ABSTRACT Herbal supplements are one of the products that are taken from outside the body derived from a mixture of herbal plant ingredients have antioxidant activity to prevent the presence of free radicals in the body. Testing natural antioxidant activity in herbal supplement products using the DPPH method (1,1-diphenyl-2-picrylhyrazil). Measurement of UV-Visible spectrophotometry absorbance with DPPH absorbance value at a wavelength of 517 nm. Sensory analysis using a hedonic test at the panelist preference level, using four attribute parameters namely taste, aroma, color and overall acceptance. Results Tests of antioxidant activity in herbal supplement products on various combination techniques and concentrations of garlic, ginger, local lemons and honey, resulting in the reduction of DPPH which is characterized by a reduction in the intensity of the color from purple to fade to yellow. Antioxidant testing obtained IC50 values from all herbal supplement samples from various treatment techniques and concentrations showed IC50 values less than 50 found in the treatment of chopped engineering materials, concentration 1 (K1) of 23.97%. While concentrations of 2 (k2) and 3 (k3) as well as in various combination techniques and other concentrations indicate that IC50 values range from 50 ppm to 100 ppm. This shows that herbal supplements have very strong antioxidants (IC50 value <50) found in the treatment of ingredients with chopped techniques with the lowest concentration. whereas herbal supplements with treatment techniques and other concentrations have a strong antioxidant value of IC50 (50-100). The results of sensory analysis indicate that the average panelist had a preference level for herbal supplement products with chopping and extraction treatment techniques. Citrus essential oils (EOs) have various bioactivities like antioxidants, with many applications. Antioxidant activities depend on the chemical compositions of the EOs, which are affected by climate, soil, and geographical region. Thus, investigations on chemical compositions and antioxidant activities of Citrus EOs in different countries are valuable. In this study, we distilled EOs from peels of Indonesian-grown Citrus, including C. nobilis, C. limon, C. aurantifolia, C. amblycarpa, and Citrus spp.Chemical compositions of EOs were analyzed using Gas Chromatography-Mass Spectrometer (GC-MS), whereas the antioxidant activities were determined by employing 2,2-diphenyl-2-picrylhydrazyl (DPPH) method. Furthermore, principal component analysis (PCA) was applied to elucidate the main contributing compounds for antioxidant activity. The results show that all EOs possess unique chemical characteristics, with limonene as the majority constituent. For antioxidant activities, C. limon and C. amblycarpa EOs are the two strongest, IC50 values below 7.00 μL/mL. PCA approach suggests that -terpinene mainly contributes to the high antioxidant activities of C. limon and C. amblycarpa. Moreover, o-cymene, thymol, p-cymene, and α-pharnesene may also be responsible for the antioxidant activity of C. limon EO. These results are valuable information for the applications of Citrus EOs as antioxidant sources.

Free radicals are formed inside and outside the body. Free radicals inside the body can be created in the cell compartments. The reactions are complex through the initiation, propagation, and termination stages. On the other hand, free radicals in the outer body can be formed from pollutions and environmental damage action. Excessive free radicals cause oxidative stress and use destructive cellular to affect health conditions. Antioxidants are known as essential agents in biological systems against diseases triggered by free radicals. One of the many antioxidants found in tea plants (Camellia sinensis) is polyphenols. A colorimetry method was employed to evaluate the dominant antioxidant compound of the total phenolic and flavonoid content in two selected tea products. The total phenolic content was determined by utilizing the Folin-Ciocalteu reagent. In addition, an aluminum colorimetric assay is applied to assess flavonoid content. Rapid screening of antioxidant activity using a spectrophotometric method is employed to determine IC50 values of tea extracts utilizing DPPH and FRAP. The FRAP method showed more sensitivity in determining antioxidant activities with the feasibility of working in low analyte concentrations than the DPPH method. The high total phenolic and flavonoid contents found in this experiment are in line with low IC50 values. It was also found out that brewed green tea has lower total phenol content than green tea bags.

The hydrolysates and peptide fractions of bigeye tuna (Thunnus obesus) skin collagen have been successfully studied. The hydrolysates (HPA, HPN, HPS, HBA, HBN, HBS) were the result of the hydrolysis of collagen using alcalase, neutrase, and savinase. The peptide fractions (PPA, PPN, PPS, PBA, PBN, PBS) were the fractions obtained following ultrafiltration of the hydrolysates. The antioxidant activities of the hydrolysates and peptide fractions were studied using the DPPH method. The effects of collagen types, enzymes, and molecular sizes on the antioxidant activities were analyzed using profile plots analysis. The amino acid sequences of the peptides in the fraction with the highest antioxidant activity were analyzed using LC-MS/MS. Finally, their bioactivity and characteristics were studied using in silico analysis. The hydrolysates and peptide fractions provided antioxidant activity (6.17–135.40 µmol AAE/g protein). The lower molecular weight fraction had higher antioxidant activity. Collagen from pepsin treatment produced higher activity than that of bromelain treatment. The fraction from collagen hydrolysates by savinase treatment had the highest activity compared to neutrase and alcalase treatments. The peptides in the PBN and PPS fractions of <3 kDa had antidiabetic, antihypertensive and antioxidant activities. In conclusion, they have the potential to be used in food and health applications.
