Superovulatory Response and Progesterone Profile in Murrah Buffaloes (Bubalus bubalis) Pretreated with GnRH Agonist

Background: Limited elite buffalo population need to be propagated at faster rate to sustain the dairy industry in India. Faster propagation could be achieved through embryo transfer technology. Low serum progesterone concentrations during the start of superstimulatory treatment is major factor that hampers the success of multiple ovulation embryo transfer (MOET) in buffaloes. This study evaluated the effect of GnRH pretreatment before superovulatory regimen on progesterone profile, superovulation and correlation of progesterone with superstimulatory parameter in Murrah buffaloes. Methods: Buffaloes (n=27) were superstimulated using 600 mg Folltropin and divided into three groups on basis of pretreatment: Group I and group II were pretreated with GnRH @ 10 µg and 06 µg, respectively while no pretreatment was subjected in group III (control). Progesterone hormone concentration, superovulatory response, ovulation rate, embryo recovery were recorded. Result: Our study revealed that superovulatory response was non-significantly different in all three groups and ranged from 6-8 in terms of number of corpus luteum (CL). Average number of recovered embryos in this study was nearly 2.0. A positive correlation between progesterone concentrations at the initiation of FSH treatment with number of CL and embryo recovery was observed. The study indicated that GnRH pretreatment before superstimulation improve the serum progesterone concentration, superovulatory response and embryo recovery in Murrah buffaloes.

The serum progesterone and estradiol profiles during synchronization of estrus by buck effect and PGF2α treatments were monitored in Surti does. Total eighteen non-pregnant does selected were evenly divided into 3 groups, 6 does in each group. The does of Group I were teased with a sexuallyactive- apronized buck; and those of Group II were treated with PGF2α, i.e., Inj. Lutalyse® @ 7.5 mg/doe IM twice 11 days apart, while the Group III served as untreated control. Blood samples were collected from all the animals on day 0 (before 1st PGF2α injection), 3rd day (during treatment), 11th day (before 2nd PGF2α injection), 14th day (after treatment) and 40th day (post-service) by jugular vein puncture. The serum separated was stored at -20°C till further analysis. In all the three groups, 83.33% does, conceived at first service in the sampling cycle. The overall mean serum progesterone concentration of Group I does (5.82±0.72 ng/ml) was significantly higher (p less than 0.01) as compared to Group II (2.93±0.38 ng/ml) and III (2.88±0.30 ng/ml). Similarly, the overall mean serum progesterone concentration of Surti does on day 0 (2.65±0.46 ng/ml), 3rd (2.56±0.80 ng/ml), 11th (4.45±0.84 ng/ml) and 14th (3.40±0.63 ng/ml) did not differ significantly, but the overall mean level at day 40 (6.31±0.45 ng/ml) was significantly (p less than 0.01) higher, because most of animals became pregnant at that time. The overall mean serum oestradiol-17β levels of Group I (24.40±2.98 pg/ ml) was significantly higher (p less than 0.01) than in Group II (15.77±1.77 pg/ml) and III (12.21±1.45 pg/ ml). On the other hand, the overall mean serum oestradiol-17β levels of Surti does on day 0 (12.89±1.21 pg/ml), 3rd (15.84±1.74 pg/ml), 11th (14.81±1.96 pg/ml), 14th (22.15±2.97 pg/ml) and 40th (21.64±5.16 pg/ml) did not differ significantly (p>0.05) and the slightly higher overall mean level found at 40th day might be the influence of the non-pregnant does at first service in the cumulative animals. The hormonal profile reflected the initiation of cyclicity and establishment of pregnancy in treated and control animals.

An investigation was taken up to study the relationship of preovulatory follicle size at the time of first postpartum AI and CL biometry with conception using transrectal ultrasonography in 70 parous Graded Murrah buffaloes maintained under rural conditions. POF sizes were measured ultrasonographically and grouped in to small preovulatory follicle (SPF), medium preovulatory follicle (MPF) and large preovulatory follicle (LPF). Serum progesterone concentration at the time of AI and pregnancy status was negatively correlated indicating that when progesterone level declined to less than 0.3 ng/ml (basal level) at the time of AI, the chances of the animal becoming pregnant was increased. The size of the POF was positively correlated to the size of the CL on day 10 in pregnant buffaloes. The overall POF diameter at the time of estrum was 12.31 ± 0.29 mm (Range 9 to 16 mm) in Graded Murrah buffaloes and 50 % of the buffaloes had POF size > 12 - £14 mm. They conceived only if the POF diameter was more than 9 mm at the time of AI however there was no significant correlation between the POF size at the time of AI and conception. Thus, it was concluded that physiological maturity rather than the diameter of the follicle influenced the fertility in Graded Murrah buffaloes under field conditions.

