Commercial probiotics have bacteria which offer scope for specific treatment as a probiotic against diarrhoea, boosting immune response and relieving stress. Research in probiotics and aquaculture has been done to promote sustainable aquaculture. Studies on microbial ecology in aquaculture and their benefits on human systems are to be assessed. Hence, the present study was aimed at the isolation and characterization of these bacteria obtained from various commercial probiotics. The commercial probiotic samples were collected for isolation using MRS agar. The colonies were selected on the basis of colonial morphology.  Isolates were put through cell morphology, physiology and different biochemical tests. Probiotic sample confirmation was done using 16srRNA molecular technique. MEGA7 was used to conduct phylogenetic evolutionary analysis and tabulate a distance matrix. In results, the isolates manifested non-identical growth patterns at dissimilar conc. of NaCl (2.0,4.0 and 6.5) oxygen and at different temperatures (150C, 300C and 450C). On the basis of sugar utilization, physiological testing, biochemical characterization and genetic identification tests, all isolates were established to different species of Lactobacillus rhamnosus, Pediococcus acidilactici and Lactobacillus Plantarum. A systematic protocol was done to identify, characterize probiotic samples and identify them by genetic analysis. Probiotic use is carefully assessed by regulations

Vibrio parahaemolyticus is the most common species among crustaceans, often causing various diseases and significant losses in aquaculture. Acute hepatopancreatic necrosis disease (AHPND) is a newly emerging shrimp disease that has severely damaged the global shrimp industry. This species of bacteria is associated with gastrointestinal illness in humans and has been implicated in foodborne disease. The present study carried out, isolation and characterization of pathogenic bacterial flora isolated from the infected hepatopancreas of vannamei, obtained from various aquafarms in Andhra Pradesh, India, on 11th June 2018. The collected samples were plated on TCBS- (Thiosulfate-Citrate-Bile salt-Sucrose) agar medium and Hi -Chrome vibrio, as described in Bergey's manual of systematic bacteriology. Isolated colonies were subjected to the following tests- microscopic examination, growth at different temperatures, growth at different NaCl concentrations, and biochemical tests. Further purity, maintenance, and propagation of purified cultures were done. The microbial culture was identified using 16s rRNA molecular technique. Phylogenetic Evolutionary analyses and distance matrix were conducted in MEGA7.In the present study, different samples were screened, a total of three green colonies (V44, V45, V46) were isolated, identified by biochemical tests and genetic identification as Vibrio parahaemolyticus. A systematic methodology has been developed to isolate and characterize Vibrio sp. from diseased shrimp and identify them by genetic analysis

Modulation of the gut microbiota through the use of probiotics has been widely used to treat or prevent several intestinal diseases. However, inconsistent results have compromised the efficacy of this approach, especially in severe conditions such as inflammatory bowel disease (IBD). The purpose of our study was to develop a personalized probiotic strategy and assess its efficacy in a murine model of intestinal inflammation. Commensal bacterial strains were isolated from the feces of healthy mice and then administered back to the host as a personalized treatment in dextran sodium sulfate (DSS)-induced colitis. Colonic tissues were collected for histological analysis and to investigate inflammatory markers such as Il-1β, Il-6, TGF-β, and Il-10, and the enzyme myeloperoxidase as a neutrophil marker. The group that received the personalized probiotic showed reduced susceptibility to DSS-colitis as compared to a commercial probiotic. This protection was characterized by a lower disease activity index and reduced histopathological damage in the colon. Moreover, the personalized probiotic was more effective in modulating the host immune response, leading to decreased Il-1β and Il-6 and increased TGF-β and Il-10 expression. In conclusion, our study suggests that personalized probiotics may possess an advantage over commercial probiotics in treating dysbiotic-related conditions, possibly because they are derived directly from the host’s own microbiota.

