Physiochemical properties and antioxidant potential of Persea Americana seed oil

Oil extracted from Persea Americana seed was assayed for its physiochemical properties and antioxidant potential using various standard methods. The oil content of the seed was found to be < 10%. Brownish-red color oil was liquid at room temperature, with specific gravity of 0.91±0.02 g/mL. Other physiochemical parameters determined were; acid value (4.51±0.08 mgKOH/g), %FFA (2.26±0.08), peroxide value (2.40±0.57 mgO2/Kg), ester value (31.26±0.03 mgKOH/g), saponification value (35.76±0.07 mgKOH/g) and iodine value (23.5±0.07). The results of the antioxidant activities of the seed oil showed that the flavonoid content (80.00±1.41 mgQE/g) was ~10 folds higher than the phenolic content (8.27±0.06 mgGAE/g). The DPPH radical scavenging value was found to be 51.54±0.25% with an IC50 value of 4.68±0.02 mg/mL and reducing power with an average absorbance of 0.85±0.01 and an IC50 value of 0.001±0.02 mg/mL. Gallic acid showed better antioxidant activities than the oil studied. The results obtained in this study showed that Persea Americana seed oil has nutritional, industrial as well as medicinal potentials.

Antioxidant potential of the 80% methanol extract of the leaves of Opuntia dillenii was evaluated using 1, 1-diphenyl, 2-picryl hydrazyl (DPPH) and nitric oxide radical scavenging, reducing power, total phenol and total flavonoid content determination assays. Preliminary phytochemical screening of the extract was also carried out, which revealed that the extract possesses flavonoids, steroids, alkaloids and tannins. The extract showed significant antioxidant activities in all antioxidant assays compared to the reference antioxidant in a dose dependent manner. In DPPH radical scavenging activity, the IC50 value of the crude extract was found to be 15.71μg/mL while the IC50 value for the reference ascorbic acid was 10.84 μg/mL. Again, the extract showed remarkable nitric oxide scavenging potential and good reducing power. Moreover, the methanol extract was found to contain high amount of phenols and flavonoids, expressed as gallic acid and rutin equivalents respectively. Based on the findings of the present study, we conclude that the methanol extract of the leaves of O. dillenii possesses remarkable antioxidant potential which may be attributed to the high amount of phenols and flavonoids present in the extract. AbstractPresent study describes rapid in vitro propagation of Caralluma tuberculata, a traditional medicinal plant, and antioxidant potential of calli and plants extracts. The highest callus induction rate (93.3%) with maximum weight of calli 5.2 g was achieved from shoot tip explants on MS medium supplemented with 9.04 μM 2,4-D and 4.44 μM BA. The maximum shoot induction rate (71.1%) with mean number of shoots 3.66 ± 1.53 and 4.6 cm average shoot length was observed on 13.32 μM BA, 4.52 μM 2,4-D and 2.89 μM GA3 appended in MS medium. The developed shoots were best rooted in the presence of 5.07 μM IAA with 3.0 ± 0.15 roots per plantlet. The plants were successfully acclimatized under in vivo conditions. The plants and calli extracts exhibited good antioxidant activities, however, plant extract activities were more pronounced. The phenolic compounds in plant and calli extracts were 0.16% and 0.057%, respectively. While the flavonoids were 0.092% in plant and 0.039% in calli extract. Total Phenolics, flavonoids; DPPH radical scavenging activity and reducing power potential distributed among different fractions depending upon polarity of the solvent. The highest DPPH scavenging activity and reducing power was exhibited by water fractions; 4.95 mg/mL and 0.729 OD at 10 mg/mL, respectively. The micropropagation protocol can be successfully used for large-scale multiplication and conservation of germplasm of this threatened plant. Furthermore, antioxidant value describes importance of this valuable plant as food and medicine.

In this study, antioxidant activities of H. fursei were investigated. Different model were used for assay. IC50 for DPPH radical-scavenging activity was 7.5±1.2 μg ml-1. The IC50 value for BHA was 92.9±4.5 µg ml-1. The extract showed very good reducing power that was comparable with vitamin C (p> 0.05) but showed weak nitric oxide scavenging activity and Fe2+ chelating ability. Extract showed good activity in scavenging of H2O2. Inhibition was 99.1% at 800 μg ml-1. The IC50 values for extract and BHA were 333.1±14.7 and 52.0±4.5 μg ml-1, respectively. Total phenol compounds, by the Folin Ciocalteu method, was 274±9.6 mg gallic acid equivalent/g of extract and the total flavonoid content, by AlCl3 method was 23.9 ± 1.4 mg quercetin equivalent/g of extract. Antioxidant activity may be attributed to the presence of phenols and flavonoids in the extract.

