This study aimed to investigate the flowering biological characteristics, floral organ characteristics, and pollen morphology of Camellia weiningensis Y.K. Li. These features of adult C. weiningensis plants were observed via light microscopy and scanning electron microscopy (SEM). Pollen viability and stigma receptivity were detected using 2,3,5-triphenyltetrazole chloride (TTC) staining and the benzidine–hydrogen peroxide reaction method. C. weiningensis is monoecious, with alternate leaves and glabrous branchlets. Its flowering period lasts 2 to 4 months, and the flowering time of individual plants lasts ≈50 days, with the peak flowering period from the end of February to the middle of March. It is a “centralized flowering” plant that attracts a large number of pollinators. Individual flowers are open for 12 to 13 days, mostly between 1230 and 1630 hr, and include four to six sepals, six to eight petals, ≈106 stamens, an outer ring of ≈24.6-mm-long stamens, an inner ring of ≈13.4-mm-long stamens, one pistil, and nine to 12 ovules. The flowers are light pink. The style is two- to three-lobed and 16.6 mm long, showing a curly “Y” shape. The contact surface of the style is covered with papillary cells and displays abundant secretory fluid and a full shape, facilitating pollen adhesion. The pollen is rhombohedral cone-shaped, and there are germ pores (tremoids). The groove of the germ pore is slender and extends to the two poles (nearly reaching the two poles). The pollen is spherical in equatorial view and trilobate in polar view. The pollen vitality was highest at the full flowering stage, and the stigma receptivity was greatest on days 2 to 3 of flowering. The best concentration of sucrose medium for pollen germination was 100 g/L. The number of pollen grains per anther was ≈2173, and the pollen-to-ovule ratio was 23,034:1. C. weiningensis is cross-pollinated. Seventy-two hours after cross-pollination, the pollen tube reached the base, and a small part entered the ovary. The time when the pollen tube reached the base after pollination was later than that in commonly grown Camellia oleifera. The results of this study might lay an important foundation for the flowering management, pollination time selection, and cross-breeding of C. weiningensis.

The secondary presentation of pollen consists of a foral mechanism where the presentation of pollen occurs in other foral structures in addition to the anther, in order to increase the precision of the dispersion of the pollen by the vectors. This study aims to describe the temporal dynamics of secondary pollen presentation, and morphological and morphometric characteristics in stages of pre- anthesis and anthesis in genotypes of fve natural Heliconia psittacorum populations. For the study of foral morphometry traits of length of the fower, stamen, stigma and height of flament insertion in the petal in bud and fower were measured. The foral morphology of pre-anthesis buds and fowers in anthesis, the presence or absence of characteristics such as herkogamy, region of flament insertion in the petal, region of stylar hairs, and of secondary pollen deposition were evaluated. Treatments of controlled pollinations, self-pollination, geitonogamy, cross-pollination, natural pollination and growth to pollen tube were sampled. Floral herkogamy and pollen transfer to the adhered hairs in the stylar region were clearly observed during anthesis, constituting the frst record of occurrence of secondary pollen presentation in Heliconiaceae. Pollen tube growth was inhibited in the stigmatic, style and basal regions of the pistil. Natural fruiting produced little or no fruit. The positioning of the stamens above the stigma, pollen viability and stigma receptivity during anthesis of H. psittacorum fowers may favor self-pollination. The stylar hairs observed in all H. psittacorum populations’ help in the retention of pollen grains. The low fruiting rate in controlled and natural pollinations suggest that the main propagation form of H. psittacorum in the study areas is based on asexual reproduction.

