The skin is the organ of the body that is frequently exposed to direct UV rays from the sun, which causes the formation of ROS (reactive oxygen species), leading to cell death and tissue damage. This health problem can be overcome by using antioxidants to stabilize the free radicals. Mustard as an organic substance which contains polysaccharide compounds, vitamin C, carotene, quercetin, routine compounds, kaempferol and β-cytostreol, all of these are beneficial to skin health and beauty. Therefore, this study aims to determine the antioxidant activity of the total flavonoids and phenol content of mustard ethanol extract using the DPPH method. Furthermore, the extract at concentrations of 2, 4, and 6% was also tested for its anti-aging activity using the following parameters: moisture and oil content, texture, collagen, wrinkles, pigment, sensitivity, and pores. The results showed that the antioxidant activity of the green mustard had an IC50 value of 170.7839 µg/ml, a total phenol content of 14.471 mg GAE/g extract and flavonoid content of 12.753 mg QE/g extract. The effectiveness of the aging activity of the extract was tested using formulation 6%, which is better than 2%, and 4%. Thus, the percentage of water content recovery, oil content, texture, collagen, wrinkles, stains, sensitivity and pores respectively was 36.41, 39.33, 64.07, 17.65, 65.28, 50.14, 49.10 and 35.09%.  Consequently, it was concluded that the extract has the potential of being developed into a herbal beauty product.

Total phenol levels and antioxidant activity in whole limes from Palopo, Luwu Regency, as well as the relationship between total phenol content and antioxidant activity, were determined. Maceration of dried whole lime fruit yielded the methanol extract (Citrus aurentifolia). Phytochemical screening was used to determine the presence of alkaloids, phenolic compounds, and flavonoids in the extract’s secondary metabolites. The total phenol extract was determined using the Folin-Ciocalteu method, yielding a value of 8.37 mgGAE/g. Antioxidant activity was determined using the DPPH method, and the IC50 value was 1793.06 g/mL. Total phenol content of whole fruit extract lime and IC50 show a linear relationship y = -168,11x + 3211 with a determination R2 = of 0.9780.

Unhealthy lifestyles and air pollution cause the number of free radicals in the body to increase. To protect the body from free radicals, there are antioxidant compounds as an antidote and stabilize free radicals. One of the Indonesian plants that can be used as antioxidants is gotu kola (Centella asiatica (L.) Urban. Objective: This study aims to determine the antioxidant activity of the ethanol extract of gotu kola herb using the DPPH (2,2-Diphenyl-1-pikrilhidrazil) method. with IC50 value. Method: Gotu kola (Centella asiatica (L.) Urban) was extracted by the soxhletation method using 96% ethanol solvent. The testing of antioxidant activity was carried out using the DPPH (2.2 Diphenyl-1-Pikrihydrazil) method. Result: Test results of antioxidant activity The ethanol extract of gotu kola herb showed an IC50 value of 78.20 ppm. Conclusion: This indicated that the ethanol extract of gotu kola herb was included in the criteria for strong antioxidants.

