AIM: This study aims to prove the superior outcome of collagen nanosilver in increasing macrophage and collagen on deep dermal burn wound healing. METHODS: This study was a randomized control trial design. All animals used were male Sprague Dawley rats aged 12–14 weeks old and weighed about 200–250 g. Each rat gets four site injuries and is given treatments. The treatment group includes: I (negative control using NaCl 0.9%), II (positive control using silver sulfadiazine cream), III (treatment with nanosilver collagen cream 1%), and IV (treatment with collagen nanosilver cream 2%). Macrophages counts were assessed using IHC F40/80 and collagen density was assessed using Masson’s trichrome staining. RESULTS: Macrophage on day 6 showed a significant difference as seen in group III and group IV. A significant difference was also obtained between macrophage on day 3 and day 6 from group IV, likewise between collagen between day 3 and day 6 from group III and IV. Collagen nanosilver acts as an antimicrobial and anti-inflammatory agent by increasing the mechanism of activation and migration of macrophages to the site of injury. Moreover, this formula modulated the collagen deposition, which forms the tissue fibers which ultimately provide a conducive environment for wound healing. CONCLUSION: Collagen nanosilver has a superior outcome in the increasing macrophage cell count and collagen density of deep dermal burn wound healing.

Burn wound healing process consists of three phases of inflammation, proliferation and remodelling. The number of fibroblasts has increased during the proliferation phase. Oxidative stress in burn may disrupt proliferation through TGF-β inhibition, result in decreased number of fibroblasts and poor burn wound healing. Genistein in edamame seeds has anti-inflammatory, antibacterial and antioxidant properties. Edamame contains vitamins A, C, and E which is also have antioxidant properties. Edamame has been suggested to protect burn wound healing from oxidative stress. This study aimed to understand the effectiveness of edamame seed (Glycine max L. Merril) ethanolic extract (ED) to fibroblast count in second degree burn wound healing. This is true experimental research with post test only control group design. The sample of this research was 24 rat models of second degree burn which was made by attaching hot metal (60 ° C) for 5 seconds on the shaved back of the rats. The sample was divided into 6 groups with topical treatment, among others: positive control group (silver sulfadiazine), negative control group (Na CMC 0.5%), group with ED 20%, ED 40%, ED 60%, and ED 80%. Treatment was given for 15 days and skin sample was taken on the 16th day. Observation of the number of fibroblasts was done by taking pictures of histhologic preparations in Hematoxylin and Eosin staining under 400x magnification of light microscope. In one way ANOVA test, significance value of 0.011 (p≤0,05) indicated that there was at least a significant difference between 2 groups. LSD’s post hoc analysis showed that there was a significant difference in the group ED 20, 40, and 60% compared with negative control. Bandotan plants (Ageratum conyzoides (L.) L.) commonly used as agent in the treatment of inflammatory, wound, and also used used as analgesic, stimulant and tonic. In this study we carried out a test of the effectiveness of the cream (w/o) of ethanolic extract of bandotan leaves in wound healing in streptozocin-induced hyperglycemia mice. Animals were divided into five groups: Group I (without induced, given cream base w/o), group II (induced, given cream base w/o), group III (induced, given cream containing 3% extract), group IV (induced, given cream containing 5% extract) and group V (induced, given cream containing 7% extract). Animals also divided into three subgroups based on time of decapitation on 7th, 14th and 21th days. The parameters observed were the percentage of wound healing and collagen fibers density scores. Results were analyzed by Two-Way ANOVA. The results showed that the cream in concentration of 3%, 5% and 7% could affect wound healing in hyperglycemia mice. The percentage of wound healing of the cream in concentration of 3%, 5% and 7% were significantly different (p<0,05) with positive control, and density scores of collagen fibers was also significantly different (p<0,05) from the positive control and negative control. However, the best results was obtained from cream containing 5% extract which perrcentage of wound healing reached 98.5% on 21th day and got a score of 3 for density of collagen fibers that had been formed on the 14th day.

Background: The process of wound healing is influenced by various factors such as age, hormones, and wound care. Wound care is done to accelerate wound healing which can be done by various methods, one of them is traditional care. Traditional wound care can use medicinal plants. Rhodomyrtus tomentosa is a medicinal plant that has an antioxidant, anti-inflammatory, antitumor and antibacterial content. Thus this study aims to evaluate the effectiveness of the antiseptic solution of the Rodhomyrtus tomentosa leaf extract on wound healing in male Wistar rats. Method: this research is pure experimental research with post test only control group design. Thirty male white rats were divided into five groups, namely negative control, positive control, Rhodomyrtus tomentosa leaf extract 15%, 30%, and 60%. Rhodomyrtus tomentosa leaf extraction was carried out by maceration method with 70% ethano solvent. The extraction results are divided into 3 concentrations (15%, 30% and 60%). The wound healing process was evaluated by measuring the length of the wound manually from 0 to 10 days in each group. Meanwhile, the number of fibroblast cells was calculated through hematoxylin eosin (HE) staining and observed using an Olympus CX41 microscope with a 10x magnification and objective lens magnification in 3 fields. Result: There was a significant difference in the reduction in wound length (p =< 0,000) between the five experimental groups (Rhodomyrtus tomentosa leaf extract solution 15%, 30% and 60%, negative control and positive control. Solution of rhodomyrtus tomentosa leaf extract accelerated the increase in the number of fibroblasts compared to the negative control group (p = 0.003), but did not make a difference (p = 0.403) with the positive control group. Rhodomyrtus tomentosa leaf extraction solution had the same microscopic effect on the number of fibroblasts with a positive control group given 0.9% NaCl solution. Conclusion: There was a significant difference in the number of fibroblasts between all groups, but no difference in wound healing length.

AbstractThe purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs.Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and immunohistochemical analyses. In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs.These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.
