Vibrio alginolyticus is a halophilic organism usually found in marine environments. It has attracted attention as an opportunistic pathogen of aquatic animals and humans, but there are very few reports on polyhydroxyalkanoate (PHA) production using V. alginolyticus as the host. In this study, two V. alginolyticus strains, LHF01 and LHF02, isolated from water samples collected from salt fields were found to produce poly(3-hydroxybutyrate) (PHB) from a variety of sugars and organic acids. Glycerol was the best carbon source and yielded the highest PHB titer in both strains. Further optimization of the NaCl concentration and culture temperature improved the PHB titer from 1.87 to 5.08 g/L in V. alginolyticus LHF01. In addition, the use of propionate as a secondary carbon source resulted in the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV). V. alginolyticus LHF01 may be a promising host for PHA production using cheap waste glycerol from biodiesel refining.

Background: In recent years, environmental pollution and heavy metal pollution caused by rapid urbanization and industrialization have become increasingly serious. Among them, mercury (II) ion (Hg2+) is one of the highly toxic heavy metal ions, and its pollution comes from various natural resources and human activities. Therefore, people attach great importance to the development of analytical methods for effective analysis and sensitive detection of Hg2+ . Objective: Using grape skin as a green and environmental friendly carbon source, to synthesize fluorescent carbon dots, and try to apply them to the detect the concentration of Hg2+ in water. Method: Using "Hutai No. 8" grape skin as carbon source, fluorescent carbon dots were synthesized by one-step hydrothermal method. Structure and fluorescent properties of the carbon dots were tested using TEM, XPS, XRD and other characterization instruments, and their utilization on detection of mercury ions in the actual water samples was explored. Results: The CDs had a particle size of about 4.8 nm and a spherical shape. There are N-H, C-N, C=O and other functional groups on the surface. It was found that Hg2+ has obvious fluorescence quenching effect on CDs, and thus CDs fluorescence quenching method to detect the concentration Hg2+ was established, and the detection limit is 3.7 μM, which could be applied to test the concentration of Hg2+ in water samples. Conclusion: Using grape skin as carbon source, fluorescent carbon dots were successfully synthesized by hydrothermal method. Carbon dots were used to detect mercury ions in water, and a method for detecting mercury ions in actual water samples was established.

Abstract After the Elizabethkingia anophelis outbreak in Wisconsin, USA, an active search for the detection of the microorganism in hospital water systems from Central and Northern Greece was performed from June to December 2016. In total, 457 water samples from 11 hospitals were analyzed. Elizabethkingia spp. was detected in three samples collected from two hospitals, both of which are located in Northern Greece. Two of the three isolated strains were identified as Elizabethkingia anophelis. No cases of Elizabethkingia infection were reported in either hospital during 2016. This is the first reported isolation of the pathogen in water supply systems in Greece.

Abstract Background Sugars and triglycerides are common carbon sources for microorganisms. Nonetheless, a systematic comparative interpretation of metabolic changes upon vegetable oil or glucose as sole carbon source is still lacking. Selected fungi that can grow in acidic mineral salt media (MSM) with vegetable oil had been identified recently. Hence, this study aimed to investigate the overall metabolite changes of an omnipotent fungus and to reveal changes at central carbon metabolism corresponding to both carbon sources. Results Targeted and non-targeted metabolomics for both polar and semi-polar metabolites of Phialemonium curvatum AWO2 (DSM 23903) cultivated in MSM with palm oil (MSM-P) or glucose (MSM-G) as carbon sources were obtained. Targeted metabolomics on central carbon metabolism of tricarboxylic acid (TCA) cycle and glyoxylate cycle were analysed using LC–MS/MS-TripleQ and GC–MS, while untargeted metabolite profiling was performed using LC–MS/MS-QTOF followed by multivariate analysis. Targeted metabolomics analysis showed that glyoxylate pathway and TCA cycle were recruited at central carbon metabolism for triglyceride and glucose catabolism, respectively. Significant differences in organic acids concentration of about 4- to 8-fold were observed for citric acid, succinic acid, malic acid, and oxaloacetic acid. Correlation of organic acids concentration and key enzymes involved in the central carbon metabolism was further determined by enzymatic assays. On the other hand, the untargeted profiling revealed seven metabolites undergoing significant changes between MSM-P and MSM-G cultures. Conclusions Overall, this study has provided insights on the understanding on the effect of triglycerides and sugar as carbon source in fungi global metabolic pathway, which might become important for future optimization of carbon flux engineering in fungi to improve organic acids production when vegetable oil is applied as the sole carbon source.

The effects of the presence of a secondary carbon source on the biodegradation of chlorinated phenols were studied with column biofilm reactors. The biodegradability of chlorinated phenols was studied with a series of batch reactors. The biodegradability of chlorinated phenols was to follow the order of phenol > 2,4-dichlorophenol > 4-chIorophenol > 2,4,6-trichlorophenol > 2-chlorophenol > 3-chloro-phenol. The presence of a relatively more biodegradable but higher chlorinated phenol, such as 2,4,6-trichlorophenol, enhanced the biodégradation of a less chlorinated butrecalcitrantphenol, such as 2-chlorophenol. The addition of phenol, an easily biodegradable compound, generally decreased the biodegradation of chlorinated phenols.
