Background: Though the neuroprotective roles of ascorbic acid are well established, the therapeutic role of nicotine in various neurological disorders is attracting increasing attention. This study evaluated the putative ameliorative role of the synergetic treatment of nicotine and ascorbic acid against neurodegenerative consequences associated with free radical species and amyloid plaques generation in adult male Wistar rats Methods: A total of 35 Wistar rats were distributed into five groups labeled A-E. Group A served as the control group; animals in group B were treated with 100mg/kg body weight of aluminium chloride (AlCl3) for 21 days. Group C animals were treated with 100mg/kg body weight of aluminium chloride for 21 days and post-treated with 14mg/kg body weight of nicotine for 21 days. Group D was treated with 100mg/kg body weight of aluminium chloride for 21 days and post-treated with 100mg/kg bodyweight of ascorbic acid for 21 days. Group E animals were treated with 100mg/kg body weight of aluminium chloride for 21 days and post-treated with 100mg/kg bodyweight of ascorbic acid and 14mg/kg body weight of nicotine. On completion of treatments, the prefrontal cortex was excised and processed for biochemical and histochemical examinations. Results: Oxidative stress was evident from the diminished level of catalase and glutathione per oxidase and elevated lipid peroxidation levels in animals administered with aluminium in addition to the presence of amyloid plaques in these animals. However, synergetic administration of ascorbic acid and nicotine attenuated these oxidative and histochemical perturbations induced by aluminium. Conclusion: Synergetic treatment with ascorbic acid and nicotine provided better ameliorative potential against aluminium-induced neurotoxicity compared to either ascorbic acid or nicotine treatments alone

The metabolic response to nutrient ingestion and the rate of digestion and absorption of nutrient molecules in bowel physiology plays an important role in the metabolic control of some human chronic non-infectious diseases. This experimentally-controlled designed nutritional study which lasted eight weeks aimed to determine the effects of proportional high-protein/low-carbohydrate (HP/LC) formulated diet on glycemic tolerance, glycemic control, body weight, organ weight and organ morphometry in healthy and diabetic adult male Wistar rats. Twenty-four male Wistar rats purchased from a disease-free stock were randomly categorized into four groups (n = 6, each) after two weeks acclimatization period in raised stainless steel cages with 6 mm2mesh floor and replaceable numbered blotters papers placed under each cage in a well-ventilated animal house. Animal groups include: Healthy control group (HC), Healthy treated group (HT), Diabetic control group (DC) and Diabetic treated group (DT. The animals were fed according to the experimental design with water ad libitumfor eight weeks. Diabetes was inducted with freshly prepared alloxan monohydrate solution (150 mg/kg bw, intraperitoneally). Body weights and fasting blood sugar concentrations were measured twice weekly, while oral glucose tolerance test was conducted on the last day of the eighth-week study and subsequently followed by organs extraction after anesthesia for weight and gross assessment. Proportional high-protein/low-carbohydrate formulated diet caused significant reduction in mean body weight of treated diabetic (DT: 22.6%; P= .001) and healthy (HT: 5.8%; P= .007) rats while the control animals on control diet recorded significant (P< .05) increase in body weight gain (DC: 12.4%; HC: 11.2%). Glycemic tolerance and control improved significantly in diabetic treated rats over that of the healthy treated rats. Gross morphometry of the extracted organs (kidneys, liver, heart, lungs, spleen and testes) revealed sustained normal morphological features without any visible lesion. In conclusion, consumption of proportional high-protein/low-carbohydrate formulated diet enhanced body weight reduction and sustained normal organ morphological features with good glycemic tolerance and control in experimental rats, suggesting its dietary potentiality, safety and suitability to ameliorate obesity-related diabetes.

This study aims to look at the histopathological picture of the dorsal arteries of the penis of the hiperlipidemic wistar rats (Rattus novergicus) induction by high-fat diet that seen in terms of lumen diameter and thickness of the arterial endotelium wall. Hyperlipidemia is a risk factor for ateriosclerosis in the penile arteries causing erectile dysfunction in men. This study is an experimental study with a randomized posttest only control goup design. The sample are  10 individuals adult male wistar rats aged 3-4 months with a range of body weight 150-200 grams. Before treatment, adaptation was carried out for 7 days. After that the sample rats in the treatment group were made hyperlidemic by feeding lard for 50 days. Then surgery is performed for histopathological preparations at the posttest. To determine the differences in endotelium thickness and arterial lumen diameter in the two groups, an independent t-test was used. Thick diameter data of the endotelium and dorsal arteries of the penis of the wistar rat between the lower treatment group and the control group. The difference that occurred was statistically significant (p <0.05). So it can be concluded that the provision of high-fat diet (hyperlipidemia) decreases the lumen diameter and endotelium thickness of dorsal arteries penis. Cell proliferation and apoptosis in the remnant rat kidney after treatment with lowdose irradiation was investigated. Material and methods. In the first group (n=9), adult male Wistar rats underwent 5/6 nephrectomy (NPX); in the second group (n=9), NPX was combined with low-dose irradiation. Rats without surgery and irradiation formed the control group (n=9). Results. Hypertension and proteinuria induced by NPX were decreased by 3-Gy irradiation. The 5/6 NPX rats showed a dramatic increase in proliferating and apoptotic cells in the glomeruli and in the distal tubules at week 2, which was significantly decreased by low-dose irradiation. Conclusion. The data demonstrate that low-dose irradiation is a factor slowing the process of chronic renal injury.