Background: Pinopods concentrations in endometrial surface is a marker of implantation. Estradiol valerate (EV) was used to change the adverse effects of Clomiphene Citrate (CC) on the endometrium. Objective: The goal was to assess whether there is a significant difference in the endometrial pinopods concentrations and other parameters after adding EV and progesterone to higher doses of CC. Materials and Methods: In this prospective randomized clinical trial, a total of 30 women who did not respond to 100 mg of CC from February 2016 to June 2016 were evaluated. They were divided into three groups: group I) received 150 mg of CC alone, group II) CC with EV, and group III) CC plus progesterone. On day 21 of the menstrual cycle, endometrial biopsy, a blood sampling, and a scanning by electron microscopy were performed. Results: On day 21 of the menstrual cycle, there was no significant difference in the pinopods concentrations (p = 0.641) and serum estrogen levels (p = 0.276) between groups. However, the Serum progesterone levels in group I was higher than the other two groups (p = 0.007) in the same day. Conclusion: Since the addition of EV and progesterone to higher dosages of CC did not change the pinopods concentration and serum estrogen levels on day 21 of the menstrual cycle, and the serum progesterone levels was higher in CC alone group (i.e. group I) compared to other groups, it can be concluded that the anti-estrogenic effects of CC just appear on the endometrium and not on the plasma levels. The objective of the study was to characterize the term Doka and to capture morphological changes in teat to find out the relationship between Doka and onset of estrus in Murrah buffaloes (Bubalus bubalis). The Murrah buffaloes maintained at of Livestock Research Centre (LRC), ICAR-NDRI, Karnal were classified into two groups based on the lactation number for the study of Doka characteristics. In group I, twenty animals up to 3rd lactation were incorporated, whereas thirty animals from 4th to 5th lactation were included in group II. The period when buffaloes showed teat engorgement was called as Doka period, is the local term used by the farmers in North India that can be taken as a visual sign for prediction of estrus. The length and diameter of teats, before and after milking were measured, for both the groups during morning and evening milking and subsequently analyzed. The present study confirmed that the teat diameter (mm) before milking during Doka was significantly (plessthan0.05) higher as compared to pre Doka period in both the groups (group-I - 37.01 ±2.13 vs 28.42 ±1.64, group-II - 53.49 ±2.82 vs 41.096±2.16). The teat diameter (mm) during Doka period after milking was significantly (plessthan0.05) more in both the groups (group-I - 26.65 ±1.26 vs 24.44 ±1.16, group-II - 39.27 ±2.41 vs 35.31 ±2.17) as compared to pre Doka period. So, it can be concluded that the changes in teat diameter can safely be considered as a sign of Doka and the forthcoming heat in Murrah buffaloes. However, the difference in teat length between different stages of Doka was found to be non significant.

The effects of two doses of cloprostenol and two doses of flurogestone acetate sponge on the onset of oestrus, and embryo recovery and quality were evaluated. Thirty-two indigenous ewes (Wera breed) were allocated into four groups of eight. All ewes were synchronised with 100 µg (Group I) or 175 µg (Group II) cloprostenol injection, 9 days apart, or insertion of intravaginal sponges containing 30 mg (Group III) or 45 mg (Group IV) flurogestone acetate (FGA) for 12 days. The ewes were superovulated with 600 IU PMSG intramuscularly 10 days after the second cloprostenol injection or immediately after sponge removal on Day 12. After the detection of oestrus, the ewes were mated naturally at 6 and 12 h, and some ewes were inseminated laparoscopically. Embryos were recovered surgically 5 or 6 days after service. All ewes exhibited oestrus. The onset of oestrus occurred significantly (P < 0.05) earlier in FGA-treated (50.0 ± 1.5 and 48.0 ± 0.00 h) than in cloprostenol-treated groups. There were no significant differences (P > 0.05) in the mean time of onset of oestrus (50.0 ± 1.5 and 48.0 ± 0.00 h) between the two doses of cloprostenol. The mean number of corpora lutea (8.1 ± 1.26) and embryos recovered (6.1 ± 1.00) was significantly (P < 0.05) higher in ewes treated with 45 mg FGA than in ewes treated with cloprostenol. Embryo recovery rate was significantly (P < 0.05) higher in ewes treated with 45 mg FGA (75.4%) than in other groups (41.7% and 51.6% in 100 μg and 175 μg cloprostenol- and 52.7% in 30 mg FGA-treated groups, respectively). Fertilisation rate was 93.3% in ewes given 100 µg cloprostenol, whereas other groups showed 100% fertilisation rate. The highest percentage (100%) of Grade 1 embryos was in FGA groups. In conclusion, despite FGA protocol presenting superior results, cloprostenol protocol was equally efficient in synchronising oestrus. The embryo recovery rate was better after 45 mg FGA than 30 mg FGA or after either dose of cloprostenol.

