Alcea L. is one of the largest genera of Malvaceae family with nearly 70 species worldwide mainly distributed in SW Asia. According to the latest revision of the family, it is represented by 34 species in the Flora of Iran, among them, 15 species are endemic. It is tough to accurate germplasm/ plant recognition by using morphological characteristics because of its propagation, growing and using. We conducted a molecular data analysis on these plant species due to their importance. We examined 156 plants from 14 species in 16 regions that were selected randomly for this investigation. It has been 119 polymorphic bands (94.33%) were resulted from 128 bands of 10 primers in amplification of genomic DNA. ISSR primers have a great capacity to detect polymorphic loci among Alcea species, as evidenced by the high average PIC and MI values found. The genetic similarity of 14 species was calculated and ranged between 0.635 to 0.990. Inter-Simple sequence repeats (ISSR) markers research revealed that Alcea tarica Pakravan & Ghahreman and Alcea kopetdaghensis lljin had the least similarity, while Alcea semnanica Pakravan and Alcea mazandaranica Pakravan & Ghahreman had the most. The current study attempts to answer three questions: 1) can ISSR markers identify Alcea species? 2) what is the genetic structure of these taxa in Iran? and 3) what is the inter-relationship between these taxa? The current study discovered that ISSR markers can be used to identify species.

Hypericum L. species are generally known locally in Iran with the names “Hofariqun” which Ebn Sina (or Bo Ali Sina) called it. Plants of the genus Hypericum have traditionally been used as medicinal plants in various parts of the world. Hypericum perforatum L. is the source to one of the most manufactured and used herbal preparations in recent years, especially as a mild antidepressant. Therefore, due to the importance of these plant species, we performed a molecular data for this species. For this study, we used 175 randomly collected plants from 17 species in 9 provinces. Amplification of genomic DNA using 10 primers produced 141 bands, of which 127 were polymorphic (95.78%). The obtained high average PIC and MI values revealed high capacity of ISSR primers to detect polymorphic loci among Hypericum species. The genetic similarities of 17 collections were estimated from 0.617 to 0.911. According to Inter-Simple sequence repeats (ISSR) markers analysis, H. androsaemum and H. hirtellum had the lowest similarity and the species of H. perforaturm and H. triquetrifolium had the highest similarity. The aims of present study are: 1) can ISSR markers identify Hypericum species, 2) what is the genetic structure of these taxa in Iran, and 3) to investigate the species inter-relationship? The present study revealed that ISSR markers can identify the species.

Identifying the accurate boundaries of a species is critical to have a better perspective of any biological studies. Therefore, species delimitation is a subject of extensive part of studies in the framework of biology. Erodium species possess significant pharmacological and biological activities. The whole plant was used as astringent and haemostatic in uterine and other bleeding Therefore, due to the importance of these plant species, we performed a molecular data analysis for this species. For this study, we used 60 randomly collected plants from 5 species in five provinces. Amplification of genomic DNA using 10 primers produced 52 bands, of which 50 were polymorphic (98.48%). The obtained high average PIC and MI values revealed high capacity of ISSR primers to detect polymorphic loci among Erodium species. The genetic similarities of five collections were estimated from 0.77 to 0.91. According to Inter-Simple sequence repeats (ISSR) markers analysis, E. cicutarium and E. malacoides had the lowest similarity and the species of E. malacoides and E. oxyrrhynchum had the highest similarity. The aims of present study are: 1) can ISSR markers identify Erodium species, and 2) to investigate the species inter-relationship? The present study revealed that ISSR markers can identify the species.

AbstractTo investigate the genetic diversity and phylogenetic relationships between polyploid Leymus and related diploid species of the Triticeae tribe, inter-simple sequence repeats (ISSR) markers was used to analyze 41 Leymus accessions representing 22 species and 2 subspecies, together with Pseudoroegneria stipifolia (St), Psathyrostachys fragilis (Ns), Australopyrum retrofractum (W), Hordeum bogdanii, H. chilense (H) and Lophopyrum elongatum (Ee). A total of 376 clear and reproducible DNA fragments were amplified by 29 ISSR primers, among which 368 (97.87%) fragments were found to be polymorphic. 8–18 polymorphic bands were amplified by each polymorphic primer, with an average of 12.69 bands. The data of 376 ISSR bands were used to generate Nei’s similarity coefficients and to construct a dendrogram by means of UPGMA. The similarity coefficients data suggested great genetic diversity in genus Leymus and related diploid Triticeae species, the genetic diversity among the different species more abundant than that of the different accessions. The dendrogram and principal coordinate analysis showed explicit interspecific relationships and demonstrated close phylogenetic relationships between Leymus species and Psathyrostachys.

Species identification is fundamentally important within the fields of biology, biogeography, ecology and conservation. The genus Paracaryum belongs to tribe Cynoglosseae of the family Boraginaceae is a herbaceous genus including approximately 67 species, mostly distributed in the Irano-Turanian phytogeographical region. In spite vast distribution of many Paracaryum species that grow in different habitats, there are not any available report on their genetic diversity, mode of divergence and patterns of dispersal. Therefore, we performed molecular (ISSR markers) of 98 accessions from 12 species of Paracaryum that were collected from different habitats. A set of 10 ISSR markers was used. The genetic distances were estimated based on Jaccard similarity coefficient and the descriptive statistics of populations for estimation of genetic parameters were also performed. A total of 90 polymorphic bands were obtained. The present study revealed that ISSR data can delimit the species. AMOVA and STRUCTURE analysis revealed that the species of Paracaryum belongs are genetically differentiated but have some degree of shared common alleles.

