A trial was performed to investigate how carbon sources can affect the welfare status of African catfish (3.8±0.2g and 8±0.11 cm) juveniles in biofloc systems. Inocula was prepared in glass tanks (5L) by adding 20g of pond bottom soil in well aerated water (1L) containing 10mg L-1 ammonium sulphate (NH)4SO4 and 400mg L-1 of four different carbon sources (tapioca, wheat offal, brewery and cassava peel) for 24 hours. Each treatment group (carbon sources and control) were replicated and each tank contained 1000 Clarias gariepinus juvenile, fed with commercial feed (Crude protein 42%) at 5% of their body weight 72 days. The water quality showed that all water parameters remained at concentrations suitable for Clarias gariepinus culture in the studied systems. The enzymes activities were noticed to be different across the biofloc and the selected organs. There were significant differences in serological content in fish between the treatment’s groups (P<0.05). The significant difference was found between the treatments in case of enzymes activity (P<0.05). The study shows that the welfare status with reference to digestive enzymes activity, oxidative status and extent of the immune system stimulation in BFT system is carbon source dependent.

Ibogaine effects are mediated by cellular receptors, ATP depletion followed by ROS production and antioxidant enzyme activity elevation in a dose and time dependent manner. Since the role of KATP channels and β-adrenoceptors in ROS cellular circuit was established here we explored their role in ibogaine pro-antioxidant effectiveness. Single dose of ibogaine (10 mg/L i.e., 28.8 μmol/L) was applied to isolated rat uterus (spontaneous and Ca2+-stimulated) and contractility and antioxidant enzymes activity were monitored during 4 h. Ibogaine increased amplitude and frequency of spontaneous active uteri immediately after addition that was prevented by propranolol (β1 and β2 adrenoceptors selective antagonists) and glibenclamide (KATP sensitive channels inhibitor; only frequency) pre-treatment. In Ca2+-stimulated uteri, ibogaine decreased both amplitude and frequency after 4 h. Pre-treatment with propranolol abolished ibogaine induced amplitude lowering, while glibenclamide had no effect. In both types of active uterus, ibogaine induced a decrease in SOD1 and an increase in CAT activity after 2 h. In Ca2+-stimulated uterus, there was also a decrease of SOD2 activity after 2 h. After 4 h, SOD1 activity returned to the baseline level, but GSH-Px activity increased. Pre-treatment with both propranolol and glibenclamide abolished observed changes of antioxidant enzymes activity suggesting that ibogaine pro-antioxidative effectiveness is β-adrenergic receptors and KATP channels mediated.

Aims: The current study was developed to investigate the influence of grape seeds (GS) and mandarin peels (MP) extracts as powerful antioxidants on the cardiotoxicity induced by cyclophosphamide (CP) in rats. Place of Study: Department of Biochemistry and Nutrition, Faculty of Women for Arts, Science and Education, Ain Shams University. Methodology: Sixty adult male Sprague-Dawley rats were divided into 6 groups. Group (1): Rats were received distilled water daily orally for 6 weeks and injected interperitoneally (i.p) with saline (0.9 %) (2.5 ml / kg BW) as single dose at the end of the sixth week of experiment. Group (2): Rats were received distilled water orally and injected with single dose of cyclophosphamide which dissolved in saline (200 mg/kg BW. i.p.) at the end of the sixth week of experiment. Groups (3 and 4):  Rats were received grape seeds extracts low and high doses (150 and 300 mg /kg BW), respectively daily orally for 6 weeks then injected with cyclophosphamide as group 2. Groups (5 and 6): Rats were received mandarin peels extracts low and high doses (150 and 300 mg /kg BW), respectively daily orally for 6 weeks then injected with cyclophosphamide as group 2. Results: Our results documented that CP caused a significant increase in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALK-P), creatine kinase (CK-MB), lactate dehydrogenase (LDH), creatine kinase (CK) enzymes activity and serum malondialdehyde (MDA) level. While total antioxidant capacity level (TAC) showed a significant decrease. On the other hand cardiac catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities and cardiac β cell lymphoma (Bcl-2) level showed a significant decrease in CP group while cardiac p53, caspase-3 and DNA fragmentation levels showed a significant increase in CP intoxicated group. Also, some histopathological changes were observed in liver and heart tissues in CP group. Oral administration of GS and MP caused an ameliorative effect in oxidative and apoptotic biomarkers, liver and heart function enzymes activity with an improvement of histopathological changes in liver and heart tissues. Conclusion: Our data proved that the protective effect of grape seeds and mandarin peels in cyclophosphamide intoxicated group may be due to their antioxidant, anti-inflammatory and anti- apoptotic properties.
