AbstractLotus (Nelumbo nucifera Gaertn), a kind of perennial aquatic plant, is widely cultivated and consumed by people in Asian countries. Lotus plumule flavonoids (LPF) have been recognized as a hypoglycemic agent. LPF was optimally obtained using novel ultrasonic-microwave assisted synergistic extraction (UMSE) method by response surface methodology (RSM) on the basis of the results of single-factor experiments. Furthermore, the hypoglycemic activity of LPF was investigated by measuring the body weight, fasting blood glucose (FBG) level, and oral glucose tolerance test (OGTT) and analyzing the physiological indexes in streptozotocin-diabetic mice model. The optimum extraction conditions consisted of microwave power 355 W, ultrasonic power 423 W, extraction time 15 min, solid-liquid ratio 1:40, ultrasound/interval time 1/0, and ethanol concentration 70% with the maximum LPF yield of 2.62%. LPF supplementation significantly decreased the body weight, FBG, OGTT, serum total cholesterol (TC), serum total triglycerides (TG), and insulin levels, indicating the antidiabetic activity of LPF. This research verified that the UMSE technique was highly efficient to extract LPF to the maximum extent and the flavonoids from L. plumule exhibited hypoglycemic activity, which showed broad development and application prospects.

Flaxseed is an oilseed (45–50% oil on a dry-weight basis) crop. Its oil has demonstrated multiple health benefits and industrial applications. The goal of this research was to evaluate the antidiabetic and anti-inflammatory potential of the free polyphenol fraction of flax (Linum usitatissimum L.) seeds (PLU), based on their use in traditional medicine. Mice with alloxan-induced diabetes were used to study the antidiabetic activity of PLU in vivo, with an oral administration of 25 and 50 mg/kg over 28 days. Measurements of body weight and fasting blood glucose (FBG) were carried out weekly, and biochemical parameters were evaluated. An oral glucose tolerance test was also performed. Inhibitory activities of PLU on α-amylase and α-glucosidase activities were evaluated in vitro. The anti-inflammatory was evaluated in vivo in Wistar rats using the paw edema induction Test by carrageenan, and in vitro using the hemolysis ratio test. PLU administration to diabetic mice during the study period improved their body weight and FBG levels remarkably. In vitro inhibitory activity of digestive enzymes indicated that they may be involved in the proposed mode of action of PLU extract. Qualitative results of PLU revealed the presence of 18 polyphenols. These findings support daily consumption of flaxseed for people with diabetes, and suggest that polyphenols in flaxseed may serve as dietary supplements or novel phytomedicines to treat diabetes and its complications.

Background: Physalis angulata have been extensively used for the management of diabetes in folklore medicine, in Nigeria. Aim: The present study aimed to investigate the antidiabetic effect of aqueous extract of Physalis angulata and its potential mechanisms in streptozotocin-induced diabetic rats. Study Duration: The period of the study was done on 30th September, 2018 at the Department of Biochemistry, Faculty of Basic Medical Sciences, university of Jos, Nigeria. Methodology: Albino rats of Wistar strain weighing between 200g to 250g were induced with single freshly prepared streptozotocin (55 mg/kg body weight). Diabetes was confirmed after forty eight hours in streptozotocin -induced rats showing fasting blood glucose levels > 10 mmol/l. The rats were randomly divided into four (4) experimental groups (n = 4). A (Control diabetic group fed with normal feed), Group B (Normal control fed with normal feed), Group Group C (Diabetic rats treated with 400 mg/Kg body weight extract of Physalis angulata and Group D, (Diabetic rats are treated with 400 mg/Kg body weight of glibenclamide). After 8 days the animals were sacrificed and blood samples were collected for biochemical and hematological analysis. Changes in the animal body weights were also measured within the period. Results: From the results, it was observed that treatment of rats with extract of Physalis angulata elevates the reduction of body weight, and caused an increase in the body weight of the treated rats. In the same order, serum glucose significantly decreased (p<0.05) after the 8-day treatment compared to diabetic control. The extent of reversal of hyperglycemia in the physalis angulata extract treated animals compared well with the glibenclamide treated group. The results, therefore, showed that Physalis angulata extract has a significant (p<0.05) hypoglycemic effect in diabetic rats and the histopathological results of treated groups showed the regenerative/protective effect on

