Transcriptomics and Transmission Ultrastructural Examination Reveals the Nephrotoxicity of Cadmium in Laying Hens

Abstract The objective of this study was to reveal the effects of cadmium (Cd) on ultrastructural changes, oxidative stress, and transcriptome expression in the kidneys of laying hens. Seventy-two healthy Hy-Line brown laying hens at 41 weeks old were randomly allocated to four treatment groups with six replicates. The control group received a basal diet without additional Cd incorporation, and the other three treatment groups received diets supplemented with 15, 30, or 60 mg Cd /kg of feed. After 6 weeks of exposure the results show that administration of 60 mg/kg Cd significantly reduced (P < 0.05) eggshell thickness. With an increase in the Cd concentration in feed, the concentrations of renal Zn, Fe also had changed. Renal histopathology and ultrastructure also showed aggravated damage to glomeruli and renal tubules, and the deformation of nuclei and mitochondria in all Cd treatment groups. With an increase in Cd in feed, the activity of GPX and CAT was significantly reduced (P<0.05), while the activity of T-AOC was decreased (P<0.05) only in the 60 mg/kg Cd group. RNA-seq analysis revealed that 410 genes displayed differential expression (≥ 1.5-fold) in the 60 mg/kg supplementation group, compared to the control group. GO and KEGG pathway analysis results showed that Cd affected many genes involved in mitochondria and ion transport. In conclusion, this study elaborates the mechanisms underlying renal toxicity caused by Cd, which might provide target candidate genes for alleviating Cd poisoning in laying hens.

A 16 week feeding trial was conducted with 140 White Leghorn layers of 22-23±0.08 weeks of age which were distributed randomly into seven experimental groups, having 4 replicates with 5 birds in each. The laying hens of control group (T1) were fed a basal diet (CP 18.04%), while in treatment groups T2, T3, T4 basal diet was supplemented with probiotic (containing 5×108 cfu/g of Lactobacillus fermentum, 1×109 cfu/g of Bacillus spp., 1×109cfu/g of Saccharomyces cerevisiae) at levels of 0.5g, 1.0 g and 2.0 g /kg feed, respectively, while in T5, T6, and T7 0.5g, 1.0g and 2.0g/kg feed of prebiotic (mannonoligosacchride of Saccharomyces cell wall-47g, formic acid-32g) was supplemented, respectively. The percent hen day egg production and feed efficiency (kg/dozen eggs and kg/kg egg mass) were improved (P less than 0.05) in probiotic and prebiotic incorporated diets. The mRNA expression levels of Toll like receptors (TLRs) TLR2 and TLR4 was increased (P less than 0.05) when graded levels of the probiotic and prebiotic were supplemented, while, TLR7 did not show any significant change. Thus, it can be inferred that the inclusion of the probiotic and prebiotic could enhance the production performance and immune status of birds by augmenting the T cell mediated immune response.

Alternative growth promoters are able to not only effectively replace the traditional use of antibiotics but also provide additional health benefits for livestock and reduce food safety concerns. This study investigated the effects of dry Hydrastis canadensis on the laying performance and fecal microbial community of laying hens. Twenty-four Lohmann (LSL, white layer strain) hens were reared from 40 to 48 weeks of age and randomly allotted to four dietary treatments (six birds/treatment). The dietary treatments comprised a basal diet with no treatment as control, a basal diet plus 0.6% powder of dry Hydrastis canadensis roots (R) or leaves (L), and a basal diet plus 0.6% powder of a mixture of dry Hydrastis canadensis roots and leaves (1:1, LR). No mortality was observed in the whole experimental period. The results indicated that albumen height in the LR group was significantly greater than that in the control group. The diet supplemented with Hydrastis canadensis had no significant effects on egg production rate, egg weight, eggshell strength, eggshell thickness, Haugh unit, or yolk height during the whole experimental phase. However, principal coordinate analysis, comparative heat map analysis, and cluster dendrogram analysis of cecal microbiota showed distinct clusters among the groups treated with Hydrastis canadensis and the control group. Regarding blood biochemical parameters, serum cholesterol levels were significantly lower in all Hydrastis canadensis-treated groups compared with those in the control group. Moreover, serum low-density lipoprotein levels were lower in hens supplemented with the leaf of Hydrastis canadensis. The abundances of the phyla Fusobacteria and Kiritimatiellaeota were increased (p < 0.05) in laying hens fed with 0.6% Hydrastis canadensis leaves, whereas the abundance of the phylum Firmicutes in cecum digesta decreased in response to treatment with Hydrastis canadensis roots and leaves. The relative abundance of the Fusobacterium genus was higher in the LR group compared with that in the control. On the contrary, we found a different trend in the Synergistes genus. The potential influences of these microbiota on the performance of laying hens were discussed. The results demonstrate that Hydrastis canadensis can improve the egg albumen height and modulate the cecum digesta microbiota composition of laying hens.