Mung bean (Vigna radiata L.) sprout is a popular fresh vegetable, tasty and high in antioxidants. To increase yield and quality after the occurrence of both abiotic and biotic stresses, the application of seaweed extracts is of great importance. Hence, this study was conducted to determine the effect of Ascophyllum nodosum extract (ANE) in the presence of salt on the antioxidant potential of V. radiata sprouts. Different concentrations of ANE viz. 0.00, 0.01, 0.05, 0.10, and 0.50% and NaCl 0, 25, 50, 75, and 100 mM alone and in combinations were tested for researching the antioxidant potential of V. radiata sprouts at 0, 24, and 36 h of sprouting. The DPPH free-radical-scavenging activity of sprouts of V. radiata was found to increase with time and peaked at 24 h of treatment. The A. nodosum extract (0.01%) could reverse the ill effect of the low level of salinity posed by up to 25 mM NaCl. The increasing salinity deteriorated the antioxidant activity using ABTS method of sprouts down to 20.45% of the control at 100 mM NaCl. The total phenolic content (TPC), total flavonoid content (TFC), and reducing power of V. radiata sprouts was found to increase till 36 h of sprouting. A slight increase in TPC, TFC and reducing power was observed when seeds were treated with low concentrations of ANE. The elevation in TPC, TFC and reducing power upon treatment with low concentrations of ANE was also noticed in sprouts in saline combinations. Alpha amylase inhibition activity was found to reach a (67.16% ± 0.9) maximum at 24 h of sprouting at a 0.01% concentration of ANE. Tyrosinase inhibition and alpha glucosidase inhibition was 88.0% ± 2.11 and 84.92% ± 1.2 at 36 h of sprouting, respectively, at 0.01% concentration of ANE. A. nodosum extract is natural, environmentally friendly, and safe, and could be used as one of the strategies to decline stress at a low level and enhance the antioxidant activities in V. radiata sprouts, thus increasing its potential to be developed as an antioxidant-based functional food.

Rheum emodiWall has been reported to possess protective effect in many inflammatory diseases and oxidative stress-related injuries. This study aims to investigate antioxidant power of stilbenoids fromR. emodiand then explore the material basis for its antioxidant potential. The most abundant stilbenoid piceatannol-4′-O-β-D-glucopyranoside (PICG) and its aglycon piceatannol (PICE) were isolated fromR. emodirhizome. Using well-accepted antioxidant chemicals as reference, antioxidant activity of these stilbenoids was examined by measuring DPPH and superoxide anion radical scavenging, ferric reducing power, and inhibition of lipid peroxidationin vitro. Both PICG and PICE displayed promising antioxidant activity in all the four assays. Comparisons among the tested compounds indicated that PICE has the most potent antioxidant activity and the presence of 3′-hydroxyl group may enhance antioxidant activity of stilbenoids. The antioxidative effect of PICE at the cellular level was further demonstrated on the model of hydrogen-peroxide-induced H9c2 rat cardiomyoblasts injury. Taking into account the rapidin vivometabolic transformation of PICG into PICE it can be inferred that the most abundant stilbenoid PICG may be an important constituent responsible for the antioxidant potential ofR. emodiand promising to be developed as an antioxidant agent for supplementary or therapeutic use.

