The aim of the study was to determine the phenolic and flavonoid content of essential oils (EOs), chloroform and ethanolic extracts of 12 Algerian Thymus species and evaluate their antioxidant and antifungal activities. EOs (1.73 ± 0.30–15.00 ± 1.24 μg/mg), chloroform extracts (33.8 ± 2.42–160.93 ± 3.88 μg/mg) and ethanol extracts (27.01 ± 3.56 –148.46 ± 4.40 μg/mg) showed considerable phenolic content. Flavonoids values of chloroform extracts ranged between 3.39± 0.17 and 20.27 ± 0.29 μg/ml while ethanolic extracts values ranged between 2.81 ± 0.11 and 26.64 ± 0.18 μg/mg. Results of DPPH showed that EOs, chloroform and ethanolic extracts exhibited strong radical scavenging activity (IC50 = 21.75 ± 6.54–338.22 ± 2.99 μg/ml, 22.91 ± 5.59–90.93 ± 1.36 μg/ml, and 33.51 ± 5.72–103.80 ± 4.54 μg/ml, respectively). Inhibition of β-carotene bleaching was potentially performed by all EOs (66.48 ± 2.41–94.06 ± 2.68 %), chloroform extracts (68.98± 1.58–95.30± 1.99%), and ethanolic extracts (62.15 ± 2.51–92.36± 1.15%). The antifungal activity of EOs and extracts was tested using the minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC). The EOs (0.1 ± 0.00 mg/ mL – 1.06 ± 0.46 mg/mL), chloroform (0.1 ± 0.00 mg/ mL –1.06 ± 0.46 mg/mL) and ethanol (0.1 ± 0.00 mg /mL–1.6 ± 0.00 mg/mL) showed remarkable antifungal activity against mycotoxigenic Aspergillus genera. The MFC of EOs (1.0 ± 0.34 mg/mL and > 4.8 mg/mL) , chloroform (0.26 ± 0.11 mg/mL and > 1.6 mg/mL) and ethanol (0.2 ± 0.00 mg/mL and > 1.6 mg/mL) were fungicidal in nature higher than MICs. The findings of the study indicated that Thymus spp. EOs and extracts could be used as natural alternatives for food industry.

ABSTRACT In this study, radical scavenging and antifungal activities of Chaerophyllum macropodum and Oliveria decumbens essential oils (EOs) extracted with microwave-assisted hydrodistillation (MAHD) were evaluated in comparison with the same EOs extracted by conventional hydrodistillation (HD). The final EO yields that were obtained using HD (after 150 min) and MAHD (after 45 min) were 1.72 and 1.67% for C. macropodum and 8.10 and 7.91% for O. decumbens, respectively. There were no significant differences between the final EO yields extracted with HD and MAHD, but MAHD could significantly reduce the duration of the extraction operation. Average rates of EO accumulation (grams per minute) with MAHD were at least three times higher than with HD. Gas chromatography–mass spectrometry analysis of EOs indicated that there were no significant differences between the composition of EOs extracted by HD and MAHD. Both plants showed high radical scavenging activity, with 50% inhibitory concentration values of 0.430 to 0.431 mg/mL for C. macropodum and 0.142 to 0.146 mg/mL for O. decumbens. Antifungal activity was performed against six fungal species, including Aspergillus niger, Aspergillus oryzae, Penicillium chrysogenum, Trichoderma harzianum, Byssochlamys spectabilis, and Paecilomyces variotii. A. niger and A. oryzae were the most resistant fungi, and T. harzianum was the most susceptible. Evaluation of MIC and minimum fungicidal concentration values showed that the O. decumbens EOs were very active against all the tested fungi, which can be attributed to the high amounts of oxygenated terpenes in the EO content. Therefore, MAHD as a fast extraction technique did not have any adverse effects on chemical composition, radical scavenging activity, and antifungal activity of C. macropodum and O. decumbens EOs.

