Global health is under constant threat due to antimicrobial drug resistance. Bacterial Infections caused by Pseudomonas aeruginosa are of importance because of their antibiotics resistance. This study aimed at evaluating the effects of extracts of Xylopia aethiopica (XA) on multidrug-resistant (MDR) Pseudomonas isolates. Fresh samples of XA leaf, stem bark and roots were collected from the botanical garden, University of Ibadan, Nigeria. Dried and pulverized samples were extracted with methanol and partitioned into n-hexane, dichloromethane and ethyl acetate. Phytochemical screening of the extracts was performed by standard methods. Antimicrobial activity and synergistic interaction were determined using microdilution and checkerboard broth dilution methods, respectively. The results revealed that crude methanol extracts of XA leaf, stem bark and root significantly (p<0.05) inhibited the growth of all tested MDR Pseudomonas isolates at 10 mg/mL. At 1 mg/mL, the ethyl acetate fraction of the leaf, and dichloromethane fraction of the roots produced clear zones of inhibition of 12 – 20 mm, and minimum inhibitory concentrations (MICs) of 1 µg/mL and 0.5 mg/mL, respectively. The modulation factor (MF) of ciprofloxacin, dichloromethane fraction of XA roots and ethyl acetate fraction of XA leaf were 4, 8, and 4 on MDR isolates E01006, OAU058 and P. aeruginosa ATCC 27853, respectively. In all tested isolates, but not E01006 and E01024, the fractional MICs of ciprofloxacin/ethylacetate fraction of XA leaf extract combination was not significantly different (p>0.05) compared with ciprofloxacin/verapamil combination. In conclusion, the root and leaf fractions Xylopia aethiopica that demonstrated antimicrobial activity against MDR P. aeruginosa and synergised with ciprofloxacin have the potential to rejuvenate the antimicrobial activity of ciprofloxacin in MDR P. aeruginosa.

Infections of the oral cavity have a broad microbial etiological profile that varies according to each microenvironment in the mouth. Such infections often require antimicrobial treatment, which can lead to the development of resistance. There is thus a need to find new therapeutic strategies based on natural plant-derived compounds. The aim of this study was to determine the phytochemical nuclei and the antimicrobial effect of Anacardium excelsum leaf and stem extracts, and of fractions derived from the leaf extract, against Streptococcus mutans ATCC 25175, Staphylococcus aureus ATCC 35548, Escherichia coli ATCC 25922, Enterococcus faecalis ATCC 19433 and Candida albicans ATCC 10231. The plant material was collected from the Quindío Botanical Garden (Calarcá, Quindío-Colombia), located at an altitude of 1500 meters above sea level. Hydroalcoholic extracts of A. excelsum leaves and stems, and fractions of the hydroalcoholic leaf extract, were obtained by percolation extraction. Phytochemical nuclei were identified by thin layer chromatography. The antimicrobial activity of the extracts and fractions (at concentrations of 2, 5, 10, 20 and 40 mg / ml) against the five ATCC reference strains was evaluated using the well diffusion technique on Mueller-Hinton agar. The leaf extract showed no antimicrobial activity against E. coli, but it did show antimicrobial activity against S. mutans, S. aureus, E. faecalis and C. albicans, at a concentration of 10 mg/ml, with zones of inhibition of 9 to 11 mm. The ethyl acetate and acetone fractions obtained from A. excelsum leaf extract had greatest antimicrobial activity at 10 mg/ml. In conclusion, (1) the A. excelsum leaf extract, and the ethyl acetate and acetone fractions obtained from the leaf extract, had the greatest antimicrobial activity on all the study microorganisms, and (2) the phytochemical nuclei in the fractions (ethyl acetate and acetone) were found to contain phenolic-type compounds, tannins, triterpene-type terpenes and steroidal-type terpenes, which might explain the antimicrobial activity observed.

