Bryophyllum pinnatum (Lam.) Oken (Crassulaceae) is used traditionally to treat many ailments. This study investigated the anti-inflammatory effect of hydro-ethanol leaf extract of Bryophyllum pinnatum on Wistar rats using acute and chronic models and also evaluates the bioactive compounds of the leaf extract. The phytochemical constituents of the plant extract were quantitatively determined by Gas Chromatography-Flame Ionization Detector (GC-FID) and acute anti-inflammatory activity was carried out with the aid of plethysmometer while chronic anti-inflammatory activity was investigated using cotton pellet. Results showed that the leaf extract of B. pinnatum was rich in kaempferol (7.006 ±0.02 μg/g), sapogenin (3.372 ±0.02 μg/g), rutin (1.837 ±0.01 μg/g) and lunamarine (1.359 ±0.01 μg/g). The findings showed that the plant had considerable anti-inflammatory effects in a dose dependent manner, returning edema in carragenean-induced and cotton pellet induced granuloma in Wistar rats to normal within 120 minutes and 7 days respectively. The findings of this work have shown that the leaf of B. pinnatum was rich in bioactive compounds which could be synthesized to produce new plant based product to fight inflammatory disorders with fewer side effects.

COVID-19 is a lethal disease caused by the pandemic SARS-CoV-2, which continues to be a public health threat. COVID-19 is principally a respiratory disease and is often associated with sputum retention, for which there are limited therapeutic options. In this regard, we evaluated the use of BromAc, a combination of Bromelain and Acetylcysteine (NAC). Both drugs present mucolytic effect and have been studied to treat COVID-19. Therefore, we sought to examine the mucolytic, antiviral, and anti-inflammatory effect of BromAc in tracheal aspirate samples from critically ill COVID-19 patients requiring mechanical ventilation. Method: Tracheal aspirate samples from COVID-19 patients were collected following next of kin consent and mucolysis, rheometry and cytokine storm analysis was performed. Results: BromAc displayed a robust mucolytic effect in a dose dependent manner. BromAc showed anti-inflammatory activity, reducing the action of cytokine storm, chemokines including MIP-1alpha, CXCL8, MIP-1b, MCP-1 and IP-10, and regulatory cytokines IL-5, IL-10, IL-13 IL-1RA and total reduction for IL-9 compared to NAC alone and control. BromAc acted on IL-6, demonstrating a reduction in G-CSF and VEGF-D at concentrations of 125 and 250ug. Conclusion: These results indicate robust mucolytic and anti-inflammatory effect of BromAc in tracheal aspirates from critically ill COVID-19 patients, indicating its potential as a therapeutic strategy to COVID-19.

Background: Inflammation is emerging as a key contributor to many vascular diseases and furthermore plays a major role in autoimmune diseases, arthritis, allergic reactions, and cancer. Lipopolysaccharide (LPS), which is a component constituting the outer membrane of Gram-negative bacteria, is commonly used for an inflammatory stimuli to mimic inflammatory diseases. Nuclear factor-kappa B (NF-κB) is a transcription factor and regulates gene expression particularly related to the inflammatory process. Stauntonia hexaphylla (Lardizabalaceae) is widely used as a traditional herbal medicine for rheumatism and osteoporosis and as an analgesic, sedative, and diuretic in Korea, Japan, and China. Objective: The purpose of this study was to investigate the anti-inflammatory activity of YRA-1909, the leaf aqueous extract of Stauntonia hexaphylla using LPS-activated rat peritoneal macrophages and rodent inflammation models. Results: YRA-1909 inhibited the LPS-induced nitric oxide (NO) and proinflammatory cytokine production in rat peritoneal macrophages without causing cytotoxicity and reduced inducible NO synthase and prostaglandin E2 levels without affecting the cyclooxygenase-2 expression. YRA-1909 also prevented the LPS-stimulated Akt and NF-κB phosphorylation and reduced the carrageenan-induced hind paw edema, xylene-induced ear edema, acetic acid-induced vascular permeation, and cotton pellet-induced granuloma formation in a dose-dependent manner in mice and rats. Conclusions: S. hexaphylla leaf extract YRA-1909 had anti-inflammatory activity in vitro and in vivo that involves modulation of Akt/NF-κB signaling. Thus, YRA-1909 is safe and effective for the treatment of inflammation.

