AbstractDuring the last 30 years, our understanding of the development and diversification of postganglionic sympathetic neurons has dramatically increased. In parallel, the list of target structures has been critically extended from the cardiovascular system and selected glandular structures to metabolically relevant tissues such as white and brown adipose tissue, lymphoid tissues, bone, and bone marrow. A critical question now emerges for the integration of the diverse sympathetic neuron classes into neural circuits specific for these different target tissues to achieve the homeostatic regulation of the physiological ends affected.

Introduction. Benign prostatic hyperplasia (BPH) is a common urological disorder in older men. It is characterized by the development of glandularstromal hyperplasia of the prostate with the formation of new glandular structures and subsequent symptoms from the lower urinary tract. It has now been established that the pathogenesis of this disease is multifactorial and one of the possible mechanisms for the development of BPH is oxidative stress. Purpose. Study of the effect of phenols with a bulky isobornyl substituent (2,6-diisobornyl-4-methylphenol and 4-hydroxymethyl-2,6-diisobornylphenol)on the growth of experimental BPH and the antioxidant balance of prostate cells in comparison with Prostamol Uno. Materials and мethods. Experiments were carried out on 50 male Wistar rats. BPH was caused by daily administration of sulpiride (60 days) to male rats of late reproductive age. After 2 months, the animals were weighed and sacrificed in a CO2 chamber. The mass, mass coefficient, volume of the lateral lobe of the pancreas were determined, morphological analysis was performed. Investigated prooxidant and antioxidant activity. The results were processed by the method of variation statistics using the Mann-Whitney nonparametric U test. Results. The efficacy of the investigated drugs in BPH decreased in the following sequence: sulpiride + substance 4-hydroxymethyl-2,6-diisobornylphenol (HDB) → sulpiride + substance Dibornol (DB) → sulpiride + Prostamol Uno (PU). When comparing the results of evaluating the anti-prooxidant status with the therapeutic effect of the studied drugs, it was found that isobornylphenols, which are highly effective as prostatotropic drugs, did not show a more significant effect, compared to PU, on the redox potential of prostatic tissue cells. Conclusions. Drugs DB, HDB, PU have a normalizing effect on the level of severity of redox reactions in the sulpiride model of BPH.

Abstract Trichomes are specialized glandular or non-glandular structures that provide physical or chemical protection against insect and pathogens attack. Trichomes in Arabidopsis, as typical non-glandular structures, have been extensively studied. However, the molecular mechanism underlying glandular trichome formation and elongation still remains largely unknown. We previously demonstrated that Hair (H) is essential for the formation of type I and type VI trichomes. Here, we found that overexpression of H increased the density and length of tomato trichomes. We revealed that H physically interacts with its close homolog SlZFP8-like (SlZFP8L) and SlZFP8L also directly interacts with Woolly (Wo) by biochemical assays. SlZFP8L overexpression plants showed increased trichome density and length. We further found that the expression of SlZFP6, encoding a C2H2 zinc finger protein, is positively regulated by H. We identified that SlZFP6, is a direct target of H through ChIP-qPCR, Y1H, and LUC assays. Similar to H and SlZFP8L, the overexpression of SlZFP6 also increased the density and length of tomato trichomes. Taken together, our results suggest that H interacts with SlZFP8-like to regulate the initiation and elongation of trichomes by modulating SlZFP6 expression in tomato.