Background: Ficus glomerata (F. glomerata) Linn. Family Moraceace is a large tree found all over India including outer Himalayan ranges, Punjab, Chota Nagpur, Bihar, Orissa, West Bengal, Rajasthan, Deccan and also as a common plant in South India. It is planted around the home and temples. It is cultivated throughout the year, distributed in evergreen forests and moist localities. Objective: The Ethanolic Extract of roots of F. Glomerata (EEFG) belonging to the family Moraceace, was investigated for its antidiabetic activity using alloxan induced diabetic rats. Methods: Thirty rats were divided into 5 groups having 6 rats in each group. The alloxan was administered to the rats of all groups except normal control group through intraperitoneal route at a concentration of 140mg/kg body weight. A dose of 100mg/kg and 200 mg/kg body weight of EEFG was administered to alloxan induced diabetic rats. The administration of the extract was lasted for 11 days. Effectiveness of the extract on glucose, cholesterol, triglycerides, and high density lipoprotein and protein concentrations was analyzed. Results: Significant (p<0.05) reduction in the levels of glucose, cholesterol, triglyceride of the diabetic rats was observed after treatment with ethanolic extract. After subjecting to oral glucose tolerance test EEFG also showed significant improvement in glucose tolerance. Conclusion: F. glomerata root ethanolic extract showed that it possesses antidiabetic effect and can be found useful for the management of diabetes mellitus.

Diabetes mellitus (DM) is a now a major global health problem and its incidence is increasing day by day in whole world. There are various medicinal plants in India those possess antidiabetic property which are traditionally used in management of diabetes. Tectona grandis Linn. (TG) plant belonging to family Verbenaceae is medicinally reported and claims to cure various diseases in Indian traditional system of medicine (Ayurveda) and also in folklore. The purpose of this present study is to examine the hypoglycemic potential of methanol fractions (50, 100 and 200mg/kg body weight) of Tectona grandis bark (MFTG) from defatted hydro-alcoholic extract in normoglycemic, streptozotocin induced (45mg.kg‒1) diabetic and glucose loaded hyperglycemic rats by single and multiple oral administration in comparison to standard drug Glibenclamide (2.5 mg/kg body weight). Initially acute oral toxicity study of MFTG was carried out in rats to estimate the dose for animal study. The study report showed that the MFTG (200mg/kg) significantly (p<0.05 to p<0.01) reduces blood glucose level both in normoglycemic and diabetic rats induced by Streptozotocin and oral glucose loaded methods till the end of 8 hour and 3hour respectively during the single dose study and from the 15th day to 30th day in multi dose study. Hence the present study reveals that MFTG possess significant hypoglycemic activity which inspires the traditional use of the plant for the treatment of diabetes mellitus.

This study aimed to establish mechanistic links between the prolonged intake of desloratadine, a common H1 receptor blocker (i.e., antihistamine), and development of obesity and metabolic syndrome. Male Sprague-Dawley rats were treated for 16 wk with desloratadine. We analyzed the dynamics of body weight gain, tissue fat accumulation/density, contractility of isolated mesenteric lymphatic vessels, and levels of blood lipids, glucose, and insulin, together with parameters of liver function. Prolonged intake of desloratadine induced development of an obesity-like phenotype and signs of metabolic syndrome. These alterations in the body included excessive weight gain, increased density of abdominal subcutaneous fat and intracapsular brown fat, high blood triglycerides with an indication of their rerouting toward portal blood, high HDL, high fasting blood glucose with normal fasting and nonfasting insulin levels (insulin resistance), high liver/body weight ratio, and liver steatosis (fatty liver). These changes were associated with dysfunction of mesenteric lymphatic vessels, specifically high lymphatic tone and resistance to flow together with diminished tonic and abolished phasic responses to increases in flow, (i.e., greatly diminished adaptive reserves to respond to postprandial increases in lymph flow). The role of nitric oxide in this flow-dependent adaptation was abolished, with remnants of these responses controlled by lymphatic vessel-derived histamine. Our current data, considered together with reports in the literature, support the notion that millions of the United States population are highly likely affected by underevaluated, lymphatic-related side effects of antihistamines and may develop obesity and metabolic syndrome due to the prolonged intake of this medication. NEW & NOTEWORTHY Prolonged intake of desloratadine induced development of obesity and metabolic syndrome associated with dysfunction of mesenteric lymphatic vessels, high lymphatic tone, and resistance to flow together with greatly diminished adaptive reserves to respond to postprandial increases in lymph flow. Data support the notion that millions of the USA population are highly likely affected by underevaluated, lymphatic-related side effects of antihistamines and may develop obesity and metabolic syndrome due to the prolonged intake of this medication.