The inflammatory response is one of natural process in the body to protect itself following tissue injury, but it can cause discomfort. Noni (Morinda citrifolia L.) leaf known as a traditional medicament to help reduce the inflammatory effect. The leaves empirically applied as a wrapping on fever or wound. The purpose of this study was to identify the anti-inflammatory effect of topical noni leaf extract paste in 5% and 10% concentration by examination of Wistar rat paw edema induced by λ-carrageenan. Twenty-four Wistar rats divided into four groups, which were negative control, positive control with diclofenac sodium 1% gel, 5% noni leaf, and 10% noni leaf paste groups. Paw edema was induced by intraplantar injection of 1% λ-carrageenan to every rat. Every treatment subsequently applied in the plantar area before injection, and the changed paw volume measured with plethysmometer at minutes 0, 30, 60, and 90. This study was at the Animal Laboratory, Faculty of Medicine, Universitas Jenderal Achmad Yani, Cimahi city in October–December 2017. The result displayed that the minimum volume after 90 minutes was on 5% and 10% noni leaf paste group, which is 1.00 mL. Kruskal-Wallis test result of inflammatory percentage was significantly different among every group in each examination time (p<0.05). Post-hoc test showed that inflammatory reduction on paw edema with noni leaf paste application on both concentrations were significantly different compared to the negative control. However, it was not different from the positive control group. This study showed that application of noni leaf paste in 5% and 10% concentration could help reduce inflammatory response on skin possibly by the active anti-inflammatory ingredients of noni leaf. EFEK PASTA EKSTRAK DAUN MENGKUDU (MORINDA CITRIFOLIA L.) TOPIKAL TERHADAP EDEMA KAKI TIKUS GALUR WISTAR YANG DIINDUKSI KARAGENANInflamasi adalah proses alami tubuh untuk melindunginya setelah cedera, namun hal tersebut dapat menyebabkan ketidaknyamanan. Daun mengkudu (Morinda citrifolia L.) dikenal sebagai obat tradisional untuk menurunkan efek inflamasi yang secara empiris digunakan untuk mengobati demam dan luka. Tujuan penelitian ini adalah mengetahui efek anti-inflamasi pasta ekstrak daun mengkudu melalui pemeriksaan edema kaki tikus yang diinduksi karagenan-λ. Dua puluh empat tikus galur Wistar dibagi menjadi 4 kelompok, yaitu kontrol negatif dengan akuades, kontrol positif (aplikasi gel Na diklofenak 1%), serta perlakuan pasta daun mengkudu 5% dan 10%. Edema dibuat dengan menginjeksi intraplantar tikus dengan 1% karagenan-λ pada tiap kelompok. Tiap-tiap perlakuan diaplikasikan sebelum tikus diinjeksi dan perubahan volume kaki tikus diukur dengan pletismometer di menit ke-0, 30, 60, dan 90. Penelitian ini dilakukan di Laboratorium Hewan, Fakultas Kedokteran, Universitas Jenderal Achmad Yani, Kota Cimahi pada Oktober–Desember 2017. Hasil pengukuran memperlihatkan penurunan volume edema kaki terkecil setelah 90 menit pada kelompok pasta daun mengkudu 10%, yaitu 1,00 mL. Hasil Uji Kruskal-Wallis terhadap persentase inflamasi berbeda nyata pada tiap kelompok dan tiap waktu pengamatan (p<0,05). Hasil uji beda menunjukkan bahwa penurunan inflamasi kaki tikus pada perlakuan pasta daun mengkudu kedua konsentrasi berbeda nyata dibanding dengan kontrol negatif, tetapi tidak berbeda dengan kontrol positif. Penelitian ini menunjukkan bahwa pasta daun mengkudu 5% dan 10% dapat membantu menurunkan reaksi inflamasi kulit dan efeknya sejalan dengan Na diklofenak karena zat aktif yang bersifat anti-inflamasi dalam daun mengkudu.