AbstractProstate cancer treatment planning is largely dependent upon examination of core-needle biopsies. In current clinical practice, the microscopic architecture of the prostate glands is what forms the basis for prognostic grading by pathologists. Interpretation of these convoluted 3D glandular structures via visual inspection of a limited number of 2D histology sections is often unreliable, which contributes to the under- and over-treatment of patients. To improve risk assessment and treatment decisions, we have developed a workflow for non-destructive 3D pathology and computational analysis of whole prostate biopsies labeled with a rapid and inexpensive fluorescent analog of standard H&E staining. Our analysis is based on interpretable glandular features, and is facilitated by the development of image-translation-assisted segmentation in 3D (ITAS3D). ITAS3D is a generalizable deep-learning-based strategy that enables tissue microstructures to be volumetrically segmented in an annotation-free and objective (biomarker-based) manner without requiring real immunolabeling. To provide evidence of the translational value of a computational 3D pathology approach, we analyzed ex vivo biopsies (n = 300) extracted from archived radical-prostatectomy specimens (N = 50), and found that 3D glandular features are superior to corresponding 2D features for risk stratification of low-to intermediate-risk PCa patients based on their clinical biochemical recurrence (BCR) outcomes.SignificanceWe present an end-to-end pipeline for computational 3D pathology of whole prostate biopsies, showing that non-destructive pathology has the potential to enable superior prognostic stratification for guiding critical oncology decisions.

Background The giant anteater belongs to the supraorder Xenarthra which occupies a systematically isolated position among placental mammals. The species is categorized as Vulnerable by the International Union for Conservation of Nature, and understanding its reproductive characteristics is critical for future conservation efforts. Methods Gross and microscopic anatomy of the genital organs of 23 male and 21 female adult and young roadkill giant anteaters in Brazil were studied. Results Male giant anteaters presented a short conical penis, intraabdominal testes, and prostate, vesicular and bulbourethral glands. A tubular remnant of the partially fused Müllerian ducts extended from the seminal colliculus through the prostate gland, continued cranially in the genital fold, bifurcated, and attached with one elongation each to the left and right epididymal corpus. The structure presented a total length of up to 10 cm and contained a yellowish liquid in its lumen. Histologically, the caudal section of this structure resembled the female vagina, the middle portion corresponded to the uterus, and the extensions showed characteristics of uterine tubes. In adult female giant anteaters, ovoid ovaries with occasional seminiferous cord-like structures were observed. The animals possessed a simple uterus, which was directly continuous with the vaginal canal. The caudal portion of the vagina had two lumina, separated by a longitudinal septum and opening into two apertures into the vaginal vestibule, cranial to the urethral opening. In the urethral and the lateral vestibular wall, glandular structures with characteristics of male prostate and bulbourethral glands, respectively, were found. The vestibule opened through a vertical vulvar cleft to the exterior. A pair of well-differentiated Wolffian ducts with a central lumen originated ventrally at the vaginal opening into the vestibule and passed in a cranial direction through the ventral vaginal and uterine wall. Each duct extended highly coiled along the ipsilateral uterine tube until the lateral pole of the ovaries where it merged with the rete ovarii. Discussion The reproductive morphology of giant anteaters reveals characteristics shared with other Xenarthrans: intraabdominal testes, a simple uterus, and a double caudal vagina. The persistence of well-differentiated genital ducts of the opposite sex in both males and females, however, singles them out among other species. These structures are the results of an aberration during fetal sexual differentiation and possess secretory functions. The possibility of a pathological degeneration of these organs should be considered in reproductive medicine of the species. Conclusion Knowledge of the unique reproductive characteristics of the giant anteater is essential for future reproductive management of the species. Additionally, further research on the peculiarities of the persisting genital duct structures might help to understand sexual differentiation in placental mammals in general.

