The oral route is the best and most popular route for the administration of drugs in the systemic circulation. There are number of drugs which are given through the oral route. Gastro-retentive drug delivery system is very important system for the drug delivery system. The gastro-retentive drugs prolonged the drug time in the git and also improve their their bioavailability. These are widely used for site specific for the treatment of git disorders and diseases. There are number of approaches for gastro retentive drug delivery system such as floating system, mucoadhesive system, swelling system, high density system etc. In this review we discussed about approaches and various perspectives of gastro retentive drug delivery system.

Oral route is most preferable and widely used route for the administration of drug. Microballoons becomes novel technology in pharmaceutical field in the floating drug delivery for achieving the gastric retention. Microballoons are also called as hollowspheres which are porous smooth in nature and thus show good floating properties in gastric fluid. Microballoons release the drug in controlled manner at the targeted site. Microballoons are spherical empty vesicles without core and that can remain buoyant in gastric region for prolong period of time without irritation in gastrointestinal tract. Multiparticulate particles having a low density system that can efficiently prolong the gastric retention time of the drugs, thus enhanced bioavailability and thus improve the dosing frequency. These are less soluble at higher pH environment. As microballoons delivery systems provide longer retention in gastric pH and enhance the solubility of drugs that are less soluble in high pH environment. The formation of cavity inside the microballoons depend on the preparation, temperature and the surface smoothness determine the floatability and the release rate of microballoons. Background:: The general purpose of this review was to briefly describe the gastro-retentive drug delivery system (GRDDS) and primary focuses on mechanisms of flotation for gastric retention. The principle mechanism of floating drug delivery systems (FDDS) is mainly based on hydro-dynamically controlled low-density system which gets swollen when come in contact with gastric environment and remain buoyant for extended period of time with improved bioavailability and therapeutic efficacy. The main perspective of this review was to focus on microbubble drug delivery due to its wide range of application. Microbubble is a new, economically viable and non-invasive technique which is alternative to conventional techniques. The size of microbubble ranges from 1-100 micrometers which contain oxygen or air and remain suspended for prolonged drug release. Due to its great potential microbubble is used in therapeutic delivery of drug molecule or genetic material at their specific targeted site in various diseases. The recent research also suggests that ultrasound mediated microbubble is more frequently used in biomedical field for imaging of site-specific molecule targeting for detection of disease and site-specific drug delivery. Conclusion:: Gastro-retentive floating drug delivery system was considered as most favorable approach for site-specific drug delivery. But nowadays, microbubble is gaining more popularity for its potential application in biomedical field. In recent years, there is much progress in microbubble for site specific delivery of drug or genetic material. Recent research also suggests that the ultrasound mediated microbubble have shown positive impact in the diagnosis and clinical management of various diseases such as Liver Cirrhosis, brain tumor, ocular and urinary tract infection.

Oral controlled release and site specific drug delivery system has been of great interest in pharmaceutical field to achieve improved therapeutic advantage. Concept of novel drug delivery system arose to overcome certain aspect related to physicochemical properties of drug molecule and the related formulations. Gastro retentive drug delivery system is one of such novel approaches to prolong gastric residence time, thereby targeting site specific drug release in the stomach for local or systemic effects. This approach is useful particularly for the drugs which have narrow absorption window in the upper part of gastro intestinal tract. In this review we have been discussed various approaches of gastro retentive drug delivery system, such as floating and non-floating systems.

One novel approach in this area is GRDDSs (Gastro Retentive Drug Delivery System). GRDDSs can improve the controlled delivery of drugs that have an absorption window by continuously releasing the drug for a prolonged period of time before it reaches its absorption site.The purpose of writing this review was to investigate, compile and present the recent as well as past literatures in more concise way with special focus on approaches which are currently utilized in the prolongation of gastric residence time. These includes floating system, swelling and expanding system, bio/mucoadhesive system, high density system and other delayed gastric emptying devices. The present review addresses briefly about the classification, formulation consideration for GRDDS, factors controlling gastric retention, merits, demerits and applications of gastroretentive drug delivery systems.

In the recent most useful route of drug delivery system is the oral route in all types of patient. Mouth dissolving strips is the type of drug delivery system which when placed in the oral cavity disintegrates or dissolves within few seconds without intake of water. The advantages of mouth dissolving strips are the administration to pediatric and geriatric patient. This technology has been used for local action, rapid release products. These films have a potential to deliver the drug systemically through intragastric, sublingual or buccal route of and also been used for local action. The mouth dissolving strips are available in various administration shapes, size and thickness. Many companies are utilizing transdermal drug delivery technology to develop thin film formats. This article is an overview of mouth dissolving strips encompassing materials used in mouth dissolving strips, manufacturing process, application, commercial technologies and future business prospects of this technology. Writing the review on gastro retentive drug delivery systems (GRDDS) was to start up the current literature with a special consequence on several gastro retentive approaches that have become main mode in the field of site-specific orally conduct sustained/controlled release drug delivery. Multi-furious ways have been made in research and development to rate-controlled oral drug delivery systems to solve physiological difficulties, like short gastric residence times (GRT) and unpredictable gastric emptying times (GET). GRDDS is a tool to prolong the GRT, thereby targeting site-specific drug release in the upper gastrointestinal tract (GIT) for local or systemic effect. Oral dosage forms as low bioavailability issues because of their swift gastric transition from the stomach, particularly in the case of drugs that are less soluble at an alkaline pH of the intestine. The drugs that produce their local action in the stomach get quickly emptied and don’t get enough residence time in the stomach. Many efforts have been made to extend the retention time of a drug delivery system to reduce the frequency of dose administration. GRDDS not only prolong dosing intervals but also increase patient compliance beyond the level of existing controlled release dosage forms. This article gives an overview of the advantages, disadvantages, and characterization of gastro retentive drug delivery systems. This also includes commercially available gastro retentive products and patents.

Novel drug delivery systems are becoming increasingly sophisticated as pharmaceutical scientists acquire a better understanding of the physicochemical and bio-chemical parameters pertinent to their performance. Despite tremendous advancements in drug delivery, the oral route remains the perfect route for the administration. Novel drug delivery system assists to achieve better patient compliance. Fast dissolving tablets are one of them.FDT have benefits such as accurate dosing, easy portability and manufacturing, good physical and chemical stability and an ideal alternative for pediatric and geriatric patients. FDDT formulation combines the advantage of both liquid and conventional tablet formulation while also offering advantage over both traditional dosage forms. Some tablets are designed to dissolve in saliva remarkably fast, within a few seconds, and are true fast- dissolving tablets. Others contain agents to enhance the rate of tablet disintegration in the oral cavity, and are more appropriately termed fast-disintegrating tablets, as they may take up to a minute to completely disintegrate.
