Abstract To understand the mechanism governing the postharvest senescence of Zizania latifolia, and the regulatory mechanism induced by 1-methylcyclopropene (1-MCP) during storage at 25°C, physiobiochemical and conjoint analyses of the transcriptome and metabolome were performed. The results indicated that 1-MCP treatment engendered changes in the expression of genes and metabolites during the postharvest storage of Z. latifolia. The 1-MCP treatment maintained a good visual appearance, preserved the cell structure, and membrane integrity of Z. latifolia by keeping the expression of membranes-related lipolytic enzymes (and related genes) low and the amount of phosphatidylethanolamine high. Compared to the control group, 1-MCP treatment enhanced the activities of antioxidant enzymes, resulting in a decrease of reactive oxygen species (ROS) and malondialdehyde (MDA) contents, and thus inhibition of oxidative damage and loss of membrane integrity. In addition, 1-MCP treatment retarded the senescence of Z. latifolia by down-regulating the expression of ethylene biosynthesis-related genes and promoting up-regulation of brassinosteroid insensitive 1 (BRI1) kinase inhibitor 1, calmodulin (CaM), glutathione reductase, jasmonate amino acid synthase, and mitogen-activated protein kinase (MAPK)-related genes. Moreover, 1-MCP retarded Z. latifolia senescence by inducing the activity of ATP-biosynthesis related genes and metabolites. Our findings should facilitate future research on the postharvest storage of Z. latifolia, and could help delay senescence and prolong the storage time for commercial applications.

Background and ObjectivesNeurofibromatosis type 1 (NF1)-associated cognitive impairments carry significant lifelong morbidity. The lack of targeted biologic treatments remains a significant unmet need. We examine changes in cognition in patients with NF1 in the first 48 weeks of mitogen-activated protein kinase inhibitor (MEKi) treatment.MethodsFifty-nine patients with NF1 aged 5–27 years on an MEKi clinical trial treating plexiform neurofibroma underwent pretreatment and follow-up cognitive assessments over 48 weeks of treatment. Performance tasks (Cogstate) and observer-reported functioning (BRIEF) were the primary outcomes. Group-level (paired t tests) and individual-level analyses (Reliable Change Index, RCI) were used.ResultsAnalysis showed statistically significant improvements on BRIEF compared with baseline (24-week Behavioral Regulation Index: t(58) = 3.03, p = 0.004, d = 0.24; 48-week Metacognition Index: t(39) = 2.70, p = 0.01, d = 0.27). RCI indicated that more patients had clinically significant improvement at 48 weeks than expected by chance (χ2 = 11.95, p = 0.001, odds ratio [OR] = 6.3). Group-level analyses indicated stable performance on Cogstate (p > 0.05). RCI statistics showed high proportions of improved working memory (24-week χ2 = 8.36, p = 0.004, OR = 4.6, and 48-week χ2 = 9.34, p = 0.004, OR = 5.3) but not visual learning/memory. Patients with baseline impairments on BRIEF were more likely to show significant improvement than nonimpaired patients (24 weeks 46% vs 8%; χ2 = 9.54, p = 0.008, OR = 9.22; 48 weeks 63% vs 16%; χ2 = 7.50, p = 0.02, OR = 9.0).DiscussionOur data show no evidence of neurotoxicity in 48 weeks of treatment with an MEKi and a potential clinical signal supporting future research of MEKi as a cognitive intervention.

Background: Black Bengal goat is one among the valuable goat breeds of India. Semen preservation and artificial insemination are practical technologies for conservation and improving livestock germplasm. Various biochemical changes that occur during cryopreservation of semen, results in loss of sperm cell structure and their functions. Inclusion of antioxidants to the semen extender has been reported to have positive effects on sperm recovery in various livestock species. The present experiment was carried out to evaluate the effect of antioxidant L-cysteine on post freeze thaw sperm characters, lipid peroxide level and superoxide dismutase (SOD) activity during cryo­preservation of Black Bengal buck semen. Methods: Twenty numbers of semen ejaculates were collected from Black Bengal bucks, antioxidant L cysteine was added to the semen extender @ 0, 1 and 2 mM in control group (CC), treatment group 1 (CT1) and treatment group 2 (CT2), respectively. Semen samples were frozen in liquid nitrogen and measured post freeze-thaw in vitro sperm characters, malondialdehyde (MDA) concentration and SOD activity. Result: Though there was no significant improvement in post thaw sperm motility, functional membrane integrity and viability count, supplementation of L-cysteine significantly improved the percentage of sperm cells with intact acrosome and significantly (P less than 0.05) reduced the level of malondialdehyde. Post thaw SOD activity was found significantly lower in the antioxidant treated groups than the control. It is concluded that supplementation of L cysteine is effective in controlling lipid peroxidation and conserving in vitro sperm characters during freezing of Black Bengal buck semen.

The genus Brassica comprises a highly diverse range of vegetable crops varying in morphology, harvestable crop product, and postharvest shelf-life that has arisen through domestication, artificial selection and plant breeding. Previous postharvest studies on the shelf-life of Brassica species has mainly focused on the variable rates of physiological changes including respiration and transpiration. Therefore, further understanding of the molecular basis of postharvest senescence in Brassica vegetables is needed to understand its progression in improving their postharvest shelf-life. The aim of this study was to better understand the trajectory of molecular responses in senescence-associated genes but not induced by ethylene and ethylene-induced genes towards altered postharvest storage conditions. After storage at different temperatures, the expression levels of the key senescence-associated genes (SAGs) and the ethylene biosynthesis, perception, and signaling genes were quantitatively analyzed in cabbage, broccoli and kale. The expression levels of these genes were tightly linked to storage temperature and phase of senescence. Expression of ORE15, SAG12, and NAC29 were continuously increased during the twelve days of postharvest storage at room temperature. Prolonged exposure of these three vegetables to cold temperature reduced the variation in the expression levels of ORE15 and SAG12, observed as mostly decreased which resulted in limiting senescence. The transcript levels of the ethylene receptor were also decreased at lower temperature, further suggesting that decreased ethylene biosynthesis and signaling in cabbage during postharvest storage would delay the senescence mechanism. These results enhanced our understanding of the transcriptional changes in ethylene-independent SAGs and ethylene-related genes in postharvest senescence, as well as the timing and temperature sensitive molecular events associated with senescence in cabbage, broccoli and kale and this knowledge can potentially be used for the improvement of postharvest storage in Brassica vegetables.

