and LLB07 isolate inhibited G. philippii (52.20%). Trichoderma sp. PNE 4 isolate is test isolates as fungal antagonistic.

Butt rot disease, caused by Thielaviopsis paradoxa (De Seynes) Hohn., is one of the major diseases in pineapple cultivation in Malaysia. The objectives of this study were to evaluate the antifungal effect of antagonist bacteria against T. paradoxa, a causal agent of butt rot disease, and to observe the mechanism of antifungal activity of tested antagonist bacteria microscopically. In this study, in vitro antifungal potential of 5 antagonist bacteria, namely B1, B2, B3, B4, and B5, were isolated from infected and non-infected soil samples and evaluated using dual culture method against T. paradoxa. The mechanisms of antifungal activities of antagonist bacteria against the pathogen were microscopically observed. All of the bacteria showed inhibitory effects against the pathogenic fungi. B1 bacteria showed the highest inhibitory potential, with 73 % inhibition, followed by B2, B3, B4, and B5, with 71, 57, 56, and 48 % of inhibition compared to control, respectively. The results also showed that B2, B3, and B4 bacteria exhibited positive inhibition towards the pathogen, with more than 50 % percentage inhibition. The development of a new product for use as a biocontrol agent, used as an additional control or used in combination with existing ones, may reduce dependency on chemical control and increase antagonistic activity efficiency.

In vitro efficacy of twenty five Trichoderma isolates (twenty were TCMS series viz., TCMS 2, 4, 5, 12, 14a, 14b, 15, 16, 24, 32, 34, 36, 43, 60, 62, 64, 65, 72, 85 and 93, and five Th series; Th 1, 3, 14, 19 and 32) were ascertained for their antagonistic activity against few major plant pathogenic oomycetes namely, Phytophthora infestans, P. parasitica and Pythium aphenidermatum using dual culture technique. P. infestans was isolated from infected potato leaves and Pythium aphenidermatum from infected brinjal. P. parasitica culture was collected from Central Potato Research Institute (CPRI), Simla. The present study was conducted at Biological Control Laboratory, Department of Plant Pathology, G.B. Pant University of Agriculture and Technology, Pantnagar. All the 25 Trichodrma isolates were found significantly effective against the test pathogens. TCMS-36 and TCMS-72 were found highly effective against P. aphinidermatum with 59.57 per cent inhibition of radial growth of the fungus. Maximum reduction in mycelial growth of P. infestans was recorded with isolate TCMS-64 (60.40%) followed by TCMS-65 (59.41%), TCMS-34 (58.42%), TCMS-24, TCMS-43 and TCMS-93 with 57.43 per cent inhibition. While, maximum inhibition of P. parasitica was recorded with TCMS-4 (92.75%) followed by TCMS-36 (92.23%), TCMS-2 (91.71%), TCMS-14a (91.17%) and TCMS-32 (90.67%). The selected potential isolates may be applied to sustainable and eco-friendly management of many major crop diseases caused by the oomycetes and other fungi.

Cocoa black pod rot is caused by pathogenic fungi, Phytophtora palmivora, which decrease the cocoa production up to 90%. The use of biological control agents, Trichoderma spp., is one of the promising P. palmivora controllers since it is low-cost, easily found and safe for the environment. The aims of this research were to understand the compatibility, antagonistic and effectiveness of biofungicide containing active ingredient of Trichoderma spp. against P. palmivora in vitro and to test the effective concentration of biofungicide containing active ingredient of T. asperellum to control P. palmivora in vitro and in vivo. T. asperellum, T. harzianum, and T. viride were grown together on PDA medium to test their compatibility. Antagonistic and effectiveness test of Trichoderma spp. against P. palmivora were tested using the in vitro dual culture method. The effectiveness of T. asperellum biofungicide was tested in vivo on cocoa pot. Compatibility test showed that all three species were compatible and the best effectiveness showed by the combination of T. asperellum and T. viride. The result also showed that T. asperellum biofungicide had an ability to inhibit P. palmivora.   Keywords: Trichoderma spp., effectivity, compatibility, antagonistic, biofungicide  ABSTRAKPenyakit busuk buah kakao disebabkan cendawan patogen Phytophtora palmivora, yang dapat menurunkan produksi kakao sampai 90%. Penggunaan agensia pengendali hayati (APH), Trichoderma spp., merupakan salah satu pengendalian P. palmivora yang menjanjikan karena murah, mudah didapat dan aman terhadap lingkungan. Penelitian ini bertujuan mengetahui kesesuaian, antagonistik, dan efektivitas biofungisida berbahan aktif Trichoderma spp. secara in vitro. Di samping itu juga bertujuan mengetahui konsentrasi efektif biofungisida berbahan aktif T. asperellum untuk mengendalikan P. palmivora secara in vitro dan in vivo. T. asperellum, T. harzianum, dan T. viride, ditumbuhkan bersama pada media PDA untuk mengetahui kesesuaian antarspesies. Antagonistik dan efektivitas Trichoderma spp. terhadap P. palmivora secara in vitro diuji menggunakan metode dual culture. Biofungisida berbahan aktif T. asperellum diuji efektivitasnya secara in vivo pada buah kakao. Hasil uji kesesuaian menunjukkan bahwa ketiga spesies yang diuji berkesesuaian dan efektifitas terbaik ditunjukkan pada kombinasi T. asperellum dan T. viride. Hasil penelitian juga menunjukkan bahwa biofungisida berbahan aktif T. asperellum dengan konsentrasi tertinggi terbukti dapat menghambat pertumbuhan P. palmivora.Kata kunci: Trichoderma spp., efektivitas, kesesuaian, antagonis, biofungisida
