Citrus is a valuable crop in Pakistan because it is rich in vitamin C and antioxidants. Huanglongbing (HLB) has an influence on citrus production around the world caused by a bacterium “ Candidatus   liberibacter asiaticus ” (CLas), africanus and americanus. The structure and diversity of bacterial species in various ecosystems can be quickly examined using NGS. This approach is considerably quicker and more precise than outdated methods. Healthy or citrus greening infected leaf samples of Grapefruit,  Citrus aurantifolia , and  Citrus reticulata  Blanco was used for diversity analysis. In this study high throughput, NGS technique was used to access the population of both cultivable and non-cultivable bacterial endophytes from citrus leaves, by using PCR amplicons of 16S rDNA sequences (V5–V7 regions) with Illumina Hi seq. As a result, a total number of 68,722 sequences were produced from the test samples. According to the NGS-based diversity classification, the most common genera of exploited bacterial endophytes were Proteobacteria, Firmicutes, Bacteroides, Cyanobacteria, and Actinobacteria.  Citrus aurantifolia  and  Citrus paradisi  showed almost equal diversity, whereas  Citrus reticulata  Blanco had a higher proportion of Proteobacteria and Cyanobacteria in their leaves. To determine alpha diversity (AD), additional data was analyzed using statistical indices such as Shannon, Chao1, and Simpson. According to the inverse Simpson diversity index, the abundance of the microbial population in six different citrus samples was 0.48, 0.567, and 0.163, respectively. The metagenomics of microbiota in plant tissues was successfully recorded by NGS technology, which can help us learn more about the interactions between plants and microbes. This research is the first step toward a better understanding of 16SrRNA-based metagenomics from citrus in Pakistan using Illumina (Hi seq) Technology.

O efeito de cinco composições de substratos na produção de mudas de laranjeira `Pêra' (Citrus sinensis, L. Osbeck) foi estudado sobre três porta-enxertos (limoeiro `Cravo', Citrus limonia, L. Osbeck; tangerineira `Cleópatra', Citrus reticulata, Blanco; e citrumeleiro `Swingle', Citrus paradisi x Poncirus trifoliata). O experimento obedeceu ao delineamento estatístico de blocos aleatorizados em esquema fatorial 5 x 3 (substratos x porta-enxertos). As composições de misturas utilizadas como substrato e respectivas combinações volumétricas foram: 1) Terra (100%); 2) Terra (33%) + areia fina (33%) + esterco bovino curtido (33%); 3) Terra (33%) + vermiculita (33%) + esterco bovino curtido (33%); 4) Terra (33%) + raspas de madeira (33%) + esterco bovino curtido (33%); 5) Terra (25%) + raspas de madeira (25%) + areia fina (25%) + esterco bovino curtido (25%). A análise dos resultados revelou que plantas de limoeiro `Cravo' e citrumeleiro `Swingle' apresentaram maior desenvolvimento do que as de tangerineira `Cleópatra'. A mistura de materiais com solo mostrou-se vantajosa e induziu à formação de plantas mais desenvolvidas do que aquelas em substrato constituído de terra exclusivamente. A composição do substrato influenciou diferentemente no desenvolvimento de cada porta-enxerto, mas de uma forma geral, composições contento 1/3 de volume de solo e 1/3 de volume de esterco bovino proporcionaram bons resultados para os três porta-enxertos estudados.

Foi instalado um experimento de seleção de porta-enxertos para a lima-ácida-'Tahiti' (Citrus latifolia Tanaka), em dezembro de 1988, na Estação Experimental de Citricultura de Bebedouro-SP, com o objetivo de conhecer seu comportamento e oferecer novas opções de plantio para as condições ecológicas semelhantes às daquela região. A variedade copa, originária do BAG-Citros do IAC, localizado no Centro de Citricultura Sylvio Moreira, Cordeirópolis-SP, é um clone nucelar de 'Tahiti', denominado IAC-5. Os porta-enxertos, que tiveram a mesma origem, foram: tangerinas-'Sunki' (Citrus sunki Hort. ex Tanaka); 'Cleópatra'(Citrus reshni Hort. ex Tan.); 'Batangas' e 'Oneco' (Citrus reticulata Blanco); trifoliata-EEL (Poncirus trifoliata Raf.); limão-'Cravo' (Citrus limonia Osbeck); limão-'Volkameriano Catania 2' (Citrus volkameriana Tan. & Pasq.); tangelo-'Orlando' (C. reticulata Blanco x Citrus paradisi Macf.); citrumelo-'Swingle' (P. trifoliata Raf. X C.paradisi Macf.); citrange-'Morton' (P. trifoliata Raf. X C. sinensis (L.) Osbeck) e laranja-'Caipira DAC' (C. sinensis (L.) Osbeck). Com relação à produção, avaliada no período de 1991 a 1998, os porta-enxertos de melhor comportamento foram o tangelo-'Orlando', citrange-'Morton' e citrumelo-'Swingle'. As mais baixas produções ocorreram nos porta-enxertos de tangerina e de laranja-'Caipira DAC'. O limão-'Cravo' apresentou produção intermediária e proporcionou curta vida útil às plantas.