Abstract Background Tetracera alnifolia Willd. (Dilleniaceae) is used in traditional African Medicine for the treatment of headache, abdominal pain, and rheumatism. Hence, this study sought to investigate the antinociceptive and anti-inflammatory effects of the hydroethanolic leaf extract of T. alnifolia (HeTA) in rodents. Methods Antinociceptive activity was evaluated using the acetic acid-induced writhing, formalin-/capsaicin-induced paw licking and hot plate tests in mice. The contribution of opioidergic, l-arginine-nitric oxide, and ATP-sensitive potassium channel pathways in HeTA-induced antinociception was also evaluated. The anti-inflammatory effect was assessed using the carrageenan-induced paw edema, xylene ear edema, cotton pellet granuloma, and complete Freund’s adjuvant (CFA)-induced arthritis in rats. Results HeTA (100, 200, and 400 mg/kg, p.o.) produced significant (p<0.05) decrease in mean number of acetic acid-induced writhing, time spent licking paw in formalin, and capsaicin tests as well as time course increase in nociceptive reaction latency in hot plate test. HeTA-induced antinociception was prevented by pretreatment of mice with naloxone (non-selective opioid receptor antagonist), l-arginine (nitric oxide precursor), or glibenclamide (ATP-sensitive potassium channel blocker). HeTA (100 mg/kg, p.o.) produced a significant anti-inflammatory effect against carrageenan-induced rat paw edema (1–5 h), xylene-induced ear edema, cotton pellet-induced granuloma formation, and CFA-induced arthritis in rats. The effects of HeTA in various models were similar to the effect of the standard reference drugs. Conclusions Findings from this study showed that HeTA possesses antinociceptive effect possibly mediated through peripheral opioid receptors with activation of l-arginine-nitric oxide and ATP-sensitive potassium channel pathway as well as anti-inflammatory activity.

Objective: The object of the present study, the methanolic leaf extract of Zanthoxylum ovalifolium was examined for anti-inflammatory activity and tested using carrageenan-induced paw edema in albino Wistar rats. Materials and Methods: The rats were grouped into five, each group consisting of six animals, Group I (negative control) received 1 ml of saline, Group II (standard) received 10 mg/kg p. o. of indomethacin, and remaining three groups received three different doses (100, 200, and 400 mg/kg p. o.) of the methanolic leaf extract after 1 h 0.1 ml of 1% carrageenan injection. The result of anti-inflammatory activity was set up to be dose-dependent in the carrageenan-induced paw edema model. Results: The methanolic leaf extract has shown significant (p<0.001) inhibition of paw edema, 66%, 66.03%, and 69.29% on the 3rd h at the doses of 100, 200, and 400 mg/kg, p. o., respectively. The results were expressed as the Mean±standard error of mean and statistical significance was analyzed by ANOVA followed by Dunnett’s test. The methanolic leaf of Z. ovalifolium extract showed significant anti-inflammatory activity compared with the standard drug indomethacin. Conclusion: Methanolic leaf extract of Z.ovalifolium showed excellent results in the anti-inflammatory experiment so, the plant can be explored as a potential medicine in suppressing inflammation.

Ammodytes personatus, known as the Pacific sand lance, thrives in cold areas of the North Pacific. In this study, the total lipid was extracted from A. personatus eggs and the fatty acid composition was determined using gas chromatography (GC)–flame ionization detection (FID). The results showed that the extracted lipid contained high content of polyunsaturated fatty acids (PUFAs). The immunomodulatory activities of the A. personatus lipid were investigated using rodent macrophages. First, immune enhancement was analyzed, and the A. personatus lipid significantly and dose-dependently increased the NO production in RAW264.7 cells, and this lipid also regulated the transcription of immune-associated genes in RAW264.7 cells by activating the NF-κB and MAPK pathways. Additionally, flow cytometry revealed that this lipid stimulated phagocytosis. Conversely, the anti-inflammatory activity of the A. personatus lipid was also analyzed and the results showed significantly decreased NO production and gene expression in a dose-dependent manner in LPS-stimulated RAW264.7 cells. In addition, the A. personatus lipid suppressed the LPS-induced phosphorylation of proteins related to the NF-κB and MAPK pathways in LPS-stimulated RAW264.7 cells. Further, flow cytometry demonstrated the lipid-regulated anti-inflammatory activity via inhibition of CD86 expression. The results indicate that A. personatus egg lipid is a potential source of immunomodulation.

