Abstract Background Obesity is considered a major public health problem in the world. At present, the association of single nucleotide polymorphisms (SNPS) Pro12Ala and C161-T in peroxisome proliferator-activated receptor gamma 2 (PPARγ2) gene with obesity is still controversial. The aim of this study is to evaluate the relationship between the Pro12Ala and C161-T polymorphisms of PPARγ2 gene and obesity among Han nationality of Yunnan Plateau in Southwest China. Methods The genotypes of 284 extremely overweight patients, 475 obese patients and 759 normal controls from Yongsheng County of Yunnan Province (China) were analyzed. The Pro12Ala and C161-T genotypes of PPARγ2 were detected by SNaPshot genotyping assay. The data were analyzed statistically. Results The overall prevalence of obesity was 4.99% and the overweight rate was 23.93% among Han nationality in Yongsheng, Yunnan Province. Statistical analysis showed that there were no significant differences in the Pro12Ala genotype, C161-T genotype and the two combined genotypes between the extremely overweight and the control 1, either between the obesity and the control 2 (P > 0.05). Further, the alleles of Pro12Ala and C161-T did not exhibit any signifificant association with extreme overweight or obesity among the complete sample population (P > 0.05). However, conditional logistic regression analysis showed that there was a statistically significant association between the two combined genotype of “CC + TT” and the extreme overweight after adjusting for covariates (Calculated with the combined genotype of “CC + CC ” as the reference category; P = 0.014; OR = 4.04; 95%CI = 1.33–12.33). Conclusions Our study indicates that the combined genotypes "CC + TT" of PPARγ2 Pro12Ala and C161-T were associated with an increased risk of extreme overweight, however, Pro12Ala and C161-T polymorphisms may not be the main cause of obesity in Han nationality of the Yunnan Plateau in Southwest China.

Abstract Backgroud To study of gut microbiota in IBS-D of Han nationality in Southwest China, explore its relationship with environmental factors and differential expression function of gut microbiota in patients. Methods 120 cases of IBS-D group and 63 cases of HCs group were recruited, baseline data such as age, height, weight, monthly income, work intensity, eating habits and exercise were collected. HAMA and HAMD scores were performed on the two groups. IBS-D group was scored with IBS-SSS and IBS-QOL, and the differences in gastrointestinal symptoms, mental and psychological status and quality of life were compared. The laboratory tests were performed such as blood routine and liver function. Fresh feces of the two groups were collected for 16S rDNA sequencing. Then analyze the differences of gut microbiota between the two groups, and look for typical biomarkers of each. According to the dominant species and differentially expressed species at different classification levels, FAPROTAX was used to predict the functional differences of gut microbiota between the two groups. Taking the above scores and laboratory tests as environmental factors, spearman correlation analysis was conducted at the phylum level to explore the impact of environmental factors on the occurrence and development of gut microbiota and IBS-D. Results IBS-D patients are mostly concentrated in low-medium income people with medium-high labor intensity. The sleep time, exercise time, intake of vegetables and fruits of IBS-D are significantly lower than those of HCs, and the proportion of IBS-D who like to eat pepper is significantly higher than those of HCs. The scores of HAMA and HAMD, Urea nitrogen, AST, ALT , GGT in IBS-D were significantly higher than those in HCs.The richness of gut microbiota in IBS-D was significantly lower than that in HCs, but not diversity and eveness. The beta diversity index of gut microbiota community was significantly different between the two groups. The biomarkers of IBS-D were Prevotella, Clostridiales and Roseburia, the biomarkers of HCs were Veillonellaceae, Bacteroides-coprocola, Bifidobacteriales, Bifidobacteriumde, etc. The functions of gut microbiota in IBS-D were significantly up-regulated in cellulolysi, xylanolysis, and significantly down-regulated in fermentation, methanogenesis, nitrite_ammonification, nitrate_reduction. Correlation analysis showed that multiple intestinal microorganisms were closely related to HAMA, IBS-SSS, IBS-QOL, inflammatory indexes and liver enzymes. Conclusion There are significant differences in richness of gut microbiota, flora structure and flora function between IBS-D and HCs in Southwest China. These differences may be closely related to environmental factors such as eating habits, living habits and mental and psychological factors.The trial was registered and approved in China Clinical Trial Registry (Registration No.ChiCTR2100045751)

Essential hypertension is a common cardiovascular disease with complex etiology, closely related to genetic and environmental factors. The pathogenesis of hypertension involves alteration in vascular resistance caused by sympathetic nervous system (SNS) and renin angiotensin system (RAS). Susceptibility factors of hypertension vary with regions and ethnicities. In this study, we conducted target capture sequencing on 54 genes related to SNS and RAS derived from a collection of Han nationality, consisting of 151 hypertension patients and 65 normal subjects in Xinjiang, China. Six non-synonymous mutations related to hypertension were identified, including GRK4 rs1644731 and RDH8 rs1801058, Mutations are predicted to affect 3D conformation, force field, transmembrane domain and RNA secondary structure of corresponding genes. Based on protein interaction network and pathway enrichment, GRK4 is predicted to participate in hypertension by acting on dopaminergic synapse, together with interacting components. RDH8 is involved in vitamin A (retinol) metabolism and consequent biological processes related to hypertension. Thus, GRK4 and RDH8 may serve as susceptibility genes for hypertension. This finding provides new genetic evidence for elucidating risk factors of hypertension in Han nationality in Xinjiang, which in turn, enriches genetic resource bank of hypertension susceptibility genes.
