The severe impairment of bone development and quality was recently described as a new target for unbalanced ultra-processed food (UPF). Here, we describe nutritional approaches to repair this skeletal impairment in rats: supplementation with micro-nutrients and a rescue approach and switching the UPF to balanced nutrition during the growth period. The positive effect of supplementation with multi-vitamins and minerals on bone growth and quality was followed by the formation of mineral deposits on the rats’ kidneys and modifications in the expression of genes involved in inflammation and vitamin-D metabolism, demonstrating the cost of supplementation. Short and prolonged rescue improved trabecular parameters but incompletely improved the cortical parameters and the mechanical performance of the femur. Cortical porosity and cartilaginous lesions in the growth-plate were still detected one week after rescue and were reduced to normal levels 3 weeks after rescue. These findings highlight bone as a target for the effect of UPF and emphasize the importance of a balanced diet, especially during growth.

Abstract Objective: Dietary transition, obesity and risky use of alcohol and tobacco are challenges to public health among indigenous peoples. The aim of the article was to explore the role of social position in dietary patterns and expenditures on food and other commodities. Design: Countrywide population health survey. Setting: Greenland. Participants: 2436 Inuit aged 15+ years. Results: Less than half of the expenditures on commodities (43 %) were used to buy nutritious food, and the remaining to buy non-nutritious food (21 %), alcoholic beverages (18 %) and tobacco (18 %). Participants were classified according to five dietary patterns. The cost of a balanced diet and an unhealthy diet was similar, but the cost per 1000 kJ was higher and the energy consumption was lower for the balanced diet. Participants with low social position chose the unhealthy pattern more often than those with high social position (40 % v. 24 %; P < 0·0001), whereas those with high social position more often chose the balanced alternative. Participants with low social position spent less money on the total food basket than those with high social position but more on non-nutritious food, alcohol and tobacco. Conclusions: Cost seems to be less important than other mechanisms in the shaping of social dietary patterns and the use of alcohol and tobacco among the Inuit in Greenland. Rather than increasing the price of non-nutritious food or subsidising nutritious food, socially targeted interventions and public health promotion regarding food choice and prevention of excessive alcohol use and smoking are needed to change the purchase patterns.

Today’s eating patterns are characterized by the consumption of unbalanced diets (UBDs) resulting in a variety of health consequences on the one hand, and the consumption of dietary supplements in order to achieve overall health and wellness on the other. Balanced nutrition is especially crucial during childhood and adolescence as these time periods are characterized by rapid growth and development of the skeleton. We show the harmful effect of UBD on longitudinal bone growth, trabecular and cortical bone micro-architecture and bone mineral density; which were analyzed by micro-CT scanning. Three point bending tests demonstrate the negative effect of the diet on the mechanical properties of the bone material as well. Addition of Spirulina algae or Pleurotus eryngii or Agaricus bisporus mushrooms, to the UBD, was able to improve growth and impaired properties of the bone. 16SrRNA Sequencing identified dysbiosis in the UBD rats’ microbiota, with high levels of pro-inflammatory associated bacteria and low levels of bacteria associated with fermentation processes and bone related mechanisms. These results provide insight into the connection between diet, the skeletal system and the gut microbiota, and reveal the positive impact of three chosen dietary supplements on bone development and quality presumably through the microbiome composition.

Abstract Hematopoietic stem cells (HSCs) interact with osteoblastic, stromal, and vascular components of the BM hematopoietic microenvironment (HM) that are required for the maintenance of long-term self-renewal in vivo. Osteoblasts have been reported to be a critical cell type making up the HSC niche in vivo. Rac1 GTPase has been implicated in adhesion, spreading, and differentiation of osteoblast cell lines and is critical for HSC engraftment and retention. Recent data suggest a differential role of GTPases in endosteal/osteoblastic versus perivascular niche function. However, whether Rac signaling pathways are also necessary in the cell-extrinsic control of HSC function within the HM has not been examined. In the present study, genetic and inducible models of Rac deletion were used to demonstrate that Rac depletion causes impaired proliferation and induction of apoptosis in the OP9 cell line and in primary BM stromal cells. Deletion of Rac proteins caused reduced trabecular and cortical long bone growth in vivo. Surprisingly, HSC function and maintenance of hematopoiesis in vivo was preserved despite these substantial cell-extrinsic changes. These data have implications for therapeutic strategies to target Rac signaling in HSC mobilization and in the treatment of leukemia and provide clarification to our evolving concepts of HSC-HM interactions.

AbstractAcrylonitrile-butadiene-styrene (ABS) has great verity applications in aerospace and automobiles industries. Mechanical strength of the ABS is superior to even that of impact resistant polystyrene. In addition metallic coatings can be applied to the surface of ABS moldings. The main aim of the present work is to investigate the mechanical properties of additives of basalt fibers (BF) to ABS with (5, 10, and 15) wt% embedded into the polymer matrix by using plastic injection molding technique. This new perceptions has been done on basalt fibers that have a potential low cost with its good mechanical performance. The ultimate tensile strength that obtained from the composite with 15 wt% is 56.67 MPa with 40.52 % increase value than neat ABS, Young’s modulus gradually increases with increasing the amount of additives. Impact un-notched strength decreases with a reported increment of 24.617 KJ.m–2. A Rockwell hardness test is also used and with the increases of additives the amount of hardness of the composite increases. A scan electron microscopy (SEM) on the fracture surface is captured to check the morphologies structure of the composite comparable with a neat ABS. and it is showed a very good distribution and bonding of the B.F. with the pure ABS. As well as the cost of the ABS and BF is reduced by a percentage of 15 %.

