Introduction: Diabetes mellitus or diabetes is a metabolic ailment which occurs as a result of insulin insufficiency or defect in insulin function, or both that leads to ihyperglycemia. Diabetes mellitus is a worldwide disease even though its prevalence in other countries vary. Kalanchoe crenata belongs to the family crassulaceae. It is also known locally as miracle plant and often utilized in Africa for medicinal purposes. The study was designed to ascertain the antidiabetic and dyslipidemic activity and effects of the ethanolic extract of both leaves and stem of Kalanchoe crenata on istreptozotocin incite diabetic rats for three (3) weeks. Methodology: 6-8 weeks old Sprague dawley rats received multiple injection of streptozotocin intraperitoneally (40mg/kg body weight) to induce diabetes melitus. Diabetes mellitus was observed and confirmed after six days of induction. The rats were given ethanolic extract of Kalanchoe crenata remarkably (10, 30 and 100mg/kg) and 5mg/kg glibenclamide orally twice daily for three weeks. Blood glucose, lipids, creatinine, urea, were then determined. Results: After week three of treatment 5mg/kg glibenclamide, 30 and 100mg/kg ethanolic extract of Kalanchoe crenata remarkably (p<0.05) decrease glycemia and improved lipidemia by decreasing overall cholesterol, LDL-C and increasing HDL-C likened to the control diabetic group. Also results from treated rats remarkably decrease blood urea nitrogen and creatinine. However, the affirmative control and the sampled treated groups showed curative and regenerative effect in the cells responsible for producing endocrine insulin “beta cells of the islets of Langerhans” located in the pancreas. Kidney and liver tissue sections of treatment groups showed a reversal of diseased insults made by the streptozotocin. Conclusion: The outcome of the research indicate that given ethanolic extract of Kalanchoe crenata remarkably contains the necessary phytochemicals for the development of a standard and effective herbal medicine for Diabetes mellitus and related complications and also with no toxic effects on the tissues of the liver, pancreas and kidney.

Objective: The aim of the present study was to examine the effect of a polyherbal formulation (PHF) on pharmacokinetics and pharmacodynamics of metformin in rats. Methods: The present study was conducted to determine the beneficial outcomes of PHF along with metformin by studying herb-drug interactions. PHF was prepared by five indigenous herbs, Those are being used traditionally as antidiabetic in India. PHF doses (100 mg/kg/day) were administered to Sprague-Dawley rats by an oral route of different groups for multiple weeks except for control. Metformin (100 mg/kg) was orally administered at 7th and 30th day to control and PHF pretreated rats for pharmacokinetics study while pharmacodynamics study was conducted in PHF treated and untreated diabetic rats. No more significant difference was found in the pharmacokinetic parameters in PHF treated at 7th day while a significant increase was found in AUC at the 30th day. Results: The hypoglycemic effect was observed with a combination of metformin and PHF, significantly more compared to control. Metformin decreased the blood glucose 1.51 fold at 7th day and 1.7 fold at 30th day respectively, compared to control. Conclusion: Thus, this finding indicates that PHF increased the AUC of metformin. It might increase bioavailability through drug-herb interaction thereby affecting the therapeutic effect. This formulation can be considered as an adjunct to metformin in the management of diabetes mellitus.

Objectives: Co-administration of chamomile and saffron are effective against diabetes and related complications. Background: Diabetes mellitus refers to comorbidities associated with reduced release of the brain-derived neurotropic factor and disruption in the metabolism of neurotransmitters leading to depression and cognitive impairment. Allopathic medications are available for the treatment of diabetes but there is no cure and multiple adverse effects adhere to it. The therapeutic effects of co-administered chamomile with saffron may reverse the diabetes and its complications. Methods: The present study sought to test hypothesis, conducted on eighty Sprague-Dawley rats randomly divided into eight groups (n=10) including healthy controls, diabetic controls, methanolic extract treatment groups and water decoction treatment groups with respective dosage once a day for two weeks. The dose of single herb group in methanolic extract and water decoction was saffron 10 mg/kg and chamomile 30 mg/kg, while co-administered groups received both herbs in half doses, saffron 5 mg/kg and chamomile 15 mg/kg. Two widely used tests for the assessment of memory (Elevated plus maze and novel object recognition) were used to assess the mood and memory (cognitive) performance after the treatment. Results: It was observed that all treatment groups exhibited antidiabetic effects with improved mood and enhanced memory, high antioxidant profile, increased brain-derived neurotropic factor and acetylcholine concentration. However, the affects were greater in the co-administered groups of saffron and chamomile especially the combined water decoction group. Conclusion : The study provides the successful results of co-administration of chamomile and saffron to alleviate the diabetes and related complications.

