Anaplasma platys is a Gram-negative, intracellular obligate bacterium that is transmitted by Rhipicephalus sanguineus ticks and has been identified in most of the country. It is the causative agent of canine infectious cyclic thrombocytopenia. The objective of this work is to report the finding of morulae compatible with A. platys in the blood smear of a 10-year-old male Weimaraner canine who was treated at the Hospital de Salud Animal of the Facultad de Ciencias Veterinarias of the Universidad Nacional del Litoral, in the city of Esperanza. The case could not be followed up, because the patient died a few days after his first consultation. The detection of obligate intracellular pathogens in blood smears has low sensitivity and specificity, therefore confirmation of the causative agent must be performed using PCR. Due to the severity of the condition presented in this case, it is necessary to differentiate the presence of Ehrliquia canis and confirm the finding of A. platys using PCR. Current scientific evidence reveals that the zoonotic potential of A. platys is very low or nil.

Ehrlichia canis, Anaplasma platys, and Babesia vogeli are frequently detected in the veterinary routine with varied pathogenesis that can lead to death, mainly in co-infections. Although canine hemotropic mycoplasmas are considered of low clinical importance, they have recently gained prominence in molecular research. The present study aimed to analyze two hospital populations of dogs, randomly selected from the year 2013 and 2015, from the municipalities of Toledo (n=68) and Cascavel (n=107). Direct examination of blood smears, serology, and PCR were used to detect the presence of E. canis, A. platys, B. vogeli and Mycoplasma sp. Direct blood smear examination was negative for the investigated agents in all samples. Serum immunochromatography (SensPERT ™, VetAll Laboratories, Korea) in 175 animals showed that only 4% (n = 7) had anti-E. canis antibodies, while 60% (n = 105) were positive for Anaplasma sp. All PCR samples were negative for E. canis, while 18.28% (n = 32) were positive for A. platys, 3.42% (n = 6) positive for Babesia vogeli, and 7.41% (n = 13) positive for Mycoplasma sp. The study revealed the presence of A. platys in Toledo and Cascavel by PCR, which accentuates the need to monitor vector populations and usual hosts and to evaluate the potential risk of infection in humans.

ABSTRACT: Hemoparasitic infections are tick-borne diseases, which affect animals and humans. Considering the importance of canine hemoparasitic infections in veterinary clinics, this study aimed to determine the occurrence of Anaplasma platys, Ehrlichia canis and Babesia vogeli in blood samples from 182 dogs not domiciled in the city of Pato Branco, southwestern region of Paraná State, Brazil, using polymerase chain reaction (PCR). The prevalence of A. platys and B. vogeli was 32.9% and 10.9% respectively, and A. platys infection prevailed (p<0.001). The number of dogs positive for A. platys was larger in Winter (p<0.05). All blood samples were negative for E. canis. In the dogs, infestation by Amblyomma cajennense predominated over that by Rhipicephalus sanguineus (p<0.001); but there was no significant association between PCR and the variables presence of ticks, sex and age. Dogs infected by A. platys and B. vogeli showed thrombocytopenia, lymphopenia and leukocytosis; but there was no correlation between such hematological changes and infection by hemoparasites. This appears to be the first molecular study that demonstrates the existence of A. platys and B. vogeli in dogs from the southwestern region of Paraná.

ABSTRACT More than 300 species have been described in the genus Hepatozoon, occurring in different vertebrates. Among these, only Hepatozoon canis and Hepatozoon americanum are seen in dogs. Different methods may be used for laboratory diagnosis. The most common of these is direct parasitological examination of parasite stages in blood smears. The aim of this investigation was to conduct a phylogenetic study on Hepatozoon isolates from symptomatic dogs in the city of Goiânia, Goiás, Brazil. Blood samples were obtained from 40 symptomatic dogs that had been referred to the Veterinary Hospital of the Federal University of Goiás. Among these, only two samples were positive for Hepatozoon spp. using the direct parasitological method. These samples were then subjected to a DNA extraction process and amplification of a fragment of the 18S rRNA by means of PCR. Subsequently, the PCR products from each sample were purified and sequenced. The sequences obtained were then analyzed using the BLASTn algorithm, which identified both sequences of this study as Hepatozoon canis. By applying the Mega4 software, it was confirmed that these isolates of H. canis from dogs in Goiânia are similar to other reference isolates of the same species from other regions of Brazil and worldwide.

