Introduction: Next Generation Sequencing (NGS) is cost-effective, capable to investigate the genes faster but the protocol has challenges throughout its steps. Objective: We investigated different adjustments of protocol to screen the BRCA genes using Ion Torrent PGM sequencing and correlate the results with number of False Positive (FP) variants. Material and methods: Library preparation process, number of FP InDels, library concentration, number of cycles in amplify targets step, purity of nucleic acid, input, and number of samples/Ion 314 chip were analyzed in association with the results obtained by NGS. Results: 51 reactions and 9 adjustments of protocols were done and 8 FP InDels were observed in homopolymer regions. No FP Single-Nucleotide Polymorphism variant was observed. Protocol variables jointly are associated in 67.5% with the quality of results obtained(p<0.05). The number of FP InDels decreased when the quality of results increased. Conclusion: Ion AmpliSeq BRCA1/BRCA2 Community Panel had better performance with 4 samples per Ion-314 chip instead of 8 and the number of cycles in the amplification step, even using DNA with high quality, was better with 23. We observed better results when the manual equalization process was done, without the Ion Library Equalizer kit. These adjustments provided higher coverage of the variants and fewer artifacts (6.7-fold). Laboratories must perform the internal validation because FP InDel variants can vary according to the quality of results. NGS assay must be validated with Sanger.

Immersive virtual environments (VEs) have the potential to provide novel cost effective ways for evaluating not only new environments and usability scenarios, but also potential user experiences. To achieve this, VEs must be adequately realistic. The level of perceived authenticity can be ascertained by measuring the levels of immersion people experience in their VE interactions. In this paper the degree of authenticity is measured via anauthenticity indexin relation to three different immersive virtual environment devices. These devices include (1) a headband, (2) 3D glasses, and (3) a head-mounted display (HMD). A quick scale for measuring immersion, feeling of control, and simulator sickness was developed and tested. The HMD proved to be the most immersive device, although the headband was demonstrated as being a more stable environment causing the least simulator sickness. The results have design implication as they provide insight into specific factors which make experience in a VE seem more authentic to users. The paper emphasizes that, in addition to the quality of the VE, focus needs to be placed on ergonomic factors such as the weight of the devices, as these may compromise the quality of results obtained when examining studying human-technology interaction in a VE.

Rank aggregation is applied on the web to build various applications like meta-search engines, consumer reviews classification, and recommender systems. Meta-searching is the generation of a single list from a collection of the results produced by multiple search engines, together using a rank aggregation technique. It is an efficient and cost-effective technique to retrieve quality results from the internet. The quality of results produced by a meta-searching relies upon the efficiency of rank aggregation technique applied. An effective rank aggregation technique assigns the rank to a document that is closest to all its previous rankings. The newly generated list of documents may be evaluated by the measurement of Spearman footrule distance. In this article, various fuzzy logic techniques for rank aggregation are analyzed and further improvements are proposed in Modified Shimura technique. Consequently, two novel OWA operators are suggested for the calculation of membership values of document ranks in a modified Shimura technique. The performance of proposed improvements is evaluated on the Spearman footrule distance along with execution time. The results show that the anticipated improvements exhibit better performance than other fuzzy techniques.

We have designed and built an electronic device which compares the resistance of a defined area of vacuum evaporated material with a variable resistor. When the two resistances are matched, the device automatically disconnects the primary side of the substrate transformer and stops further evaporation. This approach to controlled evaporation in conjunction with the modified guns and evaporation source permits reliably reproducible multiple Pt shadow films from a single Pt wrapped carbon point source. The reproducibility from consecutive C point sources is also reliable. Furthermore, the device we have developed permits us to select a predetermined resistance so that low contrast high-resolution shadows, heavy high contrast shadows, or any grade in between can be selected at will. The reproducibility and quality of results are demonstrated in Figures 1-4 which represent evaporations at various settings of the variable resistor.

It was identified that under the conditions of 2020 the most cost effective fertilizer system is an early spring application of NPK (S) (15-15-15 (10)) at a dose of 200kg/ha. The profitability level of this fertilizer was 88,9%.

Growth in telecom infrastructure and provision of modern telecom services to consumers at a cost based tariff helps growth of national economy. Modern telecoms serve as the engine of growth of national economy. Following the global trends of liberalisation and deregulation in telecoms monopolies which have thus far been providing inefficient communication at a greedily high tariffs are falling apart. Mergers in telecoms are not for increasing the size of the monopoly but to provide more efficient and cost effective services to the consumers. In Pakistan the erstwhile T&T department played a needful role at its time. Conversion of the department into a corporation and then into a company were steps necessary for following the global trends. Need now is to continue this trend further, eliminate the monopolistic approach by allowing more players in the field thus permitting the market forces to decide the provision of better quality of modern services at competitive price.

Abstract Today, failure analysis involving flip chip [1] with copper pillar bump packaging technologies would be the major challenges faced by analysts. Most often, handling on the chips after destructive chemical decapsulation is extremely critical as there are several failure analysis steps to be continued such as chip level fault localization, chip micro probing for fault isolation, parallel lapping [2, 3, 4] and passive voltage contrast. Therefore, quality of sample preparation is critical. This paper discussed and demonstrated a quick, reliable and cost effective methodology to decapsulate the thin small leadless (TSLP) flip chip package with copper pillar (CuP) bump interconnect technology.

Several methods of Deoxyribonucleic acid (DNA) extraction have been applied to extract bacterial DNA. The amount and the quality of the DNA obtained for each one of those methods are variable. The study aimed to evaluate bacterial DNA extraction using conventional boiling method followed by alcohol precipitation. DNA extraction from Gram negative bacilli was extracted and precipitated using boiling method with further precipitation by ethanol. The extraction procedure performed using the boiling method resulted in high DNA yields for both E. coli and K. pneumoniae bacteria in (199.7 and 285.7μg/ml, respectively) which was close to control method (229.3 and 440.3μg/ml). It was concluded that after alcohol precipitation boiling procedure was easy, cost-effective, and applicable for high-yield quality of DNA in Gram-negative bacteria.