ABSTRACTNanamuha (Bridelia monoica Merr.) is a plant that was used as a cancer and tumor drug by the Sangihe Islands community. This plant contains phenol compounds, flavonoids, alkaloids and tannins. The purpose of this study was to determine the potential of nanamuha leaves that grow in the Sangihe Island to have antioxidant activity and determine the total phenolic ethanol extracts of nanamuha leaf. The extraction method used is maceration with ethanol solvent p.a. The extracts obtained was determined total phenolic and antioxidant activity test using DPPH method. Based on the results obtained, the ethanol extracts of nanamuha leaves has antioxidant activity with an IC50 value of 75,03 µg / mL and the result showed that the total phenol content the ethanol extracts of nanamuha leaves of  53,34 mg / L.  Keywords : Nanamuha leaf, total phenol, antioxidant activity, DPPH  ABSTRAKNanamuha (Bridelia monoica Merr.) merupakan tanaman yang dimanfaatkan sebagai obat kanker dan tumor oleh masyarakat Kepulauan Sangihe. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol p.a. Ekstrak yang diperoleh ditentukan total fenolik dan uji aktivitas antioksidan dengan metode DPPH. ABSTRACTKersen (Muntingia calabura L.) is a plant that has begun to be eliminated and was rarely used because it is often considered to have no economic value and lack of knowledge about its utilization, whereas kersen plants contain flavonoids, saponins, and tannins which were have high benefit for health. The content of metabolites is affected by soil nutrient elements and difference place of growth. This study aims to determine the potential of kersen leaves grown in North Minahasa based on phytochemical content, ability of antioxidant activity, and toxicity. Kersen leaves were extracted using sequential maceration method with n-hexane, ethyl acetate, and ethanol as solvents. Phytochemical Screening using several reagents which tailored to the type of phytochemical test. 1.1-diphenyl-2-picrylhydrazyl (DPPH) method is used to evaluate antioxidant activity, and Brine Shrimp Lethality Test (BSLT) method is used to evaluate toxicity. The result of this study indicate that the n-hexane extract contains phenols, flavonoids, and tannins, with IC50 value 12.54 μg/mL, and LC50 value 881 μg/mL. Ethyl acetate extract contains phenols, flavonoids, tannins, and saponins, with IC50 value 61.3 μg/mL, and LC50 value 1758 μg/mL. Ethanol extract has phenol, flavonoid, tannin, saponin, and terpenoid content, with IC50 value 9.01 μg/mL, and LC50 value 106 μg/mL. Keywords : Kersen leaves, Antioxidant, Toxicity, IC50, LC50 ABSTRAKKersen (Muntingia calabura L.) merupakan tanaman yang sudah mulai tersingkirkan dan jarang dimanfaatkan karena sering dianggap tidak punya nilai ekonomis dan kurangnya pengetahuan tentang pemanfaatannya, padahal tanaman kersen memiliki kandungan flavonoid, saponin, dan tanin yang bermanfaat tinggi untuk kesehatan. Kandungan senyawa metabolit dipengaruhi oleh unsur hara tanah dan perbedaan tempat tumbuh. Penelitian ini bertujuan untuk mengetahui potensi dari daun kersen yang tumbuh di Minahasa Utara berdasarkan kandungan fitokimia, kemampuan aktivitas antioksidan, dan toksisitasnya. Ekstrak daun kersen diekstraksi dengan metode maserasi sekuensial menggunakan pelarut n-heksan, etil asetat, dan etanol.  Skrining fitokimia menggunakan beberapa reagen yang disesuaikan dengan jenis uji fitokimia. Metode 1.1-diphenyl-2-picrylhydrazyl (DPPH) digunakan untuk mengevaluasi aktivitas antioksidan, dan metode Brine Shrimp Lethality Test (BSLT) digunakan untuk mengevaluasi toksisitas. Hasil penelitian ini menunjukkan bahwa ekstrak n-heksan memiliki kandungan fenol, flavonoid, dan tanin, nilai IC50 12,54 μg/mL, dan nilai LC50 881 μg/mL. Free radicals are one of the cause of various diseases. The use of synthetic antioxidant compounds could prevent the effect of the free radicals, however may cause adverse effects on the human body such as impaired liver, lung, intestinal and poisoning. Therefore antioxidant from natural resources needs to be developed. The purpose of this research was to determine the antioxidant activity and IC50 value of the ethanol extract of ganitri (Elaeocarpus ganitrus Roxbs.) leaves. Ganitri leaf ethanol extract activity test was carried out using DPPH method with vitamin C as a standard. Antioxidant activity was determined as a decreas in the absorbance of DPPH at 517 nm wavelength after an addition of the extract with the concentrations of 20, 40, 80, and 100 ppm. The antioxidant acitivity measurement of the ganitri leaf extract showed that the linier regression equation obtained was y = 0.3669x + 29.546, r = 0.4573 while the IC50 value was 54,12 ppm. Based on the result, it is concluded that the ethanol extract of ganitri (Elaeocarpus ganitrus Roxb.) leaf showed was categorized as strong antioxidant.

Objective: The aim of this study was to investigate the chemical compound of the antioxidant activity of the extract from andaliman fruit (Zanthoxylum acanthopodium DC.) and the quality parameters and the total flavonoid determination of the highest antioxidant activity. Methods: The andaliman fruits were extracted using a solvent such as n-hexane, ethyl acetate and 70% ethanol obtained extracts, follow the antioxidant activity using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. The quality parameters and total flavonoid determination were performed from highest antioxidant activity. Fractionated was done using vacuum liquid chromatography (VLC) with the mobile phase chloroform-methanol (9:1 ~1:9). Purification with Thin Layer Chromatography (TLC) preparative, identification using UV-spectrophotometry, FTIR, Gas chromatography Mass Spectrometry (GCMS) analysis. Results: The 70% of ethanol has the highest antioxidant activity with IC50 value 84.11±0.47 ppm; the quality parameters in this study meet the quality requirements, both specific and nonspecific parameters. Total flavonoid content of the 70% ethanol extract was 2.37±0.02%. The results of the GCMS studies showed the presence of 1,2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester; squalene, methyl palmitate, methyl isopalmitate, n-tetracosanol-1 and 4-isopropyl chalcone. Conclusion: The results indicate that the 70% ethanol extracts of andaliman fruits contains 1,2-benzenedicarboxylic acid, mono (2-ethylhexyl) ester; squalene, methyl palmitate, methyl isopalmitate, n-tetracosanol-1 and 4-isopropyl chalcone have antioxidant activity with IC50 value 51.35 ppm.