Background: Canine distemper has been classified as highly contagious for most of domestic and wild carnivores, and the infection can be fatal. Canine distemper inclusion bodies, also denominated Lenz inclusion bodies, are large aggregates of viral nucleocapsid particles that can be form in red blood cells (RBCs), white blood cells (WBCs) and epithelial cells in many tissues during the acute phase of infection. Their presence in blood is transient and rarely encountered in light microscopy but are pathognomonic when identified in blood smears. The objective of this study was to investigate the frequency of distemper inclusions in erythrocytes according to the fraction of the sample used for blood smears. Materials, Methods & Results: The study was conducted with routine blood sample provided by the Veterinary Laboratory of Clinical Analysis from the Veterinary Teaching Hospital of Universidade Federal do Rio Grande do Sul. The EDTA-K2 blood sample of a 40 days old male dog, mixed breed, no immunization records, presenting diarrhea, hyporexia, myoclonus and pustules in the abdomen, was selected. In a routine peripheral blood smear examination, several distemper inclusions were observed in the erythrocytes. From this sample, ten smears were performed using a whole blood (WB) and top erythrocyte fraction combined with buffy coat, denominated of expanded buffy coat (EBC). The EBC fraction was obtained after centrifugation of EDTA whole blood in microhematocrit tubes at 9600 x g for 5 min to obtained the packed cell volume (PCV) and buffy coat. After centrifugation, the blood cells are separated into three layers based on density: platelets (adjacent to supernatant), WBCs, and RBCs in the bottom. The PCV was measured and the microhematocrit tube was ruptured 2% below the interface between leukocytes and plasma, deposited into a plastic microtubes, homogenized and used for blood smear preparation. All smears were stained with Diff-Quick Stain. The frequency of observation of RBCs with distemper inclusions bodies was performed under optical microscopy, in the immersion objective (100x), accounting for complete fields up to a minimum of 1000 RBCs, and compared between WB and EBC. In comparison between blood smears obtained from WB and EBC, a highly significant difference (P = 0.0004) was observed in the frequency distribution of distemper inclusion. The median of frequency of RBCs with distemper inclusions in a WB smears was 12.68/1000 RBCs (10.1 - 16.1/1000 RBCs), with a coefficient of variation (CV) of 12%. Median of frequency of distemper inclusions from EBC smears was 54.23/1000 RBCs (45-77.9/1000 RBCs), CV of 18% were observed. The median frequency of inclusions found in EBC smears was 4.27 times higher than the WB smears. Discussion: Buffy coat smear providing a concentrated preparation of nucleated cells and this procedure is useful to looking for low-incidence infectious organisms or other hematologic alterations. The upper fraction of the RBC column, below the buffy coat, is composed of young RBCs. Selection of these portion, and their possible formed in the bone marrow viral replication phase, could justified the increase in the frequency of RBCs containing viral inclusions in EBC, which would also increase the sensitivity of the technique. EBC was homogenized previously to make the smears, certifying the adequate cell distribution in the slide surface without interfere with the frequency of distemper inclusion in RBCs observation. These results were confirmed with the coefficients of variation. In conclusion, distemper inclusions bodies in RBCs from EBC is a recommended diagnosis method in patients suspected of canine Distemper infection. The observation being more frequent in the EBC in comparison with WB, commonly used in veterinary hematology.

Ticks parasitizing on dogs belong to mites of the order Ixodida, class Arachnida. In Poland the occurrence of four species has been recorded most frequently: Ixodes ricinus, I. hexagonus, I. crenulatus and Dermacentor reticulatus. Their importance is due to the fact that they are vectors of many dangerous diseases of humans and animals. The subject of the present study were ticks collected from dogs, patients of several veterinary clinics from the urban agglomeration of the city of Olsztyn. Ticks were collected from May to June in eight consecutive years: 2009–2016. The dominant species was I. ricinus (60.14%) in relation to D. reticulatus (39.71%). In two consecutive years, 2011–2012, single females of Rhipicephalus sanguineus were also identified, an exceptional finding of this species in our country. In D. reticulatus, nymphs predominated over females (23.74% vs 16.0%) while in I. ricinus the respective percentage was 29.6% and 30.55%. However, in subsequent years of the study the proportions between nymphs and females varied greatly in both species.