The fruit of Phoenix pusilla Garetn. has been used in herbal medicines, as it is sweet, sour, cooling and laxative, cardiotonic, aphrodisiac, carminative and roborant. The objective of the present study was to investigate the antioxidant and antidiabetic effect of ethanolic extract of unripe fruit of Phoenix pusilla in streptozotocin-induced diabetic rats. The extract was analyzed for the presence of various phytoconstituents like tannins, flavonoids, vitamin C, vitamin E, protein, carbohydrates, lipids and phenolic compounds. Streptozotocin (40 mg/kg body weight, i.p.) was administered to induce diabetes in adult rats. The extract (100 and 200 mg/kg) and glibenclamide (6 mg/kg) were administered orally for 21 days to evaluate antioxidant and antidiabetic activity. Blood glucose, serum total cholesterol and triglycerides levels were estimated. Carbohydrate and lipid metabolizing enzymes glucose-6-phosphatase, fructose-1,6-diphosphatase, glycolytic enzymes like hexokinase and liver-function enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx), production of thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALP), renal-function markers like urea and creatinine level were also measured. Histopathology of pancreas was studied. The results indicated that extract normalized the blood, liver, renal and pancreatic functions in streptozotocin-induced diabetic rats. Hence it can be concluded that the extract possesses antioxidant and antidiabetic activity. The findings support the conventional usage of Phoenix pusilla unripe fruit in treating diabetes.

Sweet potato (Ipomoea batatas L) leaf is one of well known vegetables among Indonesian people.  It is also often used as traditional medicine for diabetes mellitus.  This research aimed to investigate the protective effect of I. batatas L leaves extract on the pancreas of IDDM animal model.  Twenty five male Sprague-Dawley rats were divided into 3 treatment groups, and 2 control groups.  Streptozotocin (STZ) was injected at multiple low doses (40 mg/kg BW) intraperitoneally for 5 consecutive days,  I. batatas L leaves extract (doses 0.25, 0.8, and 2.5 g/ kg BW) were administered for 14 days after the first STZ injection.  Fasting blood glucose was analyzed after complete STZ induction (day 6), and after 14 days treatment.  At the end of the study, rats were terminated, and pancreas were removed for histological examination and immunohistochemical procedure using anti-insulin antibody.  Diabetic rats treated with 2.5 g/kg BW I. batatas L leaves extract showed lowest fasting blood glucose among treatment groups, and had approximately 50% normal Langerhans islets with functional beta cells.  These results suggest that I. batatas L leaves extract has anti diabetic activity through its protection effect on the pancreas.

The aim of this study is to determine the mechanism of sepsis-induced vascular hyperpermeability and the beneficial effect of glucocorticoid in protecting vascular endothelium. Male Sprague-Dawley rats were given either a bolus intraperitoneal injection of a nonlethal dose of LPS (Escherichia coli055:B5, 10 mg/kg, Sigma) or vehicle (pyrogen-free water). Animals of treatment groups were also given either dexamethasone (4 mg/kg, 30 min prior to LPS injection) or the matrix metalloproteinases (MMPs) inhibitor doxycycline (4 mg/kg, 30 min after LPS injection). Both activities and protein levels of MMP-2p<0.001and MMP-9p<0.001were significantly upregulated in aortic homogenates from LPS-treated rats, associated with decreased ZO-1p<0.001and syndecan-1p=0.011protein contents. Both dexamethasone and doxycycline could significantly inhibit MMPs activity and reserve the expressions of ZO-1 and syndecan-1. The inhibition of MMPs by dexamethasone was significantly lower than that by doxycycline, while the rescue of syndecan-1 expression from LPS-induced endotoxemic rat thoracic aorta was significantly higher in the dexamethasone-treated compared to the doxycycline-treatedp=0.03. In conclusion, activation of MMPs plays important role in regulating ZO-1 and syndecan-1 protein levels in LPS mediated endothelial perturbation. Both dexamethasone and doxycycline inhibit activation of MMPs that may contribute to the rescue of ZO-1 and syndecan-1 expression.