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

ABSTRACT Herbal supplements are one of the products that are taken from outside the body derived from a mixture of herbal plant ingredients have antioxidant activity to prevent the presence of free radicals in the body. Testing natural antioxidant activity in herbal supplement products using the DPPH method (1,1-diphenyl-2-picrylhyrazil). Measurement of UV-Visible spectrophotometry absorbance with DPPH absorbance value at a wavelength of 517 nm. Sensory analysis using a hedonic test at the panelist preference level, using four attribute parameters namely taste, aroma, color and overall acceptance. Results Tests of antioxidant activity in herbal supplement products on various combination techniques and concentrations of garlic, ginger, local lemons and honey, resulting in the reduction of DPPH which is characterized by a reduction in the intensity of the color from purple to fade to yellow. Antioxidant testing obtained IC50 values ​​from all herbal supplement samples from various treatment techniques and concentrations showed IC50 values ​​less than 50 found in the treatment of chopped engineering materials, concentration 1 (K1) of 23.97%. While concentrations of 2 (k2) and 3 (k3) as well as in various combination techniques and other concentrations indicate that IC50 values ​​range from 50 ppm to 100 ppm. This shows that herbal supplements have very strong antioxidants (IC50 value <50) found in the treatment of ingredients with chopped techniques with the lowest concentration. whereas herbal supplements with treatment techniques and other concentrations have a strong antioxidant value of IC50 (50-100). The results of sensory analysis indicate that the average panelist had a preference level for herbal supplement products with chopping and extraction treatment techniques. Keywords: Antioxidant, DPPH, Herbal supplements, IC50   ABSTRAK   Suplemen herbal merupakan salah satu produk yang menjadi asupan dari luar tubuh yang berasal dari hasil campuran bahan tanaman herbal yang mempunyai aktivitas antioksidan tinggi untuk mencegah adanya radikal bebas dalam tubuh. Pengujian aktivitas antioksidan alami pada produk suplemen herbal menggunakan metode DPPH (1,1-diphenyl-2-picrylhyrazil). Pengukuran absorbansi secara spektrofotometri-UV Visibel dengan Nilai absorbansi DPPH pada panjang gelombang 517 nm. Analisis sensori dengan menggunakan uji hedonik pada tingkat kesukaan panelis, menggunakan empat parameter atribut yaitu rasa, aroma, warna dan penerimaan keseluruhan. Hasil Pengujian aktivitas antioksidan dalam produk suplemen herbal pada berbagai teknik kombinasi dan konsentrasi bawang putih, jahe, lemon lokal dan madu, menghasilkan peredaman DPPH yang ditandai dengan berkurangnya intensitas warna ungu menjadi pudar sampai kekuningan. Pengujian Antioksidan didapatkan Nilai IC50 dari seluruh sampel suplemen herbal dari berbagai teknik perlakuan dan konsentrasi menunjukkan nilai IC50 kurang dari 50 terdapat pada perlakuan bahan teknik rajangan, konsentrasi 1 (K1) sebesar 23,97 %. Sementara konsentrasi 2 (k2) dan 3 (k3) serta pada berbagai teknik kombinasi dan konsentrasi lainnya menunjukan bahwa nilai IC50  berkisar 50 ppm - 100 ppm. Hal ini menunjukkan bahwa suplemen herbal memiliki antioksidan yang sangat kuat (nilai IC50 <50) terdapat pada perlakuan bahan dengan teknik rajangan dengan konsentrasi yang paling rendah, sedangkan suplemen herbal dengan teknik perlakuan dan konsentrasi lain memiliki nilai antioksidan kuat IC50 (50-100) ppm. Hasil analisis sensori menunjukan bahwa rata-rata panelis memiliki tingkat kesukaan pada produk suplemen herbal dengan teknik perlakuan rajangan dan perlakuan sari. Antioxidants are substances that can provide endogenous protection and exogenous oxidative stress by capturing free radicals. Many plants are efficacious as antioxidants, namely plants that contain polyphenols, especially flavonoids, so many are formulated as natural antioxidants. Plants such as Muntingia calabura, Syzygium cumini, Ocimum basilicum, and Eleutherine bulbosa contain polyphenol compounds, especially flavonoids which are efficacious as natural antioxidants. This research aimed to study antioxidant activity derived from some potential plants using the DPPH method by calculating the IC50 value of each plant extract. This research method starts from the determination process to prove the validity of the plants used, the extraction process using the maceration method with 70% ethanol solvent, then the antioxidant activity of extracts from each plant was carried out using the DPPH method. This research starts from the determination process to ensure the correctness of the plants used, then the extraction process is carried out using the maceration method with 70% ethanol solvent. After that the antioxidant activity was determined from the four plants using the DPPH method to see the strongest IC50 value among the four plants. IC50 is the concentration of the sample to inhibit 50% of free radicals. The results of IC50 values from ethanol extract of M. calabura leaves, Syzygium cumini leaves, Ocimum basilicum leaves and Eleutherine bulbosa bulbs, were 18.72; 63,84; 141.59 and 173.15 ppm. Ethanol extract of M. Calabura has a smaller IC50 value of 18.72 ppm which has a very strong and most powerful antioxidant from the ethanol extract of Syzygium cumini, Ocimum basilicum and Eleutherine bulbosa. Keywords : Antioxidant, Muntingia calabura, Syzygium cumini, Ocimum basilicum, Eleutherine bulbosa, DPPH Method
