Cyclic fertilin peptide (cFEE: phenylalanine, glutamic acid; glutamic acid) improves gamete interaction in humans. We investigate whether it could be via improvement of sperm movement parameters and their mitochondrial ATP production. Sperm movement parameters were studied using computer-assisted sperm analysis (CASA) in sperm samples from 38 patients with normal sperm in medium supplemented with cyclic fertilin against a control group. Sperm mitochondrial functions were studied using donor’s sperm, incubated or not with cFEE. It was evaluated by the measurement of their ATP production using bioluminescence, their respiration by high resolution oxygraphy, and of mitochondrial membrane potential (MMP) using potentiometric dyes and flow cytometry. cFEE significantly improved sperm movement parameters and percentage of hyperactivated sperm. Impact of inhibitors showed OXPHOS as the predominant energy source for sperm movement. However, cFEE had no significant impact on any of the analyzed mitochondrial bioenergetic parameters, suggesting that it could act via a more efficient use of its energy resources.

The recent introduction of various devices for computer-assisted sperm analysis allows us to have not only objective measurements of the seminal parameters but also to determine some characteristics of sperm motion such as curvilinear and linear velocity and amplitude of lateral head displacement that cannot be obtained by routine analysis. These new seminal parameters seem to be very useful in the diagnosis of male infertility and, among them, ALH seems to play a more important role; in one of our studies we found a significant correlation between this parameter and the results of the bovine cervical mucus penetration test (Penetrak). Furthermore it is possible to determine sperm subpopulations for each parameter and so it will be possible to express a more accurate judgement not only on the fertilizing capacity of semen but also to know the effects on spermatozoa of different activating and capacitating treatments.

Computer assisted sperm analysis (CASA) systems can reduce errors occurring in manual analysis. However, commercial CASA systems are frequently not applicable at the forefront of challenging research endeavors. The development of open source software may offer important solutions for researchers working in related areas. Here, we present an example of this, with the development of three new modules for the OpenCASA software (hosted at Github). The first is the Chemotactic Sperm Accumulation Module, a powerful tool for studying sperm chemotactic behavior, analyzing the sperm accumulation in the direct vicinity of the stimuli. This module was validated by comparing fish sperm accumulation, with or without the influence of an attractant. The analysis clearly indicated cell accumulation in the treatment group, while the distribution of sperm was random in the control group. The second is the Sperm Functionality Module, based on the ability to recognize five sperm subpopulations according to their fluorescence patterns associated with the plasma membrane and acrosomal status. The last module is the Sperm Concentration Module, which expands the utilities of OpenCASA. These last two modules were validated, using bull sperm, by comparing them with visual counting by an observer. A high level of correlation was achieved in almost all the data, and a good agreement between both methods was obtained. With these newly developed modules, OpenCASA is consolidated as a powerful free and open-source tool that allows different aspects of sperm quality to be evaluated, with many potential applications for researchers.

In male developing piglets, low endogenous oestrogenic hormone levels prevail. Sensitive tissues expressing high levels of oestrogen receptors, such as the epididymis, might thus be of specific relevance for early disturbances by estrogenic endocrine disruptors. We have previously shown that distinct concentrations of the natural oestrogen oestradiol-17β (E2) orally applied during the whole period of pregnancy in sows lead to a distinct shift in body composition. Therefore, we aimed at investigating whether prenatal E2 exposure had the potential to program male reproductive organs toward altered semen quality. The E2 treatment groups (n = 4 sows/group) were designed to represent doses of pharmacological relevance (PR; 1000 µg kg–1 of body weight per day), oral no-effect level (10 µg kg–1 of body weight per day), and acceptable daily intake level (0.05 µg kg–1 of body weight per day). Total RNA was extracted from the epididymis of 8-week-old piglet twins (n = 2 per sow). The RNA of littermates was pooled and subjected to microarray analysis (MA) using Agilent Porcine Genome Arrays (Agilent Technologies Inc., Santa Clara, CA, USA). Ejaculates of adult male pigs from the control group and the pharmacological group (n = 4) were collected and analysed for quantitative and qualitative parameters by using computer-assisted sperm analysis and flow cytometry. In total, 105 transcripts exhibited a significant difference of more than 1.5-fold for at least one of the treatment groups compared with the control group (P < 0.05; FDR 5%). Several transcripts revealed gradually decreasing amounts subject to the dose of E2 exposure. Eight transcripts showed significant differences in all 3 treatment groups compared with the control group. Thirty-two transcripts had reduced abundance in the PR group and the oral no-effect level group, whereas 20 transcripts were downregulated in the PR group only. Semen parameters of adult males did not differ regarding volume and semen concentration, morphology, and membrane integrity. Computer-assisted sperm analysis evaluations did not show differences in motility and other kinematic parameters. Eight weeks after E2 exposure, male prepubertal piglets exhibited significant differences in epididymal messenger RNA transcript abundance subject to an in utero E2 treatment. This points toward possible epigenetic programming of the epididymis. Although the differential transcript abundance indicates a perturbation, normal semen quality was found after adolescence. Either these transcriptional changes are of minor functional importance or flexible mechanisms account for the developmental adaptations. Considering that identical transcripts were regulated in offspring exposed to both pharmacological and very low doses, further investigations of set points obviating long-term adverse consequences are needed.

