Introduction. Exposure to lead associated with working conditions (especially in mining and metallurgical production) is usually combined with muscular work. However, the influence of this combination on the pathological process development has not been thoroughly studied. Materials and methods. During six weeks, white outbred male rats received intraperitoneal lead acetate at a dose of 11 mg / kg, corresponding to 1/20 LD50, three times a week and/or five times a week physical exercises (forced running for 10 minutes at a speed of 25 m/min). Results. As can be seen from the results, lead caused statistically significant shifts in 33.5% of the organism’s status indices, pointing to the onset of a moderate subchronic intoxication. Content of coproporphyrin in urine and percentage of reticulocytes in the peripheral blood increased, the average content of haemoglobin in the erythrocyte decreased. Under physical activity the harmful effect of lead enhanced in 23.7% of indices and weakened in 28.9% of indices. We did not find changes in 47.4% of indices. Conclusion. Lead intoxication changes ambiguously under the influence of physical exercises. The type of combined effect of lead toxicity and physical activities depends on the outcome by which this type is assessed, on this outcome’s level and the dose ratio.

Abstract Glucosylceramide (GlcCer) is an essential glycosylated lipid found in organisms ranging from fungi to mammals. It is composed of a hydrophilic β-linked glucose and a hydrophobic ceramide, with a predominant content of sphingosine in mammals (d18:1). GlcCer is the precursor of a large scale of different glycosphingolipids. This cerebrozide is synthesized from uridine diphosphate-glucose and ceramide by a GlcCer synthase (UDP-glucose:ceramide glucosyltransferase; UGCG, EC 2.4.1.80). GlcCer-based sphingolipids have been identified as important mediators of a variety of cellular functions and their disequilibrium leads to pathological process development and may induce several diseases progression. Therefore, design of UGCG inhibitor represents an important topic for pharmaceutical research. In this paper, we aimed to study effects of newly synthesized derivatives of (±)-threo-1-phenyl-2-palmitoylamino-3-morpholino-1-propanol (PPMP, known UGCG inhibitor) on: i) activity of UGCG in vitro; ii) thymocytes viability; iii) calcium transport through plasma membrane of thymocytes; iv) induction of apoptosis and autophagy in thymocytes. Thymocytes were isolated from thymus of three to seven weeks old mice (ICR strain). The key factors influencing the effect of PPMP analogues were their concentration, chemical structure and incubation time. Derivatives were able to change Ca2+ transport already after 15 min of cultivation, but their effects on cell viability were manifested at least after 12 h of cultivation. Four from fifteen studied compounds affected UGCG activity after four hour lasting cultivation, - but without correlation with data relating to effects on calcium transport and/or cell viability. Most potent UGCG inhibitor was chosen and applied for induction of apoptosis and autophagy in thymocytes. This inhibitor induced typical DNA fragmentation and upregulation of LC3B protein as autophagy marker, after 2 h and 4 h cultivation, respectively.

Tea is a rich source of polyphenolic flavonoids including catechins, which are thought to contribute to the health benefits of it. Flavonoids have been reported to have antithyroid and goitrogenic effect. The purpose of this study was to evaluate whether high doses of green and black tea have a harmful effect on thyroid physiology. Un-fractionated green and black tea extracts were administered orally to male rats for 30 days at doses of 1.25 g%, 2.5 g% and 5.0 g%. The results showed that green tea extract at 2.5 g% and 5.0 g% doses and black tea extract only at 5.0 g% dose have the potential to alter the thyroid gland physiology and architecture, that is, enlargement of thyroid gland as well as hypertrophy and/or hyperplasia of the thyroid follicles and inhibition of the activity of thyroid peroxidase and 5′-deiodinase I with elevated thyroidal Na+, K+-ATPase activity along with significant decrease in serum T3 and T4, and a parallel increase in serum thyroid stimulating hormone (TSH). This study concludes that goitrogenic/antithyroidal potential of un-fractionated green tea extract is much more than black tea extract because of the differences in catechin contents in the tea extracts.

Analysis of the HPV positive papilloma structures at different levels of pathological process development was carried out. Classical staining of preparations with hematoxylin and eosin was used for immunohistochemical determination of Ki67-positive cells and phenotyping of CD positive cells. We found that the process of papilloma formation begins with a local increase in the proliferative activity of keratinocytes which contributes to the formation of a local epithelial convex above the surface of the oropharyngeal mucosa (OPM) in the form of papilloma. The connective tissue of the OPM adjacent to the epithelium grows at the second stage. At the same time here is a decrease in immunocytes/macrophages number of the oral epithelium not only in the growth zone of the papilloma but in the adjacent neoplasm tissue. The third stage is characterized by the destruction of the basal membrane of the OPM. Apoptotic cells in the cambial layer and forming leukocytes infiltrate the OPM lamina propria. Virus-infected keratinocytes are phagocytized by macrophages or exfoliated from the surface of the epithelial layer. The emerging in the middle layers defect of tissue of the epithelial layer and the absence of Langerhans cells indicate a relationship between the migration of antigen-presenting cells expressing CD68 with impaired differentiation and specialization of keratinocytes. The conclusion is based on the analogy of "leukemic failure" in leukemia and on the absence of differentiating epithelial cells between the cambium and the specialized surface layer in the long-existing papilloma. The disappearance from the epithelium of CD68-positive cells specific to the epithelial layer is a prognostic sign of malignization in the mucous membrane of the oropharynx.