Cassava breeding is hampered by high flower abortion rates that prevent efficient recombination among promising clones. To better understand the factors causing flower abortion and propose strategies to overcome them, we 1) analyzed the reproductive barriers to intraspecific crossing, 2) evaluated pollen-pistil interactions to maximize hand pollination efficiency, and 3) identified the population structure of elite parental clones. From 2016 to 2018, the abortion and fertilization rates of 5,748 hand crossings involving 91 parents and 157 progenies were estimated. We used 16,300 single nucleotide polymorphism markers to study the parents’ population structure via discriminant analysis of principal components, and three clusters were identified. To test for male and female effects, we used a mixed model in which the environment (month and year) was fixed, while female and male (nested to female) were random effects. Regardless of the population structure, significant parental effects were identified for abortion and fertilization rates, suggesting the existence of reproductive barriers among certain cassava clones. Matching ability between cassava parents was significant for pollen grains that adhered to the stigma surface, germinated pollen grains, and the number of fertilized ovules. Non-additive genetic effects were important to the inheritance of these traits. Pollen viability and pollen-pistil interactions in cross- and self-pollination were also investigated to characterize pollen-stigma compatibility. Various events related to pollen tube growth dynamics indicated fertilization abnormalities. These abnormalities included the reticulated deposition of callose in the pollen tube, pollen tube growth cessation in a specific region of the stylet, and low pollen grain germination rate. Generally, pollen viability and stigma receptivity varied depending on the clone and flowering stage and were lost during flowering. This study provides novel insights into cassava reproduction that can assist in practical crossing and maximize the recombination of contrasting clones.

The investigations carried out in the years 2004-2005 covered five varieties of Narcissus (<i>Narcissus pseudonarcissus</i> L. x <i>Narcissus poëticus</i> L.): 'Fire Bird', 'Hardy', 'Ivory Yellow', 'Pomona' and 'The Sun'. Longevity of a flower and the flowering period of the studied Narcissus varieties determined, flower elements were compared, as well as the anatomic structure of ovary and nectary and the morphology of pollen grains. Pollen grains of 'Hardy' and 'The Sun' var. were observed in a scanning electron microscope. The flowering period of 'The Sun' plants appeared to be the longest, while the shortest in the case of 'Fire Bird' and 'Ivory Yellow'. Each perianth of the studied taxons was characterized by a different corolla appendage, from slightly corrugated ('Pomona') to greatly curled, with the notches getting inside and giving impression of a few whort in this part of the perianth ('Ivory Yellow', 'Hardy'). Monosulcate pollen grains with regards to 9 size was numbered among medium ('Pomona', 'The Sun') and large ('Fire Bird', 'Hardy', 'Ivory Yellow'). Their shape was assumed as circular elongated (<i>prolato-spheroides</i>) in polar view and flattened (<i>oblatum</i>) and slightly flat (<i>suboboblatum</i>) in equatorial view. The highest pollen vitality was shown by 'Hardy' variety (92%), whereas for 'The Sun' it proved to be the lowest (22%). In the narcissus flowers, septal nectaries are situated in the upper part of the ovary.

Abstract Adenium obesum (Forssk.) Roem. & Schult., popularly known as desert rose, has become a valuable ornamental plant. In floriculture, the production of hybrids is prioritized. Hence, knowledge on floral biology and sexual reproduction of the target species is fundamental. The objectives of this study were: (1) to test sucrose concentrations and temperatures for in vitro germination of A. obesum pollen grains; (2) to identify the effect of temperature on the viability of A. obesum pollen grains; and (3) to evaluate the viability of pollen grains and stigma receptivity in pre-anthesis, at flower opening, and 72 h post-flower opening in three accessions of A. obesum. A significant relationship between temperatures and sucrose concentrations was observed in the in vitro germination test. The highest percentage of in vitro germination of pollen grains, 39.81%, was observed at an estimated temperature of 26.05 °C. Desert rose accessions maintained in biochemical oxygen demand (BOD) chambers at 30 °C during a 16-h light photoperiod showed faster flowering, and temperatures ≥ 25 °C induced pollen grain viability percentages above 69%. Temperature is one of the most important abiotic factors, influencing mainly in pollen germination, pollen tube growing and in efficiency fertilization. The ICA-wd accession stood out and can be considered a pollen donor in artificial pollination. The stigmas of flowers were receptive from a day before flower opening until three days after. The two parameters presented above, stigma receptivity and pollen viability, allow inferences about the appropriate time for successful pollination and subsequent fertilization in desert roses.

