AbstractThe striatum is structurally highly diverse, and its organ functionality critically depends on normal embryonic development. Although several studies have been conducted on the gene functional changes that occur during striatal development, a system-wide analysis of the underlying molecular changes is lacking. Here, we present a comprehensive transcriptome profile that allows us to explore the trajectory of striatal development and identify the correlation between the striatal development and Huntington’s disease (HD). Furthermore, we applied an integrative transcriptomic profiling approach based on machine learning to systematically map a global landscape of 277 transcription factor (TF) networks. Most of these TF networks are linked to biological processes, and some unannotated genes provide information about the corresponding mechanisms. For example, we found that the Meis2 and Six3 were crucial for the survival of striatal neurons, which were verified using conditional knockout (CKO) mice. Finally, we used RNA-Seq to speculate their downstream targets.

Here, we present a comprehensive temporal transcriptome atlas of the mouse striatum at multiple developmental stages. Quantitative analysis of 15443 gene products allows us to explore the trajectory of striatal development and identify the correlation between the striatal development and Huntington’s disease (HD). More importantly, we provided the global landscape of 277 transcription factor (TF) networks based on co-expression analysis and machine learning. Furthermore, we identified the hub TFs Six3 and Meis2 which are involved in regulating the apoptosis of striatal neurons. Finally, using conditional knockout (CKO) mice and RNA-Seq data, we verified that Six3 and Meis2 indeed regulated neuronal apoptosis and inferred their downstream targets in the striatum.

During follicular development in the mammalian ovary, several functional changes occur in the granulosa cells in response to steroid hormones and gonadotropins (1,2). In particular, marked changes in the content of membrane-associated receptors for the gonadotropins have been observed (1).We report here scanning electron microscope observations of morphological changes that occur on the granulosa cell surface in response to the administration of estradiol, human follicle stimulating hormone (hFSH), and human chorionic gonadotropin (hCG).Immature female rats that were hypophysectcmized on day 24 of age were treated in the following manner. Group 1: control groups were injected once a day with 0.1 ml phosphate buffered saline (PBS) for 3 days; group 2: estradiol (1.5 mg/0.2 ml propylene glycol) once a day for 3 days; group 3: estradiol for 3 days followed by 2 days of hFSH (1 μg/0.1 ml) twice daily, group 4: same as in group 3; group 5: same as in group 3 with a final injection of hCG (5 IU/0.1 ml) on the fifth day.

Turkish dermatologist, H. Beheet described the disease as recurrent triad of iritis, oral aphthous lesions and genital ulceration. Auto immune disease is the recent focus on the unknown etiology which is still being discussed. Among the other immunosupressive drugs, CyA included in it's treatment newly. One of the important side effects of this drug is gingival hyperplasia which has a direct relation with the presence of teeth and periodontal tissue. We are interested in the ultrastructure of immunocompetent target cells that were affected by CyA in BD.Three groups arranged in each having 5 patients with BD. Control group was the first and didn’t have CyA treatment. Patients who had CyA, but didn’t show gingival hyperplasia assembled the second group. The ones displaying gingival hyperplasia following CyA therapy formed the third group. GMC of control group and their granules are shown in FIG. 1,2,3. GMC of the second group presented initiation of supplementary cellular activity and possible maturing functional changes with the signs of increased number of mitochondria and accumulation of numerous dense cored granules next to few normal ones, FIG. 4,5,6.

In studies which attempt to define the influence of various factors on recovery of hair cell integrity after acoustic trauma, an experimental and a control ear which initially have equal degrees of damage are required. With in a group of animals receiving an identical level of acoustic trauma there is more symmetry between the ears of each individual, in respect to function, than between animals. Figure 1 illustrates this, left and right cochlear evoked potential (CAP) audiograms are shown for two chinchillas receiving identical trauma. For this reason the contralateral ear is used as control.To compliment such functional evaluations we have devised a scoring system, based on the condition of hair cell stereocilia as revealed by scanning electron microscopy, which permits total stereociliar damage to be expressed numerically. This quantification permits correlation of the degree of structural pathology with functional changes. In this paper wereport experiments to verify the symmetry of stereociliar integrity between two ears, both for normal (non-exposed) animals and chinchillas in which each ear has received identical noise trauma.

Research  background  and  hypothesis.  Traditional  time  series  analysis  techniques,  which  are  also  used  for the analysis of cardiovascular signals, do not reveal the relationship between the  changes in the indices recorded associated with the multiscale and chaotic structure of the tested object, which allows establishing short-and long-term structural and functional changes.Research aim was to reveal the dynamical peculiarities of interactions of cardiovascular system indices while evaluating the functional state of track-and-field athletes and Greco-Roman wrestlers.Research methods. Twenty two subjects participated in the study, their average age of 23.5 ± 1.7 years. During the study standard 12 lead electrocardiograms (ECG) were recorded. The following ECG parameters were used in the study: duration of RR interval taken from the II standard lead, duration of QRS complex, duration of JT interval and amplitude of ST segment taken from the V standard lead.Research  results.  Significant  differences  were  found  between  inter-parametric  connections  of  ST  segment amplitude and JT interval duration at the pre and post-training testing. Observed changes at different hierarchical levels of the body systems revealed inadequate cardiac metabolic processes, leading to changes in the metabolic rate of the myocardium and reflected in the dynamics of all investigated interactions.Discussion and conclusions. It has been found that peculiarities of the interactions of ECG indices interactions show the exposure of the  functional changes in the body at the onset of the workload. The alterations of the functional state of the body and the signs of fatigue, after athletes performed two high intensity training sessions per day, can be assessed using the approach of the evaluation of interactions between functional variables. Therefore the evaluation of the interactions of physiological signals by using time series analysis methods is suitable for the observation of these processes and the functional state of the body.Keywords: electrocardiogram, time series, functional state.
