Phenolic compounds in the fruit of American cranberry (Vaccinium macrocarpon Aiton) determine the antioxidant, anti-inflammatory, anticancer, and other biological effects. The berries are used in the production of medicinal preparations and food supplements, which highlights the importance of qualitative and quantitative analysis of phenolic compounds in cranberry fruit raw material. The aim of our study was to develop and validate an efficient, cost-effective, reproducible, and fast UPLC-DAD methodology for the evaluation of the qualitative and quantitative composition of phenolic compounds in raw material and preparations of American cranberry fruit. During the development of the methodology, chlorogenic acid and the following flavonols were identified in cranberry fruit samples: myricetin-3-galactoside, quercetin-3-galactoside, quercetin-3-glucoside, quercetin-3-α-L-arabinopyranoside, quercetin-3-α-L-arabinofuranoside, quercetin-3-rhamnoside, myricetin, and quercetin. The developed and optimized UPLC-DAD methodology was validated according to the guidelines of the International Council for Harmonization (ICH), evaluating the following parameters: range, specificity, linearity (R2 > 0.999), precision (%RSD < 2%), LOD (0.38–1.01 µg/mL), LOQ (0.54–3.06 µg/mL), and recovery (80–110%). The developed methodology was applied to evaluate the qualitative and quantitative composition of phenolic compounds in fruit samples of cranberry cultivars ‘Baifay’, ‘Bergman’, ‘Prolific’, and ‘Searles’, as well as ‘Bain-MC’ and ‘BL-12′ clones. In the tested samples, the majority (about 70%) of the identified flavonols were quercetin derivatives. The greatest amount of quercetin-3-galactoside (1035.35 ± 4.26 µg/g DW) was found in fruit samples of the ‘Searles’ cultivar, and the greatest amount of myricetin-3-galactoside (940.06 ± 24.91 µg/g DW) was detected in fruit samples of the ‘Woolman’ cultivar.

The aim of this study was to determine the composition and content of phenolic compounds in ethanol extracts of eight different cultivars of American cranberry (Vaccinium macrocarpon Aiton) fruit using spectrophotometric and UPLC-ESI-MS/MS analysis and to evaluate the antioxidant activity in vitro of these extracts. The highest total amount of phenolic compounds evaluated via Folin–Ciocalteu spectrophotometry was detected in American cranberry fruit samples of the ‘Bain’ clone, and the highest total amount of flavonoids was found in samples of the ‘Drever’ and ‘Baiwfay’ cultivars. The highest total amount of the individual phenolic compounds (519.53 ± 25.12 mg/g DW) identified and quantitatively evaluated via chromatography was detected in samples of the ‘Searles’ cranberry cultivar. In the studied cranberry samples, the predominant phenolic compounds were hyperoside, quercetin, and procyanidin A2, while the amounts of other compounds were significantly lower. HCA and PCA revealed that ‘Woolman’, ‘Holliston’, ‘Pilgrim, and ‘Searles’ fruit samples had different quantitative content of phenolic compounds from other cranberry cultivars. Meanwhile, fruit of ‘Baiwfay’, ‘Drever’, ‘Bain’, and ‘Bergman’ were similar in their phytochemical profile.

Our research aim was to investigate the quantitative composition of neochlorogenic acid, chlorogenic acid, rutin, hyperoside and isoquercitrin in leaves and fruit of introduced rowan plants to Lithuania using a HPLC method. In leaf and fruit samples, quantitative estimation of neochlorogenic acid (in leaf samples 1.11–11.49 mg/g, in fruit samples 0.93–5.43 mg/g), chlorogenic acid (in leaf samples 2.74–21.91 mg/g, in fruit samples 0.55–7.50 mg/g), rutin (in leaf samples 0.03–4.88 mg/g, in fruit samples 0.02-0.39 mg/g), hyperoside (in leaf samples 0.15–7.44 mg/g, in fruit samples 0.02–1.19 mg/g) and isoquercitrin (in leaf samples 0.75–5.24 mg/g, in fruit samples 0.02-0.65 mg/g) was performed. In leaf samples, the total content of identified phenolic compounds ranged from 7.18 to 35.74 mg/g. In fruit, raw material total content of neochlorogenic acid, chlorogenic acid and identified flavonoids varied from 2.24 to 11.19 mg/g. Comparison of raw material samples of Sorbus species according to their quantitative composition of identified phenolic compounds using cluster analysis singularized S. commixta leaf and fruit samples. These contained the highest contents of chlorogenic acid, hyperoside and isoquercitrin, but also the lowest contents of neochlorogenic acid and rutin. The highest contents of neochlorogenic acid were recorded for S. lancifolia leaves, and of rutin for S. anglica leaves.

