Purpose: To investigate the antioxidant and anti-inflammatory properties of three different Rubus coreanus Miq. by-products in stimulated BV-2 microglial cells and explore its underlying physiological efficacy.Methods: Cell viability assessment was performed by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Lipopolysaccharide (LPS) was used to activate BV-2microglia. Total polyphenol and total flavonoid contents were determined by the method of Folin-Denis. As three different Rubus coreanus Miq. by-products remaining after extraction of Rubus coreanus, High performance liquid chromatography (HPLC)) finger printing, ABST (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging assay, and NO (nitric oxide) inhibitory assay were performed.Results: Three different Rubus coreanus by-product extract did not exhibit any signs of cytotoxicity to BV-2 cells up to 100 μg/ml concentration (p < 0.5). The LPS-activated excessive release of NO in BV-2 cells was significantly inhibited by Rubus coreanus by-product extract (p < 0.5) at 500 μg/mL). Total polyphenol and total flavonoid contents were highest in 50 % ethanol wine processing by-product (p < 0.5 at 30, 50, 70 and 100 %, respectively). The by-product of wine processing had the lowest RC50 radical scavenging effect (16.53 μL/ml). The quercetin content of the wine processing by-product was the highest in the 70% ethanol extract at 6.26 mg/g (p < 0.5 at 30, 50, 70 and 100%, respectively).Conclusion: These results reveal that of the three other by-products, wine processing by-product has the highest antioxidant and anti-inflammatory activities. The use of these by-products has high added value for industrial production; furthermore, they are a potential treatment for various inflammatory diseases.

Epilobium angustifolium L. is a popular and well-known medicinal plant. In this study, an attempt to evaluate the possibility of using this plant in preparations for the care and treatment of skin diseases was made. The antioxidant, antiaging and anti-inflammatory properties of ethanolic extracts from Epilobium angustifolium (FEE) were assessed. Qualitative and quantitative evaluation of extracts chemically composition was performed by gas chromatography with mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC). The total polyphenol content (TPC) of biologically active compounds, such as the total content of polyphenols (TPC), flavonoids (TFC), and assimilation pigments, as well as selected phenolic acids, was assessed. FEE was evaluated for their anti-inflammatory and antiaging properties, achieving 68% inhibition of lipoxygenase activity, 60% of collagenase and 49% of elastase. FEE also showed high antioxidant activity, reaching to 87% of free radical scavenging using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 59% using 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). Additionally, in vitro penetration studies were performed using two vehicles, i.e., a hydrogel and an emulsion containing FEE. These studies showed that the active ingredients contained in FEE penetrate through human skin and accumulate in it. The obtained results indicate that E. angustifolium may be an interesting plant material to be applied as a component of cosmetic and dermatological preparations with antiaging and anti-inflammatory properties.

The aims of this study intended to investigate the anti-inflammatory activity of the 70% ethanol extract from Scoparia dulcis (SDE) and betulinic acid on λ-carrageenan-induced paw edema in mice. The anti-inflammatory mechanism of SDE and betulinic acid was examined by detecting the levels of cyclooxygenase-2 (COX-2), nitric oxide (NO), tumor necrosis factor (TNF-α), interleukin-1β (IL-1β) and malondialdehyde (MDA) in the edema paw tissue and the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) and glutathione reductase (GRd) in the liver. The betulinic acid content in SDE was detected by high performance liquid chromatography (HPLC). In the anti-inflammatory model, the results showed that SDE (0.5 and 1.0 g/kg) and betulinic acid (20 and 40 mg/kg) reduced the paw edema at 3, 4 and 5 h after λ-carrageenan administration. Moreover, SDE and betulinic acid affected the levels of COX-2, NO, TNF-α and IL1-β in the λ-carrageenan-induced edema paws. The activities of SOD, GPx and GRd in the liver tissue were increased and the MDA levels in the edema paws were decreased. It is suggested that SDE and betulinic acid possessed anti-inflammatory activities and the anti-inflammatory mechanisms appear to be related to the reduction of the levels of COX-2, NO, TNF-α and IL1-β in inflamed tissues, as well as the inhibition of MDA level via increasing the activities of SOD, GPx and GRd. The analytical result showed that the content of betulinic acid in SDE was 6.25 mg/g extract.