AbstractMuscle force is thought to be one of the main determinants of bone development. Hence, peak muscle growth is expected to precede peak bone growth. In this study, we investigated muscle and bone development in female C57BL/6 J, DBA/2JRj, and C3H/HeOuJ mice. Femoral cortical and trabecular bone structure and the weights of selected muscles were assessed at the ages of 8, 16, and 24 weeks. Muscle mass increased from 8 to 24 weeks in all 3 strains, suggesting peak muscle development at 24 weeks or later. Bone volume fraction, trabecular number, and connectivity density of the femur decreased or remained unchanged, whereas trabecular density and trabecular thickness largely increased. These results suggest a peak in trabecular bone accrual at 8 weeks or earlier followed by further increases in density and structural reorganization of trabeculae. Cortical density, cortical thickness, and cortical cross sectional area increased over time, suggesting a peak in cortical bone accrual at 24 weeks or later. In conclusion, our data provide evidence that growth of muscle lags behind trabecular bone accrual.

We investigated how a new type of synthetic porous hydroxyapatite ceramic (HAP-TCP) acts when it is implanted in growing membranous bone. Seventy-six New Zealand White infant male rabbits (4-week-olds) were used. Rabbits received HAP-TCP block or silicone block implantation in their right nasal bone. The left nasal bone was used for a sham. Serial or cross-sectional examinations by morphometry, radiology, blood biochemistry, and histology were carried out. Both the HAP-TCP and silicone groups exhibited no systemic growth disturbance in terms of morphometry and blood biochemistry. Dual energy x-ray absorptiometry (DEXA) revealed, however, a decrease in the bone mineral content (BMC) of the right nasal bone in the silicone group. Histology revealed a superior affinity of HAP-TCP to bone tissue than that of silicone. When a HAP-TCP block was implanted under the periosteum it bonded directly to bone tissue. However, sinking of the implants into the bone tissue were noted both in the HAP-TCP and the silicone groups in longitudinal observation. These results suggest that although HAP-TCP has superior affinity to bone tissue, this by itself is not enough sufficient reason to believe that HAP-TCP can be effectively applied during the growth period.

This article discusses the problems that arise in the operation of road structures. One of the most important is rutting. Indicators of resistance to rutting depend on the method of design of non-rigid pavement and operational parameters, which allows to obtain the applied method of volumetric design of asphalt mix «Superpave» (SP-19), which in our country was called «SPAS». The authors, together with the staff of the laboratory of asphalt Concrete plant – 1, monitored asphalt concrete mixtures designed according to the «Superpave» system on the asphalt mixing plant. Then tests were carried out according to the method of PNST 181-2016 on the susceptibility of asphalt concrete materials to shear deformations arising under the action of repeated passes of the loaded wheel at ambient temperature. The result of the study of rutting showed that the mixture SP-19 have predominantly traditional. More detailed research results are presented in this article. Based on the data obtained from the results, conclusions were formulated: the Studies show that the method of «Superpeyv» is updated to the requirements of technical supervision at the objects of the road complex of the Russian Federation. In conclusion, the authors have formulated conclusions on the research: a comparison of physical and mechanical performance of asphalt concrete mix SP-19 on PNST 114 and fine-grained asphalt concrete mix type A brand I shows that the shear stability in terms of the average depth of the track is higher for the brand SP-19 by 38 %; set the required mixing temperature and compaction of asphalt concrete mixture on the system «Superpeyv»; the release of a pilot batch of asphalt concrete mix of SP-19 brand at the production site of JSC «ABZ-1» showed that the recipe selected in the laboratory is reproduced in the ABZ; cores selected within the framework of technical supervision from the experimental coating and tested in the laboratory showed corresponding to the requirements of ODM 218.4.038-2017; assessment of economic efficiency at the cost of asphalt concrete mix of SP-19 brand showed a decrease of 2.2 % relative to the cost of fine-grained asphalt concrete mix; the increase in the service life of the road surface h 30–50 % by reducing rutting increases its turnaround time. At the same time, economic efficiency can reach 50 % per m2.

Food safety is one of WHO’s primary concerns during a pandemic. The current Covid-19 pandemic requires us to boost our immune system by eating a healthy and balanced diet. Food consumed by the masses must be free of chemical and biological substances that can be harmful for the body. Nowadays, food products have developed to be more innovative, such as packaged processed food products that can be stored for a long time, generally using Food Additives. The safety of packaged processed food products must be guaranteed by the manufacturers in order to guarantee consumer protection. To ensure this, the government has established the Consumer Protection Law; the Government Regulation on Food Safety, Quality and Nutrition; and the Food and Drug Administration Division. Through the BPOM, the government supervises food products circulating in the community. The supervision carried out by BPOM are preventive and repressive. One form of supervision carried out by BPOM is granting distribution permits for packaged processed food products before they are distributed to the public. Keywords: packaged processed food products, BPOM
