The effects of microplastics (MP) are extensively studied, yet hazard data from long-term exposure studies are scarce. Moreover, for sustainable circular use in the future, knowledge on the biological impact of recycled plastics is essential. The aim of this study was to provide long-term toxicity data of virgin vs recycled (mechanical recycling) low density polyethylene (LDPE) for two commonly used ecotoxicity models, the freshwater crustacean Daphnia magna and the terrestrial crustacean Porcellio scaber. LDPE MP was tested as fragments of 39.8 ± 8.82 µm (virgin) and 205 ± 144 µm (recycled) at chronic exposure levels of 1–100 mg LDPE/L (D. magna) and 0.2–15 g LDPE/kg soil (P. scaber). Mortality, reproduction, body length, total lipid content, feeding and immune response were evaluated. With the exception of very low inconsistent offspring mortality at 10 mg/L and 100 mg/L of recycled LDPE, no MP exposure-related adverse effects were recorded for D. magna. For P. scaber, increased feeding on non-contaminated leaves was observed for virgin LDPE at 5 g/kg and 15 g/kg. In addition, both LDPE induced a slight immune response at 5 g/kg and 15 g/kg with more parameters altered for virgin LDPE. Our results indicated different sublethal responses upon exposure to recycled compared to virgin LDPE MP.

A szabadföldi trágyázási (tartam)kísérletek eredményeit talaj-, illetve diagnosztikai célú növényvizsgálatok segítségével tudjuk kiterjeszteni, általánosítani – figyelembe véve természetesen a kiterjesztés korlátait is. Kísérleti eredményeink megerősítették a Sarkadi-féle AL-P korrekciós modell helytálló voltát. Fenti megállapításunkat ugyanakkor a növényi P-tartalmakkal való összefüggéseknek is igazolniuk kell. Szükséges tehát a talajvizsgálati eredményeknek a diagnosztikai célú növényvizsgálatokkal, valamint a terméseredményekkel való összevetése. A tartamkísérletek talajai lehetőséget nyújtanak a környezetvédelmi célú P-vizsgálatok értékelésére, a talaj P-feltöltöttsége környezeti kockázatának becslésére. Ezekkel a kérdésekkel a cikksorozat további részeiben kívánunk foglalkozni.

The small GTPase RhoA, and its down-stream effector ROCK kinase, and the interacting Rac1 and mTORC2 pathways, are the principal regulators of the actin cytoskeleton and actin-related functions in all eukaryotic cells, including the immune cells. As such, they also regulate the phenotypes and functions of macrophages in the immune response and beyond. Here, we review the results of our and other’s studies on the role of the actin and RhoA pathway in shaping the macrophage functions in general and macrophage immune response during the development of chronic (long term) rejection of allografts in the rodent cardiac transplantation model. We focus on the importance of timing of the macrophage functions in chronic rejection and how the circadian rhythm may affect the anti-chronic rejection therapies.

The live genetically-engineered oral rabies virus (RABV) variant SPBN GASGAS induces long-lasting immunity in foxes and protection against challenge with an otherwise lethal dose of RABV field strains both after experimental oral and parenteral routes of administration. Induction of RABV-specific binding antibodies and immunoglobulin isotypes (IgM, total IgG, IgG1, IgG2) were comparable in orally and parenterally vaccinated foxes. Differences were only observed in the induction of virus-neutralizing (VNA) titers, which were significantly higher in the parenterally vaccinated group. The dynamics of rabies-specific antibodies pre- and post-challenge (365 days post vaccination) suggest the predominance of type-1 immunity protection of SPBN GASGAS. Independent of the route of administration, in the absence of IgG1 the immune response to SPBN GAGAS was mainly IgG2 driven. Interestingly, vaccination with SPBN GASGAS does not cause significant differences in inducible IFN-γ production in vaccinated animals, indicating a relatively weak cellular immune response during challenge. Notably, the parenteral application of SPBN GASGAS did not induce any adverse side effects in foxes, thus supporting safety studies of this oral rabies vaccine in various species.

Global concerns for the sustainability of agriculture have emphasized the need to reduce the use of mineral fertilizer. Although phosphorus (P) is accumulated in farmland soils due to the long-term application of fertilizer, most soil P is not readily available to plants. The chemical speciation of P in soils, which comprise heterogeneous microenvironments, cannot be evaluated with a high degree of specificity using only macroscopic analyses. In this study, we investigated the distribution and speciation of P accumulated in soils by using both macro- and microscopic techniques including chemical extraction, solution and solid-state 31P NMR, bulk- and micro- P K-edge X-ray absorption near edge structure (XANES), and electron probe microanalysis (EPMA). Soil samples were collected from a field in which cabbage was cultivated under three amendment treatments: i) mineral fertilizer (NPK), ii) mineral fertilizer and compost (NPK + compost), and iii) mineral fertilizer plus compost but without nitrogen fertilizer (PK + compost). Macro-scale analyses suggested that accumulated P was predominantly inorganic P and associated with Al-bearing minerals. The repeated application of compost to the soils increased the proportion of P associated with Ca which accounted for 17% in the NPK + compost plot and 40% in the PK + compost plot. At the microscale, hot spots of P were heterogeneously distributed, and P was associated with Fe and Ca in hot spots of the NPK + compost (pH 6) and PK + compost (pH 7) treated samples, respectively. Our results indicate that application of compost contributed to creating diverse microenvironments hosting P in these soils.