Some lactic acid bacteria (LAB) are observed to be potential probiotics with functional properties such as lowering fasting blood glucose (FBG), as a promising hyperglycemia management. This study investigated the ability and mechanism of Lactobacillus rhamnosus BSL and Lactobacillus rhamnosus R23 on lowering FBG in diabetic rats induced by streptozotocin (STZ). The rats were orally administered with L. rhamnosus BSL and L. rhamnosus R23 by giving 1 mL cell suspension (109 CFU/mL) daily for 30 days. The body weight (BW) was recorded once in three days, and FBG was recorded once in six days. An oral glucose tolerance test (OGTT) was measured 1 week after injection with STZ and before sacrifice. Fecal samples were collected on days 0, 15, and 30 for LAB population and identification, performed by PCR detecting 16S rRNA. Oral administration of L. rhamnosus BSL and L. rhamnosus R23 decreased FBG and improved glucose tolerance via downregulation of glucose-6-phosphatase (G6pc) expression by 0.57- and 0.60-fold change, respectively (P<0.05). The lipid profiles, BUN, creatinine, SGOT, and SGPT were significantly (P<0.05) different between normal and diabetic rats, but they were not significantly (P>0.05) different among diabetic rats. Both strains were effective in increasing fecal LAB population. Molecular identification of the isolated LAB from fecal sample indicated that they were able to survive and pass through the digestive tract. These results suggested that both strains have the ability to manage blood glucose level and become a promising agent to manage hyperglycemia and diabetes.

The purpose of this study was to investigate the protective role of orally administered taurine against diabetic retinal changes via electroretinogram (ERG) and retinal histology on rabbits. Rabbits were randomly assigned into groups: Group I (vehicle administration only); Group II (diabetes: induced by 100 mg/kg alloxan injection); Group III (diabetes and fed with 200 mg/kg taurine); and Group IV (diabetes and fed with 400 mg/kg taurine). The body weight and blood glucose levels of the rabbits were monitored weekly. The ERG was measured on weeks 5 and 15. Retinal histology was analyzed in the end of the experiment. Results revealed that a taurine supplement significantly ameliorates the alloxan-induced hyperglycemia and protects the retina from electrophysiological changes. Group II showed a significant(P<0.05)change in the mean scotopic b-wave amplitude when compared to that of Group I, whereas the diabetic rabbits treated with taurine (Group III and IV) were analogous to Group I. Histologically, the amount of Bipolar and Müller cells showed no difference(P>0.05)between all groups and when compared with those of Group I. Our study provides solid evidences that taurine possesses an antidiabetic activity, reduced loss of body weight, and less electrophysiological changes of the diabetic retina.