Mental health and physical activity practice in quarantine: a theoretical discussion essay.OBJECTIVE: To discuss the ways in which a state of quarantine and social detachment, for reasons of a pandemic, changes the habits of the population and impacts on the mental health of people in the context. METHODS: The present study intends to be a theoretical discussion essay, with explanatory / analytical designs, which uses recently published and indexed articles, to identify the negative and positive points of social detachment and quarantine, as well as to discuss its effects on mental health. It is also intended to argue in favor of the practice of physical activities, as a tool to reduce tension and anxiety, so characteristic of the current pandemic scenario.  RESULTS: Considering that a situation of social detachment, or quarantine changes the habits of the population, it is also verified that changes in routines emerge feelings of frustration and boredom, in addition, the lack of physical and social contact causes feeling of isolation from the rest of the world and distressing those who experience the phenomenon. A strategy to combat the negative effects on mental health resulting from quarantine and social isolation, is the practice of physical exercises. Among the benefits of physical exercise for mental health, it has been shown that regular physical activity is associated with decreased symptoms of depression and anxiety. CONCLUSION: Knowing the psychological impacts that are generated in this period, and making an association with the studies that demonstrate the benefits of physical exercises, it is possible to say that the practice of activities can be an efficient coping strategy, however, specific studies must still be performed.

Abstract. The results of a survey of 130 students are presented. arterial hypertension was diagnosed during the initial medical examination. The most depressive types of physical exercises, walking and running for students with arterial hypertension were established. It is shown that among students with high blood pressure, along with other physical activities, exercises of a static nature are also hypotensive. The classes that are most adequate for this contingent of persons, contributing to the restoration of functional disorders of hemodynamics, primarily blood pressure indicators, have been determined. Developed and tested a differentiated, scientifically grounded methodology of classes for students with borderline arterial hypertension.

BACKGROUND: Multiple sclerosis (MS) is a chronic degenerative neurological disease with visual, sensitive, motor, or cognitive symptoms. Physical activities are recommended for people with MS (pwMS) who are at home instead of in a rehabilitation program, to help them to maintain their autonomy. Since a lack of motivation appears to be the principal barrier for pwMS to practice exercises, it is necessary to evaluate whether mobile health (mHealth) tools can offer a solution to stimulate their motivation. OBJECTIVE: This literature review analyzes papers to investigate to how a mobile application can motivate pwMS to practice physical activities and manage their fatigue. METHODS: We performed an automatic query from digital libraries and analyzed the studies. RESULTS: We selected seven articles that responded to our criteria. These studies tested mobile applications that used different strategies to motivate pwMS to practice physical exercises at home and manage their fatigue. A lack of motivation was the main obstacle, but depending on the type of exercises chosen, some application settings seemed to resolve this issue. CONCLUSIONS: Studies in this area are limited, although this literature review highlights the need for mHealth tools in which pwMS and their therapists contribute to their use and conception.

The present work is devoted to access some of the biomarkers due to lead toxicity in experimental Wistar male rats orally exposed to graded doses of lead (200, 300 and 400 ppm of lead as lead acetate) over a period of four, eight and twelve weeks. A total of sixty Wistar male rats were equally divided into four groups A, B, C and D. Group A served as control. Groups B, C and D were exposed to 200, 300 and 400 ppm of lead as lead acetate respectively. At the end of four, eight and twelve weeks, five animals each were removed from each group; blood sample was obtained via the ocular median canthus for haematological and biochemical studies. The results indicated accumulation of blood lead which was both dose and time dependent, while there was significant decrease (P < 0.05) in packed cell volume and haemoglobin concentration. The erythrocyte indices (mean corpuscular volume, mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration) also revealed a significant decrease, revealing microcytic hypochromic anaemia. There was significant decrease (P < 0.05) in both the total plasma protein and albumin while blood urea nitrogen and creatinine revealed a significant increase, giving an indication of compromised liver and kidney functions. It was concluded that oral exposure to lead at vary doses over a period of four, eight and twelve weeks results in alterations of various haematological and biochemical parameters and thus can serve as a sensitive biomarkers in lead toxicity.