AbstractPurslane is a widespread wild vegetable with both medicinal and edible properties. It is highly appreciated for its high nutritional value and is also considered as a high-quality feed resource for livestock and poultry. In this study, Sanhuang broilers were used to investigate the effect of feeding purslane diets on the growth performance in broilers and their gut microbiota. A total of 48 birds with good growth and uniform weight were selected and randomly allocated to four treatment groups A (control), B, C and D. Dietary treatments were fed with basal diet without purslane and diets containing 1%, 2% and 3% purslane. The 16S rDNA was amplified by PCR and sequenced using the Illumina HiSeq platform to analyze the composition and diversity of gut microbiota in the four sets of samples. The results showed that dietary inclusion of 2% and 3% purslane could significantly improve the growth performance and reduce the feed conversion ratio. Microbial diversity analysis indicated that the composition of gut microbiota of Sanhuang broilers mainly included Gallibacterium, Bacteroides and Escherichia-Shigella, etc. As the content of purslane was increased, the abundance of Lactobacillus increased significantly, and Escherichia-Shigella decreased. LEfSe analysis revealed that Bacteroides_caecigallinarum, Lachnospiraceae, Lactobacillales and Firmicutes had significant differences compared with the control group. PICRUSt analysis revealed bacteria mainly enriched in carbohydrate metabolism pathway due to the additon of purslane in the diet. These results suggest that the addition of purslane to feed could increase the abundance of Lactobacillus in intestine, modulate the environment of gut microbiota and promote the metabolism of carbohydrates to improve its growth performance. This study indicates that the effect of purslane on the growth-promoting performance of broilers might depend on its modulation on gut microbiota, so as to provide a certain scientific basis for the application of purslane in the feed industry.

The aim of the experiment was to assess the effect of dietary alpha-ketoglutarate (AKG) supplementation on performance, serum hormonal indices, duodenum and jejunum histomorphometry, meat quality characteristics, bone quality traits and cartilage degradation in laying hens with a mature skeletal system. Forty-eight 30 week-old Bovans Brown laying hens were randomly assigned to a control group or the group fed the basal diet plus 1.0% AKG. The experimental trial lasted 30 weeks. The supplementation of AKG increases blood serum content of leptin, ghrelin, bone alkaline phosphatate and receptor activator of nuclear factor kappa-Β ligand, while osteoprotegerin and osteocalcin decrease. While dietary AKG was given to laying hens negatively influenced villus length, crypt depth, villus/crypt ratio and absorptive surface area in duodenum and jejunum, these changes have no effect on feed intake, weight gain, nor laying performance. In breast muscles, no significant changes in skeletal muscle fatty acid composition were observed, however, a higher shear force and decreased cholesterol content following AKG supplementation were noted, showing the improvement of muscle quality. While dietary AKG supplementation did not affect the general geometric and mechanical properties of the tibia, it increased collagen synthesis and enhanced immature collagen content. In medullary bone, an increase of bone volume fraction, trabecular thickness, fractal dimension and decrease of trabecular space were observed in AKG supplemented group. The trabeculae in bone metaphysis were also significantly thicker after AKG supplementation. AKG promoted fibrillogenesis in articular cartilage, as indicated by increased cartilage oligomeric matrix protein immunoexpression. By improving the structure and maintaining the proper bone turnover rate of highly reactive and metabolically active medullar and trabecular bones AKG showed its anti-osteoporotic action in laying hens.

