Resistance status and the rate of resistance development in Musca domestica Linnaeus (Diptera: Muscidae) to permethrin and imidacloprid in Indonesia

A previous report has indicated that in many regions of Indonesia, populations of Musca domestica Linnaeus have shown very high resistance to permethrin and low resistance to imidacloprid. In this study, the resistance status to permethrin and imidacloprid was updated using a topical application and feeding bioassay. Six housefly strains originated from six highly populated cities in Indonesia, namely Serang (SRG), Jakarta (JKT), Bandung (BDG), Semarang (SMG), Yogyakarta (JOG), and Surabaya (SBY). A seventh strain (Danish Pest Infestation Laboratory (DPIL)) served as the control. Each strain was tested for resistance to the two insecticides. In addition, the rate of development of resistance to the two insecticides was measured over ten generations. The results indicated that all field strains showed very high resistance to permethrin. The highest resistance level recorded was in the SRG strain (RR50 = 2880), and the lowest was in the JKT strain (RR50 = 520). Repeated application of permethrin over ten generations increased the resistance level by about 2.7–32.73-fold as compared to the level of their respective parental populations. On the other hand, most strains showed low to moderate resistance to imidacloprid, in which the SRG strain had the highest resistance level (RR50 = 15.5) and the SBY strain had the lowest (RR50 = 2.0). Repeated application of imidacloprid over ten generations increased the resistance level by about 3.25–17.41-fold. The findings, which is the second report of housefly resistance in Indonesia since 2016, provide a crucial foundation for developing appropriate housefly integrated pest management strategies in highly populated areas in Indonesia.

Cotton is a major crop of Pakistan, and Bemisia tabaci (Homoptera: Aleyrodidae) is a major pest of cotton. Due to the unwise and indiscriminate use of insecticides, resistance develops more readily in the whitefly. The present study was conducted to evaluate the resistance development in the whitefly against the different insecticides that are still in use. For this purpose, the whitefly population was selected with five concentrations of each insecticide, for five generations. At G1, compared with the laboratory susceptible population, a very low level of resistance was observed against bifenthrin, cypermethrin, acetamiprid, imidacloprid, thiamethoxam, nitenpyram, chlorfenapyr, and buprofezin with a resistance ratio of 3-fold, 2-fold, 1-fold, 4-fold, 3-fold, 3-fold, 3-fold, and 3-fold, respectively. However, the selection for five generations increased the resistance to a very high level against buprofezin (127-fold), and to a high level against imidacloprid (86-fold) compared with the laboratory susceptible population. While, a moderate level of resistance was observed against cypermethrin (34-fold), thiamethoxam (34-fold), nitenpyram (30-fold), chlorfenapyr (29-fold), and acetamiprid (21-fold). On the other hand, the resistance was low against bifenthrin (18-fold) after selection for five generations. A very low level of resistance against the field population of B. tabaci, at G1, showed that these insecticides are still effective, and thus can be used under the field conditions for the management of B. tabaci. However, the proper rotation of insecticides among different groups can help to reduce the development of resistance against insecticides.

Objectives. High dependency on pesticides could cause selection pressure leading to the development of resistance. This study was conducted to assess the resistance of the house fly, Musca domestica, to five insecticides, namely, permethrin, deltamethrin, beta-cypermethrin, propoxur, and dichlorvos, in Zhejiang Province. Methods. Field strains of house flies were collected from the 12 administrative districts in Zhejiang Province in 2011, 2014, and 2017, respectively. Topical application method was adopted for the bioassays. The probit analysis was used to determine the median lethal doses with the 95% confidence interval, and then the resistance ratio (RR) was calculated. The insecticides resistance in different years and the correlations of the resistance between different insecticides were also analyzed. Results. The resistance of field strains house flies to insecticides in Zhejiang Province was relatively common, especially for permethrin, deltamethrin, and beta-cypermethrin. The reversion of the resistance to dichlorvos was found, and most of the field strains in Zhejiang Province became sensitive to dichlorvos in 2017. Propoxur was much easier to cause very high level of resistance; the Hangzhou strain had the highest RR value more than 1000 in 2014, and five field strains had the RR value more than 100 in 2017. Compared to 2011 and 2014, the resistance of the house flies to propoxur and deltamethrin increased significantly in 2017. The resistance of permethrin, deltamethrin, beta-cypermethrin, and propoxur was significantly correlated with each other, and the resistance of dichlorvos was significantly correlated with beta-cypermethrin. Conclusions. Our results suggested that resistance was existed in permethrin, deltamethrin, beta-cypermethrin, and propoxur in the house flies of Zhejiang Province, while the resistance reversion to dichlorvos was found.

