The present study was aimed to investigate the composition and contents and the major lipophilic compounds, including the sterols, fatty acids, and tocols of shellfish species. Moreover, to explore the antitumor activity of these lipophilic constituents, their cytotoxicity potentials were determined against five different human cancer cells, including colon carcinoma (HCT116), epithelial melanoma (A2058), glioblastoma multiforme (T98G), lung carcinoma (A549), and adenocarcinoma (HeLa). The results show a significant variation in the contents and composition of lipophilic constituents among the studied species. The highest omega-3 (n-3) polyunsaturated fatty acids (PUFAs) were recorded from arrow squid and pacific oysters, accounting for 53.2% and 53.0% of their total fatty acids, respectively. However, the highest cholesterol content was also recorded in arrow squid (154.4 mg/100 g; 92.6% of total sterols). In contrast, in the Japanese littleneck, Yesso scallop, and common orient clam, cholesterol was just 17.1%, 18.3%, and 18.9% of total sterols, respectively, making them the richest source of non-cholesterol sterols (NCS). Lipids extracted from shellfish species showed ABTS+•- and DPPH•-scavenging activities. In the cytotoxicity analysis, lipids extracted from the Argentine red shrimp showed the highest cytotoxicity against glioblastoma multiforme T98G cells, with an IC50 value of 12.3 µg/mL. The composition and cytotoxicity data reported herein may help explore the nutritional and anticancer potentials of shellfish species.

The purpose of this paper is to detect any changes in fatty acid and cholesterol content of goat colostrum in the period of 0-72 hours after kidding. For that purpose, samples of colostrum from 16 goats, belonging to local populations in north-eastern Romania where collected. Samples were preserved by freezing and then subjected to chemical analysis. Preparation of solutions for fatty acids analysis was based on the principle of methylation, by combining alkaline methylation (NaOCH3/MeOH) with acid (BF3/MeOH). For cholesterol we used the principle of fat saponification, using potassium hydroxide, followed by filtration and separation of cholesterol with hexane. Detection of cholesterol and fatty acids content was performed by gas chromatography device. Caprinic acid (C10:0), myristic acid (C14:0), palmitic acid (C16: 0), stearic acid (C18:0) and oleic acid (C18:1) quantified more than 75% of the total fatty acids determined 72 hours after birth. Among short-medium-chain fatty acids, capronic acid (C6:0) was found in highest proportion (average concentration of 5.7% of total fatty acids), but at the same time, presented the most obvious oscillation range from 0 to 72 hours (6.56% at birth, 2.45% and 7.59% from 12 hours to 72 hours), with the differences between mean values being significant (p<0.05). In the case of polyunsaturated fatty acids, the ratio between omega 3 and omega 6 was 1/4.64 at kidding and 1/4.23 at 72 hours after birth, the differences being insignificant. In goat colostrum, the mean concentration of cholesterol was 9.43 mg per 100 g.

Abstract This experiment proposed to investigate the efficiency of omega 3 fatty acids from fish that improves autoimmune against myocarditis in the rat. Fish oil was extracted from fresh Tuna fish and performed FAME analysis and mice bioassay. The autoimmune myocarditis was induced by subcutaneous injection of porcine cardiac myosin (PCM) into the footpads of rats on the first and seventh day. Rats were dissected on the 21st day to analyze the histopathological, hemodynamic, echocardiographic factors, and immunohistochemistry expressions. In the study, 73.90% of total fatty acids were recorded. Histological analysis revealed that omega 3 fatty acids administrated groups showed tremendous development in the multifocal myocardia hyaline degeneration and necrosis with inflammatory changes. Moreover, omega 3 fatty acids inhabited the expressions of inflammatory cells (CD4, CD8 and CD11b) and suppressed the level of NF-κB. The echocardiographic factors such as heartbeat, SBP, DBP, levels of LVDs, LVDd, LVPW percentage of LVFS, EF, expression levels of inflammatory cytokines (TNF, IL-1β, IFN-ɤ, IL-2, and IL-6) also significantly suppressed by omega 3 fatty acids. Hence, the present study proved that consuming fatty acid-enriched fish might be a successful therapy for improving the inflammatory profile, regenerates the heart tissues, and controlled the production of inflammatory cells.

Maternal obesity is associated with adverse offspring outcomes. Inflammation and deficiency of anti-inflammatory nutrients like omega(n)-3 polyunsaturated fatty acids (PUFA) may contribute to these associations. Fetal supply of n-3 PUFA is dependent on maternal levels and studies have suggested that improved offspring outcomes are associated with higher maternal intake. However, little is known about how maternal obesity affects the response to n-3 supplementation during pregnancy. We sought to determine (1) the associations of obesity with PUFA concentrations and (2) if the systemic response to n-3 supplementation differs by body mass index (BMI). This was a secondary analysis of 556 participants (46% lean, 28% obese) in the Maternal-Fetal Medicine Units Network trial of n-3 (Docosahexaenoic acid (DHA) + Eicosapentaenoic acid (EPA)) supplementation, in which participants had 2g/day of n-3 (n = 278) or placebo (n = 278) from 19 to 22 weeks until delivery. At baseline, obese women had higher plasma n-6 arachidonic acid concentrations (β: 0.96% total fatty acids; 95% Confidence Interval (CI): 0.13, 1.79) and n-6/n-3 ratio (β: 0.26 unit; 95% CI: 0.05, 0.48) compared to lean women. In the adjusted analysis, women in all BMI groups had higher n-3 concentrations following supplementation, although obese women had attenuated changes (β = −2.04%, CI: −3.19, −0.90, interaction p = 0.000) compared to lean women, resulting in a 50% difference in the effect size. Similarly, obese women also had an attenuated reduction (β = 0.94 units, CI: 0.40, 1.47, interaction p = 0.046) in the n-6/n-3 ratio (marker of inflammatory status), which was 65% lower compared to lean women. Obesity is associated with higher inflammation and with an attenuated response to n-3 supplementation in pregnancy.