Background: The COVID-19 pandemic demanded a global effort towards quickly developing safe and effective vaccines against SARS-CoV-2. Objective: This review aimed to discuss the main vaccines available, their mechanisms of action, results of clinical trials and epidemiological behavior. The implications of viral variants were also debated. Methods: A non-systematic literature review was performed between February and March 2021 by searching the Pubmed, Scopus, and SciELO databases, using different combinations of the following terms: "vaccines", "clinical trials" , "SARS-CoV-2", "Coronavirus", "COVID-19", "mechanisms of action". Data regarding clinical trials of SARS-CoV-2 vaccines and epidemiological information were also searched. Results: The mechanisms of action included vector-virus, mRNA and inactivated virus vaccines. The vaccines showed positive results in phases 2/3 clinical trials. The efficacy of the mRNA 1273 and of mRNA BNT 162b2 vaccines were 94.1% and 95%, respectively. The effectiveness of the ChAdOx1 nCoV-19 vaccine varied according to the scheme, with an overall value of 70.4%. The Gam-COVID-Vac vaccine had an efficacy of 91.6%. Regarding the Ad26.COV2.S vaccine, 99% or more of seroconversion was observed in all subgroups 29 days after vaccination. The CoronaVac vaccine induced an immune response in 92% of the volunteers receiving 3ug and in 98% with 6ug, in comparison to 3% in the placebo group. Conclusion: Global efforts have resulted in vaccines available in record time, with good safety and immunogenicity profile. However, only long-term studies can provide more information on duration of immunity and the need for additional doses.

Abstract Aims The objectives of this study were to examine the long-term substitution of mineral phosphorus (P) fertilizers with manure (M) plus nitrogen (N) fertilizers and how they affect the forms of P that occur in soil, soil P distribution, and plant growth. Methods We used a solution of 31P nuclear magnetic resonance (31P-NMR) spectroscopy to study the correlations between long-term fertilization regimes and the forms of P that occur at different soil depths. Then we investigated yield, plant growth, and soil properties. Results A 40-year field experiment showed that the use of M + N fertilizers can significantly improve plant growth and yield. The proportion of organic P in the 20-40 cm soil layer was significantly increased by long-term M fertilization. The concentrations of various forms of P (orthophosphate, pyrophosphate, diesters, monoesters, and total inositol hexakisphosphate, IHP) in topsoil increased significantly with the combination of M with N + P mineral fertilization. The addition of M greatly increased the stereoisomers of IHP (myo-IHP, scyllo-IHP, neo-IHP, and D-chiro-IHP) and the proportion and concentration of corrected diesters. There were no significant differences in the pyrophosphate contents of the 40-60 cm soil layer according to fertilization type and year of fertilization. There were also no significant differences in IHP stereoisomers and diesters according to fertilization year. The P forms that contributed to corn yield were orthophosphate, diester, and IHP. Further, pyrophosphate made no significant contribution to corn growth. Conclusions Over the long-term, pig manure can significantly increase the amount of orthophosphate that is directly absorbed by crops and the amount of IHP stereoisomers that can be used by plants. Orthophosphate and IHP are the two key factors that have a positive effect on plant growth.

Phosphorus fixation in tropical soils may decrease under no-till. In this case, P fertilizer could be surface-spread, which would improve farm operations by decreasing the time spend in reloading the planter with fertilizers. In the long term, less soluble P sources could be viable. In this experiment, the effect of surface-broadcast P fertilization with both soluble and reactive phosphates on soil P forms and availability to soybean was studied with or without fertilization with soluble P in the planting furrow in a long-term experiment in which soybean was grown in rotation with Ruzigrass (Brachiaria ruziziensis). No P or 80 kg ha-1 of P2O5 in the form of triple superphosphate or Arad reactive rock phosphate was applied on the surface of a soil with variable P fertilization history. Soil samples were taken to a depth of 60 cm and soil P was fractionated. Soybean was grown with 0, 30, and 60 kg ha-1 of P2O5 in the form of triple phosphate applied in the seed furrow. Both fertilizers applied increased available P in the uppermost soil layers and the moderately labile organic and inorganic forms of P in the soil profile, probably as result of root decay. Soybean responded to phosphates applied on the soil surface or in the seed furrow; however, application of soluble P in the seed furrow should not be discarded. In tropical soils with a history of P fertilization, soluble P sources may be substituted for natural reactive phosphates broadcast on the surface. The planting operation may be facilitated through reduction in the rate of P applied in the planting furrow in relation to the rates currently applied.

Although COVID 19 infections in children are generally mild and non-fatal, there is increasing recognition of a multisystem inflammatory syndrome in children (MIS-C) associated with COVID-19, leading to severe illness and long-term sequelae. Even though there is some evidence that the MIS-C is a post-viral immunological reaction to COVID-19, understanding of the immune response induced by SARS-CoV-2 remains unclear.Various local and international guidelines are being widely practiced in the diagnosis and management of patients with MIS-C. A case report of a young child who was diagnosed and managed as MIS-C is discussed here. The diagnostic challenges with the available case definitions and currently accepted treatment options are elaborated herein.
