Objective: Both honeybees (Apis spp.) and stingless bees (Trigona spp.) produce honeys which normally taken orally, have high nutritional and therapeutics value. Until recently, phytochemical comparison of both honey is still scarce and elucidating cytotoxicity effects on human gingival fibroblast cells (HGF) in oral cavity is of interest. Materials and Methods: Kelulut honey (KH), acquired from the stingless bees and acacia honey (AH) from the sting bees honey samples were underwent GC-MS analysis to ascertain their composition. HGF were exposed to various concentrations of KH and AH from the lowest 0.015% to the highest 5% by MTT assay for 24h, 48h and 72h. Results: GC-MS analysis determined various beneficial compounds such as flavonoids, furans, pyrans, levoglucosan and hydroxymethylfurfural from both of honey samples. MTT assay showed that the HGF cells demonstrated good viability up to percentages (v/v) as high as almost 2% in both honeys. The IC50 values for both honey for all time frames fall at above 2%. Conclusion: Both honey showed good survivability of HGF cells up to 2% of concentration. Bangladesh Journal of Medical Science Vol. 21(1) 2022 Page : 158-164

Kelulut honey or stingless bee honey is a type of honey produced by stingless bees of the Trigona species where the nest is found in living trees. Objective: The aim of this study was to evaluate the cytotoxic potential of Malaysian Kelulut honey by employing MTT assay on a human gingival fibroblast cell line. Methods: Human gingival fibroblast cell line was cultured in minimal essential medium alpha (α-MEM) with 10% foetal bovine serum and 1% penicillin-streptomycin solution in a 5% CO2 incubator at 37°C in a humidified atmosphere. The cells were seeded at a cell density of 5x103 cells/well in a 96-well culture plate for 24 hours. The cells were treated with seven different concentrations (200, 100, 50, 25, 12.5, 6.25, and 3.125mg/ml) of Malaysian Kelulut honey and incubated in a CO2 incubator. The negative control comprised cells treated with growth media alone. The cell viability was assessed using MTT assay at 24, 48, and 72 hours. The test plate was shaken using a microplate shaker and the absorbance of the solution was measured at 570nm using an ELISA reader with the Magellan software. Statistical analysis of the data was carried out using Kruskal-Wallis test and SPSS 24.0.0 for Windows. A p value <0.05 was considered as statistically significant. Results: There was no cytotoxic effect of Malaysian Kelulut honey on HGF-1 based on the MTT assay at different concentrations and at different time points tested as the cell viability was above 70%. The highest percentage of cell viability at all three different durations of treatment were observed at 3.125mg/ml, whereas the lowest cell viability was observed at 200mg/ml of Kelulut honey concentration. However, statistically significant differences were seen between some of the concentrations at various time points. Conclusion: Since the cell viability of HGF-1 treated with Malaysian Kelulut honey was more than 70% at all concentrations ranging from 3.125mg/ml to 200mg/ml at three different time points (24, 48 and 72 hours), Malaysian Kelulut honey can be considered as non-cytotoxic on human gingival fibroblasts based on MTT assay under the present test conditions

Asiaticoside is a bioactive compound found in the traditional plant Centella asiatica (Asiatic pennywort or ‘Pegaga’) generally used for wound healing applications. Numerous studies have discussed the potential benefits of asiaticoside on different human cells such as keratinocytes and dermal fibroblast cells in healing of wounds. However only very few studies have been conducted to investigate its healing effect on cells originated from human oral cavity. The present study aimed to determine the potential of asiaticoside on human gingival fibroblast cells. Cytotoxic activities of the compounds were assessed by MTT assay. The wound healing was examined by scratch assay. The effect of asiaticoside on Col1A1 gene expression was also analyzed using qRT-PCR. Col1A1 is known to play a crucial role in wound healing. The MTT assay result showed that the maximum tolerable concentration of asiaticoside was 0.25 mg/ml. The scratch assay revealed that asiaticoside significantly accelerated the wound healing compared to the negative control (P<0.05). Moreover, the qRT-PCR demonstrated that asiaticoside markedly increased Col1A1 mRNA expression. These results proved asiaticoside as a potential candidate for wound healing agent in dentistry.