Aerva javanica is one of Egypt’s most important traditional medicinal plants used as antidiarrheal and anthelmintic medicine and recently as an anticancer agent. In this study, variations among ten populations of Aerva javanica in different sites in the Eastern Desert of Egypt were analyzed based on morphological and ecological attributes and molecular variation expressed by Inter-Simple Sequence Repeat (ISSR) markers. Morphological diversity was higher for populations in the Wadi El-Markh and Bir Abbady regions than others. The polymorphism revealed by ten ISSR primers was 79.4% among populations. Distance trees created using the results obtained from soil variables, morphological characterizations, and molecular data showed that the highest similarity was 0.974 between Populations 8 and 9, while the lowest similarity was 0.715 between Population 1 and Population 3 regions. In conclusion, the obtained data are important to design a plan for sustainable conservation of Aerva javanica as an important medicinal plant having a wide interspecific genetic variability within various populations.

ABSTRACT The jaboticaba tree (Plinia sp.) is a native fruit tree belonging to the Myrtaceae family, which occurs spontaneously throughout Brazil. The estimation of genetic divergence among genotypes of native populations can be useful for conservation and knowledge of available genetic resources, in the formation of gene banks and breeding species. In order to evaluate the genetic diversity of jaboticaba trees identified in the Bahia Reconcavo municipalities, 35 genotypes were characterized by ISSR primers (Inter Simple Sequence Repeats). The 18 primers generated a total of 463 amplicons, and the number of initiator fragments ranged from 14 to 36, with an average of 25.72% and 99.65% polymorphism. Polymorphic information content ranged from 0.13 to 0.33 with a mean of 0.22 and the marker resolution power from 4.34 to 15.77, with an average of 8.67. Multivariate analysis allowed the formation of five groups of genetic divergence, where the longest distance was 0.97 between JCA6 and JSF8 genotypes and the lower of JMT2 and JCA1 (0.11). From the use of ISSR markers it is possible to check the variability between genotypes on the evaluated jaboticaba trees.

Background: Crataegus spp. (Hawthorn) are used for horticultural and medicinal purposes. Objectives: In the present study, the genetic relationships among the five Iranian Crataegus species were evaluated by Inter Simple Sequence Repeats (ISSR) molecular marker Methods: PCR reactions were performed using six primers (811, 827, 834, 845, 856, and 868) and genetic similarity was calculated based on Jacquard’s similarity coefficient. Results: Six primers generated 79 products in total, of which 71 were polymorphic (89.9%), with an average of 13.1 bands per primer. The percentage of polymorphic bands ranged from 77 to 100. Primer 856 produced the highest number of bands, while the lowest was generated by primer 845. The Jacquard’s similarity coefficient, derived from ISSR marker analysis, ranged from 0.164 to 0.337, indicating high genetic variation among Crataegus species in Iran. Conclusion: This study provides important data for identifying species relationships and helps develop plant breeding strategies to improve the medicinal properties of this genus in the future.

Abstract This study was carried out to estimate the genetic diversity and relationships of 74 Iranian blackberry genotypes assigned to 5 different species using inter-simple sequence repeats (ISSR) marker analysis and morphological trait characterization. Sixteen traits including phenological, vegetative and reproductive attributes were recorded, and 10 ISSR primers were screened. Results showed that yield and leaf width have the highest and lowest genetic diversity, (diversity index = 62.57 and 13.74), respectively. Flowering and ripening date recorded as traits having the strongest correlations (r = 0.98). The selected 10 ISSR primers produced a total of 161 amplified fragments (200 to 3500 bp) of which 113 were polymorphic. The highest, lowest and average PIC values were 0.53, 0.38 and 0.44, respectively. Principle component analysis (PCA) based on morphological traits showed that the first six components explained 84.9% of the variations of traits studied, whilst the principal coordinate analysis (PCoA) based on ISSR data implied the first eight principal coordinates explained 67.06% of the total variation. Cluster analysis based on morphological traits and ISSR data classified all genotypes into two and three major groups, respectively, and the distribution pattern of genotypes was mainly based on species and the geographic origins.

Dracocephalum is the second largest genus in the family Lamiaceae with about 186 species. These species are native in temperate regions of the Northern Hemisphere and occur in the territory of the extra-tropical Asia and Europe. Eight Dracocephalum species reported in Iran; these are mainly growing in the northern and central parts of the country belonging to the Irano-Turanian phytogeographical region. Dracocephalum kotschyi is an important medicinal plant .in the country. At the same time, taxonomic position of Dracocephalum oligadenium is a challenging issue. In this work morphological, anatomical and Inter Simple Sequence Repeat (ISSR) markers were used to identify these species in Iran. MDS plot based on morphological and anatomical characters, furthermore, PCoA and MST plot based on ISSR data of species revealed hybridization between D. oligadenium and D. kotschyi.