Vibrio alginolyticus has been thought to be a halophilic marine bacterium that causes diarrhea, otitis media and wound infection through the consumption of raw or inappropriately cooked sea food. It is one of the main Vibrio pathogens affecting marine animals, such as marine fish, shrimp and shellfish which lead to large economic damage. Although there are reports on the presence of this organism in the coastal area of other countries, not so much work has been done on the isolation and characterization of this species in Bangladesh. The present study was, therefore, undertaken to isolate and characterize V. alginolyticus organisms isolated from the rivers (fresh water) and estuaries (brackish water) of Bangladesh. A total of 9 isolates of Vibrio species were obtained from different water bodies (three from Meghna river, two from Shangu river and four from estuary) and provisionally identified as Vibrio alginolyticus following standard biochemical tests. All these 9 strains showed same pattern of antibiotic resistance to ampicillin, streotomycin, penicillin, but sensitive to nalidixic acid. In the virulence properties test, two isolates showed positive results for toxR gene and none of the isolates showed positive results for tdh gene. Challenge experiments in Singhi fish (Heteropneustes fossi) with the live cells and the culture filtrate prepared from the V. alginolyticus showed high mortality of the fish population. All these studies suggest the presence of pathogenic V. alginolyticus strains in the river water and estuarine bodies of Bangladesh and the extracellular toxin(s) of the V. alginolyticus might be one of the causes for fish mortality.Bangladesh J Microbiol, Volume 29, Number 1, June 2012, pp 1-6

Marine environment remained as largely unexplored source for researchers to discover marine microorganisms with novel properties. This study aims to isolate marine bacteria from the seashore of Desaru, Malaysia. Totally, six bacterial strains were successfully obtained and were identified by complete 16S rRNA sequencing. The characterizations of bacterial strains were performed based on morphological tests, Gram-staining, biochemical tests, and antibiotic sensitivity. The 16S rRNA sequence of D-2, D-4, D-7, D-15, D-31, and D-33 revealed a high identity of 97 to 99% with taxa belong to genera of Pseudomonas, Marinomonas, Exiquobacterium, Micrococcus, Pseudoalteromonas, and Shewanella respectively. Strain D-31 exhibited higher tolerance towards antibiotics kanamycin, ampicillin, and erythromycin while the growth of other strains were retarded by at least two of these antibiotics. We further characterized strain D-4 and D-31 that belonged to Marinomonas sp. and Pseudoalteromonas sp.. Both genera are interesting as earlier researchers have discovered new antibacterial substances, industrial enzymes and unique secondary metabolites.


Certain microorganisms especially bacteria and fungi are able to use xenobiotic organic compounds as their carbon and nitrogen source for metabolism. Flower farms around lake Naivasha basin uses several agrochemicals especially pesticides to control pests and improve flower production. The aim of this study was to isolate and characterize morphologically and biochemically the main bacterial species that are able to grow and tolerate the pesticide contaminated farm soils. Soil samples were collected from randomly selected five greenhouses from each five flower farms namely Crescent, Elsamere, Karuturi, Malewa and Sewage farms around Lake Naivasha basin. The collected samples were processed for bacterial isolation using the nutrient agar, mac’ Conkey agar, blood agar, Luria-Bertani and Minimum Salt Media nutrient media. The conventional methods of swabbing and streaking were used. Pure colonies of isolates organisms were identified and characterized using standard microbiological technique. Morphological, cultural and biochemical characterization of bacterial species isolated from the flower farm soil samples identified mainly Pseudomonas auriginosa, Escherichia coli, Rhodococcus erythropolis and Bacillus subtilis species. Bacterial growth in pesticide consortia was quantified by monitoring colony growth of the species in liquid culture over time. The viable cell counts were determined turbidimetrically at O.D696nm. All the isolated bacterial species were able to grow in flower farm soil contaminated with organochloride and organophosphate pesticide residues. B. subtilis recorded the highest growth at 1.77±0.07 O.D696nm in pesticide mixture consortia. There was lower growth in organochloride pesticide consortia as compared to organophosphate pesticide consortia.

Photorhabdus bacteria are symbiotically associated with the infective juveniles of entomopathogenic nematodes of the genus Heterorhabditis. These bacteria after infecting a susceptible host, produces a host of lethal toxins that killthe insect hosts within 24 to 72 hours. Consequently they have emerged and are trending as excellent biological control agents against insect pests of agricultural crops. Therefore, this study aim to isolate, identify and test the pathogenicity of Photorhabdus bacteria isolated from Heterorhabditis bacteriophora, entomopathogenic nematode from Kashere, Gombe State, Nigeria. To this end phenotypic and biochemical tests were conducted. The tests conducted showed that the isolate exhibited characteristics similar to those of Photorhabdus bacteria that have been identified. Last instar larvae of G. mellonella exposed to different concentrations of the bacterial isolate showed some level of susceptibility of the larvae to the bacterial isolate confirming its biological control potential.

