Hairy roots (HRs) grown in vitro are a powerful platform for plant biotechnological advances and for the bio-based production of metabolites of interest. In this work, black carrot HRs able to accumulate anthocyanin as major secondary metabolite were used. Biomass and anthocyanin accumulation were improved by modulating growth medium composition—different Murashige & Skoog (MS)-based media—and H2O2-elicitation, and the level of the main antioxidant enzymes on elicited HRs was measured. Higher growth was obtained on liquid 1/2 MS medium supplemented with 60 g/L sucrose for HRs grown over 20 days. In this medium, 200 µM H2O2 applied on day 12 induced anthocyanin accumulation by 20%. The activity of superoxide dismutase (SOD)—which generates H2O2 from O2•−—increased by over 50%, whereas the activity of H2O2-scavenging enzymes was not enhanced. Elicitation in the HRs can result in a controlled oxidative burst, in which SOD activity increased H2O2 levels, whereas anthocyanins, as effective reactive oxygen species scavengers, could be induced to modulate the oxidative burst generated. Moreover, given the proven stability of the HR lines used and their remarkable productivity, this system appears as suitable for elucidating the interplay between antioxidant and secondary metabolism.

Background: Unevaluated open-pollinated germplasm represents a promising source of variability to face the problems of worldwide food production under a changing environment. In carrots, this is particularly true for black carrot accessions, which are the most relevant among Eastern carrot germplasm due to their high anthocyanin content and, concomitantly, antioxidant capacity. Methods: In the present work, a comparative characterization was conducted for the first time on 11 Eastern carrot landraces and Night Bird ‘F1’ as the reference cultivar, grown under glasshouse conditions at temperatures up to 33 °C. Results: Some landraces showed their potential for ulterior evaluation in terms of plant and taproot size, plant compactness, specific leaf area and leaf area ratio, among other traits. The highest anthocyanin and flavonoid contents were found in the reference cultivar, whereas remarkable differences in these variables were observed for the rest of accessions, which in turn may correlate with very distinct coloration patterns. Premature bolting and taproot shape abnormalities were also recorded. Mineral composition analysis showed the nutritional potential of Eastern carrot leaves, which displayed higher concentration than taproot tissue for several macro- and micronutrients. Moreover, several accessions had higher nutrient concentrations than the reference cultivar, which also highlights their profitability. Conclusions: This work contributes to the knowledge on Eastern black carrot germplasm by characterizing some of its main agricultural traits, and opens up the prospect for complementary evaluation on high-yield accessions.

Whey-protein-isolate-based composite hydrogels with encapsulated black carrot (Daucus carota) extract were prepared by heat-induced gelation. The hydrogels were blended with gum tragacanth, pectin and xanthan gum polysaccharides for modulating their properties. 1H spin-lattice relaxation experiments were performed in a broad frequency range, from 4 kHz to 30 MHz, to obtain insight into the influence of the different polysaccharides and of the presence of black carrot on dynamical properties of water molecules in the hydrogel network. The 1H spin-lattice relaxation data were decomposed into relaxation contributions associated with confined and free water fractions. The population of the confined water fraction and the value of the translation diffusion coefficient of water molecules in the vicinity of the macromolecular network were quantitatively determined on the basis of the relaxation data. Moreover, it was demonstrated that the translation diffusion is highly anisotropic (two-dimensional, 2D).
