The multifaceted and heterogeneous nature of depression presents challenges in pinpointing treatments. Among these contributions are the interconnections between the gut microbiome and neurological function termed the gut-brain axis. A diverse range of microbiome-produced metabolites interact with host signaling and metabolic pathways through this gut-brain axis relationship. Therefore, biosensor detection of gut metabolites offers the potential to quantify the microbiome’s contributions to depression. Herein we review synthetic biology strategies to detect signals that indicate gut-brain axis dysregulation that may contribute to depression. We also highlight future challenges in developing living diagnostics of microbiome conditions influencing depression.

Pakistan was one of the top 10 fastest growing developing countries between 1960 and 1990 recording an annual average growth rate of 6 per cent. The structure of the economy was also transformed during this period with the share of agriculture coming down from 50 per cent to 20 per cent. The subsequent 25 years have, however, brought about a significant decline in growth rates and in more recent seven years, it has lagged behind other South Asian countries. A combination of political instability and disruption of evolving democratic process, lack of continuity in policies and poor governance have contributed to this outcome. Pakistan has also not utilized its geographic location to take advantage of intra-regional trade and investment. Many promising opportunities were lost due to lingering tension with India. The future potential can only be realized if Pakistan is able to position itself for meeting the future challenges of integration into the regional and global economy, reaping demographic dividends because of youthful population and moving up the ladder of technology. The realization of these goals will depend upon sound macroeconomic policies, strong institutional and governance framework, investment in infrastructure and human development and political stability.

This article is the result of a research on the Kaharingan problematic issues of religious identity after being integrated into Hindu Dharma. During the ‘New Order’ (President Soeharto's government) Kaharingan religion was not included in one of the religions served by the state. The issue of state recognition and the ease of obtaining civil services for Kaharingan adherents are strong reasons for Kaharingan religious leaders to integrate Kaharingan as part of Hinduism. The research raises the issues: (1) how is the process of integrating Kaharingan religion into Hindu Dharma? (2) what are the implications of such integration? and, (3) how is the existence of Kaharingan religious identity as the original ‘Dayak tribe religion’ after integration into Hindu Dharma in the future? This study aims to portray the existence of Kaharingan religion during integration into Hindu Dharma. This type of research is qualitative-descriptive with the method of collecting data through observation and interviews with religious leaders and administrators of religious institutions namely the Hindu Kaharingan Grand Council (MB-AHK), as well as an analysis of documents related to the object of research. Theories used in this research are integration theory, identity theory and locality theory. The integration process has implications for various fields, ranging from education, social, religious, economic, political upto cultural identity. The future challenges of Kaharingan are: internal conflict, a dilemma of distortion from third parties and stigmatization as one of the Hindu Dharma sects.

ABSTRACT Background Diet is known to affect human gut microbiome composition; yet, how diet affects gut microbiome functionality remains unclear. Objective We compared the diversity and abundance/presence of fecal microbiome metabolic pathways among individuals according to their long-term diet quality. Methods In 2 longitudinal cohorts, we assessed participants’ usual diets via repeated surveys during 1996–2011 and collected a stool sample in 2015–2018. Participants who maintained a healthy or unhealthy diet (i.e., stayed in the highest or lowest quintile of a healthy diet score throughout follow-up) were selected. Participants were excluded if they reported a history of cancer, cardiovascular disease, diabetes, or hypertension; had diarrhea or constipation in the last 7 d; or used antibiotics in the last 6 mo before stool collection. Functional profiling of shotgun metagenomics was performed using HUMAnN2. Associations of dietary variables and 420 microbial metabolic pathways were evaluated via multivariable-adjusted linear or logistic regression models. Results We included 144 adults (mean age = 64 y; 55% female); 66 had an unhealthy diet and 78 maintained a healthy diet. The healthy diet group had higher Shannon α-diversity indexes of microbial gene families and metabolic pathways (both P < 0.02), whereas β-diversity, as evaluated by Bray-Curtis distance, did not differ between groups (both P > 0.50). At P < 0.01 [false discovery rate (FDR) <0.15], the healthy diet group showed enriched pathways for vitamin and carrier biosynthesis (e.g., tetrahydrofolate, acetyl-CoA, and l-methionine) and tricarboxylic acid (TCA) cycle, and increased degradation (or reduced biosynthesis) of certain sugars [e.g., cytidine monophosphate (CMP)-legionaminate, deoxythymidine diphosphate (dTDP)-l-rhamnose, and sucrose], nucleotides, 4-aminobutanoate, methylglyoxal, sulfate, and aromatic compounds (e.g., catechol and toluene). Meanwhile, several food groups were associated with the CMP-legionaminate biosynthesis pathway at FDR <0.05. Conclusions In a small longitudinal study of generally healthy, older Chinese adults, we found long-term healthy eating was associated with increased α-diversity of microbial gene families and metabolic pathways and altered symbiotic functions relevant to human nutrition and health.

The chapter provides an overview of how policy analysis takes place in Mexican Think Tanks. It focuses on two of the few organisations of this kind that currently exist in the country: the Centro de Investigación para el Desarrollo (CIDAC, or Centre for Research for Development) and the Centro de Estudios Espinosa Yglesias (CEEY, or Centre of Studies Espinosa Yglesias). The chapter is divided into four sections. The first discusses the main features of think tanks, with a particular focus on the Mexican ones. The second presents the origins and general objectives of CIDAC and CEEY, and describes how these two organizations conduct policy analysis. The third compares both cases, paying particular attention to how they define their topics of interest, how they gather relevant information, what kind of policy products they generate, what kind of communication channels they use, and how they assess the impact that their analyses may have had. The chapter closes with some conclusions and general remarks about the future challenges of policy analysis in Mexican think tanks.