ResumenEl síndrome nefrótico  se diagnóstica con base en cuatro criterios: proteinuria, hipoalbuminemia, edema e hipercolesterolemia. Generalmente se presenta entre los 2 y 10 años de edad. Este trabajo se realizó para determinar la caracterización bioquímica del síndrome nefrótico de la población infantil del Hospital del Niño de Villahermosa, Tabasco, México.Estudio descriptivo, transversal, retrospectivo realizado en el laboratorio clínico del Hospital del Niño Dr. Rodolfo Nieto Padrón de enero 2016 a diciembre 2017. Se obtuvo el rango y promedio de las pruebas bioquímicas de pacientes con síndrome nefrótico.Se analizaron 33 pacientes referidos con  diagnóstico de síndrome nefrótico,  encontrándose una media de albumina de 1.3g/dl, colesterol de 482.6 mg/dl y triglicéridos de 523.2 mg/d y una proteinuria media de 295.9 mg/dll. Palabras clave: Síndrome nefrótico,  Hipoalbuminemia, Hipercolesterolemia.   AbstractThe nephrotic syndrome is diagnosed based on four criteria: proteinuria, hypoalbuminemia, edema and hypercholesterolemia. Abstract The aim of this study was to isolate and characterize the Phosphate solubilizing bacteria from the rhizosphere of Zea mays L., Jeneponto Regency. This research was conducted in several stages; i.e, sampling, medium preparation, sample dilution, isolation, characterization in the form of gram staining, biochemical tests, and quantitative tests of phosphate solubility. Soil samples were diluted in 0.9% NaCl and soil containing microbes was isolated on the Picovskaya medium. Three isolates were obtained which could dissolve phosphate, namely J2KN1, J3KR2, and J3TG3 isolates. The isolates were generally round in shape with raised elevations, white, slimy, smooth, shiny surface, milky white, shape like coccus and bacillus, and gram-negative. Some of the isolates had positive motility, indole, voges, methyl red, glucose, and sucrose fermentation in the biochemical test. The quantitative tests of the ability to dissolve phosphate showed that J2KN1 isolate had the highest concentration of 51.1 μM, and the J3KR1 and J3TG3 isolates had a concentration of 45.2 μM and 37.6 μM, respectively.

The genus Nocardiopsis is well known to produce secondary metabolites especially antibacterial bioactive compound. Isolation and characterization of bioactive compounds producing novel isolates from unusual habitats are crucial. The present study was aimed to explore Didwana dry salt lake of Rajasthan state in India for the isolation and characterization of actinomycetes. The isolated actinomycetes isolates were characterized based on culture characteristics, biochemical tests and 16S rRNA gene sequencing. The 16S rRNA gene sequence analysis revealed that all the five isolates inhabiting soil of the said dry salt lake of Didwana, Rajasthan belonged to four species of Nocardiopsis viz., N. synnemataformans, N. potens, N. prasina and N. dassonvillei subsp. albirubida. The molecular identification based on 16S rRNA gene sequences was found accurate and robust. The phylogram generated through multiple sequence alignment of all the test isolates of Nocardiopsis revealed that the isolates aroused from a single branch and validated monophyletic association. The present study is the first report of exploring Nocardiopsis isolates from the dry salt lake. These characterized Nocardiopsis isolates isolated from Didwana dry salt lake habitat are novel stains and can be of significance in the detection and utilization of novel bioactive compounds.

ABSTRACT The isolation and characterization of mutants altered for nitrate assimilation in Neurospora crassa is described, The mutants isolated can be subdivided into five classes on the basis of growth tests that correspond to the growth patterns of existing mutants at six distinct loci. Mutants with growth characteristics like those of nit-2, nit-3 and nit-6 are assigned to those loci on the basis of noncomplementation and lack of recombination. Mutants that, from their growth patterns, appear to lack the molybdenum-containing cofactor for both nitrate reductase and xanthine dehydrogenase subdivide into three loci (nit-7, nit4 and nit-9), all of which are genetically distinct from nit-1. nit-9 is a complex locus consisting of three complementation groups and thus appears similar to the cnxABC locus of Asperillus nidulans. Extensive complementational and recombinational analyses reveal that nit-4 and nit-5 are alleles of the same locus, and two new alleles of that locus have been isolated. The results indicate that, as in A. nidulans, nitrate assimilation in N. crassa requires at least four loci (nit-1,7,8 and 9) to produce the molybdenum co-factor for nitrate reductase (and xanthine dehydrogenase), one locus (nit-3) to code for the nitrate reductase apoprotein, one locus (nit-6) to code for the nitrite reductase approtein and only one locus (nit-4/5) for the regulation of induction of the pathway by nitrate and nitrite.