Background: The dairy and livestock sector plays a very important role in national economy of India by contributing close to one third of gross income of rural households and nearly half of gross income in case of those without land. Subestrus is the most prevalent, frustrating and challenging problem encountered in postpartum buffaloes. Postpartum subestrus is one of the most prevalent, frustrating and challenging reproductive problem encountered in rural resulting into prolonged inter-calving period, reduced milk production and thus greatly affecting the economy of our farming community. Methods: The present study was carried out on Graded Murrah buffaloes during the period from September 2017 to August 2019 (includes breeding and low breeding season) which were maintained at farm and field. Treatment of sub-estrus lactating graded Murrah buffaloes (Bubalus bubalis) using Double PGF2 α (DPG) and Presynch-Ovsynch (POVS) their influence on fertility in farm and field condition during breeding and low breeding season was studied. Total cholesterol and progesterone concentration was recorded during different days/period of protocols. Further, overall per cent conception and pregnancy rates were recorded and compared between two protocols. Result: Progesterone levels increased significantly on day 21 post AI in pregnant buffaloes, whereas decreased significantly in non-pregnant buffaloes both in farm and field during breeding and low breeding season. The overall per cent conception and pregnancy rates were higher in POVS group than DPG group at both farm and field as well as during breeding and low breeding seasons.

A study was conducted in 18 healthy, fertile, non-pregnant, dioestrous female dogs to evaluate the efficacy of PGF2? analogue (cloprostenol sodium) in shortening the dioestrus. Bitches that were diagnosed non-pregnant by trans-abdominal ultrasonography, on day 30 of breeding and their dioestrual stage confirmed by serum progesterone assay, were randomly allotted to three groups of six bitches each. Group I and II bitches were treated with cloprostenol sodium @ 2.5 µg/kg b. wt., subcutaneously, thrice at 48 h interval, beginning on day 30 and 45 of last breeding, respectively. Group III animals were assigned as control and were left untreated. Fifty days from first injection of cloprostenol, serum progesterone assay was carried out among the animals in Group I and II whereas the same was done among Group III bitches on day 90 after last breeding. Among Group I and II, mean serum progesterone concentrations were 0.93 ± 0.04 and 0.87 ± 0.01ng/ mL, respectively whereas the level was 1.72 ± 0.9 ng/mL among control group; highly significant difference (p<0.01) existed between control and treatment groups. The present study indicated that administration of PGF2? analogue during dioestrus enhances the pace of luteal regression among non-pregnant bitches.

ABSTRACTDevelopments, both recent and potential, in procedures for manipulating embryos are described. The procedures considered include: embryo transfer, multiple ovulation and embryo recovery, recovery of oocytes, in vitro maturation (IVM) and fertilization (IVF) of oocytes, in vitro culture of zygotes, embryo splitting and nuclear transfer, embryo storage, embryo sexing, gene transfer and embryo stem cells. The impact of these procedures on breeding strategies such as multiple ovulation and embryo transfer (MOET) nucleus breeding schemes and progeny testing are discussed for both dairy and beef cattle.For MOET nucleus schemes all these procedures have potential applications in producing maximal rates of genetic progress for a fixed rate of inbreeding. With the current effectiveness of the procedures, embryo sexing and nuclear transfer would have the most impact. The potential for increasing genetic progress through progeny testing is enhanced using multiple ovulation, embryo recovery and transfer in cows to breed bulls, but no other procedures appeared to offer major benefits. The efficiency of beef production from the dairy herd could be increased either by using IVM and IVF to produce more beef-type calves or, potentially, by cloning and embryo transfer, to produce pure beef calves. Procedures leading to the production of clone families would make an impact on the evaluation of genotypes and environments. Gene transfer may be used to modify the composition of milk including the production of pharmaceutical proteins, and to increase milk yield or the efficiency of lean meat production.It is concluded that, although much further research is required, the procedures discussed will have major implications for the structure and organization of dairy and beef cattle herds over the next decade.