Background and Aim: Anaplasma platys is a blood parasite that infects platelets, causing thrombocytopenia. Rhipicephalus sanguineus ticks are believed to transmit A. platys. To identify A. platys, nested polymerase chain reaction (PCR) has proven to be an effective diagnostic tool. In this study, the molecular prevalence of A. platys infection in dogs was investigated for the 1st time in the Khon Kaen region of Thailand. The association between risk factors and A. platys infection was also evaluated. Materials and Methods: A total of 130 blood samples were collected from dogs in Khon Kaen, Thailand. DNA from the samples was extracted and nested PCR was applied for molecular analysis. Platelet count and packed cell volume (PCV) levels were measured. Platelet counts were categorized into four grades: Non-thrombocytopenia (platelets >200,000 cells/μL), mild thrombocytopenia (platelets 150,000-200,000 cells/μL), moderate thrombocytopenia (platelets 100,000-150,000 cells/μL), and severe thrombocytopenia (platelets <100,000 cells/μL). Four categories for PCV levels of >37%, 30-37%, 20-29%, and <20% were defined as no anemia, mild anemia, moderate anemia, and severe anemia, respectively. DNA sequencing was analyzed using BTSeq™ (Barcode-Tagged Sequencing; CELEMICS, Seoul, South Korea) for similarity index. Results: Among the 130 samples, 9 (6.9%) were positive for A. platys infection. There was an association between low platelet count and infection (p<0.05). PCV level was also associated with A. platys infection (p<0.05). DNA sequencing results of the nine positive samples showed similarity to known sequences of A. platys with 99.36-100% nucleotide identity. These results suggested low genetic diversity in A. platys infecting dogs in the Khon Kaen area. Conclusion: By amplifying 16S rRNA, A. platys infection was detected in the blood of Thai dogs. Further work should be performed to identify risk factors potentially associated with A. platys infection in dogs in Khon Kaen. Other related factors should also be considered, such as location and breeding, as well as the environmental characteristics of each locality. In addition, sampling a larger number of animals may reveal predictors for the positivity of A. platys in dogs in this region.

AbstractGram-negative bacteria in the order Rickettsiales are obligate intracellular parasites that cause human diseases such typhus and spotted fever. They have evolved a dependence on essential nutrients and metabolites from the host cell as a consequence of extensive genome streamlining. However, it remains largely unknown which nutrients they require and whether their metabolic dependency can be exploited therapeutically. Here, we describe a genetic rewiring of bacterial isoprenoid biosynthetic pathways in the Rickettsiales that has resulted from reductive genome evolution. We further investigated whether the spotted fever groupRickettsiaspeciesRickettsia parkeriscavenges isoprenoid precursors directly from the host. Using targeted mass spectrometry in uninfected and infected cells, we found decreases in host isoprenoid products and concomitant increases in bacterial isoprenoid metabolites. Additionally, we report that bacterial growth is prohibited by inhibition of the host isoprenoid pathway with the statins class of drugs. We show that growth inhibition correlates with changes in bacterial size and shape that mimic those caused by antibiotics that inhibit peptidoglycan biosynthesis, suggesting statins inhibit cell wall synthesis. Altogether, our results describe an Achilles’ heel of obligate intracellular pathogens that can be exploited with host-targeted therapeutics that interfere with metabolic pathways required for bacterial growth.ImportanceObligate intracellular parasites, which include viruses as well as certain bacteria and eukaryotes, extract essential nutrients and metabolites from their host cell. As a result, these pathogens have often lost essential biosynthetic pathways and are metabolically dependent on the host. In this study, we describe a metabolic dependency of the bacterial pathogenRickettsia parkerion host isoprenoid molecules that are used in the biosynthesis of downstream products including cholesterol, steroid hormones, and heme. Bacteria make products from isoprenoids such as an essential lipid carrier for making the bacterial cell wall. We show that bacterial metabolic dependency can represent an Achilles’ heel, and that inhibiting host isoprenoid biosynthesis with the FDA-approved statin class of drugs inhibits bacterial growth by interfering with the integrity of the cell wall. This work highlights a potential to treat infections by obligate intracellular pathogens through inhibition of host biosynthetic pathways that are susceptible to parasitism.