As the interest in heirloom cultivars of apple trees, their fruit, and processed products is growing worldwide, studies of the qualitative and quantitative composition of biological compounds are important for the evaluation of the quality and nutritional properties of the apples. Studies on the variations in the chemical composition of phenolic compounds characterized by a versatile biological effect are important when researching the genetic heritage of the heirloom cultivars in order to increase the cultivation of such cultivars in orchards. A variation in the qualitative and quantitative composition of phenolic compounds was found in apple samples of cultivars included in the Lithuanian collection of genetic resources. By the high-performance liquid chromatography (HPLC) method flavan-3-ols (procyanidin B1, procyanidin B2, procyanidin C2, (+)-catechin and (−)-epicatechin), flavonols (rutin, hyperoside, quercitrin, isoquercitrin, reynoutrin and avicularin), chlorogenic acids and phloridzin were identified and quantified in fruit samples of heirloom apple cultivars grown in Lithuania. The highest sum of the identified phenolic compounds (3.82 ± 0.53 mg/g) was found in apple fruit samples of the ‘Koštelė’ cultivar

Berries of Actinidia kolomikta (A. kolomikta) are known for high ascorbic acid content, but the diversity of phenolic compounds has been little studied. The present research aimed to investigate phenolic compounds and antioxidant activity in berries and leaves of twelve A. kolomikta cultivars. The UHPLC-ESI-MS/MS technique was used to determine differences among cultivars in the quantitative composition of individual phenolic compounds. Antioxidant activity was determined by DPPH• free radical scavenging and CUPRAC methods. In the present study, 13 phenolic compounds were detected in berries, whereas leaves contained 17 phenolic compounds. Flavonols were the primary class found in both berries and leaves; other identified phenolic compounds were flavan-3-ols, flavones and, phenolic acids; and dihydrochalcone phloridzin was identified in the leaves. The amount and variety of phenolic compounds in berries and leaves and antioxidant activity were found to be cultivar-dependent. The highest total content of phenolic compounds was found in the leaves of the cultivar ‘Aromatnaja’ and in the berries of the cultivar ‘VIR-2’. Results of this study have confirmed that berries and leaves of A. kolomikta could be a valuable raw material for both food and pharmaceutical industries.

High-performance liquid chromatography method has been used to evaluate qualitative and quantitative composition of certain phenolic compounds obtained from areal parts of Artemisia dracunculus L. introduced in Zhytomyr Polissya. 31 phenolic compounds have been detected, among which the following flavonoids were identified: rutin (1.30±0.04 mg/g), luteolin-7-glycoside (0.34±0.03), apigenin-7-glycoside (0.30±0.01) and isochlorogenic acid (0.16± 0.02 mg/g), where rutin was the dominant component. The total amount of phenolic compounds in air-dried raw material constituted 51.24±0.12 mg/g (5.12%). The employed chromatography analysis of phenolic compounds from Artemisia dracunculus areal parts shows that the plant may be considered as a valuable source of biologically active compounds of phenolic origin. Phenolic compounds are likely to determine the antimicrobial properties of the plant, established earlier. Further in-depth study and cultivation of Artemisia dracunculus in Zhytomyr Polissya have a great potential for pharmaceutical and food industries, cosmetics and development of therapeutic antioxidant diets.

The qualitative and quantitative composition of two species of Oplopanax root was studied by gas chromatography with mass spectrometric detection. Extraction of components from the investigated objects was carried out by extraction of dry ground raw material with 70% ethanol. For the analysis of polar compounds, the extracts were further derivatized to give the corresponding trimethylsilyl derivatives. Identification of components was carried out using commercial (NIST17, Wiley14) and own custom mass-spectrometer libraries. The percentage of components found was calculated using the areas of the corresponding chromatographic peaks. Found in these samples were 130 compounds of various classes: terpenes and their derivatives, alcohols, aldehydes, polyyns, polyenes, various acids and their derivatives, phenols, sterols, lignans, vitamins, nucleosides, glycosides and sugars. It has been established that the qualitative and quantitative composition of root extracts varies even within the same species. The presence of 26 compounds common to the two species of Oplopanax, including 12 acids, 3 polyyns, 6 terpenes and one representative each from the class polyenes, aldehydes, lignans, glycosides and sterols, was revealed. As a result of the comparative analysis, it was established that 4 compounds from the class of polyyns (falcarindiol acetate, oplopanediol acetate, oplopantriol A and oplopantriol B) are found only in the roots of O. horridus.

Apple (Malus domestica Borkh.) fruits are rich in phenolic glycosides, triterpenic acids and other biologically active compounds. The apples are widely used as food products due to their biologically active compounds that have specific biological effects. It is important to use high quality apples or their recycling products, so it is necessary to investigate the qualitative and quantitative composition of the bioactive compounds. The amount of triterpenic acids varies during different phenological stages of apple development and maturation. In order to determine in which phenological stage of apple growth the quantity of triterpenic acids was highest, high-performance liquid chromatography was applied. The highest total amounts of triterpenic compounds were detected at the beginning of the phenological stage of fruit development. The study showed that the highest amounts of triterpenic acids were detected at the beginning of the phenological stage of apple development, while as the apple matured, the amount of triterpenic acids decreased from 2.63 ± 0.26 mg/g to 1.6 ± 0.28 mg/g. In this study, we identified and quantified four triterpenic compounds, which by the quantitative composition of triterpenic acids could be arranged in the following ascending order: betulinic acid < corosolic acid < oleanolic acid < ursolic acid. In order to use herbal extracts for medical practice it is important to perform biological effects study in vitro and in vivo. Antioxidants with different mechanisms of action neutralize harmful reactive oxygen and nitrogen forms and enhance antioxidants protection systems. The antioxidant activity of apple extracts in vitro varied during different phenological stages of the fruit. The strongest antiradical and reductive activities were observed at the beginning of apple development. In order to determine the relationship between the antioxidant activity of the acetone extracts of apples assessed by DPPH, ABTS, CUPRAC and FRAP assays and the total amount of triterpenic acids in these extracts, a correlation analysis was carried out. The strongest positive correlation was observed between the amount of oleanolic acid and the antioxidant activity assessed by DPPH, ABTS and CUPRAC methods (respectively, r = 0.778, r = 0.784 and r = 0.720).
