With the rapid development of the automotive industry and the wide application of 5G network technology, there are more and more Telematics Box (T-Box) equipped with intelligent operating systems in vehicles and they are becoming more and more complex. Because it is connected to the on-board CAN bus internally and interconnects with mobile phone /PC through the cloud platform externally, the security of T-Box must be fully guaranteed, to make the automotive more secure. T-Box can realize remote control function, so the T-Box information security problem has been paid more and more attention. In this paper, the T-Box were tested from multiple dimensions by using various methods, and the results were statistically analyzed, and the corresponding protection strategies were proposed for the corresponding security risks.

AbstractVentral leg patterning in Drosophila is controlled by the expression of the redundant T-box Transcription factors midline (mid) and H15. Here we show that mid represses the Dpp-activated gene Daughters against decapentaplegic (Dad) through a consensus TBE site in the minimal enhancer, Dad13. Mutating the Dad13 DNA sequence results in an increased and broadening of Dad expression. We further demonstrate that the engrailed-homology-1 domain of Mid is critical for regulating the levels of phospho-Mad, a transducer of Dpp-signaling. However, we find that mid does not affect all Dpp-target genes as we demonstrate that brinker (brk) expression is unresponsive to mid. This study further illuminates the interplay between mechanisms involved in determination of cellular fate and the varied roles of mid.Summary statementVentral patterning is controlled in part by the T-box Transcription factor midline blocking Dpp signaling and Dpp-activated genes, though midline does not affect the Dpp-repressed gene brk.

Patterning of the anterior-posterior axis is fundamental to animal development. The Wnt pathway plays a major role in this process by activating the expression of posterior genes in animals from worms to humans. This observation raises the question of whether the Wnt pathway or other regulators control the expression of the many anterior-expressed genes. We found that the expression of five anterior-specific genes in Caenorhabditis elegans embryos depends on the Wnt pathway effectors pop-1/TCF and sys-1/β-catenin. We focused further on one of these anterior genes, ref-2/ZIC, a conserved transcription factor expressed in multiple anterior lineages. Live imaging of ref-2 mutant embryos identified defects in cell division timing and position in anterior lineages. Cis-regulatory dissection identified three ref-2 transcriptional enhancers, one of which is necessary and sufficient for anterior-specific expression. This enhancer is activated by the T-box transcription factors TBX-37 and TBX-38, and surprisingly, concatemerized TBX-37/38 binding sites are sufficient to drive anterior-biased expression alone, despite the broad expression of TBX-37 and TBX-38. Taken together, our results highlight the diverse mechanisms used to regulate anterior expression patterns in the embryo.

Allergic rhinitis (AR) is a complex, chronic immunoinflammatory disorder of the membrane lining of the nasal mucosa. D-Pinitol is considered a cyclic polyol with a potential effect against various allergies. In the present study, we evaluated the anti-allergic effect of pinitol on ovalbumin (OVA)-induced AR model in mice. BALB/c mice were initially sensitized with an intraperitoneal injection of OVA and divided into 5 groups (n=18, in each group) for a treating schedule of distilled water (DW), montelukast (10 mg/kg), and pinitol (5, 10, and 20 mg/kg) through the mouth. Two saline-injected groups were considered as controls by orally administrating DW and pinitol 20. Thereafter, test and control groups were intranasally challenged by OVA and saline, respectively. Our results showed that the OVA challenge caused a marked elevation in AR symptoms like nasal rubbing, sneezing, and discharge which were remarkably diminished using pinitol (10 and 20 mg/kg) and the results were comparable with montelukast. Additionally, increased levels of total and OVA-specific serum Immunoglobulin (Ig) E and IgG1 were significantly attenuated by pinitol as compared to the control group but not the montelukast group. In AR-induced mice, pinitol had significant modulatory effects on representative markers of Th2 (GATA binding protein 3), signal transducer and activator of transcription-6, Interleukins (IL)-4, IL-5, IL-13, suppressors of cytokine signaling 1, Toll-like receptor 4, and myeloid differentiation factor 88), and Type 1 T helper (Th1) immune responses (T-box protein expressed in T cells and Interferon-gamma) as well as the histopathological aberrations induced in the nasal mucosa. In conclusion, Pinitol had potential effects on OVA-induced AR mice through amelioration of nasal symptoms and balancing the Th1/Th2 immune responses during the allergic rhinitis condition.