Research Highlights Despite of many historical records from Portuguese about the physical of Melaka Malay City, its existence is still doubtful since there is no clear physical evidence that can be used as basis support to its existence. However, to prove that the city was existed, various physical and scientific evidence are required to prove its existence. Thus, this study aims to identify historical evidence that mentioned its existence which based on the first painting of A’Famosa (Malacca Fort City during Portuguese era). The study proposed three hypothesis and results indicated there is a postivie outcome to prove that the Melaka Malay City existed on and around Bukit Melaka (or St Paul Hill now) based on historical records and sketches found.   Research Objectives This paper attempts to uncover and reveal the historical evidence of Melaka Malay (Fort) City before the city was destroyed and rebuilt according to Portuguese terms. This paper was created to identify the differences between designs and layout plans between the Malacca Malay (Fort) City and the world-renowned Fort A’Famosa, thus comparing the hypothesis arguments about the existence of Melaka City. Indeed, there is a clear distinction between the design, layout, character and philosophy of the Malay and Portuguese cities. The scope focuses only on the site of the physical building (which is believed to be historical evidence) of the city at the Melaka River’s estuary where important and significant buildings during Melaka Malay empire was constructed – the palace, mosque, administration buildings, harbors, and different kind of settlements. There is a great probability of continuing this study into the use of satellite technology in determining and identifying the Melaka Malay City with more precise and accurate details. The implication of this study can be a catalyst for continuous and deeper research on the original site of the Melaka Malay City as to prove the validity of its existences.   Methodology In an effort to discuss the existence of this Melaka Malay City, various aspects and methods were conducted to find the most suitable matches and comparisons that almost accurately describe their design then prove its existence. The remains and plans of Melaka City today since the British era will be the basis of its existence. Hence, the study of historical records and sketches of the Melaka City from the Dutch government and subsequently the Portuguese will be made to seek physical evidence of the earliest design of Melaka City. Later, the historical records of the city from Malay Archipelago, Chinese, Portuguese and Dutch sources will be depicted in city design. As a reinforcement, the philosophy of urban design in the Malay World is highlighted as well as presenting the comparison between the other Malay Kingdoms and the Melaka Malay City.   Results The foundations of the Malay cities basically were designed in square or rectangular shape, no matter how far the city was set up (Abdul Halim Nasir, 1990). This is in line with the philosophy in the Malay world that associated with a simple yet sufficient form of compassion that covers the various aspects of the Malay community's facilities and their living that are synonymous with modular systems (Yusoff Hashim, 2010). The first Fort A'Famosa built by the Portuguese was rectangular (G.W.Irwin, 2006). Its position was below the foot of Bukit Melaka and along the estuary of the Melaka River which is also near the bridge stretching the mouth of the Melaka River (Portuguese Documents of Malacca, 11: 6 de Goes, para 11). The city of A'Famosa in the painting has a relatively six-tower on the corner of the city near the bridge. Whereas city fort or bastion on three other city corners are two-storey building. A’Famosa was built on the site of the Great Mosque of Melaka (Portuguese Documents of Malacca, 11: 6 de Goes, the 25th) which features a rectangular shape as the custom of mosques designs in the Malay Archipelago realm at that time, then the A'Famosa was originally built in rectangular over the ruins (or damage) of the Great Mosque Malacca after the success of Portuguese colonization.   Findings The research showed that the design of Melaka City or A'Famosa City was in square or rectangular shape as it was built on the ruins of the Great Mosque of Melaka. The rectangular fort design was synonym with the design of other Malay forts or cities as it was a fundamental design tradition in the Malay world from fourth century until the later centuries after the collapse of Melaka empire either in Malay clusters. This coincides with the urban philosophy and design of the Malay and Islamic city that breed in the Malay Archipelago region at that time.

Introduction: Colonic lipomas are low-risk lesions that require endoscopic or surgical management depending on their size and symptoms. However, diagnosing a lipoma­tous lesion in the colon is a difficult task, as the clinical or imaging findings point to a low prevalence of these lesions and a higher fre­quency in the female sex. The laparoscopic approach is the current method of choice as it represents fewer traumas and, therefore, a faster clinical recovery.Case presentation: 43-year-old female patient, Caucasian, from the city of Bogotá, housewife, without any medical history. She presented with acute abdominal pain associated with tarry stools. Diagnostic imaging tests were performed, finding colo-colonic intussusception, originated in a lipomatous lesion located in the transverse colon. This patient underwent an enlarged lap­aroscopic right hemicolectomy.Discussion: This case describes how colonic intussusception constitutes a preoperative complication that requires surgical management since its diagnosis considering the associated mortality rates.Conclusion: The analysis of this case is part of the scientific evidence indispensable to im­prove the diagnostic and therapeutic approach to colonic intussusception, as local and in­ternational literature have not addressed this condition sufficiently.

The objective of this study was to determine the species and distribution of ticks infesting domestic dogs in southern Mozambique. To this end 89 collections were taken from dogs and the geographic coordinates of the localities at which they were made noted. Fifty-eight of these collections were from animals in the city of Maputo and 16 from animals at rural settlements close to Maputo. A further 15 collections were made from dogs in rural environments north of Maputo. Ten ixodid tick species, of which one was only identified to genus level, were recovered. Rhipicephalus sanguineus was the most numerous of the ten species, and its prevalence and intensity of infestation were significantly higher on city dogs than on rural dogs (P< 0.01), whereas the converse was true for Haemaphysalis leachi (P< 0.01). Including the city of Maputo, the exact localities at which nine tick species were collected, were recorded.