ABSTRACTGas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) were employed to determine the chemical composition of the essential oil (EO) from aromatic water (AW) ofZataria multiflora. Thymol (66.9%), carvacrol (15.2%), and carvone (7.3%) were found to be the major EO constituents. Eighty laboratory BALB/c mice were infected intraperitoneally by injection of 1,500 viable protoscolices and were divided into prevention (40 mice) and therapeutic (40 mice) groups. To prove the preventive effect of theZ. multifloraAW on development of hydatid cysts, the 40 infected mice were allocated into three treatment groups, including the albendazole group (10 mice that received 150 mg/kg body weight/day for 10 days), theZ. multifloraAW group (15 mice that received 20 ml/liter in drinking water for 8 months), and a control group (15 mice that received no treatment). To estimate the therapeutic effect of theZ. multifloraAW on the hydatid cyst, after 8 months of infection, the 15 remaining mice were allocated into three experimental treatment groups of five animals each, including the albendazole group (300 mg/kg/day for 20 days),Z. multifloraAW group (40 ml/liter in drinking water for 30 days), and control group (no treatment). All mice were then euthanized, and the sizes and weights of the cysts as well as their ultrastructural changes were investigated. The weights and sizes of the hydatid cysts significantly decreased upon treatment with theZ. multifloraAW in both the preventive and therapeutic groups (P< 0.05). The results of scanning electron microscopy also showed considerable damage in the germinal layer of the hydatid cysts recovered from the treated animals.

Trichophyton rubrum is one of the most common types of dermatophyte, causing superficial skin mycosis in human populations. Although laser treatment of onychomycosis has been proven to be effective in the clinic, the underlying mechanism of the effect of the laser on fungal growth is not clear. The objective of the present study was to observe the ultrastructural changes of Trichophyton rubrum following laser irradiation and compare the transcriptome differences between the laser irradiation group and control group. In the present study, scanning electron microscopy and transmission electron microscopy were used to observe the ultrastructural changes following the laser irradiation of Trichophyton rubrum. We also performed RNA-seq to investigate the effects of laser irradiation on Trichophyton rubrum by comparing the transcriptome pattern with the control. Morphological observation with electron microscopy indicated that laser irradiation resulted in the destruction of the cell membrane system. A significant induction of apoptosis was noted compared with the control group, which was confirmed by the formation of the myeloid body and protein aggregates in the cytoplasm. RNA-seq demonstrated that the expression levels of Acyl-CoA N-acyltransferase and S-adenosyl-L-methionine-dependent methyltransferase were increased in the laser irradiation group. This result indicated that laser irradiation triggered the initiation of the damage repair pathway. In conclusion, the present study suggested that laser irradiation caused physiological injury and therefore inhibited the growth of Trichophyton rubrum.

This study was conducted to observe the effect of different administration method of garlic and neem on growth performances, feed efficiency and serum biochemical parameters for safe broiler production. The experiment was done for a period of 32 days with 384 one-day-old straight run broiler chicks. The broiler chicks were divided into six groups each of 64 birds and replicated to four subgroups each of 16 birds. The dietary groups were; control (basal diet; no additives), antibiotic (basal diet + antibiotic), garlic in feed (basal diet + 0.25% garlic powder), garlic in water (basal diet + garlic extracts), neem in feed (basal diet + 0.25% neem powder) and neem in water (basal diet + neem extracts). Results showed that the body weight and body weight gain increased significantly (P<0.05) in both the garlic and neem groups compared to the control group. Feed intake was not different (P>0.05) among the treatment groups. Garlic in feed and water and neem in feed groups showed better FCR (P<0.05) compared to the other treatment groups. There were no significant (P>0.05) difference in meat yield, bone development and dressing parameters except head and gizzard in different dietary groups. Supplementation of garlic with feed and water significantly (P<0.05) decrease cholesterol, triglyceride and LDL compared to the other groups. However, both the garlic and neem groups showed numerically higher HDL compare to the control group. There were no 0significant (P>0.05) differences in serum glucose and GPT among different treatment groups. GOT was significantly (P<0.05) lower in neem groups than that of garlic and antibiotic groups. Additive groups showed higher profitability than control group. Based on the results of the study, it may be suggested that the garlic and neem could be used both in feed and water as potential feed additives for safe broiler production. Bang. J. Anim. In this study, it was investigated the effect of clove oil supplementation at increasing levels into laying hens’ diets on performance, egg quality traits, some blood parameters and yolk TBARS (Thiobarbituric Acid Reagent) values. For this purpose 96 Lohman white laying hens, 28 weeks of age, were divided into four treatment groups. Control group was fed with basal diet (C) and treatment groups were fed with diets formed by addition of clove oil at 50 ppm (CO1), 100 ppm (CO2) and 150 ppm (CO3), respectively. During the trial, feed and water were given as ad-libitum, and poultry house was illuminated for 17 hours. Experiment lasted for 13 weeks. Addition clove oil at increasing rates into diet did not affect the live weight. The data analysed as polynomial showed that supplementation of clove oil into layer diet linearly improved feed conversion ratio and increased the egg production. But, daily feed consumption, egg weight, damaged egg ratio and egg quality parameters were not affected by treatments. Serum parameters such as triglyceride, glucose, aspartate aminotransferase, alanine aminotransferase and calcium were not affected by the clove oil supplementation. TBARS values in C, CO1, and CO2 were found higher than the CO3 group fed with diet including 150 ppm clove oil. In conclusion, clove oil at 150 ppm level in diets of laying hens could be used due to extend the egg shelf life and to decline serum cholesterol content.

