PIWI-interacting RNAs (piRNAs) repress transposons to protect the germline genome from DNA damage caused by transposon transposition. In Drosophila, the Traffic jam (Tj) mRNA is consumed to produce piRNA in its 3′ UTR. A cis element located within the 3′-UTR, Tj-cis, is necessary for piRNA biogenesis. In this study, we analyzed the structure of the Tj-cis RNA, a 100 nt RNA corresponding to the Tj-cis element, by the SHAPE and NMR analyses and found that a stable hairpin structure formed in the 5′ half of the Tj-cis RNA. The tertiary structure of the 16 nt stable hairpin was analyzed by NMR, and a novel stem-loop structure, the T-hairpin, was found. In the T-hairpin, four uridine residues are exposed to the solvent, suggesting that this stem loop is the target of Yb protein, a Tudor domain-containing piRNA biogenesis factor. The piRNA biogenesis assay showed that both the T-hairpin and the 3′ half are required for the function of the Tj-cis element, suggesting that both the T-hairpin and the 3′ half are recognized by Yb protein.

Traffic jam is increasingly aggravating in almost every urban area. Traffic forecast, traffic modeling, visualization can help to provide appropriate route and time for traveling and thus provides a significant impact on traffic jam reduction. For traffic forecasting, modeling and visualization, city-wide traffic data collection and analysis are needed, which is still challenging in many aspects. This paper aims to develop a tool for acquiring and processing traffic data from Google Maps that can be used for forecasting, modeling, and visualization. Dhaka city is used as a case study since there is no infrastructure available for traffic data collection. The traffic flow intensity of the road is analyzed to determine the congestion of the road. The flow intensity is used for traffic modeling, visualization, traffic prediction and many more.

Abstract The rapid growth of cities will certainly also increase traffic jams and emissions in the air. This study aims to analyze the increase in car volume and the CO distribution pattern in East Jakarta. Data for traffic jam patterns were recorded based on Google Maps on weekdays in the morning and evening. The spatial analysis method used to find the CO distribution pattern is the IDW interpolation, and the mathematical model calculates the moving emission based on the distance travelled (VKT). The spatial pattern of CO distribution in 2020 was scattered with high concentrations in Pasar Rebo, Ciracas, Cipayung, Kramat Jati, and Makasar Districts, with CO levels above 4,500 ppm. The spatial pattern of CO distribution from the mobile emission model differs from the air station IDW interpolation. The CO distribution pattern from the mobile emission model is very concentrated in Makassar, and Kramat Jati District was 6,740.91 tons/year. The result concluded that the increase in vehicle volume is not related to the distribution of the CO model from air station IDW interpolation, and the other hand, the congestion pattern was related to the distribution pattern of the CO model from vehicles from the level of congestion.

Mosul is the second-largest city in Iraq, the movements of people within the city have become more restricted by the crowded streets during rush hours. This issue has also become critical since it impacts most of the life aspects of the city (e.g., going to work, schools, etc.). Therefore, there is a need to mitigate this issue using low-cost strategies and solutions due to the current economic issues in the country. In this work, a network-based model is generated that represents the road network of both sides of the city (east and west coasts). The generated network is analysed based on its spatial and temporal features. Then, the elite intersections (crossroads) are extracted, which represent the most effective factors in the road network of the city. After that, low-cost sensor technologies are suggested and can contribute to mitigating the traffic jam issue in the city. Finally, the proposed solutions and suggestions can be generalized to any city that is close in the nature to the considered city in this study.

The C. elegans Wnt/β-catenin Asymmetry (WβA) pathway utilizes asymmetric regulation of SYS-1/β-catenin and POP-1/TCF coactivators. This differentially regulates gene expression during cell fate decisions, specifically by asymmetric localization of determinants in mother cells to produce daughters biased towards their appropriate cell fate at birth. Despite the induction of asymmetry, β-catenin localizes symmetrically to mitotic centrosomes in both mammals and C. elegans. Due to the mitosis-specific mobility of centrosomal SYS-1 and 'traffic jam' like enrichment of SYS-1 at kinetochore microtubules when SYS-1 centrosomal loading is disrupted, we investigated active trafficking in SYS-1 centrosomal localization. Here, we demonstrate that trafficking by microtubule motor dynein is required to maintain SYS-1 centrosomal enrichment, by dynein RNAi-mediated decreases in SYS-1 centrosomal enrichment and by temperature-sensitive allele of the dynein heavy chain. Conversely, we observe that depletion of microtubules by Nocodazole treatment or RNAi of putative dynein-proteasome adapter ECPS-1 exhibits increased centrosomal enrichment of SYS-1. Moreover, disruptions to SYS-1 or negative regulator microtubule trafficking are sufficient to significantly exacerbate SYS-1 dependent cell fate misspecifications. We propose retrograde microtubule-mediated trafficking enables SYS-1 and negative regulators to enrich at centrosomes, enhancing their interaction and perhaps implicating the centrosome as a mitotic sink for proteins targeted for degradation.

