Background: Lilium (Lilium longiflorum Thunb.) belongs to the family Liliaceae and is a native of Northern Hemisphere (up to South Canada and Siberia). Conventionally Lilium can easily be propagated by sexual and asexual methods of propagation but these prevalent methods are not capable of meeting the increasing demand in domestic and global market. Generally, Lilium is propagated through bulbs but, limited number of bulbs per plant, long dormancy period of bulbs which again results into non-availability of planting material throughout the year. Keeping in view the above facts, the present study was undertaken with the following objective: “To standardize the cost effective protocol for micro propagation of lilium to produce disease free and true to type plants at a faster rate”. Methods: The present investigation was carried out in the Tissue Culture Laboratory of the Centre for Research and Application in Plant Tissue Culture. The experiment was laid out in a C.R.D. (Factorial) with three replications. In vitro raised bulblets were separated out and were transferred on to the root regeneration media. Different levels of NAA were used in MS media for the rooting of in vitro raised bulblets and percent rooting of plantlet is recorded. Result: It was interesting to note that the media LR-3 (MS + NAA 1.0 mg/l) is most efficient for rooting in all type of cultivars. All the three cultivars used responded very poor on media LR-1 (MS basal).

Dimethylbenz(a)anthracene (DMBA)-induced and transplanted rat mammary tumours (2 lines) were examined for oestrogen receptor activity, and for sensitivity to hormones in vivo (by ovariectomy) and in vitro (by tissue culture). In vivo, the growth of all tumours induced by the administration of DMBA in random-bred Sprague–Dawley rats was found to be dependent on the ovary, whilst in all transplanted tumours (12 TG-3 and six TG-5 lines), maintained in an inbred strain of Sprague–Dawley rats, growth was found to be independent of the ovary. In vitro, the capacity for DNA synthesis in DMBA-induced tumours was better maintained after 24 h when insulin (10 μg/ml) and corticosterone (5 μg/ml) or insulin, corticosterone and prolactin (each 5 μg/ml) were present in the medium (five out of 12 and eight out of 11 tumours respectively); no effect of hormones in the media was detected after 48 h. In the transplanted tumours, no effect of hormones on DNA synthesis was detected after either 24 or 48 h of culture. Synthesis of lecithin was not detectably influenced by the presence of hormones in either DMBA-induced or transplanted tumours. Oestrogen receptor concentrations were, on average, significantly higher in the DMBA-induced tumours than in either line of transplanted tumour. For 22 DMBA-induced tumours and 15 transplanted tumours, the effect of hormones in vitro (`response') was directly correlated with receptor concentration at time 0 (Spearman's ρ = + 0·59) and inversely correlated with the rate of DNA synthesis (`basal') at time 0 (Spearman's ρ = −0·62). No single parameter or pair of parameters permitted accurate distinction between the tumour types.

The search for the occurrence of somaclonal variation of in vitro shoots and acclimatized plants of a hybrid larch (Larix × urolepis Henry) clone was performed by the analysis of eight isozyme systems. Cultures were established from short shoot buds of mature material. The effects of growth regulators in the media, subculture intervals, and periods in culture were analyzed for in vitro shoots. Variability was found in in vitro shoots but appeared to be related to a physiological response to culture conditions. Once acclimatized, most tissuecultured plants expressed the same enzymatic patterns as those of control plants (stecklings and the ortet). The variations observed for some acclimatized plants were also observed in control plants and were not related to ontogenic stage. Results from the isoenzymatic systems studied showed that hybrid larch plants regenerated from tissue culture were not significantly different from stecklings and the ortet.

Abstract In order to approach optimal micropropagation of Hypericum perforatum, it will be necessary to optimize shoot proliferation stage in in vitro culture. This study was conducted to investigate the effects of different concentrations of B group vitamins; Thiamine HCl, Pyridoxine HCl, Nicotinic acid (control and 100 fold of MS) and Sucrose (30 and 40 g.l−1) on shoot proliferation. For this purpose, Stems with one node were taken from in vitro shoots and cultured on MS medium. All cultures kept at 16h light/8h photoperiod and 25 ±2 °C in growth chamber. Results showed that the highest number of shoots and leaves were achieved when explants cultured in media containing 40 g.l−1 sucrose with 100 fold of MS vitamins. The highest shoots and leaf length were obtained with medium supplemented with 30 g.l−1 sucrose. Nicotinic acid concentrations had an important role in length of the leaves. The highest number of nodes achieved in the media containing 40 g.l−1 sucrose with both concentrations of Nicotinic acid. After two month growing plantlets, light (LGN) and dark glands number (DGN) were counted. Maximum number of LGN was observed in the media containing 30 g.l−1 sucrose with 100 fold of Thiamine and Pyridoxine. However, The Highest number of DGN achieved in the media containing 40 g.l−1 sucrose with 100 fold of Thiamine or Pyridoxine. Increasing of sucrose and vitamins concentrations were efficiently improved in vitro proliferation and some morphological attributes without negative side effects. Therefore, use of high levels of sucrose and vitamins were useful on micro-propagation of Hypericum perforatum.