The purpose of this study was to investigate the effects of electroacupuncture at Zusanli acupoint on the enteric neuropathy in diabetic rats. Sprague–Dawley rats were divided into different groups depending on the total electroacupuncture span and frequency. The expression of nitric oxide synthase (nNOS), choline acetyltransferase (CHAT), protein gene product 9.5 (PGP9.5), and doublecortin was significantly decreased in the diabetic group compared with the control group. Long-term electroacupuncture at Zusanli with either high frequency or low frequency could increase the expression levels of nNOS, CHAT, PGP9.5, and doublecortin, and the increase was greater in the high-frequency group. But no obvious changes were seen in the short-term electroacupuncture groups. These results suggest that electroacupuncture at Zusanli can restore the deficiency of enteric neurons in diabetes partly but a comparative long duration of stimuli (6 weeks) is required. The increase of doublecortin may be involved in this positive process.

Previous studies found that Kelulut Honey produced by Trigona spp. bees is able to prevent oxidative damage in various pathological conditions.  Thus, the present study aimed to determine whether Kelulut Honey could prevent the sperm and testicular damage in streptozotocin-induced diabetic rats. Male Adult male Sprague-Dawley rats were divided into four groups: Non-Diabetic (NDM), Non-Diabetic with Kelulut Honey supplementation (NDMKH), Diabetic without supplementation (DM) and Diabetic with Kelulut Honey supplementation (DMKH).  Kelulut honey was given at the dose of 2.0 g/kg weight daily via gavage for 28 consecutive days. Results showed that sperm quality produced by diabetic rats supplemented with Kelulut honey significantly improved compared to the diabetic control groups (p<0.05). SOD activity and GSH level increased significantly (p<0.05) whereas PC and MDA levels significantly decreased in sperm and testis of DMKH rats when compared to DM rats (p<0.05). Histological observation showed obvious increase in spermatozoa in the lumen of epididymis and increased spermatogenic cells density in the testis of DMKH group.  In conclusion, Kelulut Honey has a potential in preventing the damage of sperm and testis in diabetic rats.

Diet is one of the main factors related to various diseases including Diabetes Mellitus (DM). High carbohydrate, fat, protein, and low fiber diets can increase the risk of type 2 DM occurrence, especially in the patient’s family as a risk group. Previous research on students with DM families in Central Java still had a high-calorie food consumption pattern. The diversity of population, culture, and age may affect the results. Therefore, this study aimed to describe the diet in the family of patients with type 2 DM in the working area of Puskemas (Community Health Center) Garuda Bandung.This research used a quantitative descriptive method by purposive sampling technique. The respondents in this study involved 46 people who were the children of type 2 DM patients in the working area of Puskesmas Garuda. Dietary data obtained from the food record sheet for 3 days and was calculated using Nutrisurvey software in kilocalories (kcal) as the unit of measurement then the results were categorized based on Consensus Perkeni 2015. Data analysis was using frequency distribution.The results showed that 39 (84.8%) respondents in the diet category less than body requirements, 37 (80.4%) respondents in the category of sufficient carbohydrate intake, 39 (84.6%) respondents in the category of excess fat intake, 45 (97.8%) respondents in the category of adequate protein intake, and 41 (89.1%) respondents in the category of less fiber.The conclusions from this study that almost all families of patients with type 2 DM in the work area of Puskesmas Garuda were in the diet category less than body requirement but with the excess fat intake and less fiber. Based on these results, the nurses in Puskesmas are expected to optimize the outreach programs by addressing families of DM patients to make the diet as an attempt to prevent the risk in the family of patients with type 2 DM.Key words: Diet, DM patients’ families, food record. Gambaran Diet pada Keluarga Pasien Diabetes Melitus Tipe 2AbstrakDiet adalah salah satu faktor utama yang berhubungan dengan berbagai penyakit termasuk Diabetes Melitus (DM). Diet tinggi karbohidrat, lemak, protein, serta rendah serat dapat meningkatkan risiko kejadian DM tipe 2 terutama pada keluarga pasien sebagai kelompok berisiko. Penelitian sebelumnya pada mahasiswa dengan keluarga DM di Jawa Tengah masih memiliki pola konsumsi makanan yang tinggi kalori. Perbedaan populasi, budaya, serta usia mungkin akan mempengaruhi hasil. Oleh karena itu, tujuan penelitian ini untuk menggambarkan diet pada keluarga pasien DM tipe 2 di wilayah kerja Puskemas Garuda Kota Bandung.Penelitian ini menggunakan metode deskriptif kuantitatif dengan teknik purposive sampling. Responden dalam penelitian ini berjumlah 46 orang yang merupakan anak kandung pasien DM tipe 2 di Wilayah Kerja Puskesmas Garuda. Data diet diperoleh melalui lembar food record selama 3 hari dihitung mengunakan software Nutrisurvey dengan hasil ukur dalam kilokalori (kcal) yang kemudian hasilnya dikategorikan berdasarkan Konsensus Perkeni 2015. Analisis data menggunakan distribusi frekuensi.Hasil penelitian menujukkan bahwa sebanyak 39 (84,8%) responden dalam kategori diet kurang dari kebutuhan, sebanyak 37 (80,4%) responden dalam kategori asupan karbohidrat cukup, sebanyak 39 (84,6%) responden dalam kategori asupan lemak berlebih, sebanyak 45 (97,8%) responden dalam kategori asupan protein cukup, dan 41 (89,1%) responden dalam kategori serat kurang.Simpulan dari penelitian ini bahwa hampir seluruh keluarga pasien DM tipe 2 di wilayah kerja Puskesmas Garuda berada pada kategori diet kurang dari kebutuhan tetapi dengan asupan lemak berlebih dan serat yang kurang. Berdasarkan hasil tersebut, maka perawat puskesmas diharapkan dapat mengoptimalkan program luar gedung dengan menyasar keluarga penderita DM untuk menjadikan diet sebagai upaya untuk mencegah risiko DM pada keluarga pasien DM tipe 2.Kata kunci : Diet, food record, keluarga pasien DM.