AbstractUnderstanding the process of resistance development of German cockroach, Blattella germanica (L.), in detail is necessary to potentially delay the development of insecticides resistance by rotation or discontinuation of insecticides at the right time. In this study, we investigated the resistance development of the reared German cockroach to chlorpyrifos (CPF) for 23 generations from susceptible cockroaches. CPF 50% lethal dose (LD50) and resistance ratio of each generation cockroaches were determined. The CPF LD50 to each generation cockroaches was used as the insecticide selection pressure of this generation by topical application. The resistance development curve was depicted according to the CPF LD50 to all 23 generations of cockroaches. As a result, a highly resistant German cockroach cohort to CPF, which the resistance ratio was 21.63, was obtained after 23 generations’ selection. During the selection, the cockroaches developed low resistance from F1 to F5, moderate resistance from F6 to F12, and high resistance from F13 to F23. There was a rapid resistance increase every 5–7 generations. The resistance growing showed relatively slow from F1 to F11. The fastest growing phase of the resistance was from F12 to F20, in which accounted for more than 80% of the total resistance increase in 23 generations. The development of resistance to CPF tended to slow down from F21 to F23. These findings may provide a basis for the rational use of insecticides, delaying the development of resistance by rotation or discontinuation.

The factors of resistance to many insecticides were located and isolated in a homozygous condition from the diazinon-selected SKA strain of house-flies (Musca domestica L.) by inbreeding each of the five autosomes of the SKA strain into a susceptible multi-marker strain ac;ar;bwb;ocra SRS. The insecticides were: DDT, methoxychlor, tri-butyl tin, the methoxy and ethoxy analogues of parathion, chlorthion and malathion and their corresponding phosphates, diazinon and diazoxon.The SKA flies' autosomes had the following factors of resistance:—autosome I—very slight resistance to ethyl-chlorthion; autosome II—gene a for low ali-esterase activity which conferred weak to moderate resistance against all organophosphates tested, and Deh (DDT-dehydrochlorinase) which gave great resistance to DDT, but none against methoxychlor; autosome III—Pen, which delayed knock-down by slowing down the entry of insecticides into the flies and had a negligible effect at death except against tri-butyl tin and methoxychlor, and a factor of resistance to Zectran unaffected by sesamex; autosome IV—Dld, the major factor of resistance to dieldrin; and autosome V—Ses, a sesamex-suppressed factor, which gave weak to moderate resistance against ethyl-malaoxon, diazoxon, diazinon, DDT and methoxychlor, but was ineffective against the other organophosphates tested.TBTP (S,S,S tri-butyl phosphorotrithioate), an ali-esterase inhibitor, greatly synergised organophosphates only against insects with gene a. Pretreatment with sesamex, an inhibitor of microsomal activity, elicited two types of response: antagonism with most of the thioates, most pronounced in flies with gene a, and synergism with the phosphates, especially evident with flies with factor Ses. The possible reasons for differences in the response to organophosphates after pretreatment with TBTP or sesamex, and the nature of the resistance factors are discussed. High resistance in SKA flies against organophosphates arises through the interaction of resistance factors which singly give only weak to moderate resistance.

Crassocephalum crepidioides on Cornfields in Dairi Regency had been reported tobecome more difficult to control using paraquat. The objective of the research was todetermine the characteristics and the distribution of C.crepidioides resistant to paraquatin cornfields. The experiment was carried out in two steps, the first step was screeningthe population of C. crepidioides with paraquat at the recommended dose, and the secondstep, dose-response experiment for the resistance level of C. crepidioides population withdose 0, 76, 152, 304,5, 609, 1218, and 2436 g.ai /ha. In the first step experiment, paraquatdichloride was applied at 280 g.ai/ha. The treatments were arranged in a randomized blockdesign with 3 replication. The second step experiment was that the resistant populationsconfirmed in the first experiment were sprayed for their dose-response. The treatmentswere arranged in a randomized complete block design (CRBD). The results showed thatof 30 populations of C. crepidiodes, 19 populations (63.3%) were categorized to beresistant with the mortality ranging from 10.84% to 52.08%, and 11 populations (36.7%),was categorized as high resistance with mortality of 0% to 9.21%. The level ofresistance (R/S) of R-C25, R-C27, and R-C30 populations of C. crepidioides were 12,3,14,86, and 24,83 times consecutively, compared with the susceptible population. Thenumber of C. crepidioides chlorophyl leaves in susceptible populations was significantlylower than that of a resistant populations.

Maumee River, the major tributary in the western basin of Lake Erie, serves as one of major sources of freshwater in the area, supplying potable, recreational, and industrial water. In this study we collected water samples from four sites in the Maumee River Bay between 2016–2017 and E. coli was isolated, enumerated, and analyzed for antimicrobial resistance (AMR) and multidrug resistance (MDR). Strikingly, 95% of the total isolates were found to be resistant to at least one antibiotic. A very high resistance to the drugs cephalothin (95.3%), ampicillin (38.3%), tetracycline (8.8%), gentamicin (8.2%), ciprofloxacin (4.2%), cefoperazone (4%), and sulfamethoxazole (1.5%) was observed within isolates from all four sampling sites. Percentages of AMR and MDR was consistently very high in the summer and fall months, whereas it was observed to be lowest in the winter. A remarkably high number of the isolates were detected to be MDR—95% resistant to ≥1 antibiotic, 43% resistant to ≥2 antibiotics, 15% resistant to ≥3 antibiotics, 4.9% resistant to ≥4 antibiotic and 1.2% resistant to ≥5 antibiotics. This data will serve in better understanding the environmental occurrence and dissemination of AMR/MDR in the area and assist in improving and establishing control measures.