The objective of this study was to compare the effects of feed supplementation of laying hens with manganese from its inorganic and organic sources on performance and some parameters of egg quality. Ninety-six hens at 20 weeks of age were randomly allocated to 4 dietary treatments, each consisting of 6 replicates (4 birds per replicate). The control group was fed unsupplemented basal diet (BD) with only natural background Mn level of 46.4 mg/kg feed. For the three experimental treatments, the BD was supplemented with 120 mg Mn/kg either from Mn-sulphate or Mn-chelate of protein hydrolysate (Mn-Pro) or Mn-chelate of glycine hydrate (Mn-Gly). After 8 weeks of dietary treatments the egg production, egg weight, feed intake, and feed efficiency were not affected by dietary treatments. Regardless of the sources, Mn supplementation to feed resulted in significantly decreased percentages of cracked eggs compared to the unsupplemented control group. The thickness, weight, proportion, and index of eggshell were significantly elevated in all groups supplemented with Mn. The intake of Mn-Gly resulted in considerably increased Mn deposition in egg yolk compared to the control eggs. In the control and Mn-sulphate groups yolk malondialdehyde (MDA) started to increase after 20 and 30 days of egg storage respectively, whereas in eggs from hens given organic Mn-sources this parameter was not affected up to 40 days. Although there were no significant differences in MDA values between the treatments until 20 days of storage, the Mn-sulphate group showed significantly higher MDA concentration in yolks compared to the control group after 30 days of storage. These results demonstrate that supplementation of hens&rsquo; diet with Mn has positive effects on eggshell quality. Feed supplementation with Mn from organic sources appears to be more effective in preventing yolk lipid oxidation during cold storage of eggs than that from Mn-sulphate. &nbsp;

This work aimed to explore the effects of dietary supplementation of Chlorella vulgaris (CLV) on the growth performance, carcass traits, hematobiochemical variables, immunity responses, and the antioxidant status of growing rabbits. A total number of 100 rabbits were randomly distributed into four treatment groups, each of five replicates (25 rabbits/group). The experimental groups were as follows; control: a basal diet without supplementation, CLV0.5: basal diet + 0.5 g chlorella powder/kg diet; CLV1.0: basal diet + 1.0 g chlorella powder/kg diet, CLV1.5: basal diet + 1.5 g chlorella powder/kg diet. Live body weight (LBW), cumulative body weight gain (CBWG), feed intake (FI), and feed conversion ratio (FCR) were not affected by dietary CLV supplementation. Platelet count (PLT), hematocrit (HCT), means corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) values were significantly increased in the CLV0.5 group compared with the other treatment groups. Dietary supplementation of CLV (1.5 g/kg diet) significantly reduced the alanine aminotransferase (ALT) activity. The concentrations of serum triglycerides and very low-density lipoprotein (VLDL) were lower (p < 0.05) in the CLV-treated groups than those of the control. Supplemental CLV at all experimental levels gave the best values of immunoglobulins (IgG and IgM) and glutathione activities. Malondialdehyde (MDA) levels were lower in the animals that received CLV in their diet than those of the control group. Dietary supplementation of 1.0 g CLV/kg had the potential to enhance immune responses and antioxidant status, as well as reduce blood lipid accumulation. Therefore, it could be concluded that CLV supplementation to growing rabbit diets can improve the health status.