Foram amostrados à época de colheita, na Estação Experimental de Limeira, do Instituto Agronômico, frutos cítricos de nove cultivares comerciais pertencentes a diferentes espécies: Citrus sinensis - "baianinha", "hamlin", "pêra", "natal" e "valência"; Citrus reticulata -"cravo"; Citrus paradisi - "marsh-seedless"; Citrus aurantifolia - "taiti"; híbrido Citrus sinensis X Citrus reticnlata -"murcote". Os frutos foram fracionados em casca, polpa mais suco, e sementes. No material seco e moído, procederam-se às determinações dos treze elementos essenciais às plantas e mais sódio, alumínio e cobalto. As quantidades de nutrientes extraídas por tonelada de fruto fresco foram calculadas com base nas proporções e teores de umidade das partes dos frutos. Os nove cultivares estudados extraíram as seguintes quantidades médias de elementos em gramas por tonelada de fruto fresco: N-1.906; P-173; K-1.513; Ca-526; Mg-127; S-137; B-2,2; Cl-24,7; Cu-1,2; Fé-6,6; Mn-2,8; Mo-0,008; Zn-0,9; Co-0,003; Na-43,5; Al-7,6. Os dados obtidos são semelhantes aos encontrados na literatura de outros países para os citros em geral. Há, no entanto, quase sempre, uma inversão na relação nitrogênio-potássio. Os cultivares com maior capacidade de extração de macro e micronutrientes foram as laranjas natal e valência, e o de menor capacidade, o limoeiro taiti. As sementes em geral contiveram os maiores teores de nutrientes, porém a sua ocorrência nos frutos é em pequena proporção, atingindo o máximo de 3% na tangerina-cravo.

Considering the huge economic loss caused by postharvest diseases, the identification and prevention of citrus postharvest diseases is vital to the citrus industry. In 2018, 16 decayed citrus fruit from four citrus varieties—Satsuma mandarin (Citrus unshiu), Ponkan (Citrus reticulata Blanco cv. Ponkan), Nanfeng mandarin (Citrus reticulata cv. nanfengmiju), and Sugar orange (Citrus reticulata Blanco)—showing soft rot and sogginess on their surfaces and covered with white mycelia were collected from storage rooms in seven provinces. The pathogens were isolated and the pathogenicity of the isolates was tested. The fungal strains were identified as Lasiodiplodia pseudotheobromae based on their morphological characteristics and phylogenetic analyses using the internal transcribed spacer regions (ITS), translation elongation factor 1-α gene (TEF), and beta-tubulin (TUB) gene sequences. The strains could infect wounded citrus fruit and cause decay within two days post inoculation, but could not infect unwounded fruit. To our knowledge, this is the first report of citrus fruit decay caused by L. pseudotheobromae in China.

There is an increased interest in the gut microbiota as it relates to health and obesity. The impact of diet and sex on the gut microbiota in conjunction with obesity also demands extensive systemic investigation. Thus, the influence of sex, diet, and flaxseed supplementation on the gut microbiota was examined in the JCR:LA-cp rat model of genetic obesity. Male and female obese rats were randomized into four groups (n = 8) to receive, for 12 weeks, either (a) control diet (Con), (b) control diet supplemented with 10% ground flaxseed (CFlax), (c) a high-fat, high sucrose (HFHS) diet, or (d) HFHS supplemented with 10% ground flaxseed (HFlax). Male and female JCR:LA-cp lean rats served as genetic controls and received similar dietary interventions. Illumine MiSeq sequencing revealed a richer microbiota in rats fed control diets rather than HFHS diets. Obese female rats had lower alpha-diversity than lean female; however, both sexes of obese and lean JCR rats differed significantly in β-diversity, as their gut microbiota was composed of different abundances of bacterial types. The feeding of an HFHS diet affected the diversity by increasing the phylum Bacteroidetes and reducing bacterial species from phylum Firmicutes. Fecal short-chain fatty acids such as acetate, propionate, and butyrate-producing bacterial species were correspondingly impacted by the HFHS diet. Flax supplementation improved the gut microbiota by decreasing the abundance of Blautia and Eubacterium dolichum. Collectively, our data show that an HFHS diet results in gut microbiota dysbiosis in a sex-dependent manner. Flaxseed supplementation to the diet had a significant impact on gut microbiota diversity under both flax control and HFHS dietary conditions.