The objective of this study was to evaluate the influence of two organic nanofertilizers, Lithovit and Nagro, on in vitro germination, pollen tube elongation and pollen grain viability of Pisum sativum L cv. Pleven 4. The effect of their application was high and exceeded data for the untreated control (44.2 and 47.23 % regarding pollen germination and pollen tube elongation, respectively), as well as the effect of the control organic algal fertilizer Biofa (17.5 and 27.9 %, respectively). Pollen grains were inoculated in four culture media. A medium containing 15% sucrose and 1% agar had the most stimulating impact on pea pollen grains. Pollen viability, evaluated by staining with 1% carmine, was within limits of 74.72-87.97%. The highest viability of pollen grains was demonstrated after the application of Nagro organic nano-fertlizer.

Pollen viability and stigma receptivity are prerequisites for successful cross-pollination and seed set in Swainsona formosa. In this study, the pollen viabilities and stigma receptivities was assayed by in vitro pollen germination and simple hand-pollination method on glasshouse-grown plants, respectively. The viability of pollen grains was tested under three different storage conditions: 1) pollen grains were left on the plant in the glasshouse under natural conditions, 2) pollen grains were harvested and kept at a low temperature (4 o C) in total darkness, and 3) pollen grains were kept in a dry freezer (-10 o C) and in total darkness. Meanwhile, stigma receptivity was determined by hand-pollination using fresh pollen grains on flower of glasshouse-grown plants at one day before anther dehiscence up to 8 days after anther dehiscence. The results showed that pollen grains could be stored at 4 o C for up to 28 days without significantly losing their viability. Pollen longevity could be extended beyond two months when stored at -10 o C and under dry conditions. These findings provided a simple and economically sound method for storage of S. formosa pollen. In addition, stigma receptivity was found to be receptive from one day before anther dehiscence and reached its peak within four days after anther dehiscence. These results provide a valuable background to the conventional breeding of this species to create hybrids through cross-pollination

The work presented here was aimed at investigating the proper time (after spreading) for viability assessment and the effect of various sucrose concentrations on pollen germination in Swainsona formosa.  The rate of pollen tube formation was determined for freshly shed pollen grains of glasshouse-grown plants at 10, 15, 20, 25, 30, 60 and 120-minute intervals after being plated on Brewbaker and Kwack (BK) medium.  The results indicated that within 60 minutes pollen germination reached 63.70%, after which remained steady at 120 minutes (63.71%).  Under the effect of various concentrations of sucrose, i.e. 0.5, 10, 15, and 20% (w/v), the germination rate of pollen grains was assessed at 60 minutes following germination.  The results showed that sucrose concentration of 10 – 15% (w/v) produced better germination rate (64.14%) compare to lower concentrations (19.64 and 43.58% at zero and 5% sucrose, respectively).  Sucrose concentration above 15% was also found to inhibit pollen germination (48.92% at sucrose concentration of 20%).

In order to investigate the cause of differences of mature pollen in Asian pear (Pyrus phyfolia) that are collected from various sources for the artificial pollination, various factors were measured as below: the composition of nonstructural carbohydrate in bud at 30 days after full bloom, the contents of crude protein in skin, cytosol and membrane, and the affinity for lectin (CON-A: Concanavalin, type III A) of glycoprotein in cytosol of pollen were measured. Contents of sucrose and glucose in buds influenced pollen germination rate and pollen tube growth, respectively. Therefore, soluble types of carbohydrates stored in bud were regarded as influencing on pollen germination rate and pollen tube growth. Pollen, which showed low activity, had low affinity on CON-A, lectin of glycoprotein, because it had fragile membrane, proteins in cells were denatured to pollen surface and certain enzymes concerned in pollen germination lost stability and activity. Pollens that showed high activity contained 92 kDa protein while others not. This was assumed as influencing on control of pollen viability.