Objective. To To evaluate potential effects of the aqueous extract of Quassia amara L. leaves on the cardiovascular and respiratory systems of adult male Sprague- Dawley rats. Methods. The cardiovascular and respiratory effects of the Quassia amara L. leaf extract on adult male SpragueDawley rats were assessed using non-invasive blood pressure (NIBP) determination and head-out plethysmography, respectively, in a randomized, parallel group study. Mean observations of blood pressure and heart rate were recorded at different time periods after dosing. Respiratory flow and irritation effects were evaluated using mean observations of respiratory rate (RR), tidal volume (TV), mid-expiratory flow rate (EF50), time of inspiration (TI) and expiration (TE), and time of break (TB) and pause (TP). Results. There were no significant differences among the control and the treatment groups in SBP, DBP and HR parameters. The extract showed statistically significant effect on mean RR by time period (F=2.45, p=0.0234), trends over time of TV among the dose groups (F=2.00, p=0.0202), and EF50 among dose groups ((F=3.11, p=0.0422). However, these did not correlate with the changes in the time of break (TB) and time of pause (TP) which are more sensitive and specific tests for respiratory irritation. Conclusion. Aqueous leaf extract of Quassia appeared to have no significant effects on SBP, DPB, Pulse pressure, and HR. There are no conclusive dose-related respiratory flow or pulmonary irritation effects.

AIM: To evaluate the immunomodulatory effects of ethanolic extract of herb pugun tanoh on TCD4 and TCD8 cells in Doxorubicin-induced rats. METHODS: Fifteen male Sprague Dawley rats were divided into five groups consisting of six rats each as follows: Group 1, DOX-treated rats (4.67 mg/kg BW body weight on day 1 and 4) and were administered normal saline 0.9% orally once daily for 7 consecutive days, Group 2, receiving Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) of dose 150 mg/kg BW orally, Group 3, receiving dose Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) 300 mg/kg BW orally. The rats of group 2-3 were intramuscularly administered with doxorubicin at a dose of 4.67 mg/kg BW at the days 1-4 to suppress immune functions. RESULTS: Treatment of 300 mg/kg BW of Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) succeeded in reducing side effect doxorubicin based on increasing the TCD4+ and TCD8+ blood level. CONCLUSION: Ethanolic Extract of Herb Pugun Tanoh (Picria fel-terrae Lour.) could increase the level of TCD4+ and TCD8+ in rats which induced by doxorubicin.
