31 year old female presented with abdominal pain and respiratory distress in the third trimester of her second pregnancy. Her blood workup revealed a lipemic sample (Figure 1) due to markedly elevated serum triglycerides of 8178 mg/dl (Glycerol Phosphate Oxidase method). Total cholesterol and Low Density Lipoprotein were elevated at 1701 mg/dl and 788 mg/dl respectively. There was no family history of lipid disorders. Diagnosis was consistent with gestational hypertriglyceridemia with acute pancreatitis (Serum Amylase-50 U/L, Serum Lipase- 96 U/L), though genetic tests to rule out pre-existing primary hypertriglyceridemia was not feasible. In view of the life threatening condition, she was initiated on Insulin-Dextrose infusion and offered one session of Plasma Exchange. Figures 2,3 depict membrane plasma separation with the obtained effluent as lipemic plasma. Her serum triglycerides showed a declining trend and was discharged in good health (serum triglycerides at discharge-651 mg/dl).

Abstract Background Exosome-derived microRNAs (exo-miRs) as messengers play important roles, in the cross-talk between genetic [ATP-sensitive potassium channels (KATP) genetic variant rs1799858] and environmental [elevated serum low-density lipoprotein cholesterol (LDL-C) level] factors, but the plasma exo-miRs expression profile and its role in biological processes from genotype to phenotype remain unclear. Methods A total of 14 subjects with increased LDL-C serum levels (≥ 1.8 mmol/L) were enrolled in the study. The KATP rs1799858 was genotyped by the Sequenom MassARRAY system. The plasma exo-miRs expression profile was identified by next-generation sequencing. Results 64 exo-miRs were significantly differentially expressed (DE), among which 44 exo-miRs were up-regulated and 20 exo-miRs were down-regulated in those subjects carrying T-allele (TT + CT) of rs1799858 compared to those carrying CC genotype. The top 20 up-regulated DE-exo-miRs were miR-378 family, miR-320 family, miR-208 family, miR-483-5p, miR-22-3p, miR-490-3p, miR-6515-5p, miR-31-5p, miR-210-3p, miR-17-3p, miR-6807-5p, miR-497-5p, miR-33a-5p, miR-3611 and miR-126-5p. The top 20 down-regulated DE-exo-miRs were let-7 family, miR-221/222 family, miR-619-5p, miR-6780a-5p, miR-641, miR-200a-5p, miR-581, miR-605-3p, miR-548ar-3p, miR-135a-3p, miR-451b, miR-509-3-5p, miR-4664-3p and miR-224-5p. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were subsequently implemented to identify the top 10 DE-exo-miRs related specific target genes and signaling pathways. Only 5 DE-exo-miRs were validated by qRT-PCR as follows: miR-31-5p, miR-378d, miR-619-5p, miR-320a-3p and let-7a-5p (all P < 0.05). Conclusion These results firstly indicated the plasma exo-miRs expression profile bridging the link between genotype (KATP rs1799858) and phenotype (higher LDL-C serum level), these 5 DE-exo-miRs may be potential target intermediates for molecular intervention points.

Abstract Objectives This paper investigates the prevalence of hypertriglyceridemic waist in bank workers and its association with socioeconomic, labor, behavioral, anthropometric and health condition factors. Methods This is a cross-sectional study based on information from 525 bank workers. Hypertriglyceridemic waist was defined by the simultaneous presence of abdominal obesity, verified by the waist circumference, and elevated serum triglycerides. Associations were verified with sociodemographic, labor, behavioral, anthropometric and health condition variables. Results The investigation resulted in a phenotype prevalence of 19.4%, being higher in men, elderly, married and working in the bank for more than 5 years. The phenotype was also associated with overweight, low HDL-c (high-density lipoprotein), mixed hyperlipidemia, high triglyceride/HDL-c ratio and arterial hypertension. Being over 50 years of age and being overweight increased the chances of the bankers presenting the phenotype. Being female and having adequate levels of HDL-c were shown to be protective factors against the phenotype. Conclusions It was concluded that the prevalence of hypertriglyceridemic waist is high and is associated mainly with the excess weight and unfavorable lipid profile of this population. <p>The study was designed to evaluate the hepatoprotective activity of aqueous extract of cardamom in acute experimental liver injury induced by gentamicin. Twenty four male albino rats were randomly divided into four groups (six rats in each). Animals of the first group served as control and orally (p.o.) received (1 ml/kg saline). The second experimental group was given gentamicin (80 mg/kg i.p.) for 7 days. Third and fourth groups were given aqueous extract of cardamom (100 and 200 mg/kg p.o.) + gentamicin for 7 days, respectively. The degree of hepatoprotection was measured using serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), bilirubin, albumin, and lipid profile levels. In the acute liver damage induced by gentamicin, cardamom aqueous extracts (100 and 200 mg/kg, p.o.) significantly reduced the elevated serum levels of AST, ALT, bilirubin, cholesterol, triglycerides and low density lipoprotein cholesterol (LDL-chol) in gentamicin induced hepatotoxicity. Also cardamom aqueous extracts (100 &amp; 200 mg/kg, p.o.) significantly increased the lowered serum levels of albumin and high density lipoprotein cholesterol (HDL-chol) in gentamicin induced hepatotoxicity rats. Histopathological examination of the liver tissues supported the hepatoprotection. Our findings concluded that cardamom aqueous extracts possessed hepatoprotective activity against gentamicin induced hepatotoxicity in rats.</p>