The chemical composition of eight (seven shoot and one inflorescence) essential oils (EOs) of Rh. tomentosum H. plants growing in Eastern Lithuania is reported. The plant material was collected during different phases of vegetation (from April to October). The oils were obtained by hydrodistillation from air-dried aerial parts (leaves and inflorescences). In total, up to 70 compounds were identified by GC−MS and GC (flame-ionization detector, FID); they comprised 91.0 ± 4.7%–96.2 ± 3.1% of the oil content. Sesquiterpene hydrocarbons (54.1 ± 1.5%–76.1 ± 4.5%) were found to be the main fraction. The major compounds were palustrol (24.6 ± 2.6%–33.5 ± 4.4%) and ledol (18.0 ± 2.9%–29.0 ± 5.0%). Ascaridol isomers (7.0 ± 2.4%–14.0 ± 2.4% in three oils), myrcene (7.2 ± 0.3% and 10.1 ± 1.3%), lepalol (3.3 ± 0.3% and 7.9 ± 3.0%), and cyclocolorenone isomers (4.1 ± 2.5%) were determined as the third main constituents. The toxic activity of marsh rosemary inflorescence and shoot oils samples was evaluated using a brine shrimp (Artemia sp.) bioassay. LC50 average values (11.23–20.50 µg/mL) obtained after 24 h of exposure revealed that the oils were notably toxic. The oil obtained from shoots gathered in September during the seed-ripening stage and containing appreciable amounts of palustrol (26.0 ± 2.5%), ledol (21.5 ± 4.0%), and ascaridol (7.0 ± 2.4%) showed the highest toxic activity. Radical scavenging activity of Rh. tomentosum EOs depended on the plant vegetation stage. The highest activities were obtained for EOs isolated from young shoots collected in June (48.19 ± 0.1 and 19.89 ± 0.3 mmol/L TROLOX (6-hydroxy-2,5,7,8-tetra-methylchromane-2-carboxylic acid) equivalent obtained by, respectively, ABTS•+ (2,2′-amino-bis(ethylbenzothiazoline-6-sulfonic acid) diammonium salt) and DPPH•(2,2-diphenyl-1-picrylhydrazyl) assays). Agar disc diffusion assay against pathogenic yeast Candida parapsilosis revealed the potential antifungal activity of EOs. An alternative investigation of antifungal activity employed mediated amperometry at yeast Saccharomyces cerevisiae-modified electrodes. The subjection of yeast cells to vapors of EO resulted in a three to four-fold increase of electrode responses due to the disruption of yeast cell membranes.

Essential oils, also known as volatile oils, are substances produced through the secondary metabolism of plants. In this study, we determined the chemical composition and the in vitro and in vivo antifungal activity of the essential oils from four species of Eucalyptus, Eucalyptus citriodora, Eucalyptus camaldulensis, Eucalyptus grandis and Eucalyptus microcorys, against the Hemileia vastatrix fungus. The essential oils from these four species of Eucalyptus were extracted from their leaves by the hydrodistillation technique using a modified Clevenger apparatus. The chemical characterization was performed by gas chromatography coupled with a mass spectrometer detector and by gas chromatography using a flame ionization detector. The antifungal activities of the essential oils against H. vastatrix were studied by evaluating the percentage of spore germination using the microdilution test for in vitro assays. The curative and preventive effects were evaluated in in vivo tests. The principal constituents of the essential oil from E. citriodora were citronellal, citronellol and isopulegol, while E. camaldulensis produced 1,8-cineole, α-terpineol and α-pinene. 1,8-cineole, α-pinene and α-terpineol were obtained from E. grandis and 1,8-cineole, α-pinene and trans-pinocarveol were the principal components in the essential oil of E. microcorys. In vitro and in vivo antifungal activities against the fungus under study were observed for most of the essential oils, except the essential oil from E. microcorys, for which no preventive antifungal activity was observed. Only the curing of infection by the H. vastatrix fungus was observed with this oil.