Background: Medicinal plants are of great importance to health of individual and communities. About 80% of the population in Uganda relies on traditional medicine because western-trained medical personnel are limited especially in villages. Most Ugandans use Hymenoxys odorato for medicinal purposes e.g. to treat colds, fever, coughs, anti-helminthes, locally used as tea, anti-allergy and also as an anti-venom to relieve snake bites. Method: A group of 25 male wistar rats of 150 g&ndash;210 g were kept for 14 days while being fed and treated with the extract. At 14th day, anesthesia was given and blood samples collected by cardiac puncture for hematological and biochemical investigations. Serum was analyzed for Alkaline Phosphatase, Aspartate Transaminase and Alanine Transaminase while whole blood was used for complete blood count. The liver and kidney were removed and placed in 10% formalin to prepare for histology staining using haematoxylin and eosin technique. Results: The extract elevated hepatic biomarker enzymes i.e. ALP, ALT and AST. The increase was found to be significantly different (P &gt; 0.05) at 400 and 500 mg/kg doses as compared to the control group. Histological sections of the liver showed distortion of liver cytoarchitecture, steatosis, necrosis of hepatocytes and congestion of the sinusoids at high doses 300, 400 and 500 mg/kg body weight. In the sections of the kidney, there was mild distortion of the integrity of the kidney with glomerular hypercellularity at high doses (400 and 500 mg/kg per body weight). Conclusion: Hymenoxys odorato aqueous extract has toxic effects on the liver and kidney of wistar rats. The effects were observed to be in a dose dependent manner.

Background : The 1st and 2nd degree hemorrhoids is managed non-operatively with the anti-inflammatory and plebothropic drugs. Graptophyllum pictum extract (GPE) has already been used widely in Indonesia to treat hemorrhoid with good result, however, the mechanism is not supported by the molecular research.Objective : This study is intended to study the phlebothropic effects of GPE by measuring the degree of edema and extravassal leucocytes of experimental wistar hemorrhoid.Methods : Experimental study with Randomized Controlled Trial Post-test only design in male wistar rats, weight around 200 gr, induced for the development of a disease-like condition of hemorrhoids by 6% croton oil on the anus for 3 days. Random allocation was performed to divide the 14 wistar rats in 2 groups. Group I as control got normal saline solution, while group II was treated with GPE 100mg/kgbw, started on day 4th for 5 consecutive days. On 9th day blood was extracted from retroorbital fossa and anus was resected up to 2 cm from anal verge and weighted. The degree of anal edema was measured by rectoanal coefficient, that is rasio between anal weight (miligrams) and body weight (grams).Results : Until the end of study, all wistar rat were still alive. The mean (±SD) of body weight   of control group was 173.84 ­(±13.37) and the treatment group was 171.70  (±13.10), and there was no significant differences (p = 0.833). The mean (±SD) of rectoanal coefficient in the treatment group was 2.46 (±0.41) and it was significantly lower than control group  (3.13 ± 0.85) (p = 0.029). The mean (±SD) of extravassal leukocytes in the treatment group was 900.14(±48.09) and it was significantly lower than the control (1003.28 ± 99.30) (p = 0.042).Conclusions : Graptophyllum pictum extract at doses of 100 mg/Kgbw have phlebotonic effect  in decreased edema and extravassal leukocytes.

