Microsatellites or simple sequence repeats (SSRs) are highly useful molecular markers for plant improvement. Expressed sequence tag (EST)-SSR markers have a higher rate of transferability across species than genomic SSR markers and are thus well suited for application in cross-species phylogenetic studies. Our objectives were to examine the amplification of tall fescue EST-SSR markers in 12 grass species representing 8 genera of 4 tribes from 2 subfamilies of Poaceae and the applicability of these markers for phylogenetic analysis of grass species. About 43% of the 145 EST-SSR primer pairs produced PCR bands in all 12 grass species and had high levels of polymorphism in all forage grasses studied. Thus, these markers will be useful in a variety of forage grass species, including the ones tested in this study. SSR marker data were useful in grouping genotypes within each species. Lolium temulentum, a potential model species for cool-season forage grasses, showed a close relation with the major Festuca–Lolium species in the study. Tall wheatgrass was found to be closely related to hexaploid wheat, thereby confirming the known taxonomic relations between these species. While clustering of closely related species was found, the effectiveness of such data in evaluating distantly related species needs further investigations. The phylogenetic trees based on DNA sequences of selected SSR bands were in agreement with the phylogenetic relations based on length polymorphism of SSRs markers. Tall fescue EST-SSR markers depicted phylogenetic relations among a wide range of cool-season forage grass species and thus are an important resource for researchers working with such grass species.Key words: phylogeny, EST-SSR, forage grasses, tall fescue.

Cladophorus Guérin-Méneville, 1830 are endemic Papuan net-winged beetles which take part in highly diverse Müllerian mimicry rings. Available specimens were sequenced for cox1–tRNA-Leu–cox2 mitochondrial DNA fragment and the species delimitations were based on the genetic distance, phylogenetic analysis, and morphology. Three earlier described species were identified in the recently collected material and further 10 species are described: C. pallescens sp. nov., C. bicolor sp. nov., C. craterensis sp. nov., C. motykai sp. nov., C. mindikensis sp. nov., C. kailakiensis sp. nov., C. manokwarensis sp. nov., C. haiaensis sp. nov., C. humeralis sp. nov., and C. boceki sp. nov. DNA-based identifications provided some ambiguous results and closely related species could not be robustly delimited using solely molecular data. Additionally, the species limits were based on clearly defined morphological characters and the morphological differentiation was found unlinked from the genetic divergence. Colour patterns cannot be used for identification because all species available in more specimens were polymorphic and followed various local co-mimics. The Papuan fauna of Cladophorus is very diverse and the closely related species regularly occur in limited regions. Differentiation within restricted ranges is therefore considered as the main speciation mode.

Gliadins, as the major components of wheat storage proteins, determine the extensibility properties of dough and have important effects on flour processing quality. Wheat related species carries potential storage protein gene resources for quality improvement. In this study, we isolated and characterized the first complete ?-gliadin gene Omega-AS from Aegilops speltoides L. (2n = 2x = 14, SS) by allelic-specific PCR and investigated its phylogenetic relationships among Triticum and Aegilopsspecies. Molecular structure showed that Omega-AS gene consisted of 1122 bp encoding 373 amino acid residues with deduced molecular mass 41379.21 Da. Omega-AS gene was exceptionally rich in prolines and glutamines with fewer methionine and no cysteine. Sequence characterization and epitope analysis showed that three epitopes QQPIPVQPQQ, TQPQQPTPIQ and IQPQQPFPQQ were absent in Omega-AS gene encoded protein, indicating its potential value for wheat quality improvement with less toxic, or no toxic peptides. Phylogenetic analysis revealed that Omega-AS was closely related to gliadin genes of wheat and related species and its divergence from bread wheat was more recently (less than 1.243 MYA). Heterologous expression showed that Omega-AS gene could successfully express with a high level in E. coli under the control of T7promoter. The transcription expression pattern of Omega-AS gene during grain development detected by qRT-PCR revealed that the highest expression level occurred at 17 days post an thesis.

Introduction: The relationships within the Pyrrhura species complex are partly unresolved. In this study, a comprehensive phylogenetic analysis of the Pyrrhura picta-leucotis complex was carried out, covering all species except P. subandina. Material and Methods: We made a morphological analysis of 745 preserved specimens of all the taxa in different museums. Nucleotide sequences of the mitochondrial cytochrome gene were generated and used to reconstruct a molecular phylogeny of Pyrrhura. Results and Discussion: Our results show that the complex is divided into 6 main groups comprising 15 species. P. dilutissima, regarded up to now as a subspecies of P. peruviana, acquires species status and three new subspecies are described. We also provide evidence that P. roseifrons is a paraphyletic group, indicating the existence of probably 3 lineages of which 2 deserve species status.

Grouper fish (Serranidae) is a type of fish found in Indonesian waters. However, not many people have conducted further research on phylogenetics based on COI (Cytochrome Oxidase I). This study aims to explain the phylogeny of grouper fish from the Serranidae family based on COI (Cytochrome Oxidase I). This research is a literature study. The research samples were grouper fish from Lombok, Karimunjawa, Lampung, Kendari, Madura, Tanakeke, and Numfor. The research instrument was a grouper-type observation sheet and an observation sheet for the results of the MEGA 7 application test. The data were analyzed using qualitative descriptive analysis. The results showed that the level of kinship between the species tested was very close, including Epinephelus areolatus, E. merra, E. fasciatus, E. longispinis, E. coioides, E. ongus, and E. coeruleopunctatus with all genetic distance averages type. 0.02. The conclusion of this study is that in general the species relationships found are several species found in the same location with similar morphology and diet.

The genus Chilecicada Sanborn, 2014 is shown to be a complex of closely related species rather than a monospecific genus. Chilecicada citatatemporaria Sanborn & Cole n. sp., C. culenesensis Sanborn & Cole n. sp., C. curacaviensis Sanborn & Cole n. sp., C. impartemporaria Sanborn & Cole n. sp., C. magna Sanborn & Cole n. sp., C. mapuchensis Sanborn n. sp., C. oraria Sanborn & Cole n. sp., C. parrajaraorum Sanborn n. sp., C. partemporaria Sanborn & Cole n. sp., C. pehuenchesensis Sanborn & Cole n. sp., C. trifascia Sanborn n. sp., C. trifasciunca Sanborn & Cole n. sp., and C. viridicitata Sanborn & Cole n. sp. are described as new. Chilecicada occidentis Walker, 1850 is re-described to facilitate separation of the new species from the only previously known species. Song and cytochrome oxidase I analysis available for most species support the separation of the new taxa from the type species of the genus. Known species distributions and a key to the species of the genus are also provided. The new species increases the known cicada diversity 61.9% to 34 species, 91.2% of which are endemic to Chile.