The study was conducted on genitalia collected from 100 dairy cows/heifers from the Meat Technology Unit, Mannuthy. The animals brought for slaughter at Meat Technology Unit, Mannuthy were from herds of five different farms (University and Government Undertaking farms) in Kerala state. This included six animals culled on account of factors other than infertility with normal reproductive system (control group) and the remaining animals with a known history of infertility. In total, seven animals showed kinked cervix condition. Cervix was evaluated morphometrically and histologically. Grossly, the cervix was hard, kinked and S-shaped, with a mean length of 9.64 ± 1.19 cm. Average number of annular rings in the cervical canal was 4.14 ± 0.26 with an average diameter of 1.74 ± 0.18 cm. At the external os, diameter of the cervix was less, while at the uterine end, diameter was more. The opening of each cervical ring showed a misalignment instead of a straight line. Histologically, the cervix was lined by simple columnar epithelium with signs of degeneration and desquamation. However, the submucosal layer was extensively infiltrated by collagen fibres. In uterus, there was a significant difference in the number of endometrial glands and height of the glandular epithelial cells which was less when compared to the control group. Muscular layer outer to connective tissue layer was thin and uneven. Alterations of the glandular structures negatively influence the viability of spermatozoa due to the lack of secretory products. This fibrous, collagen rich kinked cervix limits the smooth deposition of the semen into the uterus during artificial insemination and the insufficiently dilated cervix affects its contractibility leading to dystocia and subfertility.

The aim of the study was to compare ecological and morphological features and evolutionary dynamics of the salivary glands in representatives of different classes of vertebrates living in different ecological conditions.Material and methods. The glandular structures of the tongue of vertebrates belonging to various taxonomic and ecological groups (fish, amphibians, reptiles, mammals) were studied. The material obtained was processed using histological and histochemical methods.The results of the study demonstrated that in the course of the evolutionary transformations of vertebrates, an increase and complication of the glandular structures of the tongue occurred due to changes in the environmental factors and in the nature of nutrition and food consumed. The evolutionary transformations of the glandular structures of the organ was directed from unicellular intraepithelial glandular structures (for example, in fish) to the complex multicellular salivary glands of higher mammals, in which there was a division into terminal secretory sections and secretory pathways. In the course of evolution, the number of functions performed by the salivary glands of the tongue has also increased. The digestive and endocrine functions were added to the function of protecting the mucous organ from damage. The serous glands of the tongue are phylogenetically younger. Their occurrence is associated with the participation of the glands of the tongue in the initial stages of chemical food processing. The preservation of more ancient mucous glands against the background of the emergence of new organisms in the course of evolution - mucoserous, seromucous and serous glands, - indicates that in the course of evolutionary development, the glandular structures of the tongue demonstrate parallelism of divergent changes.

Abstract Ticks are blood feeding ectoparasites that transmit a wide range of pathogens. The lone star tick, Amblyomma americanum, is one of the most widely distributed ticks in the Midwest and Eastern United States. Lone star ticks, like most three-host ixodid ticks, can survive in harsh environments for extended periods without consuming a blood meal. Physiological mechanisms that allow them to survive during hot and dry season include thermal tolerance and water homeostasis. Large quantity of dermal fluid secretions induced by mechanical stimulation of tick legs has been described in metastriate ticks including Amblyomma. We hypothesize that a function of tick dermal secretion is similar to the sweating in large homeothermal animals. In this study, we found that a contact with a heat probe at 45oC can trigger dermal secretion. We demonstrated that dermal secretion plays a role in evaporative cooling when ticks are exposed to high temperature. We observed that direct contact to a heat probe for 5 seconds at ~ 52oC caused an exhaustive dermal secretion with ~ 4% loss of body weight and resulted in the lethality in 24-hour, indicating that the secretion is associated with significant costs of water loss. We identified type II dermal glands having paired two cells forming large glandular structures. The secretion is triggered by an injection of serotonin and the serotonin-mediated secretion was suppressed by a pretreatment of Ouabain, a Na/K-ATPase blocker, implying that the secretion is controlled by serotonin and the downstream Na/K-ATPase.

Breast fibroadenomas are a common benign tumour in women that may need to be surgically excised. A mammary-like fibroadenoma can exceptionally be found in the pathological report after an anal polyp removal. A 60-year-old woman presented with huge anal polyp of 25 mm. It was surgically removed transanally. Histologic examination revealed a breast fibroadenoma pattern with glandular structures. Proctologist specialists must be aware that mammary-like tumours are a possible differential diagnosis for anal masses despite their rarity in the literature.