The antibacterial activities of the ethanol extracts of 35 medicinal plants were screened against Escherichia coli, Bacillus cereus, and Staphylococcus aureus by following standard antimicrobial susceptibility testing procedures. The ethanol extracts of Coptis chinensis and Glycyrrhiza uralensis showed antibacterial activity against all three test bacterial species. The screened extracts were also tested for their antioxidant activities using the DPPH free radical scavenging activity assay, and for their cytotoxic and antioxidant activity in vitro using an assessment of cell viability on mouse embryonic fibroblast cells. The ethanol extracts of both C. chinensis and G. uralensis showed good radical scavenging antioxidant properties and attenuated the rate of cell death caused by oxidative damage. Our results showed that C. chinensis and G. uralensis are promising sources of natural products with good antibacterial and antioxidant activities.

In the present study, in vitro hydroxyl radical scavenging activities, total phenols and terpene contents in 70% ethanol extracts were evaluated. Samples of crushed (CB) and non-crushed ripe juniper berries (NCB) collected at five localities in North-East Slovakia during the years 2012–2014 were compared. Standard or modified methods for determining phytochemical and antioxidant activity were used together with a novel method for the evaluation of the results after the correction of the measured values per gram of dry matter (DM). Statistically significantly higher DM contents (average values for three years) were found in the CB extracts (ranging from 18.86 to 21.91 mg/mL) in comparison to those for NCB (ranging from 2.59 to 9.90 mg/mL). Depending on the localities and years, the hydroxyl radical scavenging activity ranged from 15.52 to 32.85% for NCB and from 65.59 to 88.12% for CB, respectively. The contents of total phenols ranged from 43.75 to 246.75 mg gallic acid equivalents (GAE)/L (NCB) and from 151.84 to 278.19 mg GAE/L (CB), respectively. However, the higher content of total phenols per gram of DM was found in the NCB extracts (8.49–42.23 mg GAE/g DM) and then in CB (6.87–18.77 GAE/g DM). The results obtained in this study showed a higher efficiency for extraction from juniper berries in 70% ethanol if the pericarp was disrupted in comparison to that achieved with the maceration of intact berries.

The fruits of Heracleum aquilegifolium Wight (Apiaceae) were collected from Western Ghats of the Indian Peninsula. The essential oils were extracted by hydrodistillation. The chemical composition of the essential oils was analysed by gas chromatography and gas chromatography-mass spectrometry (GC-MS). β -Pinene (22.3%), 1,8-cineole (20.3%), and β-phellandrene (12.4%) were the main components of H. aquilegifolium fruit oils. The antioxidant properties of essential oils of H. aquilegifolium were examined by different procedures namely reducing power ability, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, nitric oxide radical scavenging activity, hydrogen peroxide scavenging activity, hydroxyl radical scavenging activity, superoxide anion scavenging activity, and metal chelating activity. The antioxidant activities were compared with those of synthetic antioxidants and standard drugs such as butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT), ascorbic acid, α-tocopherol, curcumin, and quercetin. The study confirmed the possible antioxidant potential of essential oils tested with various in vitro antioxidant methods. The presence of monoterpenes in combination with other components in the oils could be responsible for the activity

Antifungal activity of seven essential oils was studied primarily against C. albicans and E. floccosum. Four essential oils viz. turmeric, palmarosa, lemongrass and citronella, that revealed better antifungal activities during screening, were characterized further. Minimum Inhibitory concentration (MIC) and phenol coefficient values of the oils ranged between 0.97 to 62.5 µl/ml and 0125 to 0.75 respectively. The oils retained the antifungal activities when treated at high temperature (1000 C for 1h) and pressure (autoclaved), indicating presence of thermostable and barostable active components in them. The oils also resisted sonication (33,000Hz for 30min) and revealed antifungal activities against the two pathogens. Immediate killing of E. floccosum, is attributable to an irreversible damage to the cells. Antifungal activity of these oils as recorded during the study, could be attributable to the membrane inhibition mechanism and was observed to be fungicidal in nature.