Inflammation has stimulated significant worldwide scientific interest because of its implication in many human diseases. Most inflammations are caused by reactive oxygen species or free radicals. Annona muricataleaf extracts were investigated for their in-vitroantioxidant and anti-inflammatory potentials. Annona muricataleavesweredried at room temperature, blended using a mill.and extracted with solvents of varying degree of polarities. The solventsused were hexane, ethyl acetate,and ethanol. After sequential extraction, the crude extracts were examined for their in-vitroanti-inflammatory activities on lipoxygenase inhibition, proteinase inhibition, albumin denaturation inhibition,and red blood cell membrane stabilization assays,while the antioxidant activities were examined using DPPH, ABTS and hydrogen peroxide assays. The results showed that the ethanol extract had significantlyhigher albumin denaturation inhibition activity at 500 μg/mL (p &lt; 0.01). The activity of all the extracts on proteinase inhibition decreased with the increase in concentration of the extracts. Indomethacin (standard), ethanol extract,and ethyl acetate extract exhibited a dose dependent increase in lipoxygenase activity. The ethanol extract showed highred blood cell membrane stabilization activity at 500 μg/mL and the activity was comparable with that of the standard (diclofenac). Hydrogen peroxide scavenging activity of the extracts and standard (Vitamin C) were comparable at 20 –100 μg/mL. The ethanol extract showed significantly higher(p &lt; 0.01) DPPH radical scavenging activity compared with other extracts. A similar trend was also observed for ABTS radical scavenging activity. Generally,the ethanol extract exhibited higher anti-inflammatory and antioxidant activities in most of the assays, this could be attributed to the polar compounds present in the extract.

Abstract Nonsteroidal anti-inflammatory drugs (NSAIDs) are the first choice of treatment for rheumatic disorders and other degenerative inflammatory diseases. One of them, indomethacin (INDO), is highlighted in this study. With its analgesic, antipyretic, and anti-inflammatory properties, it is one of the most powerful drugs used in various clinical trials and therapies related to the mechanism of blocking prostaglandin synthesis, thus reducing and eliminating many inflammatory conditions in patients. To ensure the efficacy and safety of this drug in pharmaceutical and clinical use, precise product quality control is required. Such control is performed with routine pharmaceutical analysis using various chemical methods by which INDO is identified as a separate active ingredient in the multicomponent system of a complete pharmaceutical form. In addition, the determination of INDO is important in clinical practice, where its concentration is determined in different biological samples, ensuring better monitoring of a particular therapy. The most commonly used methods for the determination of INDO are high-performance liquid chromatography (37% of developed methods), voltammetry (16% of developed methods), and UV spectroscopy (11% of developed methods). However, each of these methods must provide precise validation parameters. A combination of analytical methods can lead to more precise results and safer application in practice.

This study examined the anthocyanin composition and antioxidant activity of various berries cultivated in Korea: blueberry, crowberry, Korean black raspberry, mulberry, and strawberry. The anthocyanins in berries were identified by high-performance liquid chromatography (HPLC) analysis, and each component was quantitatively analyzed. Furthermore, the antioxidant activity of berries was evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging, 2,2′-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation decolorization, oxygen radical absorbance capacity (ORAC), and ferric ion reducing antioxidant (FRAP) assays. The results revealed that the total content of anthocyanins in crowberry was 35.1 mg/g of extract, which was higher than that in the other four major berry species (1.9-27.7 mg/g of extract). Nineteen anthocyanins were identified in the various berries. The major anthocyanins of crowberry were cyanidin-3-galactoside and delphinidin-3-galactoside, and those from Korean black raspberry were cyanidin-3-rutinoside and cyanidin-3-sambubioside-5-rhamnoside. These two berries also had relatively strong antioxidant activity accompanied by high total polyphenol contents. Thus, consumption of crowberry and Korean black raspberry may be beneficial in reducing the risk of developing lifestyle-related chronic diseases because of their strong antioxidant activity.