Abstract Objectives This study aims to evaluate the ability of tantalum-coated titanium to improve human gingival fibroblasts’ adhesion, viability, proliferation, migration performance, and the potential molecular mechanisms. Materials and methods Titanium plates were divided into two groups: (1) no coating (Ti, control), (2) Tantalum-coated titanium (Ta-coated Ti). All samples were characterized by scanning electronic microscopy, surface roughness, and hydrophilicity. Fibroblasts’ performance were analyzed by attached cell number at 1 h, 4 h, and 24 h, morphology at 1 h and 4 h, viability at 1 day, 3 days, 5 days, and 7 days, recovery after wounding at 6 h, 12 h, and 24 h. RT-PCR, western blot were applied to detect attachment-related genes’ expression and protein synthesis at 4 h and 24 h. Student’s t test was used for statistical analysis. Results Tantalum-coated titanium demonstrates a layer of homogeneously distributed nano-grains with mean diameter of 25.98 (± 14.75) nm. It was found that after tantalum deposition, human gingival fibroblasts (HGFs) adhesion, viability, proliferation, and migration were promoted in comparison to the control group. An upregulated level of Integrin β1 and FAK signaling was also detected, which might be the underlying mechanism. Conclusion In the present study, adhesion, viability, proliferation, migration of human gingival fibroblasts are promoted on tantalum-coated titanium, upregulated integrin β1 and FAK might contribute to its superior performance, indicating tantalum coating can be applied in transmucosal part of dental implant. Clinical significance Tantalum deposition on titanium surfaces can promote human gingival fibroblast adhesion, accordingly forming a well-organized soft tissue sealing and may contribute to a successful osseointegration.

Introduction: Garlic is a natural herb which can be used to be a good alternative treatment because cheap and safe. Garlic contains allicin which may has act antibacterial and antiinflammatory effect. Moreover, garlic extract has a good biocompatibility and can stimulate cell growth. Does garlic extract biocompatible and can stimulate cell growth that is seen from the proliferation of human gingival fibroblasts and how its work will be studied.Objective: The aim of this study was to analyze the biocompatibility of garlic extract by observing the viability and proliferation of human gingival fibroblasts in vitro.Methods: Biocompatibility test was conducted using serial concentration of garlic extract. Human gingival fibroblasts was seeded into 96 microwell plate with density of 2x103 cells, added with the fourteen serial concentration of garlic extract, and incubated in 37o C and 5% CO2for 24, 48 and 72 hours. MTT assay was used to analyze the viability and proliferation of human gingival fibroblasts. Data were analyzed by the Kruskal Wallis and U Mann-Whitney test.Results: The result showed that in each time of observation, there is no significant difference in viability fibroblast (p>0,05), but there are significant difference between time of observation at 24, 48, and 72 hours (p <0.05).Data showed that all concentration of garlic extract increased the viability and proliferation of human gingival fibroblasts.Conclusions: The ethanolic garlic extract has a good biocompatibility to human gingival fibroblasts culture cell and can stimulate the proliferation of human gingival fibroblast.Bangladesh Journal of Medical Science Vol.17(4) 2018 p.556-561

The purpose of this study was to evaluate the in vitro effect of the chlorophyllin–phycocyanin mixture (Photoactive+) as a photosensitizer (PS) during antimicrobial photodynamic therapy (aPDT) on the count of Enterococcus faecalis (E. faecalis) using different light sources. The antimicrobial effect of aPDT with chlorophyllin–phycocyanin mixture using different light sources including diode laser (λ = 660 nm), diode laser (λ = 635 nm), LED (λ = 450 ± 30 nm) alone or in combination was assessed using microbial cell viability assay against E. faecalis. In addition, the cell cytotoxicity of Photoactive+ was assessed on human gingival fibroblast (HuGu) cells by MTT assay; E. faecalis growth when treated by both red wavelengths (635 nm, 660 nm) and combination of LED (420–480 nm) and red wavelengths (635 nm, 660 nm), significantly reduced compared to the control group (p < 0.05). There was no significant reduction in the number of viable cells exposed to Photoactive+ compared to the control group (p < 0.05). This study shows that the application of chlorophyllin–phycocyanin mixture and irradiation with emission of red light achieved a better result for bacterial count reduction, compared to a control. This component can be applied safely due to very negligible cytotoxicity.

BACKGROUND Daily diaries are extensively used for examining participants' daily experience in behavioral and medical science. Whether participants recall their experiences within the time frames prescribed by task has received little attention. OBJECTIVE The objectives of this study are to describe survey respondents' self-reported recall timeframe and to evaluate the impact of different daily diary items on respondents reported affective states. METHODS In this study, 577 participants completed a mood survey with one of four time frame instructions: 1) today, 2) since waking up today, 3) during the last 24 hours, or 4) in the last day. They were also asked to indicate the time periods they considered when answering these items and to recall the instructional phrases associated with the items. RESULTS Almost all participants in the "Today" (97%) and "Since waking up today" (94%)" conditions reported using time periods consistent with our expectations, while a lower proportion was observed in the "during the last 24 hours" (69%) condition. A diverse range of responses was observed in the "In the last day" condition. Furthermore, the instructions influenced the levels of some self-reported affects, although exploratory analyses were not able to identify the mechanism underlying this finding. CONCLUSIONS Overall, these results indicate that "Today" and "Since waking up today" are the most effective instructional phrases for inquiring about daily experience and that investigators should use caution when using the other two instructional phrases.