Purpose: To investigate the anti-asthmatic effect of Ping-Chuan Formula (PCF) in a mouse model, and the associated molecular mechanisms.Methods: Asthma mice were induced using ovalbumin (OVA), and PCF (600 mg/kg) was administered to the mice for 4 weeks. Sections of lung tissues were examined microscopically. The expressions of interleukins (ILs), interferon (IFN)-γ, transforming growth factor (TGF)-β were assayed, while lung tissue expressions of Toll like receptor (TLR)-4, GATA binding protein (GATA)-3, Ox40 ligand (OX40L), indoleamine 2,3-dioxygenase (IDO), and forkhead box P3 (Foxp3) determined. The T box expressed in T cells (T-bet) was evaluated using western blotting. The expressions of MHC II and co-stimulators (CD 11c, CD 80 and CD 86) of dendritic cells (DCs) were determined by flow cytometry.Results: PCF decreased inflammation in lung, and also down-regulated IL-4, -6, -17 and TGF-β (p < 0.01), whereas IL-10 and IFN-γ expressions were up-regulated (p < 0.01). Moreover, PCF decreased the expressions of TLR-4, GATA-3 and OX40L in lung tissue, and promoted Foxp3, IDO and T-bet. Besides, PCF decreased the levels of MHC II and co-stimulators (CD 80 and CD 86) on the surface of DCs.Conclusion: PCF exerts anti-asthmatic effect in mice via inhibition of inflammation, and modulation of MHC II and co-stimulators on the surface of DCs. These findings suggest that PCF is a promising candidate drug for treating asthma in humans.

Breast cancer affects women at relatively high frequency (1). We mined published microarray datasets (2, 3) to determine in an unbiased fashion and at the systems level genes most differentially expressed in the primary tumors of patients with breast cancer. We report here significant differential expression of the gene encoding T-box 15, TBX15, when comparing primary tumors of the breast to the tissue of origin, the normal breast. TBX15 was also differentially expressed in the brain metastases of patients with metastatic breast cancer. TBX15 mRNA was present at significantly lower quantities in tumors of the breast as compared to normal breast tissue. Analysis of human survival data revealed that expression of TBX15 in primary tumors of the breast was correlated with recurrence-free survival in patients with basal-like and normal-like subtype cancer, demonstrating a relationship between primary tumor expression of a differentially expressed gene and patient survival outcomes influenced by PAM50 molecular subtype. TBX15 may be of relevance to initiation, maintenance or progression of cancers of the female breast.

The node-streak border region comprising notochord progenitor cells (NPCs) at the posterior node and neuro-mesodermal progenitor cells (NMPs) in the adjacent epiblast is the prime organizing center for axial elongation in mouse embryos. The T-box transcription factor Brachyury (T) is essential for both, formation of the notochord and maintenance of NMPs, and thus is a key regulator of trunk and tail development. The T promoter controlling T expression in NMPs and nascent mesoderm has been characterized in detail. However, control elements for T expression in the notochord have not been identified yet. We have generated a series of deletion alleles by CRISPR/Cas9 genome editing in mESCs, and analyzed their effects in mutant mouse embryos. We identified a 37 kb region upstream of T essential for notochord function and tailbud outgrowth. Within that region we discovered a T binding enhancer required for notochord cell specification and differentiation. Our data reveal a complex regulatory landscape controlling cell type-specific expression and function of T in NMP/nascent mesoderm and node/notochord allowing proper trunk and tail development.