Thrombotic disorders like myocardial and cerebral infarction are fatal blood clotting related diseases. Synthetic therapeutics used in such disorders has serious adverse effects, so there is a need to investigate some more safe natural thrombolytic agents. Present study is a preliminary work towards such endeavors. During this study analysis of thrombolytic activity and anti-inflammatory activity of Thespesia populnea leaf extract using a simple and quick in vitro clot lysis assay was performed. Various concentrations of leaf extract i.e. 200μg/ml; 400μg/ml and 600μg/ml were tested at various time intervals including; 24, 48 and 72 hours duration of incubation at 37°C for observing maximum clot lysis. The result findings indicated that concentrations of leaf extract enhanced the percentage of clot lysis in dose dependent manner along with the incubation time factor. However; streptokinase SK a reference standard and water were used as a positive and negative control showed clot lysis maximum 96.35% and 35.22% in 72 hours of incubation respectively. Alcohol extract of whole plant of Thespesia populnea (Family: Malvaceae) was assessed for its anti-inflammatory activity by in vitro methods. In vitro anti-inflammatory activity was evaluated using albumin denaturation assay at different concentrations. Diclofenac sodium was used as standard drug. The results showed that Thespesia populnea alcohol extract at a concentration range of 400-1600μl significantly protects from protein denaturation.

Pluchea indica is a shrub plant found in mangrove forests. The leaves are consumed as food and herbal tea and exhibit various biological effects, such as antioxidant and anti-inflammatory activities, in macrophages and animal models. However, the inhibitory activity of P. indica leaf extract on vascular inflammation remains unknown. Therefore, this study investigated the effect of an ethanol extract from P. indica herbal tea leaves (PIE) on tumor necrosis factor-α (TNF-α)-induced human vascular endothelial EA.hy926 cells. The cytotoxic effect of PIE was determined by thiazolyl blue tetrazolium bromide assays. PIE at concentrations of 12.5 - 50 µg/mL did not show significant cytotoxicity, whereas PIE at concentrations ≥ 100 µg/mL decreased cell viability. PIE inhibited the production of reactive oxygen species (ROS) in TNF-α-stimulated endothelial cells. To evaluate the PIE’s anti-vascular inflammatory activity, the protein expression of cell adhesion molecules, including intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1), was determined by western blot. PIE significantly decreased TNF-α-induced ICAM-1 and VCAM-1 expression in a concentration-dependent manner. Furthermore, PIE upregulated heme oxygenase-1 (HO-1) in a concentration- and time-dependent manner. Inhibiting the activity of HO-1 by tin protoporphyrin IX significantly blocked the suppressive effect of PIE on ICAM-1 but not VCAM-1 expression. Therefore, PIE exerts anti-inflammatory activity on vascular endothelial cells, at least in part, by suppressing ROS production and the induction of HO-1. The obtained data suggest that PIE is a promising substance for developing therapeutic agents or as an ingredient of functional food. Traditionally, Bryophyllum pinnatum is used in the management of arthritis and inflammatory diseases. However, B. pinnatum has not been analysed previously for anti-inflammatory activity. Hence, this study is designed to determine the anti-inflammatory effects of various fractions of B. pinnatum leaf extract using rat model of formalin-induced paw edema. Treatment with various fractions showed marked decrease in formalin-induced paw volume and edema in rats. The results of BPAAF treatment were comparable to standard drug, diclofenac. These results indicate that B. pinnatum could be developed as ant-inflammatory drug after further studies.

Anti-inflammatory activity of ethanolic extract of roots, stems and leaves Mimosa hamata was investigated at the doses of 200 and 400 mg/kg using carrageenan induced paw edema and cotton pellet granuloma technique in albino rats. The stem extracts showed significant activity in dose dependent manner as compared to control group. The observations suggested that the extract of M. hamata were effective in exudative and proliferative phases of inflammation i.e. in acute and chronic inflammation. The results obtained indicate that M. hamata has an anti-inflammatory activity that supports the folk medicinal use of the plant.