Objective: The aim of the present study was to evaluate the alpha (α)-amylase and alpha (α)-glucosidase inhibitory activities and in vitro antioxidant activities of the 80 % aqueous ethanol extracts of Tinosporasinensis Lour (Merr.).Methods: The 80% aq. ethanol extract of the plant was prepared. The plant extract was examined for its antioxidant activity by using free radical 1,1-diphenyl-2-picryl hydrazyl (DPPH) scavenging method, ABTS radical scavenging ability, reducing power capacity, estimation of total phenolic content, flavonoid content and flavonol content. Different concentrations (2, 4, 8,10and 15 μg/ml) of the extract was subjected to α-amylase inhibitory and α-glucosidase inhibitory activities and IC50were calculated.Results: The study revealed that the different concentrations of the plant extract possessed a very good amount of total phenolics, flavonoid and flavonol and exhibited potent radical scavenging activity using DPPH and ABTS as a substrate. The ethanol extracts exhibited significant α-amylase and α-glucosidase inhibitory activities with an IC50 value1.093µg and 1.04µg dry extract respectively and well compared with standard acarbose drug.Conclusion: Thus, it could be concluded that due to the presence of antioxidant components the plant extracts could be used for the treatment of hyperglycemia, diabetes and the related condition of oxidative stress. This knowledge will be useful in finding more potent components from the natural resources for the clinical development of antidiabetic therapeutics.

This study was designed to examine the in vitro antioxidant DPPH free radical-scavenging activity, and the total phenolic and flavonoid content of ethanol and methanol extracts of Salvia amplexicaulis Lam. in the whole plant and different parts, leaves, stems and flowers. The largest amounts of extract yield were obtained from the flowers, 14.14% and 12.00 % (w/w) in the ethanol of methanol extracts, respectively. The ethanol extract of leaves (16.07 ?g/ml) and methanol extract of the whole plant (21.28 ?g/ml) showed the highest activity against the DPPH radical. The ethanol extract of the leaves was the richest in phenols (222.40 mg GAE/g) and flavonoids (49.81 mg QE/g), whereas the methanol extract of the whole plant contained the highest amount of phenolics (180.89 mg GAE/g) and flavonoids (38.15 mg QE/g). A very strong linear correlation between antioxidant activity and the phenolic content of the extracts was established. The obtained results suggest that S. amplexicaulis could be regarded as an important source of natural antioxidants.

The chemical composition, antioxidant activity, and antimicrobial properties of three commercially available essential oils: rosemary (REO), lavender (LEO), and mint (MEO), were determined in the current study. Our data revealed that the major components of REO, MEO, and LEO were 1,8-cineole (40.4%), menthol (40.1%), and linalool acetate (35.0%), respectively. The highest DPPH radical-scavenging activity was identified in MEO (36.85 ± 0.49%) among the investigated EOs. Regarding antimicrobial activities, we found that LEO had the strongest inhibitory efficiencies against the growth of Pseudomonas aeruginosa and Candida (C.) tropicalis, MEO against Salmonella (S.) enterica, and REO against Staphylococcus (S.) aureus. The strongest antifungal activity was displayed by mint EO, which totally inhibited the growth of Penicillium (P.) expansum and P. crustosum in all concentrations; the growth of P. citrinum was completely suppressed only by the lowest MEO concentration. The lowest minimal inhibitory concentrations (MICs) against S. enterica, S. aureus, and C. krusei were assessed for MEO. In situ analysis on the bread model showed that 125 µL/L of REO exhibited the lowest mycelial growth inhibition (MGI) of P. citrinum, and 500 µL/L of MEO caused the highest MGI of P. crustosum. Our results allow us to make conclusion that the analysed EOs have promising potential for use as innovative agents in the storage of bakery products in order to extend their shelf-life.