The variability of viability (germination rate and the length of pollen tubes) of fresh pedunculate oak (Quercus robur L.) pollen grains was studied in vitro on a medium containing 15% sucrose. Spatial variability was studied by sampling fresh pollen grains from a total of thirteen trees at four different sites in the area of Belgrade (Kosutnjak, Banovo Brdo, Ada Ciganlija and Bojcin Forest) in a single year (2011). In order to assess temporal variability and determine the effects of climate change on a small time scale, we studied the viability of the pollen grains collected from one tree at the Banovo Brdo site in six different years (2004, 2005, 2006, 2007, 2011 and 2012). Interindividual variability was tested on the pollen grains sampled from eight trees at Ada Ciganlija in 2004. The percentage values of the pollen grain germination rate and the pollen tube length showed no statistically significant differences between the sites. However, the studied characteristics of the pollen grain viability (germination rate and pollen tube length) showed statistically significant differences in both temporal (between the pollen collection years) and interindividual variability. This type of research makes a valuable contribution to pedunculate oak breeding programs through the identification of trees with stable production and a good quality of pollen. Furthermore, it can be important in defining the patterns of spatial, temporal and individual variability of pollen grain viability under the influence of climate factors, which are showing compelling changing trends from year to year.

Floral fertility of five primocane-fruiting (PF) blackberry (Rubus L. subgenus Rubus Watson) genotypes (‘Prime-Jim’®, APF-31, ‘Prime-Ark® 45’, APF-59, and APF-77) and three floricane-fruiting (FF) genotypes (‘Navaho’, ‘Natchez’, and ‘Ouachita’) were tested under field conditions using floricane flowers with four pollination treatments: undisturbed open-pollinated, emasculated self-pollinated, emasculated and cross-pollinated with pollen from a similar fruiting type (PF × PF or FF × FF), and emasculated and cross-pollinated with pollen from a different fruiting type (PF × FF or FF × PF). During primocane flowering, three pollination treatments (undisturbed open-pollinated, emasculated selfed, and emasculated cross-pollinated) were used to further test the fertility of the five PF genotypes. Significant differences between cross-pollination treatments and self-pollination were seldom noted with more differences seen in ‘Prime-Jim’® than any other genotype. Cross-pollinating primocane flowers on ‘Prime-Jim’® resulted in significantly higher fruit set, drupelet set, and average berry weight compared with self-pollination. Fruit set among genotypes ranged from 68.5 to 96.7%, and drupelet set rating ranged from 4.3 to 6.9 for floricane flowers. For open-pollinated primocane flowers, fruit set ranged from 63.9 to 92.1%, and drupelet set rating ranged from 4.3 to 7.2. The genotypes APF-31, APF-59, and APF-77 showed a marked improvement over ‘Prime-Jim’® in both percent fruit set and drupelet set of floricane and primocane flowers. The results indicated that fertility appears to be sufficient in all the genotypes evaluated and that the later-generation PF genotypes show improvement in fertility over ‘Prime-Jim’®. Pollen viability (using both chemical viability testing and in vitro germination testing), stigma receptivity, and pollen tube growth (using florescence) were evaluated in a controlled environment to determine if any improvements could be noticed when comparing a selection of later-generation PF genotypes (APF-31,' Prime-Ark® 45', APF-59, and APF-77) with ‘Prime-Jim’® and ‘Prime-Jan’®. Genotypes APF-31, APF-59, and APF-77 had significantly more viable pollen and pollen germination than ‘Prime-Jan’®. Stigma receptivity was observed in all genotypes. Pollen tube growth did not appear to be inhibited after self-pollination in any genotype studied, although significantly greater florescence resulting from pollen tube growth was seen after cross-pollination compared with self-pollination for APF-59. There were no significant differences in pollen tube growth between cross- and self-pollination for any other genotype.