Humiria balsamifera (Aubl), commonly known as “mirim”, is a plant of the Humiriaceae family, which consists of 39 species divided between eight genera: Duckesia, Endopleura, Humiria, Humiriastrum, Hylocara, Sacoglottis, Schistostemon, and Vantenea. This study aimed to characterize H. balsamifera extracts by LC-MS/MS and evaluate their antimicrobial potential through in vitro and in vivo assays. The leaves and stem bark of H. balsamifera were collected and dried at room temperature and then ground in a knife mill. The extracts were prepared with organic solvents in order to increase the polarity index (hexane, ethyl acetate, and methanol). The antimicrobial effects of these extracts were evaluated against the following bacterial strains: Escherichia coli ATCC 25922, Listeria monocytogenes ATCC 15313, Salmonella enterica Typhimurium ATCC 14028, and Staphylococcus aureus ATCC 6538. The best activity was observed in the ethyl acetate (EALE = 780 µg/mL), methanol (MLE = 780 µg/mL), and hexane (HLE = 1560 µg/mL) leaf extracts against S. aureus. Considering the results for both antimicrobial and antibiofilm activities, the EALE extract was chosen to proceed to the infection assays, which used Tenebrio molitor larvae. The EALE treatment was able to extend the average lifespan of the larvae (6.5 days) in comparison to S. aureus-infected larvae (1 day). Next, the samples were characterized by High-Performance Liquid Chromatography coupled to a mass spectrometer, allowing the identification of 11 substances, including seven flavonoids, substances whose antimicrobial activity is already well-reported in the literature. The number of bioactive compounds found in the chemical composition of H. balsamifera emphasizes its significance in both traditional medicine and scientific research that studies new treatments based on substances from the Brazilian flora.

Background: The leaves and stem bark of Anogeissus leiocarpa (DC.) Guill. & Pirr. (Combretaceae) are commonly included in recipes used for the management of sickle cell disease in western part of Nigeria. Objective: The present study was carried out to evaluate the potential of methanol extract and various fractions of A. leiocarpa leaves and stem bark to inhibit as well as reverse sodium metabisulphite induced polymerization of sickle cell haemoglobin. Method: The modified Emmel test was used to evaluate the inhibitory activity and the reversal effects of the extracts and fractions of both the leaves and the stem bark of A. leiocarpa in vitro. Results: Both the leaves and the stem bark extracts of A. leiocarpa were effective at inhibiting as well as reversing sodium metabisulphite induced polymerization of sickle cell haemoglobin, though the activity was higher in the leaves (94.52 ± 0.80% and 95.67 ± 0.67% respectively) when compared with the stem bark (51.79 ± 2.46 % and 50.45 ± 3.18 % respectively). The ethyl acetate fraction of both leaves and stem bark also gave good activities in both test methods with percentage reversal of 82.74 ± 1.82% and 67.63 ± 2.18% respectively. Conclusions: : A. leiocarpa extracts and ethyl acetate fractions exhibit good inhibition and reversal of sodium metabisulphite–induced polymerization of sickle cell haemoglobin thus justifying its inclusion in remedies used for the traditional management of sickle cell.

AbstractDue to the emergence of more and more drug resistance bacteria and the reported antibacterial activity of Ceiba pentandra, the antimicrobial activity of the partitioned ethyl-acetate and n-butanol fractions of the stem-bark of this plant were carried out on the following clinical isolates: Staphylococcus aureus; Streptococcus pyrogenes; corynebacterium ulcereans; Escherichia coli; Salmonella typhi; Shigella dysenterea; Enterococcus aerogenes; Pseudomonas aeruginosa; klebsiella pneumonia and the fungi Trichophytom rubrum, Microsporum sp., Aspergillus fumigatus and Aspergillus niger. Agar diffusion and broth delusion methods were used in this study. The ethyl acetate fraction showed wide spectrum antibacteria activity (with zones of inhibition between 27mm and 37mm) while the n-butanol extract showed activity only against the gram negative bacteria (zones of inhibition between 20mm and 21mm). The MIC ranged from 0.65 to 2.5 mg/ml and 2.5 to 5 mg/ml and MBC ranged from 2.5 mg/ml and 5 – 10 mg/ml for the ethylacetate and n-butanol respectively. Both fractions showed no activity against the fungi used in this study. The preliminary phytochemistry of the ethyl acetate showed the presence of only flavonoid and this may explain the activity against all the bacteria. The n-butanol showed the presence of carbohydrates, saponins, tannins and cardiac glycoside and the absence of flavonoid and alkaloids. This study justifies the use of this plant in herbal medicine.

