ABSTRACT: Corticosteroid therapy has been used for ruminants to allow lung maturation and the birth of premature babies. However, when considering laboratory analyses of these animals, very little data is available regarding hematological and biochemical patterns, especially for premature goats, and the effects of corticotherapy on these parameters are unknown. In this context, the objective of this study was to evaluate the hematological and biochemical parameters during the first hours of life of premature kids from goats subjected to different dexamethasone protocols. For this, the goats were divided into four groups: group I, goats that received 20 mg of dexamethasone at 139 days of gestation; group II, 2 mg of dexamethasone from the 133rd to 136th day of gestation, 4 mg from the 137th to 139th, and 20 mg on the 140th; group III, 16 mg of dexamethasone from the 139th day, with repeated doses every 12 h until elective surgery; and group IV, goats that received 4, 8, 16, and 20 mg of dexamethasone at 137, 138, 139, and 140 days of gestation, respectively. Blood samples were obtained at birth (T0h) and after 1 (T1h), 12 (T12h), 24 (T24h), and 48 h (T48h) of life for hemogram and serum biochemistry assessment of urea, creatinine, total protein (PT), and gamma-glutamyltransferase (GGT). PT levels and GGT activity were lower at birth in all groups and rose after colostrum ingestion. The creatinine values for all the experimental groups did not differ between T0h and T1h; however, they decreased in the subsequent moments. Except for group I, urea concentrations were higher at T48h than at T1h. The red blood cell, hemoglobin, hematocrit, and mean corpuscular hemoglobin counts decreased over time. The total leukocyte count behaved differently in different experimental groups, and was influenced by the levels of dexamethasone, mainly due to the change in the counts of segmented neutrophils and lymphocytes. It was concluded that significant changes in the hematological and biochemical parameters occur in the first hours of life of premature kids, and that the treatment of goats with dexamethasone can affect these parameters in a dose-dependent manner.

Aim: This research project investigates the effect of mastitis on some hematological and biochemical parameters of Red Sokoto goats (RSGs). Materials and Methods: In this investigation, 16 clinically and subclinically diagnosed mastitic and 20 non-mastitic RSGs, within Sokoto metropolis, were sampled. Blood samples were collected both in ethylenediaminetetraacetic acid and anticoagulant free sample bottles for hematology and biochemical analysis, respectively. Results: A statistical analysis of the results revealed no significant difference in all the hematological parameters analyzed for both the mastitic and non-mastitic goats except mean corpuscular hemoglobin where significant difference (p<0.05) was observed. Similarly, no significant difference was recorded in the serum biochemistry except for the increase in total protein (p<0.001), globulin (p<0.05), and alanine aminotransferase activity (p<0.05). Conclusion: This finding is a pointer to the fact that mastitis could be regarded as a localized problem affecting the udder without serious systemic or metabolic involvement in RSGs.

This study is conducted to evaluate the therapeutic and antioxidant effect of lemon juice on some hematological and biochemical parameters. Thirty female mice used in this study were exposed to oxidative stress through giving them hydrogen peroxide in drinking water for 30 days. Animals randomly distributed over 3 groups, each group contained 10 animals and treated as follows: T1 control group (drinking distilled water only), T2 (0.75% hydrogen peroxide in drinking water) and T3 (0.75% hydrogen peroxide in drinking water with daily drenching with 1 mL lemon juice). At the end of the experiment, blood samples were collected from animals for evaluating the following hematological and biochemical parameters: Haemoglobin concentration (Hb), red blood cells count (RBC), white blood cells count (WBC), packed cell volume (PCV), mean corpuscular volume (MCV), mean corpuscular haemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), platelet count (PLT), level of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and total protein. The results showed that T3 exhibited an enhancement in RBC count, Hb concentration, WBC, lymphocyte and total protein and reduction in the level of AST and ALT compared to T2. These findings clearly revealed the advance protective and antioxidant features of lemon juice on hematological and biochemical parameters of the oxidatively stressed female mice.

Aim: The present study was designed to evaluate the plausible efficacy of ethanolic extract of Rauwolfia serpentina to mitigate copper induced toxicity by investigating cytogenetic biomarkers, i.e., chromosome aberrations (CA) and micronucleus (MN) formation; assessment of biochemical changes in liver enzymes (SGOT, SGPT and ALP) and estimation of protein levels in a freshwater spotted snakehead fish(Channa punctatus). Methodology: The experiment was carried out in six groups, each having 15 specimens for 24, 48, 72, and 96 hr. Group I served as control. Groups II and III were maintained with 3 mg l-1 of Rauwolfia root extract and 0.4 mg l-1 of Cu2+, respectively. Groups IV, V, and VI were simultaneously co-exposed with 0.4 mg l-1 of Cu2+ and three different concentrations of Rauwolfia root extract 1, 2 and 3 mg l-1, respectively. For the genotoxicity assessment, blood and kidney tissues were used. Hepatic impairments were assessed after each exposure period. Results: A significant increase (p<0.05) in chromosomal aberrations, micronuclei frequency, activity of liver enzymes (SGOT, SGPT and ALP) and a decrease in protein level were recorded in Group III in comparison to the control. Groups co-exposed with Cu2+and Rauwolfia serpentine showed a significant (p<0.05) decrease in cytogenetic biomarkers, activity of liver enzymes and an increase in protein levels, as compared to Group III, with respect to control in a dose dependent manner. Interpretation: Thus, result of the present investigation establish the efficacy of R. serpentina root extract against Cu2+ induced toxicity in spotted snakehead (Channa punctatus).