Abstract Adrenocortical Carcinoma (ACC) is a rare endocrine tumor with poor overall prognosis and 1.5-fold overrepresentation in females. In children, ACC is associated with inherited cancer syndromes with 50–80% of childhood-ACC associated with TP53 germline variants. ACC in adolescents and young adults (AYA) is rarely due to germline TP53, IGF2, PRKAR1A and MEN1 variants. We analyzed exome sequencing data from 21 children (<15y), 32 AYA (15-39y), and 60 adults (>39y) with ACC, and retained all pathogenic, likely pathogenic, and highly prioritized variants of uncertain significance. We engineered a stable lentiviral-mutant ACC cell line, harboring an EGFR variant (p.Asp1080Asn) from a 21-year-old female without germline-TP53-variant and with aggressive ACC. We found that 4.8% of the children (P = 0.004) and 6.2% of AYA (P < 0.0001), all-female participants, harbored germline EGFR variants, compared to only 0.3% of the control group. Expanding our analysis to the RTK-RAS-MAPK pathway, we found that the RTK genes have the highest number of highly prioritized germline variants in these individuals amongst all three arms of this pathway. We showed EGFR mutant cells migrate faster and are characterized by a stem-like phenotype compared to wild type cells. While EGFR inhibitors did not affect the stemness of mutant cells, Sunitinib, a multireceptor tyrosine kinase inhibitor, significantly reduced their stem-like behavior. Our data suggest that EGFR could be a novel underlying germline predisposition factor for ACC, especially in the Childhood-AYA (C-AYA) population. Further clinical validation can improve precision oncology management of this disease, which is known to have limited therapeutic options.

Abstract Background Many people exercise because they know it is good for their health. Although this is true, it can make us feel deserving of a reward and lead us to eat more indulgent, less healthy food than if we had not done any exercise. Generally, lower energy-dense (LED) foods are recognised as healthier choices than higher energy-dense (HED) options. Despite our intention to make healthy choices, seeing tempting higher-calorie foods on offer often side-tracks us. Priming is a psychological tool that makes specific changes to our environment that remind us of our motivation to be healthy. This makes it easier to choose a healthier option, by nudging us towards it without us even realising. However, it is currently unclear which method of priming achieves the best results. Aims Our study explores whether priming people to expect they will receive LED food leads them to make this healthier choice after exercise, even when also offered tempting less healthy HED foods at the moment of selection. Methods Our study observed the foods selected by university athletes after their sports matches. Before the match, half of the participants were primed by asking them to choose a LED snack from the options we offered, which they would receive after the match. The remaining half of participants were not asked this same question. To distract the athletes from our observation of their food choices, participants completed a task prior to choosing their snack, which was disguised as a ‘thank you’ for taking part. Results Overall, we found the priming group did not choose LED foods significantly more than the control group, hence priming did not increase LED food selection. Conclusion Importantly, our results indicate that priming must be more noticeable to achieve its goal. Additionally, we demonstrated that priming may be less successful for young athletic individuals, compared to older and more overweight adults recruited in other studies. This highlights the importance of studying a broader demographic range of individuals from the general population. We support future research into this area, which will help us to tweak priming to achieve the best outcomes. Trial registration ISRCTN Registry, ISRCTN74601698. Abstract Introduction Bacterial adherence to restorative materials such as composite resin is one of the aetiology of secondary caries. This study evaluated the antibacterial efficacy of fifth generation bonding agent (BA) modified with nisin, against Streptococcus mutans based on its growth, adherence and membrane integrity. Methods Adhesive eluents of the experimental bonding agents were obtained using 250 μl Brain Heart Infusion (BHI) broth and the groups were control (BA with 0% Nisin), bonding agent with 1 wt% (NBA 1) and 5 wt% nisin (NBA 5). To this, 10 μl S. mutans culture was added and incubated at 37 °C. Bacterial growth was estimated by changes in optical density using spectrophotometer every 20 min for 2 h. The results were statistically analysed using one way ANOVA followed by Tukey Post Hoc test. For adherence and membrane integrity test, 10 μl of BHI supplemented with 1% sucrose and 50 μl of bacterial suspension were inoculated onto the cured specimens, and incubated for 4 h. After rinsing, 1 ml of Live/Dead BacLight bacterial viability stain was added and incubated in the dark for 15 min and observed under confocal laser scanning microscope (CLSM) for intact (green/live) and damaged (red/dead) bacterial membranes. Results Mean optical density was significantly higher in control group at all time intervals with maximum value at 2 h (0.83 ± 0.008), while there was a concentration dependant reduction in bacterial growth with the NBA groups (0.50 ± 0.007). Correspondingly, the NBA groups showed higher amount of dead than live bacteria, while live bacteria were predominant in the control group. Significance Addition of an antibacterial agent nisin in dentin bonding agent may render the resin dentin interface more resistant to bacterial penetration, and adherence of cariogenic bacteria like S. mutans.