Third junction of Sungai Bengkel was a Signalized Intersection which a hasn’t have traffic jam ,which has three approach, are North approach, West approach and East approach. However, the alinyemen for the North approach has problems that make it difficult for vehicles, especially four-wheelers to turn because the effect bottle neck resulting in the slow motion of the vehicle and change of phase time . the problem solving, the evalution of traffic light and intersection geometry were carried out by widening  the alinyemen for the North approach and East approaches and doing changes in cycle time. The purpose of evaluating the performance of traffic light and Geometrics at the third intersection of Sungai Bengkel is to determine the value of the degree of saturation and to know the performance of the intersection.At the intersection planning, it refers to the PKJI 2014 for Intersection Performance, while for the Geometrics it refers to Module 4 of the Ministry of Public Works and Public Housing on Geometric Planning of Plane Intersections.Based on  the calculation results of the Sungai Bengkel junction performance indicate that, the degree of  saturation  < 0.85,which is 0.529 for North,  0.537 for East, and 0.570  for West .With the radius is 15 m, and the cycle  time of traffic light 50 second, with the delay time  as long as 17 second/pcu

The imbalance population and number of vehicles increasing from year to year with number of roads causing traffic jam. Build toll road is the government's effort to solve traffic jam. The construction of toll roads that carried out by Government is expected to be able reduce traffic jam on arterial roads but in reality, does not reduce traffic jam even though traffic jam still exist on toll roads. Traffic jam causes stress and fatigue and can lead to traffic accidents. Indonesia has a much higher mortality rate compared to other countries. Traffic accidents are influenced by three main factors: human factors, vehicle factors, and road factors. To improve road safety and high number of deaths each year, it will be carried out research by direct observation using the Road Safety Audit (RSA) form of the Kunciran-Serpong Toll Road. The result of this research by direct observation on Kunciran-Serpong Toll Road in general there are still many shortcomings, such as various types of road damage and pavement, there are no signs to bend to the left or right, the drainage system is not good on several roads, and the median is not good because not all segments get fenced. Keywords: Traffic Jam; Accident, Toll; Road Safety AuditAbstrakSeiring dengan pertambahan jumlah penduduk maka jumlah kendaraanpun terus berkembang dari tahun ke tahun, sedangkan panjang ruas jalan tidak dapat menyusul secara signifikan, hal ini mengakibatkan terjadinya kemacetan lalu lintas. Upaya pemerintah mengurangi kemacetan dengan membangun jalan Tol yang diharapkan untuk mampu mengurangi kemacetan yang ada di jalan arteri. namun pada kenyataannya tidak mengurangi kemacetan bahkan jalan tol saat ini juga mengalami kemacetan. Beberapa jalan tol dibangun untuk mengurangi kemacetan yang ada di ruas jalan tol yang lama, karena apabila tidak dikurangi maka kemacetan menyebabkan stres dan lelah fisik, mengurangi produktifitas yang akhirnya mengakibatkan kecelakaan lalu lintas. Indonesia memiliki tingkat kematian yang jauh lebih tinggi dibandingkan dengan negara lainnya di ASEAN. Kecelakaan lalu lintas dipengaruhi tiga faktor utama yaitu faktor manusia, faktor kendaraan, dan faktor jalan. Dalam rangka upaya meningkatkan keselamatan jalan dan mengurangi angka kematian setiap tahunnya maka dilaksanakan observasi langung dengan mengunakan formulir Audit Keselamatan Jalan (AKJ) atau Road Safety Audit (RSA) pada ruas jalan tol operasional. Hasil dari observasi langung pada ruas jalan tol operasional di bawah 1 tahun secara umum masih terdapat banyak kekurangannya, seperti berbagai jenis kerusakan jalan terutama pada perkerasan, tidak adanya rambu peringatan tikungan ke kiri maupun ke kanan dan beberapa rambu lainnya, kecepatan eksisting yang lebih besar dari kecepatan rencana, dan median yang kurang baik karena tidak semua ruas terdapat pembatas

The common of urban city problem is a traffic jam, and it is heavily affect the public transportation service quality. Many cities in tropical countries suffer of heavy traffic jam due to the populaition of motorbike. According to Central Java Province Government, in 2014 the population of motorcycle is 439.418 units with the ownership ratio at 0.75 unit/person. Solo is one of typical middle size city in Central Java, Indonesia. Solo suffers from traffic jam in particular time of the day and it affect the quality of service of batik solo as the mass transportation service in Surakarta. This research would like to help the community get the optimal travel route with the fastest time, especially at the busiest times. The method used is Generate and Test algorithm. This method is a combination of Depth First Search and Backtracking. This research is conducted to get information about the influence of traffic volume, service road level and traffic jam level toward the time needed for Batik Solo Trans to find the optimal route. There is a route that been selected to be the most optimal route by considering the factors that influence it and the algorithm applied