Tagetes erecta (T. erecta) is an herbaceous species, having wide ethno medicinal and traditional uses. It demonstrates different pharmacological activities like antioxidant, antimicrobial and anti depressant. The present study deals with the estimation of antioxidant properties of four solvent extracts of flowers of T. erecta. The study aims to discover potent extracts as a novel source of natural antioxidant. The crude extracts; Petroleum ether (PE), Chloroform (CF), Ethyl Acetate (EA) and Methanol (MET) were screened for their free radical scavenging capacity and reducing powers using DPPH, Superoxide and Reducing power assay. Ascorbic acid was used as standard. All the extracts show considerable antioxidant potential. However, methanolic extract can strongly scavenge DPPH and superoxide radical with significant IC50 value range 30.08 ± 0.98 µg/mL and 64.22±0.04 µg/mL respectively. The IC50 value of the extracts follows the order; MET less than EA less than PE less than CF. Lowest IC50 value indicates highest antioxidant potential. The reducing power of extracts were in the order; CF less than PE less than EA less than Met

Poultry waste is an abundant renewable source for the recovery of several value-added metabolites with potential industrial applications. This study describes the production of protease on poultry waste, with the subsequent use of the same poultry waste for the extraction of antioxidants. An extracellular protease-producing strain was isolated from Cuddalore coast, India, and identified asStreptomycessp. MAB18. Its protease was purified 17.13-fold with 21.62% yield with a specific activity of 2398.36 U/mg and the molecular weight was estimated as 43 kDa. The enzyme was optimally active at pH 8–10 and temperature 50–60°C and it was most stable up to pH 12 and 6–12% of NaCl concentration. The enzyme activity was reduced when treated with Hg2+, Pb2+, and SDS and stimulated by Fe2+, Mg2+, Triton X-100, DMSO (dimethyl sulfoxide), sodium sulphite, andβ-mercaptoethanol. Furthermore, the antioxidant activities of protease were evaluated usingin vitroantioxidant assays, such as DPPH radical-scavenging activity, O2scavenging activity, NO scavenging activity, Fe2+chelating activity, and reducing power. The enzyme showed important antioxidant potential with an IC50value of78±0.28 mg/mL. Results of the present study indicate that the poultry waste-derived protease may be useful as supplementary protein and antioxidant in the animal feed formulations.

  Objective: The study was carried out to evaluate the antioxidant potential of quercetin and hesperidin.Methods: The antioxidant potential of quercetin and hesperidin was estimated using free radicals in in vitro condition. Antioxidant activity of quercetin and hesperidin was estimated using 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide, nitric oxide, hydroxyl radical, hydrogen peroxide radical, and reducing power assay.Results: Quercetin and hesperidin have high efficiency in scavenging free radicals. Both the compounds are more efficient in scavenging DPPH (IC50 value 36.15±0.30 and 41.55±0.49 mg/ml) and superoxide radicals (IC 50 value 19.3±0.26 and 28.08 ±0.18 mg/ml) have high reducing capacity when compared to the standards ascorbic acid (IC50 value 31.45±1.31 and 23.2±0.20 mg/ml).Conclusion: The present study suggests that quercetin and hesperidin have significant antioxidant activities and could be easily accessible to the immune system.

Background: NSAIDs are the most widely prescribed medications worldwide for their anti-inflammatory, antipyretic, and analgesic effects However, their chronic use can lead to several adverse drug events including GI toxicity. The selective COX-2 inhibitors developed as gastro-sparing NSAIDs also suffer from serious adverse effects which limit their efficacy. Objective: Local generation of reactive oxygen species is implicated in NSAID-mediated gastric ulceration and their combination with H2 antagonists like famotidine reduces the risk of ulcers. The objective of this work was to design and synthesize novel methanesulphonamido isoxazole derivatives by hybridizing the structural features of NSAIDs with those of antiulcer drugs (ranitidine, famotidine, etc.) to utilize a dual combination of anti-inflammatory activity and reducing (antioxidant) potential. Method: The designing process utilized three dimensional similarity studies and utilized an isoxazole core having a potential for anti-inflammatory as well as radical scavenging antioxidant activity. The compounds were assayed for their antiinflammatory activity in established in vivo models. The in vitro antioxidant activity was assessed in potassium ferricyanide reducing power (PFRAP) assay employing ascorbic acid as the standard drug. Results: Compounds (5, 6, 9 and 10) showed anti-inflammatory activity comparable to the standard drugs and were also found to be non-ulcerogenic at the test doses. Compounds 6-10 exhibited good antioxidant effect in the concentration range of 1.0-50.0 µmol/ml. The test compounds were also found to comply with the Lipinski rule suggesting good oral absorption. Conclusion: A new series of isoxazole based compounds is being reported with good anti-inflammatory activity coupled with antioxidant potential as gastro-sparing anti-inflammatory agents.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.