Several of Calophyllums genus have been searched and proven as medicinal plants and Calophyllum macrophyllum is one of its genus. We have been isolated a compound from ethyl acetate fraction of the stem-bark. The compound was flavan-3-ol (5,7,2’,5’-tetrahydroxy flavan-3-ol). This research aimed to determine inhibition antidiabetic activity and affinity of its compound on α-glycosidase enzyme. In vitro antidiabetic effect shown by IC50 9.10 µg/ml and docking by Arguslab 4.01 shown by ΔG -10,24 kkal/mol as in silico test. The inhibition activity of flavan-3-ol against the α-glucosidase enzyme that is very strong shows its promising potential as a candidate for antidiabetic drugs.

During the course of our continuing search for novel anti-malarial compounds from Indonesian plants, the methanol extract of the bark of E. variegata showed significant anti-malarial activity toward Plasmodium falciparum in vitro using the lactate dehydrogenase (LDH) assay. The methanol extract of the bark of E. variegata  was separated by using bioassay-guide fractionation. The ethyl acetate fraction showed the most activity, exhibiting equipotency against both strains of parasite with IC50 of 23.8 µg/mL against 3D7 and 9.3 µg/mL against K1. Furthermore, by using the anti-malarial activity to follow separation, the ethyl acetate fraction was separated by combination of column chromatography to yield an active compound. The chemical structure of active compound was determined on the basis of spectroscopic evidences and comparison with those previously reported and identified as an isoflavonoid, warangalone. The warangalone showed anti-malarial activity against both strains of parasite used with IC50 of 4.8 µg/mL against 3D7 and 3.7 µg/mL against K1.   Keywords: Antimalarial, Erythrina variegata, warangalone

AbstractThe antimicrobial activity of the dichloromethane extract from aerial parts of Leuzea carthamoides DC. was tested in vitro against 19 Staphylococcus aureus strains (ATCC 25923, CNCTC Mau 43/60, clinical isolates). The extract was fractionated by column chromatography on silica gel into six fractions (petroleum ether, toluene, dichloromethane, ethyl acetate, methanol and water). The minimum inhibitory concentrations (MICs) of the fractions ranged from 64 to 1024 µg/mL. An ethyl acetate fraction (EA 1) with the widest range of activity inhibited all of the strains with MIC in the range 128–512 µg/mL. This fraction exhibited potent activity against strains which showed associated resistance to oxacillin, ciprofloxacin and erythromycin.

Introduction. The stem bark of Maerua angolensis DC. (Capparaceae) is traditionally used for management of epilepsy. Our aim was to evaluate the antiseizure potential and identify possible mechanisms by which the effects are registered. Methods. The petroleum ether/ethyl acetate extract (100–1000 mg kg−1) was administered per os to male Sprague-Dawley rats after pretreatment with flumazenil (0.3 mg kg−1) or L-arginine (150 mg kg−1) or sildenafil (5 mg kg−1) and they subsequently received a subcutaneous injection of pentylenetetrazole (65 mg kg−1). Rats were observed for latency to and duration of myoclonic seizures and additionally the level of protection against oxidant markers and products was assessed in vitro and in vivo. Results. The extract (300 and 1000 mg kg−1, p.o.) significantly delayed the onset and decreased the duration and frequency of PTZ-induced convulsions. The anticonvulsant effect of MAE (300 mg kg−1, p.o.) was reversed by pretreatment with flumazenil, L-arginine, or sildenafil. Also, MAE (300 mg kg−1) treatment reversed significantly PTZ-induced oxidative stress in rat brain tissue. Conclusion. The petroleum ether/ethyl acetate fraction exhibits antiseizure activity by affecting GABAergic and nitric oxide-cGMP pathways. In addition, the extract protects against the generation of free radicals and the oxidative products of the PTZ-induced seizures.

The fruit of Rosa multiflora has been used as traditional herbal medicine in Asian countries. The present investigation was undertaken to study the antimicrobial activity and total polyphenol content of hexane, ether, ethyl acetate, water fraction of methanol extract of fruit and flower from Rosa multiflora and Rosa wichuraiana. Antimicrobial activity of the mentioned fractions against 3 gram-positive and 4 gram-negative bacteria using disk diffussion method. The measurement of minimal inhibition concentration (MIC) showed that the ethyl acetate fraction of the two Rosa species is the most effective against the tested bacteria. The total polyphenol content of ethyl acetate fraction of the two Rosa species is higher than the other fractions. The results indicate the antimicrobial activity was related with the total polyphenol content and the fruit and flower of the two Rosa species can be considered as a natural source of antimicrobial agents.