Inflammation is all a pervasive phenomenon, which is elicited by the body in response to obnoxious stimuli as a protective measure. However, sustained inflammation leads to several diseases including cancer. Therefore it is necessary to neutralize inflammation. Sonapatha (Oroxylum indicum), a medicinal plant, is traditionally used as a medicine in Ayurveda and other folk systems of medicine. It is commonly used to treat inflammatory diseases including rheumatoid arthritis and asthma. Despite this fact its anti-inflammatory and analgesic effects are not evaluated scientifically. Therefore, the anti-inflammatory and analgesic activities of Sonapatha (Oroxylum indicum) were studied in Swiss albino mice by different methods. The hot plate, acetic acid, and tail immersion tests were used to evaluate the analgesic activity whereas xylene-induced ear edema and formalin induced paw edema tests were used to study the anti-inflammatory activity of Sonapatha. The administration of mice with 250 and 300 mg/kg b.wt. ofO. indicumreduced pain and inflammation indicating that Sonapatha possesses analgesic and anti-inflammatory activities. The maximum analgesic and anti-inflammatory activities were observed in mice receiving 300 mg/kg b.wt. ofO. indicumethanol extract.Our study indicates thatO. indicumpossesses both anti-inflammatory and analgesic activities and it may be useful as an anti-inflammatory agent in the inflammation related disorders.

The aim of this study was to elucidate the correlation between total flavonoid content and total phenolic content on antioxidants activity of ethanol extracts from three cultivars of papaya leaves: 'Holland,' 'Khak Dam' and 'Red Lady.' All crude extracts were investigated to find antioxidant capacity in DPPH radical scavenging. The result indicated that the ethanol extract of ‘Red Lady’ papaya leaves exhibited the highest level of DPPH radical scavenging activity with the IC50 of 0.18 mg/mL, followed by the ‘Khak Dam’ and ‘Holland’ papaya leaves having an IC50 value of 0.24 and 0.44 mg/mL, respectively. The ethanol extract from 'Red Lady' papaya leaves showed the highest level of total flavonoid content (TFC) of 276.72 ± 1.04 µgQE/g DW and total phenolic content (TPC) of 169.85 ± 6.54 mgGAE/g DW. All three cultivars showed a distinctive correlation between IC50 and the content of both total flavonoid and total phenolic with a negative relationship of Pearson’s correlation of -0.922 and -0.940, respectively.

In this study, the extracts of green Coffea arabica bean by country of origin were tested as a potential source for natural ingredients in cosmetic products. Antioxidative activities were measured by total polyphenol, 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), and 2,2 -azino-bis (3- ethylbenzothiazoline-6-sulphonic acid) (ABTS) assay. Anti-inflammatory activities were checked by nitric oxide (NO) assay. The total polyphenol content was found to be higher in ethanol extract than in water extracts, and in particular Ethiopia ethanol extracts had 852.39 mg/100 g, making them the highest in content. The antioxidant measurement was measured with DPPH and ABTS, indicating that antioxidant strength increased dependent on the concentration of all extracts to be superior. The cell survival rate of the extracts of green Coffea arabica beans by country of origin was confirmed by MTT assay and was close to 100 ㎍/㎖ to 100%. Anti-inflammatory activity by NO assay showed lipopolysacchride (LPS)-induced NO was significantly inhibited following treatment with extract of green Coffea arabica beans by country of origin and decreased in a concentration-dependent manner. This study confirmed the efficacy of green Coffea arabica beans by country of origin through research related to antioxidants and anti-inflammatory, and confirmed its availability as a natural material for cosmetics.

The present study was carried out to investigate anti-inflammatory, antioxidant and anti-diarrheal effect of ethanol extract of Stephania japonica. This study showed that the plant extract has significant (p<0.05) anti-inflammatory effect at all phases of carrageenan induced inflammation at a dose level 2g/kg. The DPPH free radical scavenging effect of the extract was compared with standard antioxidant ascorbic acid. IC50 values were found 33.57 μg/ml for the extract and 15.57 μg/ml for ascorbic acid. S. japonica extract at dosage level 2g/kg and 1g/kg decreased the gastrointestinal motility 36.56 and 21.53 %, respectively, in rats. The ethanol extract of the plant also reduced the total number of feces as well as wet feces of rats in castor oil-induced diarrheal model. The results revealed that the extract possesses promising anti-inflammatory, antioxidant and antidiarrheal activity.