The present study was intended to evaluate the effects of Paeonia emodi rhizome extracts on serum triglycerides (TGs), total cholesterol (TC), low density lipoprotein cholesterol (LDL-c), high density lipoprotein cholesterol (HDL-c), atherogenic index (AI), superoxide dismutase (SOD), and glutathione peroxidase (GPx). The plant was extensively examined for its in vitro antioxidant activity, and the preliminary phytochemical screening was carried out using standard protocols. Male Wistar rats were induced with hyperlipidemia using high-fat diet and were treated orally with hydroalcoholic and aqueous extracts at the dose of 200 mg/kg bw for 30 days. TGs, TC, LDL-c, and AI were significantly reduced while HDL-c, SOD, and GPx levels rose to a considerable extent. After subjecting to acute toxicity testing, the extracts were found to be safe. The observations suggest antihyperlipidemic and antioxidant potential of P. emodi in high-fat diet induced hyperlipidemic/oxidative stressed rats.

As part of the Dutch Nutrition Surveillance System, cardiovascular risk factors and food consumption (24 h recall) as well as haematological, Fe and vitamin status (A, B6, C) were assessed in 126 Dutch boys aged 10–11 years (response 71 %). Body mass index (BMI) and the sum of four skinfolds were strongly associated (r0.85,P< 0.01) and 8% of the boys were overweight (BMI > 20.1 kg/m2). Elevated serum total cholesterol levels (> 4.4 mmol/l) were observed in 38 %; total cholesterol and low-density-lipoprotein-cholesterol levels were strongly associated (r0.88,P< 0.001). Intake of fat was high (38 % of energy) and too much fat was saturated (polyunsaturated: saturated 0.44, guideline: 0.5–1.0), whereas intake of carbohydrate (49% of energy) and dietary fibre was low. About 12% of the boys had insufficient Fe stores (plasma ferritin < 12.0 μg/l) and the mean Fe intake (9.0 mg/d) was below recommended daily allowance (10.0 mg/d). Plasma ferritin was, however, not associated with haematological indices and no frank anaemias were observed. No marginal values were observed for vitamins A, B6and C status. In conclusion, the main nutritional risks in boys aged 10–11 years are cardiovascular risk factors and Fe nutrition.

Atherosclerosis, the underlying cause of cardiovascular disease, is characterized by chronic inflammation and altered immune response. Cholesterol is a well-known risk factor associated with the development of cardiovascular diseases. Elevated serum cholesterol is unique because it can lead to development of atherosclerosis in animals and humans even in the absence of other risk factors. Modifications of low-density lipoproteins mediated by oxidation, enzymatic degradation, and aggregation result in changes in their function and activate both innate and adaptive immune system. Oxidized low-density lipoprotein (LDL) has been identified as one of the most important autoantigens in atherosclerosis. This escape from self-tolerance is dependent on the formation of oxidized phospholipids. The emerging understanding of the importance of immune responses against oxidized LDL in atherosclerosis has focused attention on the possibility of development of novel therapy for atherosclerosis. This review provides an overview of immune response to lipoproteins and the fascinating possibility of developing an immunomodulatory therapy for atherosclerosis.

Berberine is a plant alkaloid that has several pharmacological effects such as antioxidant, antilipidemic, and anti-inflammatory effects. Nonalcoholic steatohepatitis (NASH) triggers different aspects of disorders such as impaired endogenous lipid metabolism, hypercholesterolemia, oxidative stress, and neurotoxicity. In this study, we examined the mechanism by which NASH induces neurotoxicity and the protective effect of berberine against both NASH and its associated neurotoxicity. NASH induced rats showed significant impairments in lipid metabolism with increased serum triglycerides, cholesterol, and low-density lipoprotein (LDL). The NASH induced group also demonstrated a significant oxidative stress which is characterized by increased TBARs level and decreased antioxidant capacity such as GSH and SOD levels. Moreover, the NASH induction was associated with inflammation which was demonstrated by increased TNFαand nitric oxide levels. Hyperglycemia and hyperinsulinemia were observed in the NASH induced group. Also, our results showed a significant increase in the expression of the acetylcholine esterase (AChE) and amyloid beta precursor protein (AβPP). These changes were significantly correlated with decreased insulin degrading enzyme (IDE) and beta-amyloid40(Aβ40) and increased beta-amyloid42(Aβ42) in the hippocampal region. Daily administration of berberine (50 mg/kg) for three weeks ameliorated oxidative stress, inflammation, hyperlipidemia, hyperglycemia, hyperinsulinemia, and the observed neurotoxicity.