Initiation of tissue culture of many plant species is a very difficult stage due to appearance of many contaminations. The other problem might be a choice of media for regeneration. Initiation of grass species tissue cultures are thought to be very difficult. Therefore, a research was undertaken to evaluate the use of nano-silver particles for plant material disinfection and to estimate a medium Pennisetum alopecuroides. The plant material were buds and nodal explants that were disinfected in 2% NaOCl for 30 min or 0.1% HgCl2 for 1 min. Half of the explants disinfected with NaOCl were soaked in 50, 100 or 250 mg·dm Ag–3 NPs for 1 hour. Explants not soaked in nano-silver were placed on media with Ag NPs at concentrations of 4, 8 or 16 mg·dm–3. An influence of growth regulators on Pennisetum alopecuroides was evaluated in vitro. Regenerated shoots were placed on MS media with: 3 mg·dm–3 BA + 0.3 mg·dm–3 IBA, 3 mg·dm–3 KIN + 0.3 mg·dm–3 IAA, 1 mg·dm–3 BA + 0.1 mg·dm–3 IBA. It was observed that the use of nano-silver particles lowered the level of contamination. The best results were obtained when Ag NPs was used at concentration of 100–250 mg·dm–3 alone or as a supplementation of the media, at concentration of 4 mg·dm–3 for nodes and 16 mg·dm–3 for adventitious buds. The use of nodal explants allowed to obtain less contamination. Regeneration depended on a media content. The most regenerated shoots were obtained on the MS media supplemented with 1 mg·dm–3 BA and 0.1 mg·dm–3 IBA.

Abstract There are some limitations in the practical applications of in vitro date palm tissue culture, such as low multiplication efficiency, low rooting rate, and high mortality experienced by in vitro raised plantlets during laboratory to soil transfer. The objective of the present study is to determine the effect of polyamines (putrescine "PUT" and spermidine" SPD") and silver thiosulfate (STS) on enhancing propagation of date palm cv Quntar in vitro. Media supplemented with 75 mg L-1 SPD in combination with 10 mgL-1 STS gave the highest percentage of callus producing buds (83.34%) and average bud formation (16.3) per jar. The addition of PUT and STS to the medium was most effective in root regeneration and the number of roots per shoot, where the best result 91.67% and 6.37 roots per shoot, respectively, were obtained using 75 mgL-1 PUT and 10 mgL-1 STS, resulting in fast-growing plantlets during acclimatization phase, reaching 90% of plant survival. The genetic fidelity assessment of plants derived from micropropagation was confirmed by RAPD analysis. Four operon primers were used, and all of them showed amplified unambiguous (OPA02, OPC-04, OPD-07, and OPE-15). All generated bands were monomorphic and had no variation among the tissue culture-derived plants tested. Accordingly, these results indicate that adding polyamines and silver thiosulfate to the nutrient medium of date palm cv. Quntar is beneficial in improving shoot organogenesis, rooting, and production of genetically stable date palm plants.

Vanilla has a potential to be developed through tissue culture techniques to anticipate the limitations of the parent plant as a source of planting material. The in vitro propagation ability of vanilla shoots needs to be controlled with the regulation of Kinetin and Benzyl Amino Purines. The interests of this study are 1) analysis of the response of vanilla explants at several Kinetin concentrations; 2) analysis of the response of vanilla explants at several concentrations of BAP and 3) analysis of the interaction of Kinetin and BAP on the response of vanilla explants to form shoot multiplication. The research was conducted at the Tissue Culture Laboratory Politeknik Negeri Jember from June to December 2020 using a factorial Completely Randomized Design (CRD). Factor 1 was the Kinetin concentration of 0.0, 1.0, 2.0 mg.L-1 and the second factor was the concentration of BAP 0.5, 1.5, 2.5 mg.L-1. The results proved that the fastest shoot multiplication occurred on MS medium + Kinetin 2 mg.L-1 with a mean of 8.7 days after inoculation. The mean number of shoots was 7.6 shoots/explant with the highest average wet weight of 0.9 grams/explant at the addition of BAP 1.5 mg. L-1 at measurement 70 days after inoculation.

The demand for Hass avocado in the global market exceeds the supply by over 50%. Colombia has a remarkable advantage as a producer in the region due to its high yields. However, the productivity of this crop can be seriously affected by diseases such as root rot, caused by Phytophthora cinnamomi, postharvest body rot and stem end rot, caused by Colletotrichum sp. and Phomopsis sp., respectively. The potential of 76 bacterial isolates obtained from avocado rhizosphere to produce inhibitory metabolites against avocado´s pathogens was evaluated. The antagonistic effect of the rhizobacteria against P. cinnamomi, Colletotrichum sp. and Phomopsis sp. was tested through dual cultures. Thirty-six percent of the tested isolates presented inhibition halos against P. cinnamomi, 36% against Colletotrichum sp. and 67% against Phomopsis sp. Additionally, three isolates were selected for fermentation tests using different broth cultures. The extracts obtained from fermentations in the minimal medium of isolates ARP5.1 and AED06 showed inhibitory activity against the evaluated pathogens, but this effect was not observed with the AED26 extract. The media supplemented with copper chloride did not enhance activity of the extracts. These results suggest that using microbial metabolic extracts is a viable alternative for controlling avocado pathogens in vitro.