The objective of this study was to determine the chemical composition of five different food seeds (sunflower, poppy, hemp, flax and sesame) regarding fatty acid, mineral (Fe, Cu, Zn, Na, Mg, K, Ca, Al) and protein content. In addition, the total antioxidant capacity of the seeds was evaluated using the photochemiluminescent assay. The food seeds were subjected to lipid extraction and converted into fatty acid methyl esters before the gas chromatography analysis. In all food seeds, the saturated (SFAs), monounsaturated (MUFAs) and polyunsaturated fatty acids (PUFAs) were identified, respectively. PUFAs were the most abundant fatty acids (61.2% ± 0.07% and 84.8% ± 0.08% of total fatty acids), with the highest content in flax and hemp seed oil. Also, high amounts of omega-3 from PUFAs were determined in flax and hempseed oil. Based on the obtained results the sunflower, sesame and poppy seeds are good sources of omega-6, while flax and hemp seeds are good sources of omega-3. All samples are rich in minerals (Na, K, Ca, Mg) and have more than 20% protein content.

Abstract Fish are a major dietary source of n-3 and n-6 polyunsaturated fatty acids (PUFAs) in humans since the human body lacks the necessary enzymes required to synthesize them. Diets rich in fish oil containing n-3 fatty acids have been shown to lower the incidence rates of cancer. They have also been shown to have cytotoxic or anti-proliferative effects on a variety of human cancer cell lines, and they are known to nurture overall human health without any observable side effects. The present review focused on in-vitro research findings regarding the apoptotic and anti-proliferative role of n-3 fatty acids FAs that highlight the important mechanisms and pathways of their action in inhibiting the progression of cancer. A survey of the literature indicated the paucity of information on the use of n-3 FAs in combination with other anticancer substances. Therefore, further research needs to be undertaken on the use of n-3 FAs in combination with other anticancer phytochemicals and drugs, the use of antibodies as specific carriers of n-3 FAs, and different increasing ratios of n-3/n-6 FAs that not only control the side effects of chemotherapy but also enhance its efficacy and selectivity. These FAs might also prove to be a pioneer therapy against cancer.

Background: The erythrocyte ratio of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) over total fatty acids, the omega-3 index (O3I), has been suggested as an overall health marker and to motivate corporate health recommendations. We set out to assess the O3I status in a working population, the differences between normal and rotating shift employees, the consumption of omega-3 rich food and whether recommendations to increase intake of omega-3 rich foods can improve the O3I. Methods: Employees registered for their occupational health check-up were offered to participate in a pre-post study at the Ludwigshafen (Germany) site including an assessment of their O3I at baseline and after 4 months (follow-up) and two subsequent food frequency questionnaires. For those with O3I below 8%, a recommendation was provided to increase the intake of omega-3 fatty acid rich food and to take advantage of the employees’ catering service with its enhanced fatty seafood offer during the study period. Dietary intake of EPA and DHA, erythrocyte fatty acid profiles, clinical and lifestyle parameters were assessed. Results: In 500 employees (26.6% female, 21–64 years, median age: 47 years [IQR: 37–53]), at baseline the overall mean O3I was 4.1 ± 1.1% (99.6% of O3I assessed were below 8%), higher in women, in participants with “normal” body weight, upper employment grade, and non-smokers, but not different between regular and rotating shift workers. The three fifths of the cohort also participating in the follow-up increased their EPA and DHA intake by 0.1 g/d and their O3I by 0.5 percentage points. Conclusion: This study provides essential data on omega-3 erythrocyte concentrations in a clinically healthy German working population and the challenges of increasing the O3I with dietary recommendations even in study participants motivated to follow up on their omega-3 status.

Green vegetable consumption has long been considered to have health benefits mainly due to the vitamins, minerals and phytonutrients (such as vitamin C, folate, antioxidants etc) contained in a vegetable-rich diet. Additionally, green vegetables are known to contain a relatively high proportion of omega-3 polyunsaturated fatty acids (PUFAs), primarily in the form of alpha-linolenic acid (18:3n-3). However, there are no data available on the fatty acid composition and concentration of green vegetables commonly consumed in Australia. The present study determined the fatty acid content of 11 green vegetables that are commonly available in Australia. The total fatty acid concentrations of the vegetables under study ranged from 44 mg/100 g wet weight in Chinese cabbage to 372 mg/100 g in watercress. There were three PUFAs in all vegetables analyzed; these were 16:3n-3, 18:2n-6, and 18:3n-3 fatty acids. Sample vegetables contained significant quantities of 16:3n-3 and 18:3n-3, ranging from 23 to 225 mg/100g. Watercress and mint contained the highest amounts of 16:3n-3 and 18:3n-3, and parsley had the highest amount of 18:2n-6 in both percentage composition and concentration. Mint had the highest concentration of 18:3n-3 with a value of 195 mg/100 g, while watercress contained the highest concentration of 16:3n-3 at 45 mg/100 g. All 11 green vegetables contained a high proportion of PUFAs, ranging from 59 to 72% of total fatty acids. The omega-3 PUFA composition ranged from 40 to 62% of total fatty acids. Monounsaturated fatty acid composition was less than 6% of total fatty acids. The proportion of saturated fatty acids ranged from 21% in watercress and mint to 32% of total fatty acids in Brussels sprouts. No eicosapentaenoic and docosahexaenoic acids were detected in any of the samples. Consumption of green vegetables could contribute to 18:3n-3 PUFA intake, especially for vegetarian populations.