A study was conducted to characterize the common Pythium spp. in greenhouses in Oman and their level of resistance to hymexazol, a widely used fungicide in the country. Pythium isolates were obtained from soil samples, cocopeat bags, and cucumber roots collected from seven regions in the country. Identification of 80 Pythium isolates to the species level using sequences of the internal transcribed spacer region of the ribosomal RNA showed that they belong to four species: Pythium aphanidermatum (77 isolates), P. spinosum (1 isolate), P. myriotylum (1 isolate), and P. catenulatum (1 isolate). Investigating the aggressiveness of three Pythium spp. on cucumber showed that P. aphanidermatum, P. myriotylum, and P. spinosum are pathogenic. Phylogenetic analysis of P. aphanidermatum isolates showed that most of the isolates obtained from cocopeat clustered separately from isolates obtained from soil and roots. This may indicate a difference in the origin of the cocopeat isolates. Evaluating the resistance of 27 P. aphanidermatum isolates to hymexazol showed that most isolates were sensitive (0.9 to 31.2 mg liter−1) whereas one isolate was resistant (142.9 mg liter−1). This study is the first to report P. myriotylum and P. catenulatum in Oman. It is also the first to report the development of resistance to hymexazol among P. aphanidermatum populations from greenhouses. Growers should use integrated disease management strategies to avoid further development of resistance to hymexazol.

ABSTRACT In this study we investigated the interplay of antibiotic pharmacokinetic profiles and the development of mutation-mediated resistance in wild-type and hypermutable Pseudomonas aeruginosa strains. We used in vitro models simulating profiles of the commonly used therapeutic drugs meropenem and ceftazidime, two agents with high levels of antipseudomonal activity said to have different potentials for stimulating resistance development. During ceftazidime treatment of the wild-type strain (PAO1), fully resistant mutants overproducing AmpC were selected rapidly and they completely replaced wild-type cells in the population. During treatment with meropenem, mutants of PAO1 were not selected as rapidly and showed only intermediate resistance due to the loss of OprD. These mutants also replaced the parent strain in the population. During the treatment of the mutator P. aeruginosa strain with meropenem, the slowly selected mutants did not accumulate several resistance mechanisms but only lost OprD and did not completely replace the parent strain in the population. Our results indicate that the commonly used dosing regimens for meropenem and ceftazidime cannot avoid the selection of mutants of wild-type and hypermutable P. aeruginosa strains. For the treatment outcome, including the prevention of resistance development, it would be beneficial for the antibiotic concentration to remain above the mutant prevention concentration for a longer period of time than it does in present regimens.

This study was carried out across the six geopolitical zones to assess knowledge and health seeking behaviors of Nigerians during the first wave of COVID-19 pandemic lockdown in order to understand the seemingly low cases of COVID-19 in Nigeria. Structured and pretested short questionnaires were employed to obtain information electronically and physically across the six geopolitical zones on socio-demographic characteristics of participants; the knowledge of participants on COVID-19 disease; availability and accessibility to testing and isolation centers in the communities, attitudes and health seeking behaviors. Atotal of 1023 respondents; 705 (68.9%) males and 318 (31.1%) females across the six geopolitical zones in Nigeria participated in this study with 477(46.6%) within the 26-45 years age category, and 6(0.6%) above 66 years. Atotal of 985(96.3%) had heard about COVID-19 at the time of survey while a total of  859(84.0%) of the respondents were aware of the presence of COVID-19 laboratory testing centers. Across the geopolitical zones, majority of the respondents 487(47.6%) claimed there are no routine testing for COVID-19 at their community level, while 303 (29.6%) affirmed otherwise and 216(21.1%) did not know if there were routine testing going on or not. On health seeking behaviors across the  country, majority of the respondents 558(54.5%) affirmed they would visit the hospital as the first point of contact if the opportunitypresented itself, 244(23.9%) claimed they would call the NCDC toll line, 2(0.2%) said they would use prayers, while 1(0.1%) respectively claimed they would use home management strategies through visiting chemist stores and checking the internet for solutions. Findings from this study confirm that there is very high awareness of the COVID-19 disease across the country, and limited number of testing centers at the community level. The implications include the possibility of unreported COVID19 cases in the cities as well as in the  communities. Keywords: COVID-19, knowledge, health seeking behaviors, community level