Abstract Background Propofol injection pain (PIP) is common and may decrease patient comfort. The aim of this study was to compare the effects of dezocine with lidocaine on prevention of PIP. Methods 235 patients, who scheduled for elective surgery, aged 18-80 years and ASA (American Society of Anesthesiologists) I or II, were randomly assigned into five groups (n = 47 each). All patients were induced through the dor­sal hand vein or antecubital vein. The five groups were given the following medication intravenously: saline (Group I), lidocaine 20 mg (Group II), lidocaine 40 mg (Group III), dezocine 2 mg (Group IV) and dezocine 4 mg (Group V). Twenty seconds later all patients received a propofol infusion and were asked to grade pain or discomfort in the hand or forearm according to a four-point scale until anesthesia. Results Three groups showed a significantly lower incidence of total PIP than Group I: Group III (OR (Odds ratio): 0.39 (0.16, 0.93)); Group IV (OR: 0.21 (0.09, 0.51)) and Group V (OR: 0.12 (0.05, 0.30)). Group IV showed a significantly lower incidence of total PIP than Group II (OR: 0.32 (0.13, 0.77)). Group V showed a significantly lower incidence of total PIP than Group II (OR: 0.15 (0.06, 0.39)) or Group III (OR: 0.30 (0.13, 0.72)). Conclusion Dezocine is a novel agent to alleviate PIP and appears to be more effective than lidocaine.

Functional and immunologic levels of antithrombin III (ATIII) and plasminogen were assessed in three groups of nine patients each: elective surgery, moderate and severe trauma. Patients otherwise healthy were classified as follows: I, elective extremity surgery, no blood transfusions, normal coagulation screening tests (14-50y); II, isolated, moderately severe trauma (e.g., replantation or revascularization of one or more digits), no transfusions (14-56y); and III, Isolated, severe trauma to extremities, with replantation and revascularization of a severed arm or leg, receiving 1-4 units of whole CPD blood (20-59y). Preoperative, intraoperative (2 hr into surgery) and 24 hr postoperative values were measured. Group III only had intraoperative sampling, with the exception of two patients who had preoperative values prior to any transfusions. A detailed statistical comparision was made (at the 95% confidence level) of the differences in ATIII activity, ATIII antigen level, the ratio of ATIII activity-to-antigen and plasminogen activity among categories within and between groups. The data suggest that elective surgery and moderate trauma had virtually the same effect on ATIII and plasminogen. Activity and antigen values decreased intraoperatively but on the average not enough to be out of the acceptable range. During the convalescent period most ATIII values returned to their preoperative level while most plasminogen values continued to decline somewhat (although still in the acceptable range). By contrast, both ATIII and plasminogen values in the severe trauma group were markedly depressed compared to preoperative levels In either group I or II. Four of the patients in this group suffered postoperative thrombosis and loss of an extremity even though the limb was initially viable. The transient lowering of both ATIII and available plasminogen is undoubtedly most critical when surgical stress is superimposed on existing trauma.

