Single chain variable fragments (scFv) have widely been used in research, diagnosis and treatment, but the scFv is considered as difficult protein for expression in E. coli. In previous studies, we expressed a construction of recombinant single chain variable fragments again antigen specific for blood type A (antiA-scFv) individually or fused with Trx or SUMO. However, soluble fraction was low abandant and only approximately 40% when fused with Trx, the other cases were expressed in form of inclusion body. Therefore, it was difficult for purification, refolding and activity assesment. In thispaper, we demonstrated a suitable construction for soluble production of antiA-scFv fused with SUMO (SM/antiA-scFv) in presence of chaparones. Under fermentation with 0.1 mM IPTG at 20oC, the SM/antiA-scFv was entirely expressed in soluble form. Importantly, after cleavage from SUMO with SUMOprotease, antiA-scFv was still maintained in the supernatant fraction. Therefore, it can help ensure bioactivity and is useful for purification process. To the best of our knowledge, this is the first report showing soluble recombinant scFv fused with SUMO in presence of chaperone for determination of blood group antigens. Thus, this result facilitates the optimal study of soluble expression, purification and bioactivity determination of the antiA-scFv recombinant antibody.

This article was under review for ICELTICS 2018 -- In the medical world there is still service dissatisfaction caused by lack of blood type testing facility. If the number of tested blood arise, a lot of problems will occur so that electronic devices are needed to determine the blood type accurately and in short time. In this research we implemented an Artificial Neural Network on Xilinx Spartan 3S1000 Field Programable Gate Array using XSA-3S Board to identify the blood type. This research uses blood sample image as system input. VHSIC Hardware Discription Language is the language to describe the algorithm. The algorithm used is feed-forward propagation of backpropagation neural network. There are 3 layers used in design, they are input, hidden1, and output. At hidden1layer has two neurons. In this study the accuracy of detection obtained are 92%, 92%, 92%, 90% and 86% for 32x32, 48x48, 64x64, 80x80, and 96x96 pixel blood image resolution, respectively.

Malaria is known as an endemic disease that often causes death in Indonesia, especially in Papua. The malaria cases control in Papua has not been carried on based on data studies, therefore bionomic of Anopheles sp is important to be studied. Bionomics data are consisted of breeding places, resting places and feeding habits are from direct observation. Interviews and questionnaires were conducted to gain information about respondent behavior. Descriptive and qualitative data were then analyzed. The breeding places of Anopheles sp. were mostly found in swampy areas. Based on the feeding habit, the feeding activity of Anopheles sp. inside the house has only one biting peak at 23.00-02.00 WIT, while outside the house, biting peaks occurred at 21.00-22.00 WIT and 00.00-01.00 WIT. Resting place data shown that Anopheles sp. mostly found in piles of clothes and shoe racks. Recorded factors that affect mosquitos bionomics are temperature, humidity, salinity, pH, community behavior, and the presence of livestock. Environmental factors (temperature, humidity, salinity, and pH), habitual behavior of host (3M action, the habit of using insect repellent, mosquito repellent, and mosquito nets), the presence of livestock, and the type of bait blood type affect mosquito activity.

<p>Penelitian ini membangun suatu sistem pakar untuk menentukan menu makanan sehat berdasarkan golongan darah dan tingkat kadar kolesterol pasien dengan metode Forward Chaining. Tujuan untuk membantu orang awam dalam menentukan menu makanan sehat untuk pasien kolesterol. Sistem ini menganalisa masalah penentuan menu makanan sehat berdasarkan golongan darah dan tingkat kadar kolesterol pasien. Hasil yang diperoleh dari sitem pakar ini adalah berupa informasi makanan sehat yang akan dikonsumsi oleh pasien kolesterol dengan jenis golongan darah dan tingkat kadar kolesterol yang berbeda. Analisa dilakukan dengan cara mengetahui jenis golongan darah dan tingkat kadar kolesterol pasien yang ditampilkan oleh program sistem pakar ini, rancangan sistem ini menggunakan inference forward chaining, dengan implementasi sistem menggunakan sistem database Microsoft Office Access dan bahasa pemrograman Visual Basic 6.0. Dari rancangan aplikasi sistem pakar yang dibuat, maka orang awam yang memderita kolesterol dapat menentukan menu makanan sehat untuk di konsumsi berdasarkan golongan darah dan tingkat kadar kolesterol dengan menjalankan aplikasi sistem pakar.</p><p><em><br /></em></p><p><em><em>This study builds an expert system to determine the healthy food menu based on blood type and cholesterol levels of patients with Forward Chaining method. The goal is to help the layman in determining a healthy diet for cholesterol patients. This system analyzes the problem of determining healthy food menu based on blood group and patient cholesterol level. The results obtained from this expert system is in the form of healthy food information that will be consumed by cholesterol patients with the type of blood group and different cholesterol levels. From the design of expert system applications created, the layman who memderita cholesterol can determine the healthy diet to be consumed by blood type and cholesterol level by running an expert system application.<br /> <br /> </em></em></p>

Abstract Background Although criteria for liver transplantation, such as the Milan criteria and Hangzhou experiences, have become popular, criteria to guide adjuvant therapy for patients with hepatocellular carcinoma after liver transplantation are lacking. Methods We collected data from all consecutive patients from 2012 to 2019 at three liver transplantation centers in China retrospectively. Univariate and multivariate analyses were used to analyze preoperative parameters, such as demographic and clinical data. Using data obtained in our center, calibration curves and the concordance Harrell’s C-indices were used to establish the final model. The validation cohort comprised the patients from the other centers. Results Data from 233 patients were used to construct the nomogram. The validation cohort comprised 36 patients. Independent predictors of overall survival (OS) were identified as HbeAg positive (P = 0.044), blood-type compatibility unmatched (P = 0.034), liver transplantation criteria (P = 0.003), and high MELD score (P = 0.037). For the validation cohort, to predict OS, the C-index of the nomogram was 0.874. Based on the model, patients could be assigned into low-risk (≥ 50%), intermediate-risk (30–50%), and high-risk (≤ 30%) groups to guide adjuvant therapy after surgery and to facilitate personalized management. Conclusions The OS in patients with hepatocellular carcinoma after liver transplantation could be accurately predicted using the developed nomogram.