ABSTRACT Excessive energi consumption of high-fat diet increase the body’s adipose tissue which also affect the increased secretion of leptin, this is because leptin secretion is proportional to the mass of adipose tissue. Leptin activates the lateral hypothalamus as the center of hunger and feeding activity. The aim of this study was to obtain the relationship between high trans fat feeding on male Wistar rats and feeding activity. This method uses male Wistar rats (Rattus norvegicus) as an animal model. Sixteen rats were grouped into 4 treatment groups. The first group was given additional feed 20% trans fat, the second group was given additional feed 40% trans fat, a third group was given additional feed 60% trans fat, and the last as a control group. This feed was given once a day for 70 days. Observation of activities perform using Opto-varimex® auto-track system ver. 4.31. Abstract: Weight is one measure that gives description of the tissue mass including body fluids. Weight change is nfluenced by nutriment consumption. Sago contains high carbohydrates that can increase body weight. This study aimed to obtain the effect of sago compared to rice to body weight of wistar rats. This was a random laboratory experimental pre-post test with control group design. Subjects were male wistar rats, aged 5-6 months. The rats were divided in two groups: rice group as control and sago group. Sago was cooked in papeda form as much as 75 g of dried sago and 300 mL water. The rats were fed for 2 weeks. Data were analyzed by using the Wilcoxon test. The results showed that the sago group showed an increase of body weight significantly (p=0,001) meanwhile the rice group lose body weight significantly (p=0,001). Conclusion: Sago can increase body weight of wistar rats significantly.Keywords: body weight, sago, riceAbstrak: Berat badan merupakan salah satu ukuran yang memberikan gambaran massa jaringan termasuk cairan tubuh. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian sagu terhadap berat badan dibanding nasi pada tikus wistar. Desain penelitian ini ialah eksperimental laboratorium acak pre-post test with control group design. Subjek penelitian ialah tikus wistar jantan berusia 5-6 bulan dibagi atas 2 kelompok: kelompok nasi (kontrol) dan kelompok sagu. Sagu dimasak dalam bentuk papeda sebanyak 75 g sagu kering dan 300 mL air. Tikus diperlihara selama 2 minggu. Berat badan diukur dengan menggunakan timbangan. Data dianalisis dengan uji Wilcoxon. Hasil penelitian menunjukkan bahwa pemberian sagu sebanyak 2 cc setiap hari selama seminggu mampu meningkatkan berat badan secara bermakna (p = 0,001) sedangkan pemberian nasi menurunkan berat badan (p=0,001). Simpulan: Pemberian sagu dapat meningkatkan berat badan tikus wistar secara bermakna.Kata kunci: berat badan, sagu, nasi

Seaweed has potential nutrient content such as carotenoids, vitamins, fatty acids, carbohydrates, minerals, and other essential substances. Carotenoids have important biological functions as an antioxidant, and immunostimulatory which can prevent the disease, anti-inflammatory, anti-stress, anti-aging, and protect the skin from the harmful effects of ultraviolet radiation. Seaweed generally consumed as a vegetable by people in Bali, known as the local name Bulung Boni (Caulerpa spp.) and Bulung Sangu (Gracilaria spp.).. So far there has been no report or results of research on the effects of extract ethanol of Bulung Boni (Caulerpa sp.) and Bulung Sangu (Gracilaria sp.) as an antioxidant that can prevent lipid peroxidation which can be seen in decreased level of MDA in liver tissue or blood plasma. Therefore it is necessary to determine of plasmaMDA level of Wistar rat after fed high cholesterol diet treated with extract ethanol of Caulerpa sp. and Gracillaria sp. This experimental study used completely randomized design. Research using total of 24 Wistar rats divided into six sample groups of equal size, all fed with a diet high in cholesterol especially in negative control. The study consisted of negative control group (standard diet), positive control group (high cholesterol diet), high-cholesterol diet with Caulerpa sp. extract dose of 20 mg and 60 mg/100 g, high cholesterol diet with Gracilaria sp. extract dose of 20 mg and 60 mg/100 g body weight rat per day.The study resulted that rats fed high cholesterol diet with treated extract ethanol Caulerpa sp. and Gracilaria sp. with a dose of 20 mg and 60 mg per 100 g body weight rat / day had plasma MDA level significantly lower (p <0.05) compared with rats fed high cholesterol diet without treated with extract of Caulerpa sp. and Gracilaria sp.

Background: Rats are commonly used animals in development of newer drugs, rectification of toxicity and to record the various alterations in physiological parameters following pharmacological and non pharmacological interventions.Aim and Objectives: The purpose of this study was to identify the best physiological window during anesthesia. Therefore, we compared the effect of anesthesia using combination of ketamine and xylazine (KX) and thiopental sodium (intraperitoneally) on blood pressure and heart rate in adult male Wistar rats. Material and Methods: Twelve, male Wistar rats with a mean body weight of 260 ± 15 g were acquired. Thiopental sodium and cocktail of ketamine and xylazine (KX) were administered (ip) in group- I and group-II respectively. The systolic blood pressure and heart rate was recorded in both the groups till the awakening phase.Results: We found that there was a constant SBP and HR in Ketamine/Xylezine groups that are from 30 to 90 minutes after injection of anesthesia while this window was not observed in thiopental group.Conclusion: Our study concludes that the best time to observe the effect of newer drug during period between 30- 90 minutes after anesthesia.Asian Journal of Medical Sciences Vol.9(6) 2018 37-39

AbstractMale Wistar rats were deprived of food for 3 days and then forced to swim continuously to a criterion of exhaustion. Controls swam after ad libitum feeding. In the first experiment a load equal to 11% of the rat's body weight was attached to the dorsal skin near the tail during the swim. In a second study, two matched groups, treated identically as those in the first study swam without a load in water treated with a wetting agent, "Aerosol O.T." In a third experiment, controls as well as experimentals were deprived of food for three days. The control group, however, was fed for 30 minutes, immediately before the swim. A fourth experiment was conducted in a similar manner as the third, except both groups were deprived 6 hours. In each of the experiments the deprived groups swam significantly longer than the fed groups.