Accumulating evidences have reinforced the use of medicinal plants in the treatment of various ailments as a result of negative side effects associated with conventional drugs. Plant components such as phenols and flavonoids with antioxidant potential have confirmed protective roles against oxidative stress-induced degenerative diseases like diabetes mellitus (DM). The current study was carried out to investigate the effect of seed pod ethanol extract from Copaifera salikounda (SPEECS) in alloxan-induced diabetic rats. SPEECS was obtained by maceration of seed pod powder in absolute ethanol for 72 h, filtered, concentrated and dried in-vacuo. Gas chromatography-mass spectrometry (GC–MS) technique was used to quantitatively elucidate the chemical constituents of SPEECS. Twenty-four male albino rats were randomly allocated into four groups (n=6): normal control, DM control, DM + 200 mg/kg SPEECS and DM + 400 mg/kg SPEECS groups. DM was induced in the Wistar albino rats through intraperitoneal injection of 200 mg/kg body weight of alloxan. After 14 days of treatment, the body weight changes and the fasting blood glucose level were determined in the different groups. Also, serum biochemical parameters such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), albumin (ALB), total protein (TP), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were estimated. The GC-MS results confirm nine bioactive compounds with 9-octadecenoic acid (55.75%) being most abundant. SPEECS (200 and 400 mg/kg) administration significantly (P 0.05) caused gain in weight, decreased fasting blood glucose and reversed the elevated liver function enzymes (ALT, AST, ALP) while total TP and ALB were markedly elevated relative to DM control group. Furthermore, SPEECS attenuated the activities of SOD and CAT while the level of MDA was significantly (P 0.05) decreased in dose dependent manner in comparison to the DM control. Rastlinske sestavine kot so fenoli in flavonoidi z antioksidativnim potencialom, imajo po nekaterih raziskavah zaščitno vlogo pred degenerativnimi boleznimi, ki jih povzroča oksidativni stres, kot je sladkorna bolezen diabetes mellitus (DM). Študija je bila izvedena z namenom raziskovanja učinka etanolnega semenskega ekstrakta iz rastline Copaifera salikounda (SPEECS) pri podganah s sladkorno boleznijo, ki jo je povzročil alloxan. SPEECS je bil pridobljen z maceracijo praška semen v prahu v absolutnem etanolu 72 ur ter nadaljnjo filtracijo, koncentracijo in sušenjem v vakuumu. Za kvantitativno ugotavljanje kemijskih sestavin SPEECS je bila uporabljena tehnika plinske kromatografije in masne spektrometrije (GC-MS). Štiriindvajset samcev podgan Wistar je bilo naključno razporejenih v štiri skupine (n=6): normalna kontrola, kontrola DM, DM + 200 mg/kg SPEECS in DM + 400 mg/kg SPEECS. DM je bil pri podganah sprožen z intraperitonealno injekcijo 200 mg/kg telesne mase alloxana. Po 14 dneh zdravljenja so bile pri različnih skupinah določene spremembe telesne teže in nivo glukoze v krvi (na tešče). Poleg tega so avtorji raziskave izmerili še nekatere serumske biokemične parametre kot so ravni alaninske aminotransferaze (ALT), aspartatne aminotransferaze (AST), alkalne fosfataze (ALP), albumina (ALB), skupnih proteinov (TP), malondialdehida (MDA), superoksiddismutaze (SOD) in katalaze (CAT). Rezultati GC-MS so v izvlečku SPEECS pokazali devet bioaktivnih spojin, v katerih je največ 9-oktadecenojske kisline (55,75%). SPEECS (200 in 400 mg/kg) je povzročil znatno (P 0,05) povečanje telesne mase, znižanje glukoze v krvi na tešče in znižal raven encimov pokazateljev jetrne funkcije (ALT, AST, ALP), medtem ko je bila raven TP in ALB pri podganah, ki so prejemale SPEECS izrazito povišana v primerjavi z DM kontrolno skupino. Zdravljenje s  SPEECS je tudi oslabilo aktivnosti SOD in CAT, medtem ko se je raven MDA znatno zmanjšala (P 0,05) v primerjavi s kontrolno skupino DM. Objective: The main purpose of the present study was to analyze the antidiabetic activity of methanolic extract of the leaf of Kalanchoe pinnata in alloxan-induced diabetic rats. Methods: Diabetes mellitus (DM) is a chronic metabolic and endocrine disease regarded as a serious global public health problem. Albino Wistar rats were divided into four groups. Group I (normal) received drinking water throughout the course till 20 days. Groups II-IV received alloxan (120 mg/kg b.w) i.p on the 1st day of the study period. Group III animals received glibenclamide (10 mg/kg p.o) and Group IV received K. pinnata (200 mg/kg b.w p.o) for 20 days. The body weight, blood glucose level, serum urea, blood urea nitrogen (BUN), creatinine, cholesterol, total protein (TP), uric acid, and tissue (liver) antioxidant parameters: Malondialdehyde (MDA) and glutathione (GSH) were measured. Results: K. pinnata treated rats showed the percentage increase in the body weight, decrease in the blood sugar level, creatinine, TP level, urea, uric acid, and BUN, MDA, and GSH when compared to alloxan-induced diabetic mellitus control rats. Thus, K. pinnata could be possibly employed to treat DM. Conclusion: This preliminary analysis tested the most promising profile. It seems that methanolic extract of the leaf of K. pinnata improved general, blood serum, and liver antidiabetic agent. However, further studies confirming its potential is certainly warranted.

The part of the plant to curing the diabetes mellitus and improves available data on plants with hypoglycemic effects. Antidiuretic potential of eucalyptus extracts has been evaluated in alloxan-induced diabetic rabbit. Here we used, plants extract about 256 kg were mix with 1000 ml methanol and keep in Flask. After filtering the extracts were exposed to rotary evaporator machine that methanol separated from plant extract. In the Nine rabbit were divided into three different groups. The body weight was recorded on every 6 days interval. The data obtained revealed that eucalyptus extracts reduced the glucose level effectively. Such treatment with extracts decreased the SGPT level. Therefore, it is concluded that the eucalyptus possesses significant antidiabetic activity. And finally, we recommend this plant for medicinal uses and for pharmaceutical industries to isolate pure drugs.