Background: Stress and anxiety in patients planned for surgery under anaesthesia may change pH of gastric secretion. Premedication of surgical patients with pH altering drugs may modify the pH favourably. With the advent of newer agents, premedication has been carried out with different agents. Most of the time choice of drug is made by the perioperative physician on his/her own. Objective: To study gastric pH in patients premedicated either with Esomeprazole or Famotidine. Methods: This is a randomized controlled double blind prospective study conducted in 150 patients of American Society of Anesthesiologists grade I and II posted for elective surgery under general anaesthesia. The patients enrolled in the study were randomly assigned to three groups having 50 patients in each. Group I (control group) did not receive any pH altering drug, Group II (Famotidine Group) received 40 mg of Famotidine and Group III (Esomeprazole Group) received 40 mg of Esomeprazole the night before surgery. The observer was totally blind about the groups or drugs given to the patients. On the day of surgery, after induction of anaesthesia gastric juice was obtained via nasogastric tube and was checked for pH using pH indicator paper. Results: The pH raised by Esomeprazole was statistically significant (p<0.001) when compared to that of control group or Famotidine group. The mean pH value in control group was less than 2.5 whereas the pH value was higher than 2.5 in patients premedicated with either Famotidine or Esomeprazole. Conclusions: Gastric pH is raised by Famotidine or Esomeprazole premedication prior to routine surgery, Esomeprazole being superior to Famotidine, p<0.001. Patients (84%) not premedicated with either of the drugs had pH less than 2.5. Objective: We investigated the liver protective activity of 2-3-6 trimethyloct-6-enal from the methanol extract of Pajanelia longifolia (Willd.) K. Schuman. The liver protective activity of 2,3,6 trimethyloct-6-enal was evaluated against paracetamol (2 mg/kg body weight per orally) induced liver toxicity in swiss albino mice.Methods: Considering the Spectral data (IR spectrum, 1HNMR spectrum and 13C NMR spectrum) the predictable structure of 2,3,6 trimethyloct-6-enal was elucidated. To study the liver protective activity of the compound, Swiss albino mice of either sex were divided into six groups and treated for 5 d. Group I and II served as normal and toxic control, Group III were treated with Silymarin as a standard drug (50 mg/kg), and Group IV to VI was treated with 2-3-6 trimethyloct-6-enal at the dose of 50 mg/kg, 150 mg/kg and 250 mg/kg b.w. p. o. respectively. The liver protective activity of the compound was measured on biochemical parameters such as aspertate amino transferase (AST), alanine amino transferase (ALT), alkaline phosphatase (ALP), total bilirubin (TB), triglycerides (TGL), total cholesterol (TC) and protein. Further antioxidant activity of the compound was also measured on antioxidant enzymatic and non-enzymatic levels such as reduced glutathione (GSH), lipid peroxidation (LPO), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx)).Results: The study revealed that the compound has protective activity at the dose of 50, 150 and 250 mg/kg b.w. p. o. against paracetamol induced toxicity. In some biochemical parameters such as aspartate amino transferase and bilirubin, the compound has showed better result at a dose of 150 mg/kg compared to standard drug silymarin (value of aspartate amino transferase (compound) =71.10±0.12, (toxic) = 173.43±1.21, (silymarin) =79.86±0.02and total bilirubin (compound) = 1.04±0.11), (toxic) = 2.69±0.02, (silymarin) ==1.11±0.01. The findings were also confirmed by histopathological observations.Conclusion: 2,3,6 trimethyloct-6-enal from Pajanelia longifolia may be considered as a potent liver protective agent.

Trigonella foenum-graecum L. is enriched with many active ingredients. TFG oil was evaluated for its protective effect against deltamethrin toxicity in rats. Rats of the control group were administered saline. The 2nd group was administered deltamethrin (DLM) orally at a concentration of 15 mg/kg body mass. The 3rd and 4th groups were administered DLM at a concentration of 15 mg/kg body mass and were fed diets containing 2.5% and 5% TFG oil, respectively. DLM intoxication reduced red blood cell and platelet counts, hemoglobin concentration, and hematocrit value while it induced leucocytosis. Furthermore, it increased serum levels of lactate dehydrogenase, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, γ-glutamyltransferase, triglycerides, cholesterol, uric acid, urea, and creatinine; increased hepatic, renal, and brain lipid peroxidation; decreased serum acetylcholine esterase level; and decreased hepatic, renal, and brain antioxidant markers’ activities. However, TFG oil kept the studied hematological and biochemical parameters within normal ranges. In addition, it prevented lipid peroxidation and oxidative stress induced by DLM intoxication in a dose-dependent manner. Therefore, these results indicated that TFG oil inhibited the toxic effects of DLM on hematological and biochemical parameters as well as oxidative status by its free radical scavenging and potent antioxidant activities, and it appeared to be a promising protective agent against DLM-induced toxicity.

Purpose The aim of this study was to evaluate the effects of propofol and alfentanil on the increase in intraocular pressure (IOP) due to succinylcholine and intubation, in comparison with thiopental sodium and vecuronium bromide. Methods Forty patients aged 20–50 years, scheduled for elective surgery requiring endotracheal intubation, were assigned to four groups of ten. General anesthesia was induced with 2.5 mg/kg propofol in Group I, 2.5 mg/kg propofol and 10 μg/kg alfentanil in Group II and 5 mg/kg thiopental in Groups III and IV; muscle relaxation was obtained with either 1.5 mg/kg succinylcholine (Group I, II and III) or 0.1 mg/kg vecuronium bromide (Group IV). In all patients mean arterial pressure, heart rate, oxygen saturation and IOP were recorded before (baseline) and after induction, after the muscle relaxant and after endotracheal intubation. Results Compared with their baseline values in Group I IOP decreased significantly after propofol (p<0.01) and increased significantly after intubation (p<0.01). In Group II IOP decreased significantly after propofol and alfentanyl (p<0.001), remained low after succinylcholine (p<0.01) and did not change after intubation. In Group III IOP decreased significantly after thiopental (p<0.001) and increased significantly after intubation (p<0.001). In Group IV it decreased significantly after thiopental (p<0.001), remained low after vecuronium (p<0.001) and increased significantly after intubation (p<0.05). Conclusions In all Groups, IOP did not increase significantly after succinylcholine, but only anesthesia induced with propofol and alfentanil prevented the increase in IOP due to intubation.