Практической частью репродуктивной технологии является прогноз и оценка фертильности самцов. Существующие методы оценки семени не дают достаточно корректного прогноза результатов осеменения и, следовательно, репродуктивного потенциала того или иного производителя. В работе приведены данные комплексной оценки биологической полноценности семени чистопородных и гибридных хряков-производителей с использованием компьютерной технологии CASA (computer-assisted sperm analysis). Оценивали активность сперматозоидов в образцах классическим способом (микроскопирование) и с помощью компьютерной технологии, программа «Зоосперм 1.0». Исследования проводили в 2014 – 2015 гг. в Лаборатории репродуктивной криобиологии животных (Россия). Объектом исследования служила сперма чистопородных и гибридных хряков-производителей, разводимых в разных регионах Российской Федерации: крупная белая (n = 24), дюрок (n = 28), ландрас (n = 18), пьетрен (n = 8), гибридные (n = 6). Установлено, что высокими репродуктивными качествами отличаются хряки-производители породы ландрас и гибридов, от них в среднем за эякулят получено 290,2 и 290,6 мл спермы соответственно; наименьшими — хряки-производители породы пьетрен (270 мл). Частота встречаемости сперматозоидов с аномальной морфологией высокая у хряков-производителей породы дюрок (12 %) и пьетрен (15 %). Частота встречаемости аномалии в разных сегментах сперматозоидов зависит от индивидуальной особенности хряков-производителей. У одних производителей наиболее часто встречаются морфологические отклонения головки, например, у хряка породы ландрас оно составило 58,60 %, у других часто встречались отклонения от нормы в строении жгутика (12,1 – 29,3 %). Использование компьютерной технологии при оценке биологической полноценности сперматозоидов хряков-производителей является наиболее точным и информативным.

Genetic resource banks (GRB) preserve the genetic material of endangered, valuable individuals or genetically relevant breeds. Semen cryopreservation is a crucial technique to reach these goals. Thus, we aimed to assess the sperm parameters of semen doses from the native pig breed Gochu Asturcelta stored at the GRB of Principado de Asturias (GRB-PA, Gijón, Spain), focusing on intrinsic and extrinsic (boar, season) factors. Two straws per boar (n = 18, 8–71 months of age) were thawed, pooled, and assessed after 30 and 150 min at 37 °C by CASA (computer-assisted sperm analysis system; motility and kinematic parameters) and flow cytometry (viability, acrosomal status, mitochondrial activity, apoptosis, reactive oxygen species, and chromatin status). The effects of age, incubation, and season on post-thawing quality were determined using linear mixed-effects models. Parameters were on the range for commercial boar breeds, with chromatin status (SCSA: fragmentation and immaturity) being excellent. Incubation decreased sperm quality and functionality. The boar age did not have a significant effect (p > 0.05), but the between-boar variability was significant (p < 0.001). The season significantly affected many parameters (motility, kinematics, viability, acrosomal status, mitochondrial activity), especially after 150 min of incubation. In general, samples collected in spring and summer showed higher quality post-thawing, the lowest in winter. In conclusion, the sperm doses from the Gochu Asturcelta breed stored at the GRB-PA showed excellent chromatin status and acceptable characteristics after thawing. Therefore, boar and seasonal variability in this autochthonous breed could be relevant for cryobank management.
